| 3 | Research and development of new neodymium laser glasses显示文摘This work presents a brief introduction on three kinds of newly developed Nd^(3+)-doped laser glasses in Shanghai Institute of Optics and Fine Mechanics(SIOM), China. Two Nd^(3+)-doped phosphate glasses with lower thermal expansion coefficient and thermal shock resistance 4 times higher than that of N31 glass are developed for laser processing.Nd:Silicate and Nd:Aluminate glasses with peak emission wavelength at 1061 and 1065 nm, effective emission bandwidth of 34 and 50 nm, respectively, are developed for Exawatt-class laser system application. Fluorophosphate glasses with low nonlinear refractive index(n_2=0.6–0.86) and long fluorescence lifetime(430–510 μs) are investigated for the purpose of decreasing B integral in high-power laser system. | 4 | Characterization of a Novel Anti-DR5 Monoclonal Antibody WD1 with the Potential to Induce Tumor Cell Apoptosis显示文摘TNF-related apoptosis-inducing ligand (TRAIL) is a TNF family member capable of inducing apoptosis. Death receptor 5 (DR 5) is a key receptor of TRAIL and plays an important role in TRAIL-induced apoptosis. To prepare monoclonal antibodies (mAbs) against DR5,cDNA encoding soluble DR5 (sDR5) was firstly amplified by reverse transcriptase-polymerase chain reaction (RT-PCR) with specific primers,and then inserted into a prokaryotic expression vector pET-30a. The recombinant plasmid was expressed in Escherichia coli strain BL21 (DE3),and sDR5 was purified by nickel affinity chromatography. As an antigen,sDR5 was used to immunize mice. Hybridomas secreting antibodies against sDR5 were identified. One positive clone was selected to produce antibody,WD1. ELISA and immunofluorescence demonstrated that WD1 could bind recombinant sDR5 and membrane-bound DR5 (mDR5) on Jurkat and Molt-4 cells. ATPLite assays showed that Jurkat and Molt-4 cells were sensitive to the antibody in a dose dependent manner. The Annexin V/PI assays and Giemsa's staining both showed that WD1 could induce Jurkat cell apoptosis efficiently. Transient transfection of 293T cells and indirect immunofluorescence assay demonstrated that mAb (WD1) couldn't cross-react with DR4. Our findings indicated that the novel antibody,WD1 could act as a direct agonist,bind DR5 characteristically,and initiate efficient apoptotic signaling and tumor regression. | 5 | Nano-and microphytoplankton community characteristics in brown tide bloom-prone waters of the Qinhuangdao coast,Bohai Sea, China显示文摘Since 2009, the newly recorded species Aureococcus anophagefferens has successively induced brown tides in the Qinhuangdao coastal waters of the Bohai Sea. Here, we focus on the phytoplankton community structure in this region and try to point out the biological background characteristics of the outbreaks of successive brown tides. We analyzed species composition,cell abundance, diversity indices and size-fractionated chlorophyll a(Chl-a) concentration monthly from March 2013 to January2014. The phytoplankton community, as observed by microscopy, underwent succession from nano-celled chained diatoms to dinoflagellates in the spring and summer, and then to micro-celled diatoms and nano-celled chained diatoms as the most prominent groups in the subsequent autumn and winter. Canonical correspondence analysis revealed that changes in the abundance of diatoms were related to nutrient availability, especially of nitrogen, while water temperature was the crucial factor influencing the abundance of dinoflagellates and A. anophagefferens. Considering the contribution of nano-celled chained species to the micro-sized Chl-a,small phytoplankton(<20 μm) composed the majority of total Chl-a and were generally abundant during our study. | 7 | Influence of the replacement of alkoxyl with alkylthienyl on photovoltaic properties of two small molecule donors for organic solar cells显示文摘Two benzo[1,2-b:4,5-b¢]dithiophene(BDT)-based small molecule(SM) donor materials with identical conjugated backbones but different substitution groups, named as DRTB-O and DRTB-T, were well explored to demonstrate the influence of the replacement of alkoxy with alkylthienyl on their photovoltaic properties in fullerene-based and fullerene-free organic solar cells(OSCs). The study shows that the two SM donors possess similar absorption spectra and energy levels but different crystalline structures in solid films. The carrier transport property and phase separation morphologies of the blend films have also been fully investigated.By employing PC71 BM as the acceptor, the power conversion efficiency(PCE) of DRTB-O:PC71BM and DRTB-T:PC71BM based devices were 4.91% and 7.08%, respectively. However, by blending with IDIC, the two SM donors exhibited distinctly different photovoltaic properties in fullerene-free OSCs, and the PCE of DRTB-O:IDIC and DRTB-T:IDIC based devices were 0.15% and9.06%, respectively. | 12 | Paclitaxel liposome for injection (Lipusu) plus cisplatin versus gemcitabine plus cisplatin in the first-line treatment of locally advanced or metastatic lung squamous cell carcinoma: A multicenter, randomized, open-label, parallel controlled clinical study显示文摘Background:Lipusu is the first commercialized liposomal formulation of pacli-taxel and has demonstrated promising efficacy against locally advanced lung squamous cell carcinoma(LSCC)in a small-scale study.Here,we conducted a multicenter,randomized,phase 3 study to compare the efficacy and safety of cis-platin plus Lipusu(LP)versus cisplatin plus gemcitabine(GP)as first-line treat-ment in locally advanced or metastatic LSCC.Methods:Patients enrolled were aged between 18 to 75 years,had locally advanced(clinical stage IIIB,ineligible for concurrent chemoradiation or surgery)or metastatic(Stage IV)LSCC,had no previous systemic chemother-apy and at least one measurable lesion as per the Response Evaluation Criteria in Solid Tumors(version 1.1)before administration of the trial drug.The primary endpoint was progression-free survival(PFS).The secondary endpoints included objective response rate(ORR),disease control rate(DCR),overall survival(OS),and safety profiles.To explore the possible predictive value of plasma cytokines for LP treatment,plasma samples were collected from the LP group at baseline and first efficacy evaluation time and were then subjected to analysis by 45-Plex ProcartaPlex Panel 1 to detect the presence of 45 cytokines using the Luminex xMAP technology.The correlation between treatment outcomes and dynamic changes in the levels of cytokines were evaluated in preliminary analyses.Results:The median duration of follow-up was 15.4 months.237 patients in the LP group and 253 patients in the GP group were included in the per protocol set(PPS).In the PPS,the median PFS was 5.2 months versus 5.5 months in the LP and GP group(hazard rtio[HR]:1.03,P=0.742)respectively.The median OS was 14.6 months versus 12.5 months in the LP and GP group(HR:0.83,P=0.215).The ORR(41.8%versus 45.9%,P=0.412)and DCR(90.3%versus 88.1%,P=0.443)were also similar between the LP and GP group.A significantly lower proportion of patients in the LP group experienced adverse events(AEs)leading to treatment interruptions(10.9%versus 26.4%,P<0.001)or treatment termination(14.3%versus 23.1%,P=0.011).The analysis of cytokine levels in the LP group showed that low baseline levels of 27 cytokines were associated with an increased ORR,and 15 cytokines were associated with improved PFS,with 14 cytokines,including TNF-a,IFN-y,IL-6,and IL-8,demonstrating an overlapping trend.Conclusion:The LP regimen demonstrated similar PFS,OS,ORR and DCR as the GP regimen for patients with locally advanced or metastatic LSCC but had more favorable toxicity profiles.The study also identified a spectrum of different cytokines that could be potentially associated with the clinical benefit in patients who received the LP regimen. | Jie Zhang Yueyin Pan Qin Shi Guojun Zhang Liyan Jiang Xiaorong Dong Kangsheng Gu Huijuan Wang Xiaochun Zhang Nong Yang Yuping Li Jianping Xiong Tienan Yi Min Peng Yong Song Yun Fan Jiuwei Cui Gongyan Chen Wei Tan Aimin Zang Qisen Guo Guangqiang Zhao Ziping Wang Jianxing He Wenxiu Yao Xiaohong Wu Kai Chen Xiaohua Hu Chunhong Hu Lu Yue Da Jiang Guangfa Wang Junfeng Liu Guohua Yu Junling Li Jianling Bai Wenmin Xie Weihong Zhao Lihong Wu Caicun Zhou | 2022 | Cancer Communications2022,42,1: | 1 |

| 15 | Variation in Sunshine Duration and Related Aerosol Influences at Shangdianzi GAW Station, China: 1958–2021显示文摘Sunshine duration(SD) is adopted widely to study global dimming/brightening. However, long-term simultaneous measurements of SD and closely related impact factors require further analysis to elucidate how and why SD has varied during the past decades. In this study, a long-term(1958–2021) SD data series obtained from the Shangdianzi Global Atmosphere Watch(GAW) station in China was analyzed to detect linear trends, climatic jumps, and climatic periods in SD using linear fitting, the Mann–Kendall trend test, and the continuous wavelet transform method. Annual SD exhibited steady dimming(-67.3 h decade-1) before 2010, followed by a period of brightening(189.9 h decade-1)during 2011–2020. An abrupt jump in annual SD occurred in 1995, and the annual SD anomaly exhibited significant oscillation with ~3-yr periodicity during 1960–1978. Partial least squares analysis revealed that annual SD anomaly was associated with variations in relative humidity, gale days, cloud cover, and black carbon(BC). Further analysis of the clear-sky daily sunshine percentage(DSP) and simultaneous measurements of aerosol properties, including aerosol optical depth, aerosol extinction coefficient, single scattering albedo(SSA), BC, and total suspended particulates, suggested that variation in DSP was affected primarily by aerosol scattering and absorption. Furthermore, the hourly clear-sky SD at high aerosol loading was approximately 60% and 56% of that at middle and low aerosol loadings, respectively. | 20 | Targetable elements in SARS-CoV-2 S2 subunit for the design of pan-coronavirus fusion inhibitors and vaccines显示文摘The ongoing global pandemic of coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS‐CoV‐2),has caused devastating impacts on the public health and the global economy.Rapid viral antigenic evolution has led to the continual generation of new variants.Of special note is the recently expanding Omicron subvariants that are capable of immune evasion from most of the existing neutralizing antibodies(nAbs).This has posed new challenges for the prevention and treatment of COVID-19.Therefore,exploring broad-spectrum antiviral agents to combat the emerging variants is imperative.In sharp contrast to the massive accumulation of mutations within the SARS-CoV-2 receptor-binding domain(RBD),the S2 fusion subunit has remained highly conserved among variants.Hence,S2-based therapeutics may provide effective cross-protection against new SARS-CoV-2 variants.Here,we summarize the most recently developed broad-spectrum fusion inhibitors(e.g.,nAbs,peptides,proteins,and small-molecule compounds)and candidate vaccines targeting the conserved elements in SARS-CoV-2 S2 subunit.The main focus includes all the targetable S2 elements,namely,the fusion peptide,stem helix,and heptad repeats 1 and 2(HR1-HR2)bundle.Moreover,we provide a detailed summary of the characteristics and action-mechanisms for each class of cross-reactive fusion inhibitors,which should guide and promote future design of S2-based inhibitors and vaccines against new coronaviruses. | Liyan Guo Sheng Lin Zimin Chen Yu Cao Bin He Guangwen Lu | 2023 | Signal Transduction and Targeted Therapy2023,8,6: | 0 |
