2β-Cypermethrin promotes the adipogenesis of 3T3-L1 cells via inducing autophagy and shaping an adipogenesis-friendly microenvironment显示文摘The toxicity of synthetic pyrethroids has garnered attention, and studies have revealed that pyrethroids promote fat accumulation and lead to obesity in mice. Nevertheless, the effect of β-cypermethrin (β-CYP) on adipogenesis and its underlying mechanism remains largely unknown. In this study, mouse embryo fibroblasts 3T3-L1 cells were exposed to β-CYP, and the cell viability, intracellular reactive oxygen species (ROS) level, autophagy, and adipogenesis were assessed to investigate the roles of oxidative stress and autophagy in the toxic effects of β-CYP on adipogenesis. The results demonstrated that treatment with 100 μΜ β-CYP elevated the ROS level, decreased mitochondrion membrane potential, stimulated autophagy, and enhanced the adipogenesis induced by the mixture of insulin, dexamethasone, and 3-isobutyl-1-methylxanthine. However, co-treatment with N-acetyl-L-cysteine partially blocked the abovementioned effects of β-CYP in 3T3-L1 cells. In addition, co-treatment with rapamycin, an autophagy agonist, enhanced the inductive effect of β-CYP on adipogenesis, whereas co-treatment with 3-methyladenine blocked the enhancement of adipogenesis caused by β-CYP. Moreover, β-CYP also altered the microenvironment of 3T3-L1 cells to an adipogenesis-friendly one by reducing the extracellular expression of miR-34a, suggesting that the culture media of β-CYP-treated 3T3-L1 cells could shift macrophages to M2 type. Taken together, the data obtained in the present study demonstrated that β-CYP promoted adipogenesis via oxidative stress-mediated autophagy disturbance, and it caused macrophage M2 polarization via the alteration of miR-34a level in the microenvironment. 3Xiyanping injection therapy for children with mild hand foot and mouth disease:a randomized controlled trial显示文摘OBJECTIVE: To evaluate the clinical effects of Xiyan-ping injection intervention in pediatric patients with mild hand foot and mouth disease(HFMD).METHODS: A total of 329 patients were stratified and block-randomized for symptomatic treatment of HFMD and assigned to one of the following groups: Western Medicine(group A, n = 103), Xiyanping injection(group B, n = 109), or Xiyanping injection and symptomatic treatment using Western Medicine(group C, n = 117). During the trial, fever,rash, ulcers of the mouth were observed among participants in each group before and after treatment, and conversion rates from mild to severe HFMD were measured.RESULTS: After 3-7 days' treatment, no significant differences in the conversion rates from mild to severe HFMD were observed among the three groups(P > 0.05). There was a significantly low number of patients with the onset time of antifebrile effect,vanished time of hand and foot rashes and cumulative time for the ulcers in the mouth vanished,among the three groups(P < 0.05, P < 0.01). The onset time of antifebrile effect of patients in groups B and C were markedly shorter compared with those in group A(P < 0.05, P < 0.01); there was no statically significant difference observed between groups B and C(P > 0.05). No significant differences were observed in the time for body temperature recovery among the three groups(P > 0.05). There were no significant differences in adverse effects among the three groups(P > 0.05).CONCLUSION: Xiyanping injection together with use of Western Medicine is most effective for symptomatic treatment of mild HFMD. 4Behavior and adrenal activity in a group of zoo golden snub-nosed monkeys,Rhinopithecus roxellana,following social structure change显示文摘The effects of individual removal by zoo managers on behavior and fecal glucocorticoid changes in snub-nosed monkeys(Rhinopithecus roxellana) were investigated at Beijing Zoo.The results showed that the effect of the removal of three subordinate individuals(DD,male,4 years;XZZ,male,3 years and LL,female,3 years) from a group varied across the remaining group members that may be related to individual's social status,gender and age.The dominant male was impacted less than subordinate individuals in both the frequency and time duration for the four main behavioral categories.The frequency of stereotypic behavior significantly decreased in the dominant female after the removal of three animals(Z= 2.862,P=0.003).Fluctuations were found in both the behavioral frequency and time duration in other remaining individuals.These findings suggest that small,socially stable groups were somewhat resilient to changes in member composition.Moreover,the cortisol level did not change significantly in the remaining members.An alternative method of hormone assay using fecal samples for vulnerable animals was presented,and that may be useful for monitoring wild golden monkeys and other endangered species of animal.ZHANG XiaoLii HE Xini LIU DingZheni YAO XiuLini ZHANG YiZhuo ZHANG EnQuan ZHANG JinGuo JIANG GuoHua ZHAO ChangQi 2013Chinese Science Bulletin2013,58,17:4
5Modeling and Control of an Isolated Module Multilevel DC/DC Converter for DC Grid显示文摘This paper presents an isolated DC/AC/DC converter using a middle frequency transformer coupling two modular multilevel converters(MMC),suitable for interconnecting DC transmission lines of different voltage levels in high voltage direct current(HVDC)system.The basic operational principle of the isolated module multilevel DC/DC converter(IMMDCC)is analyzed.The dynamic model of IMMDCC is studied in detail and the transient relationship between DC side and AC side of IMMDCC is revealed,which is physically straightforward for understanding the power transfer in IMMDCC.The control strategy in D-Q coordinate system is put forward,and the fault characteristic and corresponding protection method is analyzed.Finally,computer simulation using Matlab/Simulink is performed to verify the dynamic model and the proposed control strategy.The simulation results show good performances and the quick response ability of the proposed control strategy.Xinying Wang Guangfu Tang Zhiyuan He Xiaoguang Wei Hui Pang Xiangning Xiao 2017CSEE Journal of Power and Energy Systems2017,3,2:3
6Comparison of gut microbiota in autism spectrum disorders and neurotypical boys in China:A case-control study显示文摘Background:Autism spectrum disorders(ASDs)are a set of complex neurobiological disorders.Growing evidence has shown that the microbiota that resides in the gut can modulate brain development via the gut–brain axis.However,direct clinical evidence of the role of the microbiota–gut–brain axis in ASD is relatively limited.Methods:A case-control study of 71 boys with ASD and 18 neurotypical controls was conducted at China-Japan Friendship Hospital.Demographic information and fecal samples were collected,and the gut microbiome was evaluated and compared by 16S ribosomal RNA gene sequencing and metagenomic sequencing.Results:A higher abundance of operational taxonomic units(OTUs)based on fecal bacterial profiling was observed in the ASD group.Significantly different microbiome profiles were observed between the two groups.At the genus level,we observed a decrease in the relative abundance of Escherichia,Shigella,Veillonella,Akkermansia,Provindencia,Dialister,Bifidobacterium,Streptococcus,Ruminococcaceae UCG_002,Megasphaera,Eubacterium_coprostanol,Citrobacter,Ruminiclostridium_5,and Ruminiclostridium_6 in the ASD cohort,while Eisenbergiella,Klebsiella,Faecalibacterium,and Blautia were significantly increased.Ten bacterial strains were selected for clinical discrimination between those with ASD and the neurotypical controls.The highest AUC value of the model was 0.947.Conclusion:Significant differences were observed in the composition of the gut microbiome between boys with ASD and neurotypical controls.These findings contribute to the knowledge of the alteration of the gut microbiome in ASD patients,which opens the possibility for early identification of this disease.Fang Ye Xinying Gao Zhiyi Wang Shuman Cao Guangcai Liang Danni He Zhitang Lv Liming Wang Pengfei Xu Qi Zhang 2021Synthetic and Systems Biotechnology2021,6,2:2

10Extraction Process and Quality Standard of Danshen Jianxin Capsules显示文摘[Objectives]This study was conducted to establish the quality standard of Danshen Jianxin Capsules.[Methods]Orthogonal design was adopted to optimize the water extraction process by taking the amount of water added,extraction time and extraction times as factors,and the content of tanshinone I as the index.Salviae Miltiorrhizae,Notoginseng Radix Et Rhizoma and Radix Astragali in the capsules were qualitatively identified by TLC,and the content of tanshinone I in the capsules was determined by HPLC.[Results]The best water extraction process included the steps of adding 12 times of water each time,extracting the materials twice,for 1 h each time.In TLC identification,the test samples showed spots of the same color at the positions corresponding to tanshinone II A,ginsenoside Rg1,notoginsenoside R1 and astragaloside IV reference samples.[Conclusions]Danshen Jianxin Capsules was prepared by the water extraction method,which is characterized by high efficiency and being suitable for mass production.The quality control method is reliable,fast and accurate,which can effectively control the quality of the product.Chengtong LIU Xinying MO Silu HE Donglian CHEN Guilin YANG Hongsheng TAN Peng LIANG Haisheng ZENG 2023Medicinal Plant2023,14,1:1

14MEIC-global-CO_(2):A new global CO_(2)emission inventory with highly-resolved source category and sub-country information显示文摘CO_(2)emission inventory provides fundamental data for climate research and emission mitigation.Currently,most global CO_(2)emission inventories were developed with energy statistics from International Energy Agency(IEA)and were available at country level with limited source categories.Here,as the first step toward a high-resolution and dynamic updated global CO_(2)emission database,we developed a data-driven approach to construct seamless and highly-resolved energy consumption data cubes for 208 countries/territories,797 sub-country administrative divisions in 29 countries,42 fuel types,and 52 sectors,with the fusion of activity data from 24 international statistics and 65 regional/local statistics.Global CO_(2)emissions from fossil fuel combustion and cement production in 1970–2021 were then estimated with highly-resolved source category(1,484 of total)and sub-country information(797 of total).Specifically,73%of global CO_(2)emissions in 2021 were estimated with sub-country information,providing considerably improved spatial resolution for global CO_(2)emission accounting.With the support of detailed information,the dynamics of global CO_(2)emissions across sectors and fuel types were presented,representing the evolution of global economy and progress of climate mitigation.Remarkable differences of sectoral contribution were found across sub-country administrative divisions within a given country,revealing the uneven distribution of energy and economic structure among different regions.Our estimates were generally consistent with existing databases at aggregated level for global total or large emitters,while large discrepancies were observed for middle and small emitters.Our database,named the Multiresolution Emission Inventory model for Climate and air pollution research(MEIC)is publicly available through http://gffzz6c575fb1d0a54d36hwbcovou5wo906qun.ffgz.tsg.suse.edu.cn with highly-resolved information and timely update,which provides an independent carbon emission accounting data source for climate research.Ruochong XU Dan TONG Qingyang XIAO Xinying QIN Cuihong CHEN Liu YAN Jing CHENG Can CUI Hanwen HU Wenyu LIU Xizhe YAN Huaxuan WANG Xiaodong LIU Guannan GENG Yu LEI Dabo GUAN Kebin HE Qiang ZHANG 2024Science China Earth Sciences2024,67,2:0
15Impact of body mass index,weight gain,and metabolic disorders on survival and prognosis in patients with breast cancer who underwent chemotherapy显示文摘Background: Weight gain during chemotherapy in patients with breast cancer contributes to their poor prognosis. However, a growing number of studies have found that metabolic disorders seem to play a more important role in breast cancer prognosis than weight gain. This study aimed to explore the prognostic effects of body mass index (BMI), weight gain, and metabolic disorders on the overall survival (OS) and prognosis of patients with breast cancer who underwent chemotherapy.Methods: Data from the inpatient medical records of patients with breast cancer who underwent chemotherapy at the Beijing Cancer Hospital Breast Cancer Center from January to December 2010 were retrospectively collected, and the patients were followed up until August 2020.Results: A total of 438 patients with stages I to III breast cancer met the inclusion and exclusion criteria. Forty-nine (11.19%) patients died, while 82 (18.72%) patients had tumor recurrence and metastasis at the last follow-up (August 2020). From the time of diagnosis until after chemotherapy, no significant differences were observed in the body weight (t=4.694,P<0.001), BMI categories (χ^(2)=19.215,P=0.001), and incidence of metabolic disorders (χ^(2)=24.841,P<0.001);the BMI categories and weight change had no effect on the OS. Both univariate (χ^(2)=6.771,P=0.009) and multivariate survival analyses (hazard ratio=2.775, 95% confidence interval [CI]: 1.326-5.807,P=0.007) showed that low high-density lipoprotein cholesterol (HDL-C) levels at diagnosis had a negative impact on the OS. The multivariate logistic regression analysis showed that the HDL-C level at diagnosis (odds ratio [OR]=2.200, 95% CI: 0.996-4.859,P=0.051) and metabolic disorders after chemotherapy (OR=1.514, 95% CI: 1.047-2.189,P=0.028) are risk factors for poor prognosis in patients with breast cancer.Conclusions: Chemotherapy led to weight gain and aggravated the metabolic disorders in patients with breast cancer. 16Insights into the Functionalization of the Methylsalicyclic Moiety during the Biosynthesis of Chlorothricin by Comparative Kinetic Assays of the Activities of Two KAS Ⅲ-like Acyltransferases显示文摘Summary of main observation and conclusion Chlorothricin (CHL),an archetypal member of the family of spirotetronate antibiotics,possesses a tetronate-containing pentacyclic aglycone that is conjugated with a modified methylsalicyclic acid (MSA) moiety through a disaccharide linkage.MSA is a polyketide product assembled by the iterative type Ⅰ polyketide synthase ChlB1.incorporation of this pharmaceutically important moiety into CHL relies on the activities of two distinct β-Ketoacyl-ACP synthase Ⅲ(KAS Ⅲ)-Iike acyltransferases,ChlB3 and ChlB6,which function together to coordinate the transfer of MSA through ChlB2,a discrete acyl carrier protein (ACP).During the maturation of CHL,MSA needs to be further functionalized by C2-O-methylation and C5-chlorination;however,timing of this functionalization process remains poorly understood.In this study,we report comparative kinetic assays of the activities of the two KAS Ⅲ-like acyltransferases ChlB3 and ChlB6 using substrates that vary in substitution extent and ACP carrier.ChlB3 prefers to transfer the immediately assembled 6-methyI-MSA moiety from ChlB1-ACP to the discrete ACP ChlB2,from which this moiety is preferred to be transferred directly onto the molecule desmethylsalicyl-CHL prior to C2-O-methylation and C5-chlorination.Consequently,MSA functionalization appears to occur at the molecule level rather than at the covalently tethered protein level,i.e.,ChlB1-ACP or ChlB2.Both ChlB3 and ChlB6 are flexible in substrate tolerance,holding promise for CHL engineering-based structural diversity by using variable MSA moiety.Xuan Yi Qunfei Zhao Zhenhua Tian Xinying Jia Weiguo Cao Wen Liu Qing-Li He 2019Chinese Journal of Chemistry2019,37,8:0

18Computation Rate Maximization in Multi-User Cooperation-Assisted Wireless-Powered Mobile Edge Computing with OFDMA显示文摘In the era of Internet of Things(Io T),mobile edge computing(MEC)and wireless power transfer(WPT)provide a prominent solution for computation-intensive applications to enhance computation capability and achieve sustainable energy supply.A wireless-powered mobile edge computing(WPMEC)system consisting of a hybrid access point(HAP)combined with MEC servers and many users is considered in this paper.In particular,a novel multiuser cooperation scheme based on orthogonal frequency division multiple access(OFDMA)is provided to improve the computation performance,where users can split the computation tasks into various parts for local computing,offloading to corresponding helper,and HAP for remote execution respectively with the aid of helper.Specifically,we aim at maximizing the weighted sum computation rate(WSCR)by optimizing time assignment,computation-task allocation,and transmission power at the same time while keeping energy neutrality in mind.We transform the original non-convex optimization problem to a convex optimization problem and then obtain a semi-closed form expression of the optimal solution by considering the convex optimization techniques.Simulation results demonstrate that the proposed multi-user cooperationassisted WPMEC scheme greatly improves the WSCR of all users than the existing schemes.In addition,OFDMA protocol increases the fairness and decreases delay among the users when compared to TDMA protocol.Xinying Wu Yejun He Asad Saleem 2023China Communications2023,20,1:0
19Alternative splicing of a carboxyl/choline esterase gene enhances the fenpropathrin tolerance of Tetranychus cinnabarinus显示文摘Detoxification plays a crucial role in agricultural pests to withstand pesticides,and cytochrome P450s,carboxyl/choline esterases(CCEs),and glutathione-S-transferases are the main proteins responsible for their detoxification ability.The activity of CCEs can be upregulated,downregulated,or modified by mutation.However,few studies have examined the role of alternative splicing in altering the properties of CCEs.We identified 2 variants of TcCCE23 in Tetranychus cinnabarinus:a long version(CCE23-V1)and a short version that is 18 nucleotides shorter than CCE23-V1(CCE23-V2).Whether splicing affects the activity of TcCCE23 remains unclear.Overexpression of CCE23-V2 in fenpropathrin-resistant T.cinnabarinus revealed that splicing affected the detoxification of fenpropathrin by CCE23-V2.The mortality of mites was significantly higher when the expression of CCE23-V2 was knocked down(43.2%±3.3%)via injection of CCE23-dsRNA(double-stranded RNA)compared with the control group injected with green fluorescent protein-dsRNA under fenpropathrin exposure;however,the downregulation of CCE23-V1(61.3%±6.3%)by CCE23-small interfering RNA had no such effect,indicating CCE23-V2 plays a greater role in xenobiotic metabolism than CCE23-V1.The tolerance of flies overexpressing CCE23-V2 to fenpropathrin(50%lethal dose[LD_(50)]=19.47μg/g)was significantly higher than that of Gal4/UAS-CCE23-V1 transgenic flies(LD_(50)=13.11μg/g).Molecular docking analysis showed that splicing opened a“gate”that enlarges the substrate binding cavity of CCE23-V2,might enhance the ability of CCE23-V2 to harbor fenpropathrin molecules.These findings suggest that splicing might enhance the detoxifying capability of TcCCE23.Generally,our data improve the understanding of the diversity and complexity of the mechanisms underlying the regulation of CCEs.Peng Wei Xinying Zeng Haonan Han Yiqing Yang Youjun Zhang Lin He 2023Insect Science2023,30,5:0
