2Discriminated sgRNAs-Based SurroGate System Greatly Enhances the Screening Efficiency of Plant Base-Edited Cells显示文摘The development of CRISPR/Cas9-mediated base editing has made genomic modification more efficient. However, selection of genetically modified cells from millions of treated cells, especially plant cells, is still challenging. In this study, an efficient surrogate reporter system based on a defective hygromycin resistance gene was established in rice to enrich base-edited cells. After step-by-step optimization, the Discriminated sgRNAs-based SurroGate system (DisSUGs) was established by artificially differentiating the editing abilities of a wild-type single guide RNA (sgRNA) targeting the surrogate reporter gene and an enhanced sgRNA targeting endogenous sites. The DisSUGs enhanced the efficiency of screening base-edited cells by 3- to 5-fold for a PmCDA1-based cytosine-to-tyrosine base editor (PCBE), and 2.5- to 6.5-fold for an adenine base editor (ABE) at endogenous targets. These targets showed editing efficiencies of <25% in the conventional systems. The DisSUGs greatly enhanced the frequency of homozygous substitutions and expanded the activity window slightly for both a PCBE and an ABE. Analyses of the total number of single-nucleotide variants from whole-genome sequencing revealed that, compared with the no-enrichment PCBE strategy, the DisSUGs did not alter the frequency of genome-wide sgRNA-independent off-target mutations, but slightly increased the frequency of target-dependent off-target mutations. 3Genome-wide prediction for hybrids between parents with distinguished difference on exotic introgressions in Brassica napus显示文摘Extensive exotic introgression could significantly enlarge the genetic distance of hybrid parental populations to promote strong heterosis.The goal of this study was to investigate whether genome-wide prediction can support pre-breeding in populations with exotic introgressions.We evaluated seed yield,seed yield related traits and seed quality traits of 363 hybrids of Brassica napus (AACC) derived from two parental populations divergent on massive exotic introgression of related species in three environments.The hybrids presented strong heterosis on seed yield,which was much higher than other investigated traits.Five genomic best linear unbiased prediction models considering the exotic introgression and different marker effects (additive,dominance,and epistatic effects) were constructed to test the prediction ability for different traits of the hybrids.The analysis showed that the trait complexity,exotic introgression,genetic relationship between the training set and testing set,training set size,and environments affected the prediction ability.The models with best prediction ability for different traits varied.However,relatively high prediction ability (e.g.,0.728 for seed yield) was also observed when the simplest models were used,excluding the effects of the special exotic introgression and epistasis effect by5-fold cross validation,which would simplify the prediction for the trait with complex architecture for hybrids with exotic introgression.The results provide novel insights and strategies for genome-wide prediction of hybrids between genetically distinct parent groups with exotic introgressions.Dandan Hu Yusheng Zhao Jinxiong Shen Xiangxiang He Yikai Zhang Yong Jiang Rod Snowdon Jinling Meng Jochen C.Reif Jun Zou 2021The Crop Journal2021,9,5:2
4National Cancer Data Linkage Platform of China:Design,Methods,and Application显示文摘Background:The National Cancer Center(NCC)and China CDC cooperatively designed a National Cancer Data Linkage(NCDL)Platform to fulfill the task of sharing cancer outcome data through an automatic web-based system.Methods:NCC and China CDC established a web-based NCDL Platform to link death information from China CDC with the cancer database from NCC.Overall,76,708 cancer patients’data were analyzed to assess the feasibility and match rate of the NCDL Platform for 7 major cancers.Results:The function of the platform includes a data application and approval system,data linkage module,and results visualization system.Through the platform,38.9%cases were identified as deaths cases from the NCDL Platform in the first 3 years after cancer diagnosis.The linkage rate was highest in liver cancer and lowest in breast cancer.Conclusions:The NCDL Platform provides a powerful and efficient way to link national vital statistics with national cancer programs’data.Expanding cancer outcome data linkage may not only improve data collection efficiency,but also improve data use.Hongmei Zeng Yunning Liu Lijun Wang Peng Yin Baohua Wang Ruiying Fu Xianhui Ran Rongshou Zheng Siwei Zhang Jiangmei Liu Jinling You Kexin Sun Shaoming Wang Li Li Ru Chen Wenqiang Wei Maigeng Zhou Jing Wu Jie He 2022China CDC weekly2022,4,13:1

6Accumulation, transportation, and distribution of tetracycline and cadmium in rice显示文摘Co-exposure to heavy metal and antibiotic pollution might result in complexation and synergistic interactions,affecting rice growth and further exacerbating pollutant enrichment.Therefore,our study sought to clarify the influence of different Tetracycline (TC) and Cadmium(Cd) concentration ratios (both alone and combined) on rice growth,pollutant accumulation,and transportation during the tillering stage in hydroponic system.Surprisingly,our findings indicated that the interaction between TC and Cd could alleviate the toxic effects of TC/Cd on aerial rice structures and decrease pollutant burdens during root elongation.In contrast,TC and Cd synergistically promoted the accumulation of TC/Cd in rice roots.However,their interaction increased the accumulation of TC in roots while decreasing the accumulation of Cd when the toxicant doses increased.The strong affinity of rice to Cd promoted its upward transport from the roots,whereas the toxic effects of TC reduced TC transport.Therefore,the combined toxicity of the two pollutants inhibited their upward transport.Additionally,a low concentration of TC promoted the accumulation of Cd in rice mainly in the root tip.Furthermore,a certain dose of TC promoted the upward migration of Cd from the root tip.Laser ablation-inductively coupled plasma mass spectrometry demonstrated that Cd mainly accumulated in the epidermis and stele of the root,whereas Fe mainly accumulated in the epidermis,which inhibited the absorption and accumulation of Cd by the rice roots through the generation of a Fe plaque.Our findings thus provide insights into the effects of TC and Cd co-exposure on rice growth.Chunfeng He Jini Zhou Chen Yang Zhiyi Song Junheng He Ziqing Huang Yurong Deng Jinling Wang Yu Xiong Zhi Dang 2023Journal of Environmental Sciences2023,,4:1

11Deformation and failure behavior of heterogeneous Mg/SiC nanocomposite under compression显示文摘The heterogeneous magnesium(Mg) matrix nanocomposite with dispersed soft phase exhibits high strength and toughness. Herein, the deformation behavior and failure process were investigated to reveal the unique mechanical behavior of the heterogeneous microstructure under compression. The extensive plastic deformation is accompanied by the flattening and tilting of the soft phase, inhibiting strain localization and leading to strain hardening. Moreover, a stable crack multiplication process is activated, which endows high damage tolerance to the heterogeneous Mg matrix nanocomposites. The final failure of the composite is caused by crack coalescence in the shear plane along a tortuous path. The presence of dispersed soft phases within the hard matrix induces a noticeable change in mechanical response. Especially,the malleability of the heterogeneous Mg matrix nanocomposite is two and ten times higher than that of pure Mg and the homogeneous Mg matrix nanocomposite, respectively. 16Gate voltage control of helicity-dependent photocurrent and polarization detection in(Bi_(1-x)Sb_(x))_(2)Te_(3)topological insulator thin films显示文摘Optical helicity provides us with an effective means to control the helicity-dependent photocurrent in the spinmomentum-locked surface states of topological insulators(TIs).Also,the TIs show potential in polarization detection as an intrinsic solid-state optical chirality detector for easier integration and fabrication.However,the complex photoresponses with the circular photogalvanic effect,the linear photogalvanic effect,and the photon drag effect in the TIs prevent them from direct chirality detection of the elliptically polarized light.Here,by fitting with the theoretical models to the measured photocurrents,the microscopic origin of different components of the helicity-dependent photocurrent has been demonstrated.We show a comprehensive study of the helicitydependent photocurrent in(Bi_(1-x)Sb_(x))_(2)Te_(3)thin films of different thicknesses as a function of the light incident angle and the gate-tuned chemical potential.The observation of the light incident angle dependence of the helicitydependent photocurrent provides us with a polarization detection strategy using a TI thin film without the use of any additional optical elements,and the detection accuracy can be enhanced by gate tuning.Additionally,the Stokes parameters can be extracted by arithmetic operation of photocurrents measured with different incident angles and gating voltages for complete characterization of the polarization states of a light beam.Using this means,we realize the polarization detection and the Stokes parameters analysis with a single device.Our work provides an alternative solution to develop miniaturized intrinsic polarization-sensitive photodetectors.SHENZHONG CHEN JINLING YU XIYU HONG KEJING ZHU YONGHAI CHEN SHUYING CHENG YUNFENG LAI KE HE QIKUN XUE 2023Photonics Research2023,11,11:0
17Bimetallic Au/Pd catalyzed aerobic oxidation of alcohols in the poly(ethylene glycol)/CO_(2) system显示文摘Bimetallic Au/Pd nanoparticles were prepared and used to catalyze oxidation of alcohols in the poly(ethylene glycol) (PEG)/CO_(2) biphasic system using O2 as the oxidant without adding any base.The catalytic activity of Au/Pd bimetal with different mole ratios was studied using benzyl alcohol as the substrate.It was found that bimetallic Au/Pd nanoparticles with Au:Pd=1:3.5 had higher catalytic activity than monometallic Au,Pd and the bimetallic Au/Pd nanoparticles with other molar ratios.The effect of CO_(2) pressure on the oxidation of benzyl alcohol and 1-phenylethanol in PEG/CO_(2) was investigated.It was demonstrated that CO_(2) pressure could be used to tune the conversion and selectivity of the reactions effectively.α,β-Unsaturated alcohols were also studied and found to be more reactive than benzyl alcohol and 1-phenylethanol.Recycling experiments showed that the Au/Pd/PEG/CO_(2) catalytic system could be recycled at least four times without reducing the activity.In addition,the catalytic system is clean and the products can be separated easily.HE JinLing WU TianBin HU BaoJi CHENG Yan WU SuXiang JIANG Tao HAN BuXing 2010Science China Chemistry2010,53,7:0
18Micromechanical simulation on strength and ductility of two kinds of Al-based nanostructural materials显示文摘The nanostructured Al-based composites possess the combination of high yield strength and good ductility. In this paper, a micromechanical model is presented to simulate the mechanical response of bimodal nanostructured Al and the particle-reinforced aluminum matrix composite(PAMC). The constitutive relations for different phases are addressed in the model, as well as the contribution of microcracks. Numerical results show that the model can successfully describe the enhanced strength and ductility of the bimodal nanostructured Al, and the predictions of the PAMC are in good agreement with the experimental data. It is worth noting that the strength and ductility are sensitive to the volume fraction of constituents and the distribution of microcracks in both bimodal nanostructured Al and PAMC.
