2Hes1 Knockdown Exacerbates Ischemic Stroke Following tMCAO by Increasing ER Stress-Dependent Apoptosis via the PERK/ eIF2a/ATF4/CHOP Signaling Pathway显示文摘Apoptosis induced by endoplasmic reticulum(ER)stress plays a crucial role in mediating brain damage after ischemic stroke.Recently,Hes1(hairy and enhancer of split 1)has been implicated in the regulation of ER stress,but whether it plays a functional role after ischemic stroke and the underlying mechanism remain unclear.In this study,using a mouse model of ischemic stroke via transient middle cerebral artery occlusion(tMCAO),we found that Hes1 was induced following brain injury,and that siRNA-mediated knockdown of Hes1 increased the cerebral infarction and worsened the neurological outcome,suggesting that Hes1 knockdown exacerbates ischemic stroke.In addition,mechanistically,Hes1 knockdown promoted apoptosis and activated the PERK/eIF2a/ATF4/CHOP signaling pathway after tMCAO.These results suggest that Hes1 knockdown promotes ER stress-induced apoptosis.Furthermore,inhibition of PERK with the specific inhibitor GSK2606414 markedly attenuated the Hes1 knockdown-induced apoptosis and the increased cerebral infarction as well as the worsened neurological outcome following tMCAO,implying that the protection of Hes1 against ischemic stroke is associated with the amelioration of ER stress via modulating the PERK/eIF2a/ATF4/CHOP signaling pathway.Taken together,these results unveil the detrimental role of Hes1 knockdown after ischemic stroke and further relate it to the regulation of ER stress-induced apoptosis,thus highlighting the importance of targeting ER stress in the treatment of ischemic stroke.Yueyong Li Yingjun Zhang Huangde Fu Huadong Huang Qifeng Lu Houji Qin Yingning Wu Huatuo Huang Guizhen Mao Zhongheng Wei Pinhu Liao 2020Neuroscience Bulletin2020,36,2:12
3Ecological provinces of spring phytoplankton in the Yellow Sea:species composition显示文摘Phytoplanktonic ecological provinces of the Yellow Sea(31.20°–39.23°N, 121.00°–125.16°E) is derived in terms of species composition and hydrological factors(temperature and salinity). 173 samples were collected from 40 stations from April 28 to May 18, 2014, and a total of 185 phytoplanktonic algal species belonging to 81 genera of 7phyla were identified by Uterm?hl method. Phytoplankton abundance in surface waters is concentrated in the west coast of Korean Peninsula and Korea Bay, and communities in those areas are mainly composed of diatoms and cyanobacteria with dominant species of Cylindrotheca closterium, Synechocystis pevalekii, Chroomonas acuta,Paralia sulcata, Thalassiosira pacifica and Karenia mikimotoi, etc. The first ten dominant species of the investigation area are analyzed by multidimensional scaling(MDS) and cluster analysis, then the Yellow Sea is divided into five provinces from Province I(P-I) to Province V(P-V). P-I includes the coastal areas near southern Liaodong Peninsula, with phytoplankton abundance of 35 420×10~3–36 163×10~3 cells/L and an average of 35 791×10~3 cells/L, and 99.84% of biomass is contributed by cyanobacteria. P-II is from Shandong Peninsula to Subei coastal area. Phytoplankton abundance is in a range of 2×10~3–48×10~3 cells/L with an average of 24×10~3cells/L, and 63.69% of biomass is contributed by diatoms. P-III represents the Changjiang(Yangtze River) Diluted Water. Phytoplankton abundance is 10×10~3–37×10~3 cells/L with an average of 24×10~3 cells/L, and 73.14% of biomass is contributed by diatoms. P-IV represents the area affected by the Yellow Sea Warm Current.Phytoplankton abundance ranges from 6×10~3 to 82×10~3 cells/L with an average of 28×10~3 cells/L, and 64.17% of biomass is contributed by diatoms. P-V represents the cold water mass of northern Yellow Sea. Phytoplankton abundance is in a range of 41×10~3–8 912×10~3 cells/L with an average of 1 763×10~3 cells/L, and 89.96% of biomass is contributed by diatoms. Overall, structures of phytoplankton community in spring are quite heterogeneous in different provinces. 4Grain-Size Distribution of Surface Sediments in the Bohai Sea and the Northern Yellow Sea: Sediment Supply and Hydrodynamics显示文摘The grain-size distribution of surface sediments in the Bohai Sea(BS) and the northern Yellow Sea(NYS), and its relationship with sediment supply and hydrodynamic environment were investigated based on grain-size compositions of surface sediments and modern sedimentation rates. The results showed that the surface sediments in the BS and the NYS were primarily composed of silty sand and clayey silt with a dominant size of silt. In addition, the Yellow River delivered high amount of water and sediments to the BS, and they are dominated in surface sediments(mainly silt) in the Bohai Bay, the Yellow River mouth, the center of the BS, and the north coast of Shandong Peninsula. The coarse-grained sediments were mainly deposited at the river mouth due to the estuarine filtration and physical sorting. Meanwhile, there was a significant relationship among the modern sedimentation rate, the surface sediment grain size distribution and sediment transport pattern. The areas with coarser surface sediments generally corresponded low sedimentation rates because of strong erosion;whereas the sedimentation rate was relatively high at the place that the surface sediments were fine-grained. 6Rapid shifts of the river plume pathway off the Huanghe (Yellow) River mouth in response to water-sediment regulation scheme in 2005显示文摘Freshwater plume represents a key passage for the river to deliver sediment to the coastal ocean. A wa- ter-sediment regulation scheme was implemented for the Huanghe River in 2005; in order to examine the effect of such an activity on the river plume dynamics, three cruises were carried out off the Huanghe River mouth. The prelimi- nary results of the in-situ measurements suggested that the plume pathway shifted rapidly during the period of less than twenty days, which was confirmed by satellite remote sensing data in the same period. The rapid shifts were resulted from intensive interactions between river discharge and the vary- ing bathymetry in the river mouth area. 8Geochemical and Grain-Sized Implications for Provenance Variations of the Central Yellow Sea Muddy Area Since the Middle Holocene显示文摘Based on high-resolution analysis to a 280-cm long sediment core obtained from the muddy area in the central Yellow Sea, we examined the provenance of muddy sediments and discussed the changing marine sedimentary environment since the middle Holocene. The results indicated that fine-grained sediments in the muddy area were mainly derived from the Huanghe(Yellow River) and Changjiang(Yangtze River) with considerable stepwise variations during the past 6.6 kyr. The Yellow Sea Warm Current was initiated at 6 kyr when the sea level was high together with the enhanced East Asian Winter Monsoon. These in combination established the framework of shelf circulation in the Yellow Sea that began to trap the river-derived fine-grained sediments. From 4.9 kyr to 2.8 kyr, both the Kushiro Current and East Asian Monsoon were significantly weakened, reducing the delivery of Changjiang sediments to the muddy area. As a result, the sediments were mainly originated from the Huanghe. From 2.8 kyr to 1.5 kyr the continuously weakened East Asian Winter Monsoon and enhanced Yellow Sea Warm Current entrapped more fine-grain sediments. Whereas the enhanced East Asian Winter Monsoon and the human caused increase in sediment load of the Huanghe since 1.5 kyr, and direct delivery of Huanghe sediments to the Yellow Sea during 1128–1855 AD might dominated the sedimentation in the study area. 9The Ap-2α/Elk-1 axis regulates Sirpα-dependent tumor phagocytosis by tumor-associated macrophages in colorectal cancer显示文摘The inhibitory receptor signal regulatory protein-α(Sirpα)is a myeloid-specific immune checkpoint that engages the“don’t eat me”signal CD47,which is expressed on tumor and normal tissue cells.However,the profile and regulatory mechanism of Sirpαexpression in tumor-associated macrophages(TAMs)are still not clear.Here,we found that the expression of Sirpαin TAMs increased dynamically with colorectal cancer(CRC)progression.Mechanistically,CRC cell-derived lactate induced the nuclear translocation of the transcription factor Ap-2αfrom the cytoplasm in TAMs.Ap-2αfunctioned as a transcription factor for Elk-1 by binding to the conserved element GCCTGC located at−1396/−1391 in the mouse Elk-1 promoter.Subsequently,the Elk-1 protein bound to two conserved sites,CTTCCTACA(located at−229/−221)and CTTCCTCTC(located at−190/−182),in the mouse Sirpαpromoter and promoted Sirpαexpression in TAMs.Functionally,the macrophage-specific knockout of Ap-2αnotably promoted the phagocytic activity of TAMs and suppressed CRC progression,whereas these effects were prevented by the transgenic macrophage-specific expression of Elk-1,which regulated TAM phagocytosis and CRC development in a Sirpα-dependent manner.Furthermore,we showed that Elk-1 expression was positively correlated with Sirpαexpression in TAMs and was associated with poor survival in CRC patients.Taken together,our findings revealed a novel mechanism through which CRC evades innate immune surveillance and provided potential targets for macrophage-based immunotherapy for CRC patients.Xiaojiao Wang Xi Luo Chuan Chen Ye Tang Lian Li Banghui Mo Houjie Liang Songtao Yu 2020Signal Transduction and Targeted Therapy2020,5,1:2

13MOTION-BASED REGION GROWING SEGMENTATION OF IMAGE SEQUENCES显示文摘This paper proposes a motion-based region growing segmentation scheme for the object-based video coding, which segments an image into homogeneous regions characterized by a coherent motion. It adopts a block matching algorithm to estimate motion vectors and uses morphological tools such as open-close by reconstruction and the region-growing version of the watershed algorithm for spatial segmentation to improve the temporal segmentation. In order to determine the reliable motion vectors, this paper also proposes a change detection algorithm and a multi-candidate pro- screening motion estimation method. Preliminary simulation results demonstrate that the proposed scheme is feasible.
