2Outcomes in refractory diffuse large B-cell lymphoma:results fromamulticenter real-world study in China显示文摘Background:Diffuse large B-cell lymphoma(DLBCL)patients refractory to rituximab-based immunochemotherapy have a dismal prognosis.However,the definition of refractory DLBCL remains inconsistent and no large cohort study data is available from Asian countries.To validate the definition and outcomes of refractory DLBCL in China,we conducted a multicenter,retrospective cohort study.Methods:The REtrospective AnaLysis of Treatment REspoNse of refractory DLBCL(REAL-TREND)study was performed using real-world data from 8 centers in China.DLBCL patients with curative intent were included in the REAL-TREND dataset.Overall survival(OS)was estimated using the Kaplan-Meier method and compared by the log-rank test.Due to heterogeneity in response rates among different centers,the response rates of refractory patients were pooled using random-effect models.Multivariate survival analysis was performed using the Cox regression model.Results:A total of 2778 DLBCL patients diagnosed between January,2010 and December,2015 were enrolled to this study.After validating previous definitions,the SCHOLAR-1 study was most suitable to define refractory DLBCL.The estimated 5-year cumulative incidence of refractory patients was 20%(95% confidence Interval[CI]=18%-22%).After the determination of refractory disease,overall response rate and complete remission rate were 30%(95%CI=22%-38%)and 9%(95%CI=4%-15%),respectively.Patients with either no response to immunochemotherapy or relapse within 12 months after stem-cell transplantation had inferior survival with a median OS of 5.9 months(95%CI=5.5-7.1 months)and 2-year OS rate of 16%(95%CI=12%-20%).International prognostic index score 4-5(hazard ratio[HR]=2.22;95%CI=1.47-3.35),central nervous systemrelapse(HR=1.43;95%CI=1.04-1.97),and best response status(HR=2.68;95%CI=1.42-5.03 for partial remission.HR=5.97,95%CI=3.21-11.11 for stable disease/progressive disease)were independent unfavorable prognostic factors.Conclusions:This is the first large-scale Asian cohort study focusing on outcomes of refractory DLBCL.The definition of the SCHOLAR-1 study identifies patients with homogenously inferior survival,thus is appropriate to select refractory DLBCL.Due to poor clinical outcomes in the rituximab era,patients with refractory DLBCL may be potential candidates for novel treatment modalities.Shuo Wang Li Wang Jianda Hu Wenbin Qian Xi Zhang Yu Hu Qi Zhu Bobin Chen Depei Wu Chung-Chou H.Chang Pengpeng Xu Xiaoyun Zheng Juying Wei Yao Liu Guohui Cui Yong Tang Yan Ma Haiwen Huang Hongmei Yi Weili Zhao 2021Cancer Communications2021,41,3:10

4Decrement of Sugar Consumption in Rice Young Panicle Under High Temperature Aggravates Spikelet Number Reduction显示文摘Two rice genotypes Huanghuazhan(HHZ, heat-resistant) and IR36(heat-susceptible) were subjected to high-temperature(HT, 40℃) and normal-temperature(NT, 32℃) treatments at the spikelet differentiation stage. HT treatment inhibited spikelet differentiation, aggravated spikelet degeneration, reduced spikelet size, and disordered carbohydrate allocation. Meanwhile, HT treatment increased nonstructural carbohydrate content in leaves, but decreased that in stems and young panicles, and the same tendencies of sucrose and starch contents were observed in leaves and stem. However, HT treatment significantly increased the sucrose content and sharply decreased the glucose and fructose contents in young panicles. Lower activity levels of soluble acid invertase(EC3.2.1.26) and sucrose synthase(EC2.4.1.13) were observed under HT treatment. Moreover, HT treatment reduced the activities of key enzymes associated with glycolysis and the tricarboxylic acid cycle, which indicated sucrose consumption was inhibited in young panicles under HT treatment. Exogenous glucose and fructose applied under HT treatment increased the spikelet number more than exogenous sucrose. In conclusion, the results demonstrated that the reduction of spikelet number under high temperature was more affected by the decrease in sugar consumption than the blocking of sucrose transport. 6Rapid growth and properties of large-aperture 98%-deuterated DKDP crystals显示文摘In this paper,a highly deuterated potassium dihydrogen phosphate(DKDP)crystal with sizes up to 318 mm×312 mm×265 mm was grown by the rapid-growth method.The synthesis tank device was specially designed to synthesize a higher deuterium concentration and high-purity DKDP solution.The deuterium content of the as-grown crystal,which was 97.9%,was determined by two methods,including infrared(IR)spectroscopy and thermo-gravimetric analysis(TGA)measurements.The performances of the 97.9%DKDP crystal,including transmission,absorption coefficient,and laserinduced damage threshold(LIDT)were measured.The results indicate that,in the near-infrared band,the transmission of the 97.9%DKDP crystal is higher than that of KDP and 70%DKDP crystals,and the absorption coefficient is lower.The LIDT of the crystal reached 23.2 J·cm^-2(R-on-1,1064 nm,3 ns),which meets the engineering requirements for use in optical applications.Xumin Cai Xiuqing Lin Guohui Li Junye Lu Ziyu Hu Guozong Zheng 2019High Power Laser Science and Engineering2019,7,3:4
7Lacustrine Shale Oil Resource Potential of Es3L Sub-Member of Bonan Sag, Bohai Bay Basin, Eastern China显示文摘Following shale gas, shale oil has become another highlight in unconventional hydrocarbon exploration and development. A large amount of shale oil has been produced from a host of marine shale in North America in recent years. In China, lacustrine shale, as the main source rock of conventional oil and gas, should also have abundant oil retained in place. In this study, geochemical and geologic characteristics of lacustrine shale from Es_3~L sub-member in Bonan sag were characterized by using total organic carbon(TOC), Rock-Eval pyrolysis, X-ray diffraction, and ?log R method. The results show that the Es_3~L sub-member shale have TOC contents ranging from 0.5 wt.% to 9.3 wt.%, with an average of 2.9 wt.%. The organic matter is predominantly Type I kerogen, with minor amounts of Type II1 kerogen. The temperature of maximum yield of pyrolysate(Tmax) values ranges from 424 to 447 °C, with an average of 440 °C, and vitrinite reflectance(Ro%) ranges from 0.7% to 0.9%, indicating most of shales are thermally mature. The dominant minerals of Es_3~L shale in Bonan sag are carbonates(including calcite and dolomite), averaging 51.82 wt.%, and the second minerals are clay(mostly are montmorillonite-illite-mixed layer and illite) and quartz, averaging about 18 wt.%. Finally, its shale oil resources were evaluated by using the volumetric method, and the evaluation result shows that the shale oil resource is up to 5.94 billion tons, and mostly Class I resource. 8Exosomal PD-L1 induces osteogenic differentiation and promotes fracture healing by acting as an immunosuppressant显示文摘A moderate inflammatory response at the early stages of fracture healing is necessary for callus formation.Over-active and continuous inflammation,however,impairs fracture healing and leads to excessive tissue damage.Adequate fracture healing could be promoted through suppression of local over-active immune cells in the fracture site.In the present study,we achieved an enriched concentration of PD-L1 from exosomes(Exos)of a genetically engineered Human Umbilical Vein Endothelial Cell(HUVECs),and demonstrated that exosomes overexpressing PD-L1 specifically bind to PD-1 on the T cell surface,suppressing the activation of T cells.Furthermore,exosomal PD-L1 induced Mesenchymal Stem Cells(MSCs)towards osteogenic differentiation when pre-cultured with T cells.Moreover,embedding of Exos into an injectable hydrogel allowed Exos delivery to the surrounding microenvironment in a time-released manner.Additionally,exosomal PD-L1,embedded in a hydrogel,markedly promoted callus formation and fracture healing in a murine model at the early over-active inflammation phase.Importantly,our results suggested that activation of T cells in the peripheral lymphatic tissues was inhibited after local administration of PD-L1-enriched Exos to the fracture sites,while T cells in distant immune organs such as the spleen were not affected.In summary,this study provides the first example of using PD-L1-enriched Exos for bone fracture repair,and highlights the potential of Hydrogel@Exos systems for bone fracture therapy through immune inhibitory effects.Ze Lin Yuan Xiong Weilin Meng Yiqiang Hu Lili Chen Lang Chen Hang Xue Adriana CPanayi Wu Zhou Yun Sun Faqi Cao Guodong Liu Liangcong Hu Chenchen Yan Xudong Xie Chuanchuan Lin Kaiyong Cai Qian Feng Bobin Mi Guohui Liu 2022Bioactive Materials2022,7,7:4

11Genes in the serotonin pathway are associated with bipolar affective disorder in a Han Chinese population显示文摘Serotonin plays an important role in mood regulation, but the involvement of serotonin pathway genes in the development of bipolar I disorder (BP-I), a mood disorder, is not clear. We selected 21 singlenucleotide polymorphisms (SNPs) within the HTR2A gene, 8 within the SLC6A4 gene and 23 within the TPH2 gene for genotyping using the GoldenGate genotyping assay. A total of 375 patients with BP-I and 475 normal controls were recruited. Two out of 21 SNPs (rs1475196 and rs9567747) in the HTR2A gene and 1/23 SNPs (rs17110566) in the TPH2 gene were significantly associated with BP-I, both genotype-wise and allele-wise. Furthermore, a specific haplotype in the HTR2A gene showed a significant association with BP-I. 14P granules phase transition induced by cytoplasmic streaming in Caenorhabditis elegans embryo显示文摘P granules are germ granules contained in Caenorhabditis elegans germ cells.The first germ cell is specified by the one-cell embryo in which P granules localize to the posterior.Previous studies suggested that the mechanism of the localization phenomena is induced by liquid-liquid phase transition(LLPT),in which the polarity proteins control the saturation point of P granules.In the present study,we propose that the P granules phase transition can be triggered by the cytoplasmic streaming.A two-phase flow model is employed to simulate the localization of P granules,i.e.,the cytoplasm is considered as a liquid phase,and the droplet-like P granules are another liquid phase.With the presence of the cytoplasmic streaming,P granules,initially distributing uniformly in the entire one-cell embryo,eventually condense/dissolve in the cytoplasm phase,regulated by difference between the saturation pressure and the hydrodynamic pressure.The numerical results reveal that the cytoplasmic streaming has significant effects on the localization of P granules,as well as the embryo division.Hang Wang GuoHui Hu 2017Science China(Physics,Mechanics & Astronomy)2017,60,1:1

18Comparisons of the Paleo-Mesoproterozoic large igneous provinces and black shales in the North China and North Australian cratons显示文摘Comparisons of large igneous provinces(UPs)and black shales from different cratons can provide important constraints on Precambrian paleogeographic reconstructions and a better understanding of the environmental effects of large-scale volcanic events.A comparison of intraplate mafic events mostly interpreted as LIPs or portions of LIPs(LIP fragments/remnants due to continental breakup or erosion)from the North China Craton(NCC)and North Australian Craton(NAC)shows good correlation in the age range from 1800 Ma to 1300 Ma,and four robust age matches at ca.1790-1770 Ma,ca.1730 Ma,ca.1680-1670 Ma and ca.1320 Ma have been identified.Most notably,the coeval ca.1320 Ma Yanliao LIP in the eastern-northern NCC and the Derim Derim-Galiwinku LIP in the NAC are also characterized by similar field occurences and dominantly subalkaline tholeiitic basalts and intraplate geochemical compositions,and are interpreted as portions of the same LIP,separated by continental breakup.Subsequent to 1300 Ma,the NCC and NAC exhibit very different magmatic histories,indicating that separation of these two cratons occurred,likely subsequent to the ca.1320 Ma LIP event.A comparison of Paleo-Mesoproterozoic black shales from the NCC and NAC provides further evidence for close connections between these regions during this period.Black shales of the Chuanlianggou Formation in the northern NCC and the Cuizhuang Formation in the southern NCC were deposited in the age range ca.1650-1635 Ma and can be correlated with ca.1640-1635 Ma black shales in the Barney Creek Formation of the NAC.Deposition of black shales within the Xiamaling Formation in the NCC and the Velkerri and Kyalla formations of the McArthur Basin in the NAC occurred synchronously at ca.1380-1360 Ma.Our results from matching of LIP ages and black shales combined with paleomagnetic data show that the northern-northeastern margin of the NCC was connected to the northern margin of the NAC from ca.1800 Ma to 1300 Ma.This long-lived late Paleoproterozoic to mid-Mesoproterozoic connection lasted for at least 500 million years until separation of the NCC from the NAC between ca.1320 and ca.1230-1220 Ma.Shuan-Hong Zhang Richard E.Ernst Tim J.Munson Junling Pei Guohui Hu Jian-Min Liu Qi-Qi Zhang Yu-Hang Cai Yue Zhao 2022Fundamental Research2022,2,1:1

20Morphology of cylindrical cell sheets with embedded contractile ring显示文摘The behavior of large deformations of cellular tissues is usually affected by the local properties of cells and their interactions,resulting in folding which acts as an important role in the embryonic development,as well as growing and spreading of a tumor,which can rapidly promote the stereo complexity of the architecture of the tissues.In the present study,a cylindrical vertex model is constructed to explore the morphology of the tubular cell sheets subject to an embedded contractile ring.It is found that an inner region of the contractile ring in equilibrium will protrude from the tube wall,and it will suddenly collapse when the contractile strength exceeds a threshold,indicating the occurrence of a bifurcation.These results on the effect of embedded contraction in the tubular shell are quite different from the planar cases,which can reveal the importance of the interaction between the geometric and material non-linearity in cylindrical geometry.The dependence of the large deformation on the bending modulus parameters and contraction strength is also analyzed for the cylindrical cell shell.Nan NAN Guohui HU 2019Applied Mathematics and Mechanics(English Edition)2019,40,12:1
