| 6 | Correlation of the temporal and spatial expression patterns of HQT with the biosynthesis and accumulation of chlorogenic acid in Lonicera japonica flowers显示文摘Hydroxycinnamoyl-CoA quinate transferase(HQT)is one of the key enzymes in the biosynthesis of chlorogenic acid(CGA)in the flowers of Lonicera japonica.However,the spatiotemporal expression patterns of HQT and its relationship to the dynamics of CGA biosynthesis,transport,and storage remain largely unknown.In this study,we collected L.japonica flower samples at different growth stages(S1–S6)and examined the spatiotemporal expression pattern of HQT and the dynamic accumulation patterns of CGA using a combination of molecular and cytological techniques.Our results suggest that the spatiotemporal expression pattern of HQT is directly correlated with dynamic changes in CGA accumulation and distribution in L.japonica flowers.We further show that CGA is synthesized primarily in the cytoplasm and chloroplasts.CGA synthesized in the cytoplasm first accumulates in specialized vesicles and is then transferred to large central vacuoles for storage by fusion of CGA-containing vesicles with vacuoles.Furthermore,CGA synthesized in the chloroplasts appears to be transferred into the vacuoles for storage by direct membrane fusion between the tonoplast and the disrupted chloroplast membranes.Collectively,our results suggest that CGA is synthesized in chloroplasts and cytoplasm and finally transferred to the vacuole for long-term storage. | Yanqun Li Dexin Kong Mei Bai Hanjun He Haiyang Wang Hong Wu | 2019 | Horticulture Research2019,6,1: | 7 |

Herein, we developed an environmental friendly route to produce HMDA via catalytic reductive amination of 1,6-hexanediol(HDO) in the presence of hydrogen. The activities of several heterogeneous metal catalysts such as supported Ni, Co, Ru, Pt, Pd catalysts were screened for the present reaction in supercritical ammonia without any additives. Among the catalysts examined, Ru/Al_2O_3 presented a high catalytic activity and highest selectivity for the desired product of HMDA. The high performance of Ru/Al_2O_3 was discussed based on the Ru dispersion and the surface properties like the acid-basicity. | 10 | Conversion of a normal maize hybrid into a waxy version using in vivo CRISPR/Cas9 targeted mutation activity显示文摘Waxy maize is a specialty maize that produces mainly amylopectin starch with special food or industrial values. The objective of this study was to overcome the limitations of wx mutant allele acquisition and breeding efficiency by conversion of parental lines from normal to waxy maize. The intended mutation activity was achieved by in vivo CRISPR/Cas9 machinery involving desired-target mutation of the Wx locus in the ZC01 background,abbreviated as ZC01-DTM^(wx). Triple selection was applied to segregants to obtain high genome background recovery with transgene-free wx mutations. The targeted mutation was identified, yielding six types of mutations among progeny crossed with ZC01-DTM^(wx).The amylopectin contents of the endosperm starch in mutant lines and hybrids averaged94.9%, while those of the wild-type controls were significantly(P < 0.01) lower, with an average of 76.9%. Double selection in transgene-free lines was applied using the Bar strip test and Cas9 PCR screening. The genome background recovery ratios of the lines were determined using genome-wide SNP data. That of lines used as male parents was as high as98.19% and that of lines used as female parents was as high as 86.78%. Conversion hybrids and both parental lines showed agronomic performance similar to that of their wild-type counterparts. | 12 | Severity of early allograft dysfunction following donation after circulatory death liver transplantation:a multicentre study显示文摘Background:Early allograft dysfunction(EAD)is associated with decreased graft and patient survival rates.This study aimed to identify the severity of EAD and develop a predictive model for EAD after donation after circulatory death(DCD)liver transplantation(LT).Furthermore,the influence of operative time on EAD incidence was also evaluated.Methods:In this retrospective,multicentre cohort study,nomograms were established based on a single-centre training cohort(n=321)and validated in a 3-center validation cohort(n=501).Results:The incidence rate of EAD was 46.4%(149/321)in the training cohort and 40.5%(203/501)in the validation cohort.Of the 149 EAD patients in the training cohort,77 patients with either elevated alanine aminotransferase(ALT)or aspartate aminotransferase(AST)were classified as having EAD type A,and the rest of the EAD patients were classified as having EAD type B.Recipients with EAD type B had lower graft and patient survival rates than recipients with EAD type A(P=0.043 and 0.044,respectively).We further developed a nomogram to predict EAD(graft weight,cold ischemia time,donor age,model for end-stage liver disease(MELD)score)and another nomogram to predict EAD type B(graft weight,cold ischemia time,MELD score).The nomograms for the prediction of EAD and EAD type B had good discrimination[concordance index(C-index)=0.712(0.666-0.758),0.707(0.641-0.773)]and calibration[Hosmer-Lemeshow(HL)P=0.384,P=0.425]in the validation cohort.An increased operative time(>6 h)was associated with increased EAD and EAD type B incidence in the high-risk group(P=0.005,P=0.020,respectively).Conclusions:EAD type B was associated with decreased graft and patient survival rates.The novel nomograms effectively predicted the incidence of EAD and EAD type B in DCD LT patients. | Kun Wang Di Lu Yuhui Liu Wangyao Li Li Zhuang Zhenyu Ma Qinfen Xie Binhua Pan Yichao Wu Junli Chen Lidan Lin Xiaowen Feng Qiang Wei Xuyong Wei Haiyang Xie Zhengxin Wang Shusen Zheng Xiao Xu | 2021 | Hepatobiliary Surgery and Nutrition2021,10,1: | 4 |

| 15 | Serological Investigation of Laboratory-Confirmed and Suspected Ebola Virus Disease Patients During the Late Phase of the Ebola Outbreak in Sierra Leone显示文摘This study aimed to investigate the serological characteristics of Ebola virus(EBOV) infection during the late phase of the Ebola outbreak in Sierra Leone. In total, 877 blood samples from 694 suspected Ebola virus disease(EVD) cases assessed from March to December 2015, were analyzed via real-time reverse transcription polymerase chain reaction(RT-PCR) for viral RNA and enzyme-linked immunosorbent assay(ELISA) and Luminex to detect antibodies against EBOV. Viral load and EBOV-specific IgM/IgG titers displayed a declining trend during March to December 2015. Viral RNA load decreased rapidly at earlier stages after disease onset, while EBOV-specific IgM and IgG still persisted in 58.1%(18/31) and 93.5%(29/31) of the confirmed EVD patients and in 3.8%(25/663) and 17.8%(118/663) of the RNA-negative suspected patients in the later phase, respectively. | 16 | Protective effect of Tongdu Tiaoshen acupuncture combined with Xiaoxuming decoction(小续命汤)on dopaminergic neurons in Parkinson’s disease model显示文摘OBJECTIVE:To explore the possible mechanism of Tongdu Tiaoshen acupuncture combined with Xiaoxuming decoction(小续命汤,XXMD)in the treatment of Parkinson’s disease(PD).METHODS:C57BL/6 mice were randomly divided into eight groups(n=12),including blank group,model group,medication group,acupuncture group,high-dose XXMD group(XXMD-H),low-dose XXMD group(XXMD-L),acupuncture combined with high-dose XXMD group(A+H),and acupuncture combined with low-dose XXMD group(A+L).After treatment for 6 weeks,dopamine(DA)neurons and the pathological changes of tyrosine hydroxylase(TH)positive cells were observed.The enzyme-linked immunosorbent assay(ELISA)was used to measure the content of DA and the level of interleukin-1β(IL-1β),interleukin-6(IL-6),interleukin-10(IL-10)and tumor necrosis factor alpha(TNF-α).The m RNA level of PINK1 and Parkin and the protein expression of Nix,PINK1 and Parkin in the substantia nigra were also detected.RESULTS:Combination treatment effectively ameliorated the symptoms of PD.Compared with model group,combined treatment significantly up-regulated the protein expression of Nix,Parkin and PINK1 and the m RNA levels of PINK1 and Parkin in the substantia nigra(P<0.0001,P<0.001,P<0.01 or P<0.05).Furthermore,the levels of pro-inflammation cytokines were obviously decreased after combination therapy,while IL-10 content was increased remarkably(P<0.01).CONCLUSION:Compared with each treatment alone,combination therapy improved the pathological damage of DA neurons of PD mice more effectively.The possible mechanism may be attributed to the up-regulated level of mitochondrial autophagy and improved mitochondrial function.These results provide fresh insight into the mechanism of co-treatment with Tongdu Tiaoshen acupuncture and XXMD for PD. | WU Haiyang WANG Ying HAN Wei LI Huihui JI Haisheng LIU Xiuxiu | 2023 | Journal of Traditional Chinese Medicine2023,43,3: | 2 |
| 17 | Analysis of risk factors associated with endoscopic retrograde cholangiopancreatography for patients with liver cirrhosis: a multicenter, retrospective, clinical study显示文摘Background:Endoscopic retrograde cholangiopancreatography(ERCP)is the endoscopic modality of choice for the treatment of biliary and pancreatic diseases.However,patients with cirrhosis,particularly those with decompensated cirrhosis,are believed to be at increased risk for complications associated with ERCP.There is a paucity of literature describing the outcomes of ERCP for patients with cirrhosis.This study aimed to investigate the outcomes of ERCP for cirrhosis patients,especially adverse events,and evaluated its safety and efficacy.Methods:We performed a multicenter,retrospective study of all patients at Beijing Friendship Hospital affiliated to Capital Medical University,Xijing Hospital affiliated to Air Force Military Medical University,Beijing Youan Hospital affiliated to Capital Medical University,and the Fifth Medical Center of the People’s Liberation Army General Hospital from June 2003 to August 2019.The adverse events of inpatient ERCP for patients with(n=182)and without liver cirrhosis(controls;n=385)were compared.Results:A total of 567 patients underwent ERCP between January 2003 and December 2019 were enrolled in this study.Compared to patients without cirrhosis,patients with cirrhosis were at higher risk for postoperative complications(odds ratio[OR],4.172;95%confidence interval[CI],1.232–7.031;P<0.001)such as postoperative pancreatitis(OR,2.026;95%CI,1.002–4.378;P=0.001)and cholangitis(OR,3.903;95%CI,1.001–10.038;P=0.036).The main indications for ERCP for patients with cirrhosis in this study included choledocholithiasis(101 cases;55.5%),benign bile duct strictures(46 cases;25.3%),and malignant bile duct strictures(28 cases;15.4%).Among them,23 patients(12.6%)underwent balloon dilation and 79 patients(43.4%)underwent sphincterotomy.Of the patients with cirrhosis,delayed bleeding occurred in ten patients(5.5%),postoperative pancreatitis occurred in 80 patients(44.0%),and postoperative cholangitis occurred in 25 patients(13.7%).An additional multivariate analysis showed that the total bilirubin(TBIL)level(OR,4.58;95%CI,2.37–6.70)and Child-Pugh score of C(OR,3.11;95%CI,1.04–5.37)were risk factors for postoperative complications in patients with cirrhosis.Conclusions:Compared with the general population of patients undergoing ERCP,patients with cirrhosis were more prone to postoperative pancreatitis and cholangitis.TBIL levels and Child-Pugh scores were risk factors for postoperative complications in patients with cirrhosis. | Jielin Li Jiexuan Hu Peng Li Yongdong Wu Yongjun Wang Ming Ji Haiyang Hua Wenbin Ran Yanglin Pan Shutian Zhang | 2022 | Chinese Medical Journal2022,135,19: | 2 |
| 18 | The development from hyperuricemia to gout:key mechanisms and natural products for treatment显示文摘Gout is a common of inflammatory arthritis and is caused by the deposition of monosodium urate(MSU)crystals as a result of hyperuricemia(HUA).Although HUA is considered to be the main risk factor for gout,only approximately 10%of the individuals with HUA will eventually experience a gout attack.In this review,we first briefly introduce the development of gout and then summarize several possible reasons for its development.Genetic factors play a more prominent role in gout than in other diseases;functional mutations related to urate control and innate immunity components have been found to be associated with gout.Here,we list some of the most prominent genes involved in the pathogenesis of gout.In joints with MSU deposition,mature macrophages may uptake MSU crystals without causing inflammation,and this helps to maintain joints in an asymptomatic state.As an auxiliary inflammation pathway,the ATP-P2X7R-NLRP3 axis may contribute to the amplification of MSU-induced inflammation to affect the development of gout.Finally,this review summarizes the research progress on natural products that can be used in the treatment of HUA and gout. | Lin Liu Dan Wang Mengyang Liu Haiyang Yu Qian Chen Yuzheng Wu Ruixia Bao Yi Zhang Tao Wang | 2022 | Acupuncture and Herbal Medicine2022,2,1: | 2 |
