| 1 | SHP2 inhibition triggers anti-tumor immunity and synergizes with PD-1 blockade显示文摘Tyrosine phosphatase SHP2 is a promising drug target in cancer immunotherapy due to its bidirectional role in both tumor growth promotion and T-cell inactivation. Its allosteric inhibitor SHP099 is known to inhibit cancer cell growth both in vitro and in vivo. However, whether SHP099-mediated SHP2 inhibition retards tumor growth in vivo via anti-tumor immunity remains elusive. To address this, a CT-26 colon cancer xenograft model was established in mice since this cell line is insensitive to SHP099.Consequently, SHP099 minimally affected CT-26 tumor growth in immuno-deficient nude mice, but significantly decreased the tumor burden in CT-26 tumor-bearing mice with intact immune system.SHP099 augmented anti-tumor immunity, as shown by the elevated proportion of CD8tIFN-γtT cells and the upregulation of cytotoxic T-cell related genes including Granzyme B andPerforin, which decreased the tumor load. In addition, tumor growth in mice with SHP2-deficient T-cells was markedly slowed down because of enhanced anti-tumor responses. Finally, the combination of SHP099 and antiPD-1 antibody showed a higher therapeutic efficacy than either monotherapy in controlling tumor growthin two colon cancer xenograft models, indicating that these agents complement each other. | 2 | GYY4137 protects against myocardial ischemia and reperfusion injury by attenuating oxidative stress and apoptosis in rats显示文摘Hydrogen sulfide(H_2S) is a gasotransmitter that regulates cardiovascular functions.The present study aimed to determine the protective effect of slow-releasing H_2S donor GYY4137 on myocardial ischemia and reperfusion(I/R) injury and to investigate the possible signaling mechanisms involved.Male Sprague-Dawley rats were treated with GYY4137 at 12.5 mg/(kg·day),25 mg/(kg·day) or 50 mg/(kg·day) intraperitoneally for7 days.Then,rats were subjected to 30 minutes of left anterior descending coronary artery occlusion followed by reperfusion for 24 hours.We found that GYY4137 increased the cardiac ejection fraction and fractional shortening,reduced the ischemia area,alleviated histological injury and decreased plasma creatine kinase after myocardial I/R.Both H_2S concentration in plasma and cystathionine-γ-lyase(CSE) activity in the myocardium were enhanced in the GYY4137 treated groups.GYY4137 also decreased malondialdehyde and myeloperoxidase levels in serum,attenuated superoxide anion level and suppressed phosphorylation of mitogen activated protein kinases in the myocardium after I/R.Meanwhile,GYY4137 increased the expression of Bcl-2 but decreased the expression of Bax,caspase-3 activity and apoptosis in the myocardium.The data suggest that GYY4137 protects against myocardial ischemia and reperfusion injury by attenuating oxidative stress and apoptosis. | Guoliang Meng Jing Wang Yujiao Xiao Wenli Bai Liping Xie Liyang Shan Philip K Moore Yong Ji | 2015 | The Journal of Biomedical Research2015,29,3: | 18 |
| 3 | Adaptive Robust Motion Trajectory Tracking Control of Pneumatic Cylinders with LuGre Model-based Friction Compensation显示文摘Friction compensation is particularly important for motion trajectory tracking control of pneumatic cylinders at low speed movement. However, most of the existing model-based friction compensation schemes use simple classical models, which are not enough to address applications with high-accuracy position requirements. Furthermore, the friction force in the cylinder is time-varying, and there exist rather severe unmodelled dynamics and unknown disturbances in the pneumatic system. To deal with these problems effectively, an adaptive robust controller with LuGre model-based dynamic friction compensation is constructed. The proposed controller employs on-line recursive least squares estimation(RLSE) to reduce the extent of parametric uncertainties, and utilizes the sliding mode control method to attenuate the effects of parameter estimation errors, unmodelled dynamics and disturbances. In addition, in order to realize LuGre model-based friction compensation, the modified dual-observer structure for estimating immeasurable friction internal state is developed. Therefore, a prescribed motion tracking transient performance and final tracking accuracy can be guaranteed. Since the system model uncertainties are unmatched, the recursive backstepping design technology is applied. In order to solve the conflicts between the sliding mode control design and the adaptive control design, the projection mapping is used to condition the RLSE algorithm so that the parameter estimates are kept within a known bounded convex set. Finally, the proposed controller is tested for tracking sinusoidal trajectories and smooth square trajectory under different loads and sudden disturbance. The testing results demonstrate that the achievable performance of the proposed controller is excellent and is much better than most other studies in literature. Especially when a 0.5 Hz sinusoidal trajectory is tracked, the maximum tracking error is 0.96 mm and the average tracking error is 0.45 mm. | 4 | Cardiomyocyte-like differentiation of human bone marrow mesenchymal stem cells after exposure to 5-azacytidine in vitro显示文摘Objective To investigate the potential of adult mesenchymal stem cells (MSCs) derived from human bone marrow to undergo cardiomyogenic differentiation after exposure to 5-azacytidine (5-aza) in vitro. Methods A small bone marrow aspirate was taken from the iliac crest of human volunteers, and hMSCs were isolated by 1.073g/mL Percoll and propagated in the right cell culturing medium as previously described. The phenotypes of hMSCs were characterized with the use of flow cytometry. The hMSCs were cultured in cell culture medium (as control) and medium mixed with 5-aza for cellular differentiation. We examined by immunohistochemistry at 21 days the inducement of desmin, cardiac-specific cardiac troponin I (cTnI), GATA 4 and connexin-43 respectively. Results The hMSCs are fibroblast-like morphology and express CD44+ CD29+ CD90+ / CD34- CD45- CD31- CD11a. After 5-aza treatment, 20-30% hMSCs connected with adjoining cells and coalesced into myotube structures after 14days. Twenty-one days after 5-aza treatment, immunofluorescence showed that some cells expressed desmin,GATA4, cTnI and connexin-43 in 5,10 μmol/L 5-aza groups, but no cardiac specific protein was found in neither 3μmol/L 5-aza group nor in the control group. The ratio of cTnI positively stained cells in 10 μmol/L group was higher than that in 5 μmol/L group (65.3 ± 4.7% vs 48.2 ± 5.4%, P < 0.05). Electron microscopy revealed that myofilaments were formed. The induced cells expressed cardiac-myosin heavy chain (MyHC) gene by reverse transcription-polymerase chain reaction (RT-PCR). Conclusions Theses findings suggest that hMSCs from adult bone marrow can be differentiated into cardiac-like muscle cells with 5-aza inducement in vitro and the differentiation is in line with the 5-aza concentration. | 6 | Aggregate binding agents improve soil aggregate stability in Robinia pseudoacacia forests along a climatic gradient on the Loess Plateau,China显示文摘The distribution of binding agents(i.e.,soil organic carbon(SOC)and glomalin-related soil protein(GRSP))in soil aggregates was influenced by many factors,such as plant characteristics and soil properties.However,how these factors affect binding agents and soil aggregate stability along a climatic gradient remained unclear.We selected the Robinia pseudoacacia L.forests from semi-arid to semi-humid of the Loess Plateau,China to analyze the plant biomass,soil physical-chemical properties,SOC and GRSP distribution in different sized soil aggregates.We found that from semi-arid to semi-humid forests:(1)the proportion of macro-aggregates(>0.250 mm)significantly increased(P<0.05),whereas those of micro-aggregates(0.250–0.053 mm)and fine materials(<0.053 mm)decreased and soil aggregate stability was increased;(2)the contents of SOC and GRSP in macro-aggregates and micro-aggregates significantly increased,and those in fine materials decreased;(3)the contribution of SOC to soil aggregate stability was greater than those of total GRSP and easily extractable GRSP;(4)soil properties had greater influence on binding agents than plant biomass;and(5)soil aggregate stability was enhanced by increasing the contents of SOC and GRSP in macro-aggregates and soil property was the important part during this process.Climate change from semi-arid to semi-humid forests is important factor for soil structure formation because of its positive effect on soil aggregates. | JING Hang MENG Min WANG Guoliang LIU Guobin | 2021 | Journal of Arid Land2021,13,2: | 4 |
| 7 | D2 lymphadenectomy with complete mesogastrium excision vs. conventional D2 gastrectomy for advanced gastric cancer显示文摘Background: The complete mesogastrium excision (CME) based on D2 radical gastrectomy is believed to significantly reduce the local-regional recurrence compared with D2 radical gastrectomy in advanced gastric cancer, and it is widely used in China. This study aimed to explore whether D2 + CME is superior to D2 on surgical outcomes during gastrectomy from Chinese data.Methods: Feasible studies comparing the D2 + CME (D2 + CME group) and D2 (D2 group) published up to March 2020 are searched from electronic databases. The data showing surgical and complication outcomes are extracted to be pooled and analyzed.Results: Fourteen records including 1352 patients were included. The D2 + CME group had a shorter mean operative time (weighted mean difference [WMD] = -16.72 min, 95% confidence interval [CI]: -26.56 to - 6.87 min,P < 0.001), lower mean blood loss (WMD = -39.08 mL, 95% CI: -49.94 to -28.21 mL,P < 0.001), higher mean number of retrieved lymph nodes (WMD = 2.13, 95% CI: 0.58-3.67,P = 0.007), shorter time to first flatus (WMD =-0.31 d, 95% CI: -0.53 to - 0.10 d,P = 0.005), and postoperative hospital days (WMD =-1.09, 95% CI: -1.92 to -0.25,P = 0.010) than the D2 group. Subgroup analysis suggested that the advantages from the D2 + CME group were obvious in traditional open radical gastrectomy, proximal gastrectomy, and distal gastrectomy compared with D2 group. The evaluations of post-operative complications showed that the patients who underwent D2 + CME had a lower incidence of post-operative complications than the patients who underwent D2 surgery alone (relative risk [RR] = 0.65, 95% CI: 0.45-0.87,P = 0.003). The D2 radical gastrectomy plus CME improved 3-year overall survival (OS) (RR = 1.16, 95% CI: 1.02-1.32,P = 0.020) and lowered the local recurrence rate (RR = 0.51, 95% CI: 0.28-0.94,P = 0.030). | 8 | Assessing current stocks and future sequestration potential of forest biomass carbon in Daqing Mountain Nature Reserve of Inner Mongolia, China显示文摘Assessment of regional forest carbon stocks and underlying controls is critical for guiding forest management in the context of carbon sequestration. We investigated the variations in tree biomass carbon stocks relating to forest types, and estimated the total tree biomass carbon stocks and projected gains through natural stand development by 2020 and 2050 in the Daqing Mountain Nature Reserve based on Category II data of the Forest Inventory of Inner Mongolia for the period ending 2008. Over a total area of 388,577 ha,this nature reserve currently stores an estimated 2221 Gg C in tree aboveground biomass alone, with potential to grow by more than 30 % to reach 2938 Gg C by 2020 and nearly double to 4092 Gg C by 2050 through natural development of the existing forest stands. The tree biomass carbon density and potential gain in tree biomass carbon stocks vary markedly among forest types and with stand development.The variations in the potential change of tree biomass carbon density for the periods 2008–2020 and 2008–2050 among forest types partly reflect the varying relationships of tree biomass carbon density with stand age for different tree species, and partly are attributable to variations in the stand age structure among different forest types. Of the major forest types, the ranking of projected changes in tree biomass carbon density are not consistent with variations in the relationship between tree biomass carbon density and stand age, neither are they explainable by variations in stand age structures, implying the interactive effect between forest type and stand dynamics on temporal changes in tree biomass carbon density. Birch rank highest for future biomass carbon sequestration because of its dominance in cover area and better age structure for potential gain in tree biomass carbon stocks. Poplar and larch were out-performers compared to other forest types given their greater contribution to total tree biomass carbon stocks relative to their distributional areas. Findings in this study illustrate that protection and proper management of under-aged forests can deliver marked gains in biomass carbon sequestration. | 10 | The protective effect and crucial biological pathways analysis of Trametes lactinea mycelium polysaccharides on acute alcoholic liver injury in mice based on transcriptomics and metabonomics显示文摘Trametes lactinea mycelia polysaccharides(TLMPS)have a wide range of bioactivities.The potential mechanisms of action of TLMPS against acute alcohol-induced liver injury in vivo were investigated by analyzing the physical and chemical properties of TLMPS and its protective effects on a mouse model of alcoholic liver injury.TLMPS protected the liver against alcohol-induced injury,as evidenced by the reduced alcohol-induced elevation of the liver index,serum biochemical indices,and maintenance of hepatic morphology.Potential mechanisms were analyzed using transcriptome and metabolome analyses.The transcriptome data revealed the involvement of many differentially expressed genes in chemical carcinogenesis,drug metabolism,and metabolism of xenobiotics by cytochrome P450.The metabolome analysis revealed that TLMPS significantly regulated specific metabolites in the liver,including organic acids,lipids,nucleosides,and organic oxygen compounds.KEGG enrichment analysis revealed the signifi cant involvement of different metabolites in choline metabolism,ATP-binding cassette transporters,and glycerophospholipid metabolism.Assessment of the changes in gene expression and metabolites revealed the significantly different expression of several genes encoding key enzymes and metabolites in choline metabolism pathway.The collective fi ndings confi rmed that choline metabolism plays an important role in the protective effects of TLMPS against acute alcoholic liver injury. | Jinbin Hao Liyun Ye Guoliang Meng Yanjiao Song Junsheng Fu Xiaoping Wu | 2021 | Food Science and Human Wellness2021,10,4: | 2 |
