1A Kelch Motif-Containing Serine/Threonine Protein Phosphatase Determines the Large Grain QTL Trait in Rice显示文摘A thorough understanding of the genetic basis of rice grain traits is critical for the improvement of rice (Oryza sativa L.) varieties.In this study,we generated an F 2 population by crossing the large-grain japonica cultivar CW23 with Peiai 64 (PA64),an elite indica small-grain cultivar.Using QTL analysis,17 QTLs for five grain traits were detected on four different chromosomes.Eight of the QTLs were newly-identified in this study.In particular,qGL3-1,a newly-identified grain length QTL with the highest LOD value and largest phenotypic variation,was fine-mapped to the 17 kb region of chromosome 3.A serine/threonine protein phosphatase gene encoding a repeat domain containing two Kelch motifs was identified as the unique candidate gene corresponding to this QTL.A comparison of PA64 and CW23 sequences revealed a single nucleotide substitution (C→A) at position 1092 in exon 10,resulting in replacement of Asp (D) in PA64 with Glu (E) in CW23 for the 364th amino acid.This variation is located at the D position of the conserved sequence motif AVLDT of the Kelch repeat.Genetic analysis of a near-isogenic line (NIL) for qGL3-1 revealed that the allele qGL3-1 from CW23 has an additive or partly dominant effect,and is suitable for use in molecular marker-assisted selection.Zejun Hu Haohua He Shiyong Zhang Fan Sun Xiaoyun Xin Wenxiang Wang Xi Qian Jingshui Yang Xiaojin Luo 2012Journal of Integrative Plant Biology2012,54,12:41

4Adhesive anastomosis for organ transplantation显示文摘The recent development of tough tissue adhesives has stimulated intense interests among material scientists and medical doctors.However,these adhesives have seldom been tested in clinically demanding surgeries.Here we demonstrate adhesive anastomosis in organ transplantation.Anastomosis is commonly conducted by dense sutures and takes a long time,during which all the vessels are occluded.Prolonged occlusion may damage organs and even cause death.We formulate a tough,biocompatible,bioabsorbable adhesive that can sustain tissue tension and pressurized flow.We expose the endothelial surface of vessels onto a gasket,press two endothelial surfaces to the adhesive using a pair of magnetic rings,and reopen the bloodstream immediately.The time for adhesive anastomosis is shortened compared to the time for sutured anastomosis.We have achieved adhesive anastomosis of a great vein in transplanting the liver of a pig.After the surgery,the adhesive is absorbed,the vein heals,and the pig lives for over one month.Kang Liu Hang Yang Gaobo Huang Aihua Shi Qiang Lu Shanpei Wang Wei Qiao Haohua Wang Mengyun Ke Hongfan Ding Tao Li Yanchao Zhang Jiawei Yu Bingyi Ren Rongfeng Wang Kailing Wang Hui Feng Zhigang Suo Jingda Tang Yi Lv 2022Bioactive Materials2022,7,7:3
5miR-495 andmiR-5688 are down-regulated in non-small cell lung cancer under hypoxia to maintain interleukin-11 expression显示文摘Background:Hypoxia is a hallmark of cancer and is associated with poor prognosis.However,the molecular mechanism by which hypoxia promotes tumor progression remains unclear.MicroRNAs dysregulation has been shown to play a critical role in the tumor and tumor microenvironment.Here,we investigated the roles ofmiR-495 and miR-5688 in human non-small cell lung cancer(NSCLC)and their underlying mechanism.Methods:The expression levels of miR-495 and miR-5688 in human NSCLC tissue specimens were measured by quantitative real-time polymerase chain reaction(qRT-PCR).Deferoxamine(DFO)was used to determine whether the regulation of miR-495 and miR-5688 under hypoxia was dependent on hypoxia-inducible factor 1-alpha(HIF-1α).Furthermore,the functions of miR-495 and miR-5688 in tumor progression were evaluated using colony formation,3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2Htetrazolium(MTS),wound healing,transwell assays,and xenograft model.Two algorithms,PicTAR and Targetscan,were used to predict the target gene of these two miRNAs,and dual-luciferase reporter assay was conducted to confirm the target.The unpaired two-tailed t test,Pearson correlation analysis,and Fisher’s exact probability test were performed for statistical analyses.Results:Two miRNAs,miR-495 and miR-5688,were found to participate in NSCLC progression under hypoxia.They were down-regulated in NSCLC tissues compared with normal tissues.We determined that hypoxia led to the down-regulation of miR-495 and miR-5688 in NSCLC cells,which was independent of HIF-1αand cellular metabolic energy.In addition,miR-495 and miR-5688 suppressed cell proliferation,migration,and invasion in vitro.The NSCLC xenograft model showed that miR-495 and miR-5688 inhibited tumor formation in vivo.Interestingly,we found that miR-495 and miR-5688 had the same target,interleukin-11(IL-11).Recombinant human IL-11 counteracted the effects of miR-495 and miR-5688 on NSCLC cells,suggesting that miR-495 and miR-5688 executed their tumor suppressive role by repressing IL-11 expression.Conclusion:We found that hypoxia down-regulated the expression levels of miR-495 and miR-5688 in NSCLCto enhance IL-11 expression and tumor progression,indicating that the miR-495/miR-5688/IL-11 axismay serve as a therapeutic target and potential biomarker for NSCLC.Meng Zhao Jiao Chang Ran Liu Yahui Liu Jin Qi Yanhui Wang Xinwei Zhang Lu Qiao Yu Jin Haohua An Li Ren 2020Cancer Communications2020,40,9:2

9Natural variation of an autophagy-family gene among rice subspecies affects grain size and weight显示文摘Elucidating the genetic basis of natural variation in grain size and weight among rice varieties can help breeders develop high-yielding varieties.We identified a novel gene,GW3a(Grain Weight 3a)(LOC_Os03g27350),that affects rice grain size and weight.gw3a mutants showed higher total starch content and dry matter accumulation than the wild type(WT),Nipponbare,suggesting that GW3a negatively regulates grain size and weight.Moreover,our study found that GW3a interacted with OsATG8 by cleaving it,suggesting that GW3a may be involved in the assembly of autophagosomes and starch degradation in plants.The haplotype analysis of GW3a showed functional differences between indica and japonica rice.Taken together,we conclude that GW3a is expressed in the autophagosome pathway regulating starch metabolism in rice,affecting yield-related traits,such as grain size,grain weight and thousand grain weight(TGW).Our findings also shed new light on autophagy-mediated yield trait regulation,proposing a possible strategy for the genetic improvement of high-yield germplasm in rice.Shiying Huang Peng Wang Caijing Li Qingwen Zhou Tao Huang Yicong Cai Qin Cheng Hui Wang Qi Zhong Zhihao Chen Liping Chen Haohua He Jianmin Bian 2024The Crop Journal2024,12,1:0
10An Optimization System for Intent Recognition Based on an Improved KNN Algorithm with Minimal Feature Set for Powered Knee Prosthesis显示文摘In this article,a new optimization system that uses few features to recognize locomotion with high classification accuracy is proposed.The optimization system consists of three parts.First,the features of the mixed mechanical signal data are extracted from each analysis window of 200 ms after each foot contact event.Then,the Binary version of the hybrid Gray Wolf Optimization and Particle Swarm Optimization(BGWOPSO)algorithm is used to select features.And,the selected features are optimized and assigned different weights by the Biogeography-Based Optimization(BBO)algorithm.Finally,an improved K-Nearest Neighbor(KNN)classifier is employed for intention recognition.This classifier has the advantages of high accuracy,few parameters as well as low memory burden.Based on data from eight patients with transfemoral amputations,the optimization system is evaluated.The numerical results indicate that the proposed model can recognize nine daily locomotion modes(i.e.,low-,mid-,and fast-speed level-ground walking,ramp ascent/decent,stair ascent/descent,and sit/stand)by only seven features,with an accuracy of 96.66%±0.68%.As for real-time prediction on a powered knee prosthesis,the shortest prediction time is only 9.8 ms.These promising results reveal the potential of intention recognition based on the proposed system for high-level control of the prosthetic knee.Yao Zhang Xu Wang Haohua Xiu Lei Ren Yang Han Yongxin Ma Wei Chen Guowu Wei Luquan Ren 2023Journal of Bionic Engineering2023,20,6:0
11Bioactive Components of Cordyceps taii and Their Immune Functions in Mice显示文摘The polysaccharide,adenosine and mannitol components of Cordyceps taii collected from the wild were determined,and the effects of aqueous C.taii extracts on immune functions in mice were investigated using carbon clearance and delayed-type hypersensitivity testing(DTH),and by determining thymic and splenic indices.Polysaccharide,adenosine and mannitol levels in C.taii were 36.20 mg/g,0.27 mg/g and 80.20 mg/g,respectively.Aqueous C.taii extracts partially alleviated the suppressive effects on the thymic and splenic indices caused by cyclophosphamide,and increased nonspecific immunity and cellular immune functions.Toxicity tests revealed that the LD50 for aqueous C.taii extract on oral administration was >69.42 g/kg.ZHONG Han GAN Lixia ZHANG Weimin PAN Qingling LI Haohua TAO Meihua WANG Lei TAN Yuzhi LIU Yanping 2008食用菌学报2008,15,3:0
12Xenogeneic hepatic progenitor cell transplantation ameliorate CCl_4 /partial hepatectomy-induced rat acute liver failure显示文摘Objective: Hepatic progenitor cell transplantation has shed light on the treatment of liver failure. The present study was designed to evaluate whether xenogeneic liver epithelial progenitor cells(LEPCs) transplantation could promote liver recovery in a rat model of acute liver failure. The engraftment and hepatocytic differentiation of transplanted hepatic progenitor cells in the rat spleen was also investigated. Methods: LEPCs were propagated in vitro for long and transduced with lentiviral vector carrying mCherry gene. Intraperitoneal injection of CCl4 followed by 2/3 partial hepatectomy three days later were used to establish rat models of acute liver failure. Rats were intrasplenically injected with mCherry modified LEPCs(n=20, 1×107cells/0.5 mL) or the same volume of medium(n=20). Serum liver enzymes(ALT, AST) and liver histology were evaluated for 21 days after transplantation. The engraftment of transplanted LEPCs in the spleens was tested by polymerase chain reaction(PCR) amplification targeting mCherry gene. The differentiation into hepatocytic lineage of transplanted LEPCs was investigated using immunohistochemistry staining against Alb. Results: LEPCs were effectively transduced with lentiviral vector showing a transduction efficiency of 90%. Compared with control, cell-injected group displayed significantly lower levels of ALT and AST(P<0.05) and better histological features including less swelling change and hepatocyte death. PCR amplification of mCherry sequences confirmed the engraftment of LEPCs in the spleens. Alb-positive cells first appeared 5 days after cell transplantation and the number of Alb-positive cells increased substantially(P<0.05), which revealed the hepatocytic differentiation process. Conclusion: Xenogeneic hepatic progenitor cells can engraft and differentiate into hepatocytes in the splenic parenchyma. 13Mechanism of hepatocellular damage in rat caused by low serum selenium显示文摘The aim of this paper is to investigate the mechanism of hepatocellular damage in rats caused by low serum selenium.Thirty six rats were randomly divided into 2 groups:group A(fed with low-selenium diet from the Keshan Disease area with the content of selenium being 0.017 mg/kg);group B[fed with sel-enium-supplemented diet and 0.3 mg/L selenium(Na_(2)SeO_(3))was added to the drinking water].Both were respectively fed for 12 weeks.At the end of the 12th week,the levels of serum selenium,glutathione peroxidase(GPX)and malondialdehyde(MDA)in hepatic tissue were measured;the hepatocellular ultrastructure and apoptosis were observed as well.The levels of serum sel-enium and GPX in group A were markedly lower than those in group B.MDA level in group A was significantly higher than that in group B.Under the electron micro-scope(EM),the mitochondria were remarkably changed in group A.The rate of liver cell apoptosis appeared much higher in group A as well.It indicated that the damage caused by selenium deficiency was through the process of oxidation.Selenium deficiency led to apoptosis of hepa-tocytes where oxidative damage to mitochondria might be the cause.Yi LU Bo QU Chang LIU Liang YU Xuemin LIU Haohua WANG An JIANG Xiaogang ZHANG 2008Frontiers of Medicine2008,2,3:0
14USH2A mutation and specific driver mutation subtypes are associated with clinical efficacy of immune checkpoint inhibitors in lung cancer显示文摘This study aimed to identify subtypes of genomic variants associated with the efficacy of immune checkpoint inhibitors(ICIs)by conducting systematic literature search in electronic databases up to May 31,2021.The main outcomes including overall survival(OS),progression-free survival(PFS),objective response rate(ORR),and durable clinical benefit(DCB)were correlated with tumor genomic features.A total of 1546 lung cancer patients with available genomic variation data were included from 14 studies.The Kirsten rat sarcoma viral oncogene homolog G12C(KRAS^(G12C))mutation combined with tumor protein P53(TP53)mutation revealed the promising efficacy of ICI therapy in these patients.Furthermore,patients with epidermal growth factor receptor(EGFR)classical activating mutations(including EGFRL858Rand EGFRΔ19)exhibited worse outcomes to ICIs in OS(adjusted hazard ratio(HR),1.40;95%confidence interval(CI),1.01-1.95;P=0.0411)and PFS(adjusted HR,1.98;95%CI,1.49-2.63;P<0.0001),while classical activating mutations with EGFR^(T790)Mshowed no difference compared to classical activating mutations without EGFR^(T790)Min OS(adjusted HR,0.96;95%CI,0.48-1.94;P=0.9157)or PFS(adjusted HR,0.72;95%CI,0.39-1.35;P=0.3050).Of note,for patients harboring the Usher syndrome type-2A(USH2A)missense mutation,correspondingly better outcomes were observed in OS(adjusted HR,0.52;95%CI,0.32-0.82;P=0.0077),PFS(adjusted HR,0.51;95%CI,0.38-0.69;P<0.0001),DCB(adjusted odds ratio(OR),4.74;95%CI,2.75-8.17;P<0.0001),and ORR(adjusted OR,3.45;95%CI,1.88-6.33;P<0.0001).Our findings indicated that,USH2A missense mutations and the KRAS^(G12C)mutation combined with TP53 mutation were associated with better efficacy and survival outcomes,but EGFR classical mutations irrespective of combination with EGFR^(T790)Mshowed the opposite role in the ICI therapy among lung cancer patients.Our findings might guide the selection of precise targets for effective immunotherapy in the clinic.DEXIN YANG YUQIN FENG HAOHUA LU KELIE CHEN JINMING XU PEIWEI LI TIANRU WANG DAJING XIA YIHUA WU 2023Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2023,24,2:0
