| 2 | Class A scavenger receptor activation inhibits endoplasmic reticulum stress-induced autophagy in macrophage显示文摘Macrophage death in advanced atherosclerosis promotes plaque necrosis and destabilization.Involvement of autophagy in bulk degradation of cellular components has been recognized recently as an important mechanism for cell survival under endoplasmic reticulum(ER) stress.We previously found that the engagement of class A scavenger receptor(SR-A) triggered JNK-dependent apoptosis in ER-stressed macrophages.However,pro-apoptotic mechanisms mediated by SR-A are not fully understood.Therefore,we sought to see if SR-A mediated apoptosis was associated with autophagy in macrophages.Here,we showed that fucoidan inhibited microtubule-associated protein light chain 3-phospholipid conjugates(LC3-Ⅱ) formation as well as the number of autophagosomes under ER stress.The inhibition of LC3-Ⅱ formation was paralleled by the activation of the mTOR pathway,and the inhibition of mTOR allowed LC3-Ⅱ induction in macrophages treated with thapsigargin plus fucoidan.Furthermore,apoptosis induced by fucoidan was prevented under ER stress by the mTOR inhibitor.We propose that fucoidan,a SR-A agonist,may contribute to macrophage apoptosis during ER stress by inhibiting autophagy. | Hanpeng Huang Xiaoyu Li Yan Zhuang Nan Li Xudong Zhu Jin Hu Jingjing Ben Qing Yang Hui Bai Qi Chen | 2014 | The Journal of Biomedical Research2014,28,3: | 9 |
| 3 | Experimental research into the effect of gas pressure,particle size and nozzle area on initial gas-release energy during gas desorption显示文摘Coal and gas outburst is a violent disaster driven by released energy from gas desorption.The initial expansion energy of released gas(IEERG)is a new method to predict coal and gas outburst.In this paper,an instrument for IEERG measurement was developed.Compared with previous setups,the new one which is equipped with three convergent nozzles and quick-release mechanism gets improved in data acquisition and gas sealing and releasing performance.To comprehensively know the effect of gas pressure,particle size,and nozzle area on IEERG,a series of experiments were carried out with this new setup.The variable control test results indicated that the gas pressure-IEERG curves remain the linear trend and the particle size-IEERG curves maintain the negative exponential trend for nozzle areas at 1.13,2.26,and3.39 mm2,respectively.The increase in nozzle area leads to deceases in value of IEERG and absolute value of slope of fitting curves in each test.In addition,the orthogonal experiment showed that the influence of gas pressure,nozzle area,and particle size on IEERG decreases in turn.Only gas pressure had a marked impact on IEERG.This work offers great importance in improving the accuracy of prediction of coal and gas outburst. | Weitao Hou Hanpeng Wang Liang Yuan Wei Wang Yang Xue Zhengwei Ma | 2021 | International Journal of Mining Science and Technology2021,31,2: | 4 |

| 6 | Experimental and theoretical study on the dynamic effective stress of loaded gassy coal during gas release显示文摘In the process of mining coalbed methane(CBM),an unsteady state often arises due to the rapid extraction,release and pressure relief of CBM.In this case,the effective stress of coal changes dynamically,affecting the stability of the gassy coal seam.In this paper,gas release tests of gassy coal under conventional triaxial compression were performed,and the dynamic effective stress(DES)during gas release was obtained indirectly based on a constitutive equation and deformation of coal.The results show that the maximum increases in DES caused by the release of free gas and adsorbed gas under the stress of 1.1 MPa were 0.811 and 5.418 MPa,respectively,which seriously affected the stress state of the coal.During the gas release,the free gas pressure and the adsorbed gas volume were the parameters that directly affected the DES and showed a positive linear relationship with the DES with an intercept of zero.The DES of the coal sample increased exponentially with time,which was determined by the contents of free and adsorbed gas.Based on the experimental results and theoretical analysis,an effective stress model was obtained for loaded gassy coal during gas release.The results of verification indicated accuracy greater than 99%. | Bing Zhang Hanpeng Wang Peng Wang Guofeng Yu Shitan Gu | 2023 | International Journal of Mining Science and Technology2023,33,3: | 2 |
| 7 | Correlation of obstructive sleep apnea hypopnea syndrome with metabolic syndrome in snorers显示文摘Though obstructive sleep apnea hypopnea syndrome(OSAHS) and metabolic syndrome(MS) are correlated;the contributing factors for the occurrence of MS in Chinese snorers remain largely undefined.We aimed to investigate the associated pathogenesis of coexistence of OSAHS and MS in Chinese snorers.A total of 144 Chinese habitual snorers were divided into 3 groups,the control group(simple snorers)(n = 36),the mild OSAHS group(n= 52)and the moderate-to-severe OSAHS group(n = 56).The incidence of MS in the moderate-to-severe OSAHS group(26.8%) was significantly higher than that in the control group(8.3%),the mild OSAHS group(11.1%) and all the OSAHS patients(19.45%)(all P < 0.05).Homeostatic model assessment(HOMA) index and proinsulin(PI) were negatively correlated with nocturnal meanSpO_2 and miniSpO_2.Meanwhile,nocturnal SpO_2 were negatively correlated with body mass index,waist and neck circumferences and diastolic blood pressure,but positively correlated with total cholesterol and high-density lipoprotein cholesterol.The study indicated that in Chinese snorers,moderate-to-severe OSAHS was closely associated with MS via nocturnal hypoxemia. | Qun Zhang Xiaofeng Zhang Mengmeng Zhao Hanpeng Huang Ning Ding Xilong Zhang Hong Wang | 2014 | The Journal of Biomedical Research2014,28,3: | 2 |

The detection and quantification of RNA modifications generally are challenging tasks since most of the modifications exist in low abundance in vivo. Here we developed an on-line trapping/capillary hydrophilic-interaction liquid chromatography/electrospray ionization-mass spectrometry(on-line trapping/cHILIC/MS) method for sensitive and simultaneous quantification of RNA modifications of N^6-methyladenosine(m^6A) and 5-methylcytosine(5-mC) from human blood. The hydrophilic organic-silica hybrid monolith was prepared using sol-gel combined with 'thiol-ene' click reaction for the separation of nucleosides. A poly(MAA-co-EGDMA) monolithic capillary was used as the on-line trapping column.With the developed on-line trapping/cHILIC/MS analytical platform, the detection limits of m^6A and 5-mC can reach to 0.06 fmol and 0.10 fmol. We then investigated the contents of m^6A and 5-mC in human blood RNA from healthy persons at the age of 6-14 and 60-68 years. The realized structure and fabrication are the key elements of the micro-hotplate. Directly fabrication of micro-pellsitor catalyst is very difficult because of the small dimensions of the active area. In this paper, a novel micro-pellistor was designed by combining micro fabrication technique and nano technology. The supporting beams and micro-hotplate of the micro-pellistor were made of nonoporous alumina film. The active area of the designed and fabricated micro-pellistors ranges from 200×200 μm2 to 450×300 μm2. The micro- pellistor was heated by platinum thin film heater and the Pd catalyst was deposited by dipping the PdCl2 solution on the detecting element. The lowest power consumption is 50 mW at 500 °C and the maximum temperature can reach 900 °C before rupture. The response of the devices to methane is also tested. | 14 | Two-stage matrix-assisted glare suppression at a large scale显示文摘Scattering-induced glares hinder the detection of weak objects in various scenarios.Recent advances in wavefront shaping show one can not only enhance intensities through constructive interference but also suppress glares within a targeted region via destructive interference.However,due to the lack of a physical model and mathematical guidance,existing approaches have generally adopted a feedback-based scheme,which requires timeconsuming hardware iteration.Moreover,glare suppression with up to tens of speckles was demonstrated by controlling thousands of independent elements.Here,we reported the development of a method named twostage matrix-assisted glare suppression(TAGS),which is capable of suppressing glares at a large scale without triggering time-consuming hardware iteration.By using the TAGS,we experimentally darkened an area containing 100 speckles by controlling only 100 independent elements,achieving an average intensity of only 0.11 of the original value.It is also noticeable that the TAGS is computationally efficient,which only takes 0.35 s to retrieve the matrix and 0.11 s to synthesize the wavefront.With the same number of independent controls,further demonstrations on suppressing larger scales up to 256 speckles were also reported.We envision that the superior performance of the TAGS at a large scale can be beneficial to a variety of demanding imaging tasks under a scattering environment. | DAIXUAN WU JIAWEI LUO ZHIBING LU HANPENG LIANG YUECHENG SHEN ZHAOHUI LI | 2022 | Photonics Research2022,10,12: | 0 |
| 15 | Evaluation of heat stress effects on cellular and transcriptional adaptation of bovine granulosa cells显示文摘Background:Heat stress is known to affect follicular dynamics,oocyte maturation,and fertilization by impairing steroidogenic ability and viability of bovine granulosa cell(bGCs).The present study explored the physiological and molecular response of bGCs to different heat stress intensities in-vitro.We exposed the primary bGCs to heat stress(HS)at 39℃,40℃ and 41℃ along with control samples(38℃)for 2 h.To evaluate the impact of heat stress on bGCs,several in vitro cellular parameters including cell apoptosis,intracellular reactive oxygen species(ROS)accumulation and HSP70 kinetics were assessed by flow cytometry,florescence microscopy and western blot,respectively.Furthermore,the ELISA was performed to confirm the 17β-estradiol(E2)and progesterone(P4)levels.In addition,the RNA sequencing(RNA-Seq)method was used to get the molecular based response of bGCs to different heat treatments.Results:Our findings revealed that the HS significantly decreased the cell viability,E2 and P4 levels in bGCs,whereas,increased the cellular apoptosis and ROS.Moreover,the RNA-Seq experiments showed that all the treatments(39℃,40℃ and 41℃)significantly regulated many differentially expressed genes(DEGs)i.e.BCL2L1,STAR,CYP11A1,CASP3,SOD2,HSPA13,and MAPK8IP1 and pathways associated with heat stress,apoptosis,steroidogenesis,and oxidative stress.Conclusively,our data demonstrated that the impact of 40°C treatment was comparatively detrimental for cell viability,apoptosis and ROS accumulation.Notably,a similar trend of gene expression was reported by RT-qPCR for RNA-seq data.Conclusions:Our study presented a worthy strategy for the first time to characterize the cellular and transcriptomic adaptation of bGCs to heat stress(39,40 and 41°C)in-vitro.The results infer that these genes and pathways reported in present study could be useful candidates/indicators for heat stress research in dairy cattle.Moreover,the established model of bGCs to heat stress in the current study provides an appropriate platform to understand the mechanism of how heat-stressed bGCs can affect the quality of oocytes and developing embryo. | Adnan Khan Jinhuan Dou Yachun Wang Xiaolong Jiang Muhammad Zahoor Khan Hanpeng Luo Tahir Usman Huabin Zhu | 2020 | Journal of Animal Science and Biotechnology2020,11,2: | 0 |

| 17 | Clinical significance of serum IgM and IgG levels in COVID-19 patients in Hubei Province,China显示文摘Background:There have been many studies about coronavirus disease 2019(COVID-19),but the clinical signifi-cance of quantitative serum severe acute respiratory syndrome-coronavirus-2(SARS-CoV-2)-specific IgM and IgG levels of COVID-19 patients have not been exhaustively analyzed.We aimed to investigate the time profiles of these IgM/IgG levels in COVID-19 patients and their correlations with clinical features.Methods:A multicenter clinical study was conducted from February 20 to March 52020.It involved 179 COVID-19 patients(108 males and 71 females)from five hospitals in Huangshi in Hubei Province,China.To detect SARS-CoV-2-specific IgM/IgG,quantitative antibody assays(two-step indirect immunoassays with direct chemilu-minescence technology)based on the nucleocapsid protein(NP)and spike protein 1(S1)were used.For normally distributed data,means were compared using the t-test,��2-test,or exact probability method.For categorical data,medians were compared using Mann-Whitney U test.Results:The median age was 57(44-69)years(58[38-69]for males and 57[49-68]for females).The median duration of positive nucleic acid test was 22.32(17.34-27.43)days.The mortality rate was relatively low(3/179,1.68%).Serum SARS-CoV-2-specific IgG was detected around week 1 after illness onset,gradually increased until peaking in weeks 4 and 5,and then declined.Serum IgM peaked in weeks 2 and 3,then gradually declined and returned to its normal range by week 7 in all patients.Notably,children had milder respiratory symptoms with lower SARS-CoV-2-specific IgM/IgG levels.The duration of positive nucleic acid test in the chronic obstructive pulmonary disease(COPD)group was 30.36(18.99-34.76)days,which was significantly longer than that in the non-COPD group(21.52[16.75-26.51]days;P=0.025).The peak serum SARS-CoV-2-specific IgG was sig-nificantly positively correlated with the duration of positive nucleic acid test.The incidence rate of severe and critical cases in the IgM hi group(using the median IgM level of 29.95 AU/mL as the cutofffor grouping)was about 38.0%(19/50),which was twice as much as that in the IgM lo group(18.4%;9/49).The patients with positive chest imaging and lymphocytopenia(<1×10^(9)/L)had a higher SARS-CoV-2-specific IgM level.Conclusions: Quantitative SARS-CoV-2-specific IgM and IgG levels are helpful for the diagnosis, severity classifi- cation, and management of COVID-19 patients, and they should be monitored in each stage of this disease. | Zhenjiang Bai Qing Li Qinghui Chen Changming Niu Yu Wei Hanpeng Huang Wei Zhao Nian Chen Xin Yao Qiang Zhang Chuanyong Mu Jian Feng Chuanlong Zhu Zhuo Li Ming Ding Binhui Feng Chaochao Jin Xiang Lu Yi Yang Shuiyan Wu Xiaochen Shu Haibo Qiu YingZi Huang | 2022 | Journal of Intensive Medicine2022,2,1: | 0 |
| 18 | Weighted single-step GWAS and RNA sequencing reveals key candidate genes associated with physiological indicators of heat stress in Holstein cattle显示文摘Background: The study of molecular processes regulating heat stress response in dairy cattle is paramount for developing mitigation strategies to improve heat tolerance and animal welfare. Therefore, we aimed to identify quantitative trait loci(QTL) regions associated with three physiological indicators of heat stress response in Holstein cattle, including rectal temperature(RT), respiration rate score(RS), and drooling score(DS). We estimated genetic parameters for all three traits. Subsequently, a weighted single-step genome-wide association study(WssGWAS) was performed based on 3200 genotypes, 151,486 phenotypic records, and 38,101 animals in the pedigree file. The candidate genes located within the identified QTL regions were further investigated through RNA sequencing(RNA-seq) analyses of blood samples for four cows collected in April(non-heat stress group) and four cows collected in July(heat stress group).Results: The heritability estimates for RT, RS, and DS were 0.06, 0.04, and 0.03, respectively. Fourteen, 19, and 20 genomic regions explained 2.94%, 3.74%, and 4.01% of the total additive genetic variance of RT, RS, and DS, respectively. Most of these genomic regions are located in the Bos taurus autosome(BTA) BTA3, BTA6, BTA8, BTA12, BTA14, BTA21, and BTA24. No genomic regions overlapped between the three indicators of heat stress, indicating the polygenic nature of heat tolerance and the complementary mechanisms involved in heat stress response. For the RNA-seq analyses, 2627 genes were significantly upregulated and 369 downregulated in the heat stress group in comparison to the control group. When integrating the WssGWAS, RNA-seq results, and existing literature, the key candidate genes associated with physiological indicators of heat stress in Holstein cattle are: PMAIP1, SBK1, TMEM33, GATB, CHORDC1, RTN4IP1, and BTBD7.Conclusions: Physiological indicators of heat stress are heritable and can be improved through direct selection. Fifty-three QTL regions associated with heat stress indicators confirm the polygenic nature and complex genetic determinism of heat tolerance in dairy cattle. | 20 | Effects of silver-doping on properties of Cu(In,Ga)Se_(2) films prepared by Cu In Ga precursors显示文摘The AgCuInGa alloy precursors with different Ag concentrations are fabricated by sputtering an Ag target and a CuInGa target.The precursors are selenized in the H_(2)Se-containing atmosphere to prepare(Ag,Cu)(In,Ga)Se_(2)(ACIGS)absorbers.The beneficial effects of Ag doping are demonstrated and their mechanism is explained.It is found that Ag doping significantly improves the films crystallinity.This is believed to be due to the lower melting point of chalcopyrite phase obtained by the Ag doping.This leads to a higher migration ability of the atoms that in turn promotes grain boundary migration and improves the film crystallinity.The Ga enrichment at the interface between the absorber and the back electrode is also alleviated during the selenization annealing.It is found that Ag doping within a specific range can passivate the band tail and improve the quality of the films.Therefore,carrier recombination is reduced and carrier transport is improved.The negative effects of excessive Ag are also demonstrated and their origin is revealed.Because the atomic size of Ag is different from that of Cu,for the Ag/(Ag+Cu)ratio(AAC)≥0.030,lattice distortion is aggravated,and significant micro-strain appears.The atomic radius of Ag is close to those of In and Ga,so that the continued increase in AAC will give rise to the Ag;or Ag;defects.Both the structural and compositional defects degrade the quality of the absorbers and the device performance.An excellent absorber can be obtained at AAC of 0.015. | Chen Wang Daming Zhuang Ming Zhao Yuxian Li Liangzheng Dong Hanpeng Wang Jinquan Wei Qianming Gong | 2022 | Journal of Energy Chemistry2022,31,3: | 0 |
