1A novel colloidal gold immunochromatography assay strip for the diagnosis of schistosomiasis japonica in domestic animals显示文摘Background:Schistosomiasis remains a major public health concern in China and an epidemiological survey has revealed that schistosome-infected bovines and goats are the main transmission sources for the disease.Therefore,development of a sensitive technique for the diagnosis of schistosomiasis in domestic animals is necessary.Method:A novel colloidal gold immunochromatography assay(GICA)strip was developed for detecting Schistosoma japonicum in domestic animals.The colloidal gold was conjugated with recombinant streptococcal protein G(rSPG).As the test and control lines,the schistosome soluble egg antigen and rSPG,respectively,were blotted on nitrocellulose membrane.Results:The lowest detectable serum dilution was 1∶640 for schistosome-infected buffaloes.The cross-reaction rate of GICA was 14.29%with Paramphistomum sp.in buffaloes,16.67%with Haemonchus sp.in goats,and 33.33%with Orientobilharzia sp.in goats.These results were slightly lower and similar to those obtained through ELISA.Moreover,the strips for detecting S.japonicum in mice,rabbits,buffaloes,and goats showed high sensitivity(100.00%,100.00%,100.00%,and 100.00%,respectively)and specificity(100.00%,100.00%,94.23%,and 88.64%,respectively).And the sensitivity or specificity of the GICA strips did not present any significant differences after storage for 12 months at room temperature.When compared with ELISA,the GICA strips exhibited similar sensitivity and specificity in the diagnosis of schistosomiasis in mice,rabbits,buffaloes,and goats.Besides,only 5μl of serum are required for the test and the detection can be completed within 5 min.Conclusion:This study is the first to develop a GICA strip using gold-rSPG conjugate for the diagnosing of schistosomiasis in domestic animals,and preliminary results showed that the developed strip may be suitable for large-scale screening of schistosomiasis in endemic areas.Rui Xu Jintao Feng Yang Hong Chao Lv Dengyun Zhao Jiaojiao Lin Ke Lu Hao Li Jinming Liu Xiaodan Cao Tao Wang Jinli Zai Zhaozhe Wang Bingguang Jia Qian Han Chuangang Zhu 2017Infectious Diseases of Poverty2017,6,1:12
2Disinfection efficiency of positive pressure respiratory protective hood using fumigation sterilization cabinet显示文摘Concerns have been raised about both the disinfection and the reusability of respiratory protective equipment following a disinfection process.Currently,there is little data available on the effects of disinfection and decontamination on positive pressure respiratory protective hoods(PPRPH).In this study,we evaluated the effect of vaporized hydrogen peroxide(VHP)on the disinfection of PPRPH to determine applicability of this method for disinfection of protective equipment,especially protective equipment with an electric supply system.A hydrogen peroxide-based fumigation sterilization cabinet was developed particularly for disinfection of protective equipment,and the disinfection experiments were conducted using four PPRPHs hung in the fumigation chamber.The pathogenic microorganism Geobacillus stearothermophilus ATCC 7953 was used as a biological indicator in this study and the relationship between air flow(the amount of VHP)and disinfection was investigated.Both function and the material physical properties of the PPRPH were assessed following the disinfection procedure.No surviving Geobacillus stearothermophilus ATCC 7953,both inside and outside of these disinfected PPRPHs,could be observed after a 60 min treatment with an air flow of 10.5–12.3 m^(3)/h.Both function and material physical properties of these PPRPHs met the working requirements after disinfection.This study indicates that air flow in the fumigation chamber directly influences the concentration of VHP.The protective equipment fumigation sterilization cabinet developed in this paper achieves the complete sterilization of the PPRPHs when the air flow is at 10.5–12.3 m^(3)/h,and provides a potential solution for the disinfection of various kind of protective equipment.Limei Hao Jinhui Wu Enlei Zhang Ying Yi Zongxing Zhang Jinming Zhang Jiancheng Qi 2019Biosafety and Health2019,1,1:5

4Development of a negative pressure hood for isolation and transportation of individual patient with respiratory infectious disease显示文摘The frequent and sudden occurrence of both known and unknown infectious diseases can cause global social panic.If the source of infection can be effectively controlled in the early stages of an outbreak,the spread of infectious diseases can be prevented.In view of this situation,this study developed for infectious or suspected infectious patients a negative pressure isolation hood which effectively achieves direct individual isolation during the early stages of disease outbreak,and facilitates long-distance transport.The hood body is made of flexible transparent polyvinyl chloride(PVC)material,and the combination of the hood material is airtight.The unique inflatable column support structure and the design of the inflatable neck sleeve effectively ensure both stiffness and air tightness of the hood body.The electrical exhaust system maintains a stable negative pressure environment inside the hood,and polluted air inside the hood can be purified by a high efficiency filter.Test results showed that the internal noise of the hood was 68±1 dB(A),the air exhaust volume of the electric exhaust system was not<200 L/min,and the filtration efficiency of the filter to 0.3μm particles was>99.99%,indicating that the hood achieved effective isolation protection for patients with respiration infectious diseases.Limei Hao Jinhui Wu Jinming Zhang Zhangyi Liu Ying Yi Zongxing Zhang Enlei Zhang Jiancheng Qi 2019Biosafety and Health2019,1,3:3

6A polyphenol-assisted IL-10 mRNA delivery system for ulcerative colitis显示文摘With the development of synthesis technology, modified messenger RNA(mRNA) has emerged as a novel category of therapeutic agents for a broad of diseases. However, effective intracellular delivery of mRNA remains challenging, especially for its sensitivity to enzymatic degradation. Here, we propose a polyphenol-assisted handy delivery strategy for efficient in vivo delivery of IL-10 mRNA.IL-10 mRNA binds to polyphenol ellagic acid through supramolecular binding to yield a negatively charged core, followed by complexing with linear polyetherimide and coating with bilirubin-modified hyaluronic acid to obtain a layer-by-layer nanostructure. The nanostructure specifically up-regulated the level of IL-10, effectively inhibited the expression of inflammatory factors, promoted mucosal repair, protected colonic epithelial cells against apoptosis, and exerted potent therapeutic efficacy in dextran sulfate sodium salt-induced acute and chronic murine models of colitis. 12Lenvatinib-and vadimezan-loaded synthetic high-density lipoprotein for combinational immunochemotherapy of metastatic triple-negative breast cancer显示文摘Metastatic triple-negative breast cancer(TNBC)is the most aggressive type of breast cancer.Combination of systemic chemotherapy and immune checkpoint blockade is effective but of limited benefit due to insufficient intratumoral infiltration of cytotoxic T lymphocytes(CTLs)and the accumulation of immunosuppressive cells.Herein,we designed a lenvatinib-and vadimezan-loaded synthetic high-density lipoprotein(LV-sHDL)for combinational immunochemotherapy of metastatic TNBC.The LV-sHDL targeted scavenger receptor class B type 1-overexpressing 4T1 cells in the tumor after intravenous injection.The multitargeted tyrosine kinase inhibitor(TKI)lenvatinib induced immunogenic cell death of the cancer cells,and the stimulator of interferon genes(STING)agonist vadimezan triggered local inflammation to facilitate dendritic cell maturation and antitumor macrophage differentiation,which synergistically improved the intratumoral infiltration of total and active CTLs by 33-and 13-fold,respectively.LV-sHDL inhibited the growth of orthotopic 4T1 tumors,reduced pulmonary metastasis,and prolonged the survival of animals.The efficacy could be further improved when LV-sHDL was used in combination with antibody against programmed cell death ligand 1.This study highlights the combination use of multitargeted TKI and STING agonist a promising treatment for metastatic TNBC.Chao Zheng Wen Zhang Jinming Wang Yihui Zha Fengqin Xiong Ying Cai Xiang Gong Binyu Zhu Helen He Zhu Hao Wang Yaping Li Pengcheng Zhang 2022Acta Pharmaceutica Sinica B2022,12,9:1
13Down-staging depth score could be a survival predictor for locally advanced gastric cancer patients after preoperative chemoradiotherapy显示文摘Objective:The predictive effect of preoperative chemoradiotherapy(CRT)is low and difficult in guiding individualized treatment.We examined a surrogate endpoint for long-term outcomes in locally advanced gastric cancer patients after preoperative CRT.Methods:From April 2012 to April 2019,95 patients with locally advanced gastric cancer who received preoperative concurrent CRT and who were enrolled in three prospective studies were included.All patients were stage T_(3/4) N_(+).Local control,distant metastasis-free survival(DMFS),disease-free survival(DFS)and overall survival(OS)were evaluated.Clinicopathological factors related to long-term prognosis were analyzed using univariate and multivariate analyses.The down-staging depth score(DDS),which is a novel method of evaluating CRT response,was used to predict long-term outcomes.Results:The median follow-up period for survivors was 30 months.The area under the curve(AUC)of the receiver operating characteristic(ROC)curve predicted by the DDS was 0.728,which was better than the pathological complete response(pCR),histological response and ypN0.Decision curve analysis further affirmed that DDS had the largest net benefit.The DDS cut-off value was 4.pCR and ypN0 were associated with OS(P=0.026 and 0.049).Surgery and DDS are correlated with DMFS,DFS and OS(surgery:P=0.001,<0.001 and<0.001,respectively;and DDS:P=0.009,0.013 and 0.032,respectively).Multivariate analysis showed that DDS was an independent prognostic factor of DFS(P=0.021).Conclusions:DDS is a simple,short-term indicator that was a better surrogate endpoint than pCR,histological response and ypN0 for DFS.Ning Li Xin Wang Yuan Tang Dongbin Zhao Yihebali Chi Lin Yang Liming Jiang Jun Jiang Jinming Shi Wenyang Liu Hua Ren Hui Fang Yu Tang Bo Chen Ningning Lu Hao Jing Shunan Qi Shulian Wang Yueping Liu Yongwen Song Yexiong Li Jing Jin 2021Chinese Journal of Cancer Research2021,33,4:1
14Fish Assemblage Responses to a Low-head Dam Removal in the Lancang River显示文摘Dam removal is becoming an effective approach for aquatic biodiversity restoration in damming river in order to balance the aquatic ecosystem conservation with large-scale cascade damming. However, the effects of dam removal on fish communities in Asian mountainous rivers, which are dominated by Cypriniformes fishes, are still not well known. To determine whether dam removal on a mountainous river benefit restoration of fish diversity, we investigated the response of fish assemblage to dam removal using a before-after-control-impact design in two tributaries of the Lancang River(dam removal river: the Jidu River, and control river: the Fengdian River). Fish surveys were conducted one year prior to dam removal(2012) and three years(2013–2015) following dam removal. We observed rapidly and notably spatio-temporal changes in fish biodiversity metrics and assemblage structure, occurring in the Jidu River within the first year after dam removal. Overall, fish species richness, density and Shannon-Wiener diversity all increased immediately in above-and below-dam sites, and maintained a stable level in subsequent years, compared to unchanged situation in the control river. All sites in the Jidu River experienced shifts in fish composition after dam removal, with the greatest temporal changes occurred in sites below-and above-the former dam, resulting in a temporal homogenization tendency in the dam removed river. 16A General Strategy toward Synthesis of Well-Defined Polypeptides with Complex Chain Topologies显示文摘The precise synthesis of polypeptides with varying chain topologies has attracted significant interest due to their diverse functional applications.However,conventional polymerization of environment-susceptible N-carboxylic anhydride monomers using primary amine initiators suffers from inevitable side reactions,which greatly compromise chain-end fidelity,thereby disabling access to polypeptides with high-order architectures.Herein,we have developed a general and robust strategy based on normal aminemechanism for the synthesis of various topological polypeptides via polymerization of moisture-insensitive and airtolerant N-phenoxycarbonyl-functionalizedα-amino acid precursors using primary amine hydrochloride as initiators.This strategy enabled the synthesis of a block,star,star-block,brush-type,and multiblock(co)polypeptides with desired sequences,predictable molecular weights,low polydispersity,and high-fidelity chain ends even under open-air conditions.Remarkably,the robustness of this approach has been exemplified by the precise synthesis of welldefined photoresponsive decablock copolypeptides,exhibiting a spontaneous morphological evolution from polymersomes to lamellae nanostructures upon light irradiation.This work provides a general and reliable tool for synthesizing polypeptides with varying topologies,shedding light on the development of synthetic polypeptide-based materials.Lei Li Jie Cen Wenjin Li Wenhao Pan Yuben Zhang Hao Yin Jinming Hu Shiyong Liu 2022CCS Chemistry2022,4,12:0

18Evaluation of global navigation satellite system spoofing efficacy显示文摘The spoofing capability of Global Navigation Satellite System(GNSS)represents an important confrontational capability for navigation security,and the success of planned missions may depend on the effective evaluation of spoofing capability.However,current evaluation systems face challenges arising from the irrationality of previous weighting methods,inapplicability of the conventional multi-attribute decision-making method and uncertainty existing in evaluation.To solve these difficulties,considering the validity of the obtained results,an evaluation method based on the game aggregated weight model and a joint approach involving the grey relational analysis and technique for order preference by similarity to an ideal solution(GRA-TOPSIS)are firstly proposed to determine the optimal scheme.Static and dynamic evaluation results under different schemes are then obtained via a fuzzy comprehensive assessment and an improved dynamic game method,to prioritize the deceptive efficacy of the equipment accurately and make pointed improvement for its core performance.The use of judging indicators,including Spearman rank correlation coefficient and so on,combined with obtained evaluation results,demonstrates the superiority of the proposed method and the optimal scheme by the horizontal comparison of different methods and vertical comparison of evaluation results.Finally,the results of field measurements and simulation tests show that the proposed method can better overcome the difficulties of existing methods and realize the effective evaluation.WANG Yue SUN Fuping HAO Jinming ZHANG Lundong WANG Xian 2022Journal of Systems Engineering and Electronics2022,33,6:0
