| 1 | Whole-genome sequencing provides insights into the genetic diversity and domestication of bitter gourd(Momordica spp.)显示文摘Bitter gourd(Momordica charantia)is a popular cultivated vegetable in Asian and African countries.To reveal the characteristics of the genomic structure,evolutionary trajectory,and genetic basis underlying the domestication of bitter gourd,we performed whole-genome sequencing of the cultivar Dali-11 and the wild small-fruited line TR and resequencing of 187 bitter gourd germplasms from 16 countries.The major gene clusters(Bi clusters)for the biosynthesis of cucurbitane triterpenoids,which confer a bitter taste,are highly conserved in cucumber,melon,and watermelon.Comparative analysis among cucurbit genomes revealed that the Bi cluster involved in cucurbitane triterpenoid biosynthesis is absent in bitter gourd.Phylogenetic analysis revealed that the TR group,including 21 bitter gourd germplasms,may belong to a new species or subspecies independent from M.charantia.Furthermore,we found that the remaining 166 M.charantia germplasms are geographically differentiated,and we identified 710,412,and 290 candidate domestication genes in the South Asia,Southeast Asia,and China populations,respectively.This study provides new insights into bitter gourd genetic diversity and domestication and will facilitate the future genomics-enabled improvement of bitter gourd. | Junjie Cui Yan Yang Shaobo Luo Le Wang Rukui Huang Qingfang Wen Xiaoxia Han Nansheng Miao Jiaowen Cheng Ziji Liu Changyuan Zhang Chengcheng Feng Haisheng Zhu Jianwen Su Xinjian Wan Fang Hu Yu Niu Xiaoming Zheng Yulan Yang Dai Shan Zhensheng Dong Weiming He Narinder P.S.Dhillon Kailin Hu | 2020 | Horticulture Research2020，7，1： | 7 |
| 2 | The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. | 4 | Enhanced magnetic properties and improved corrosion performance of nanocrystalline Pr-Nd-Y-Fe-B spark plasma sintered magnets显示文摘Recently it is a hot topic to make full use of high abundant Y element in Nd_(2)Fe_(14)B-type permanent magnets.In contrast to Pr and Nd elements,Y shows different metallurgical behaviors during preparation process.In this paper,we have explored the magnetic properties,microstructures and corrosion performance of Pr-Nd-Y-Fe-B magnets fabricated by spark plasma sintering(SPS)technique from the ribbons of nanocrystalline and amorphous precursors,respectively.The coercivity and maximum energy product were improved for the magnets prepared from amorphous precursor materials(denoted as SPS-A hereafter)compared with the magnets prepared from crystalline precursor materials(denoted as SPS-C hereafter).Magnetic properties of Jr=0.79 T,Hci=864 k A/m,and(BH)_(max)=102 k J/m^(3)were obtained for SPS-A magnets.In contrast with SPS-C magnets,the magnetic properties of SPS-A magnets are not so sensitive to the preparation conditions,which is quite beneficial to the homogeneity of microstructure and enhancement of coercivity for large-scale production of the designated magnets.Aggregated(Pr,Nd,Y)-rich phase was found out in SPS-C magnets.Pr and Nd elements are rich at grain boundary while Y is distributed uniformly at main phase and grain boundary phase.The strip grains and equiaxed grains exist in SPS-C and SPS-A magnets,respectively.The enhanced magnetic properties for SPS-A magnets are accredited to the uniform distribution of rare-earth-rich phase and low demagnetization factor.It is revealed by electrochemical test and dipping test that the corrosion potential is more positive and the corrosion rate is slower for the SPS-A magnets in 3.5 wt.%Na Cl solution.The work is also expected to shed light on developing the nanocrystalline Pr-Nd-Y-Fe-B SPSed high-performance magnets in industry. | Qingzheng Jiang Jie Song Qingfang Huang Sajjad Ur Rehman Lunke He Qingwen Zeng Zhenchen Zhong | 2020 | Journal of Materials Science & Technology2020，58，23： | 1 |

| 6 | An Extracellular Oligopeptide Permease May Be a Potential Virulence Factor of Vibrio harveyi显示文摘An oligopeptide permease A(OppA)was purified from the extracellular product of Vibrio harveyi SF-1.The molecular weight of the purified protein was estimated to be 58 kDa on SDS-PAGE.The purified protein showed phospholipase C activity at the optimal values of temperature 50℃ and pH 8.0.The enzymatic activity decreased when the temperature increased to 40℃.The N-terminal sequence of the purified protein was determined as ADVPAGTKLA,which is similar to that of OppA.The OppA pre-cursor gene was cloned from the genome of V.harveyi SF-1.The gene consisted of 1665 base pairs and encoded a 554 amino acid polypeptide,which showed a high similarity to those of the OppAs of V.harveyi and other Vibrio species.The gene was subcloned into pET-28a(+)and expressed in Escherichia coli.The expressed recombinant protein was purified by Ni-NTA metal affinity chro-matography.The expressed recombinant protein showed a 58 kDa band on SDS-PAGE and exhibited phospholipase C activity with the optima of temperature 50℃ and pH 8.0.The purified protein was toxic to the flounder gill cells.An OppA mutant of V.harveyi SF-1 was constructed by homologous recombination.The mutant strain was less virulent when it was intraperitoneally inoculated to zebra fish,with the LD50 of 5.46×105 CFU fish-1,compared to 3.11×104 CFU fish-1 of the wild-type strain,which indicated that the OppA might play an important role in the pathogenicity of V.harveyi. | HE Qingfang CHEN Jixiang LI Caifeng | 2011 | Journal of Ocean University of China2011，10，4： | 0 |
