A Chromosome-Scale Genome Assembly of Paper Mulberry(Bmussonetia papyrifera)Provides New Insights into Its Forage and Papermaking Usage显示文摘Paper mulberry(Broussonetia papyrifera)is a well-known woody tree historically used for Cai Lun papermaking, one of the four great inventions of ancient China.More recently,Paper mulberry has also been used as forage to address the shortage of feedstuff because of its digestible crude fiber and high protein contents. In this study,we obtained a chromosome-scale genome assembly for Paper mulberry using integrated approaches, including Illumina and PacBio sequencing platform as well as Hi-C,optical,and genetic maps. The assembled Paper mulberry genome consists of 386.83 Mb,which is close to the estimated size, and 99.25%(383.93 Mb)of the assembly was assigned to 13 pseudochromosomes.Comparative genomic analysis revealed the expansion and contraction in the flavonoid and lignin biosynthetic gene families, respectively, accounting for the enhanced flavonoid and decreased lignin biosynthesis in Paper mulberry. Moreover, the increased ratio of syringyl-lignin to guaiacyl-lignin in Paper mulberry underscores its suitability for use in medicine, forage, papermaking, and barkcloth making. We also identified the rootassociated microbiota of Paper mulberry and found that Pseudomonas and Rhizobia were enriched in its roots and may provide the source of nitrogen for its stems and leaves via symbiotic nitrogen fixation.Collectively, these results suggest that Paper mulberry might have undergone adaptive evolution and recruited nitrogen-fixing microbes to promote growth by enhancing flavonoid production and altering lignin monomer composition. Our study provides significant insights into genetic basis of the usefulness of Paper mulberry in papermaking and barkcloth making, and as forage. These insights will facilitate further domestication and selection as well as industrial utilization of Paper mulberry worldwide. | Xianjun Peng Hui Liu Peilin Chen Feng Tang Yanmin Hu Fenfen Wang Zhi Pi Meiling Zhao Naizhi Chen Hui Chen Xiaokang Zhang Xueqing Yan Min Liu Xiaojun Fu Guofeng Zhao Pu Yao Lili Wang He Dai Xuming Li Wei Xiong Wencai Xu Hongkun Zheng Haiyan Yu Shihua Shen | 2019 | Molecular Plant2019,12,5: | 18
Rhizosphere and bulk soil enzyme activities in a Nothotsuga longibracteata forest in the Tianbaoyan National Nature Reserve,Fujian Province,China显示文摘The rhizosphere, distinct from bulk soil, is defined as the volume of soil around living roots and influenced by root activities. We investigated protease, invertase, cellulase, urease, and acid phosphatase activities in rhizosphere and bulk soils of six Nothotsuga longibracteata forest communities within Tianbaoyan National Nature Reserve, including N. longibracteata + either Phyllostachys pubescens, Schima superba, Rhododendron simiarum, Cunninghamia lanceolata, or Cyclobalanopsis glauca, and N. longibracteata pure forest. Rhizosphere soils possessed higher protease, invertase, cellulase, urease, and acid phosphatase activities than bulk soils. The highest invertase, urease, and acid phosphatase activities were observed in rhizosphere samples of N. longibracteata+ S.superba. Protease was highest in the N. longibracteata + R. simiarum rhizosphere, while cellulase was highest in the pure N. longibracteata forest rhizosphere. All samples exhibited obvious rhizosphere effects on enzyme activities with a significant linear correlation between acid phosphatase and cellulase activities(p \ 0.05) in rhizosphere soils and between protease and acid phosphatase activities(p \ 0.05) in bulk soils. A principal component analysis, correlating 13 soil chemical properties indices relevant to enzyme activities, showed that protease, invertase, acid phosphatase, total N, and cellulase were the most important variables impacting rhizosphere soil quality. | Shihong Xiao Huiming You Weibin You Jinshan Liu Changtang Cai Jianqin Wu Zhirong Ji Shihua Zhan Zhesen Hu Zhongrui Zhang Dongjin He | 2017 | Journal of Forestry Research2017,28,3: | 7

Modeling of maize gross primary production using MODIS imagery and flux tower data显示文摘Maize is one of the most important crops cultivated on the global scale.Accurate estimation of maize Gross Primary Production(GPP)can provide valuable information for regional and global carbon budget studies. From site level to regional/global scales,GPP estimation depends on remote sensing or eddy covariance flux data.In this research, the 8-day composite GPP of maize was estimated by Moderate Resolution Imaging Spectroradiometer(MODIS)and flux tower data at eight study sites using a Regional Production Efficiency Model(REG-PEM).The performance of the model was assessed by analyzing the linearly regression of GPP estimated from the REG-PEM model(GPPEST)with the GPP predicted from the eddy covariance data(GPPEC).The coefficient of determination, root mean squared error and mean absolute error of the regression model were calculated. The uncertainties of the model are also discussed in this research. The seasonal dynamics(phases and magnitudes)of the GPPEST reasonably agreed with those of GPPEC,indicating the potential of the satellite-driven REG-PEM model for up-scaling the GPP in maize croplands.Furthermore, the maize GPP estimated by this model is more accurate than the MODIS GPP products(MOD17A2).In particular,MOD17A2 significantly underestimated the GPP of maize croplands. The uncertainties in the REG-PEM model are mostly contributed by the maximum light use efficiency and the fraction of photosynthetically active radiation. | Li Shihua He Ping Liu Baosheng Ni Ping Han Xing | 2016 | International Journal of Agricultural and Biological Engineering2016,9,2: | 2

Compression and shearing force on kernel rupture in shelling fresh lotus seeds显示文摘The mechanical properties of fresh lotus seeds are still poorly understood, which complicates the design of mechanical shelling machinery. Therefore, this work carried out four-factor orthogonal tests to determine the maximum permitted compressive force and minimum necessary shearing force to shell fresh lotus seeds without rupturing the kernel. It was found that the mean compression force that cracked the fresh lotus seed and led to kernel rupture was 213.03 N.Both the compressive force and the seed deformation upon kernel rupture were affected,in descending order of significance,by loading mode, seed grade, loading rate, and seed standing time.On the other hand, the shearing force needed to shell the seeds had a mean value of 7.84 N,far less than the compressive force that cracked the seed shell. The shearing force was affected,in descending order of significance,by seed standing time, tip angle of cutter blade, and loading rate, but not significantly affected by seed grade. The results suggested that mechanical shelling of fresh lotus seeds should ideally be carried out for fresh lotus seeds with a standing time of no more than 6 h using a cutter blade with an angle of about 40°at a loading rate of 30-90 mm/min. | Jincheng He Zhiying Tao Shihua Liang Dapeng Ye | 2021 | International Journal of Agricultural and Biological Engineering2021,14,1: | 1

Effect of oxygen concentration on resistive switching behavior in silicon oxynitride film显示文摘SiO_xN_y films with different oxygen concentrations were fabricated by reactive magnetron sputtering, and the resistive switching characteristics and conduction mechanism of Cu/SiO_xN_y/ITO devices were investigated. The Cu/SiO_xN_y/ITO device with SiO_xN_y deposited in 0.8-sccm O_2 flow shows a reliable resistive switching behavior, including good endurance and retention properties. As the conductivity of SiO_xN_y increases with the increase of the oxygen content dynamical electron trapping and detrapping is suggested to be the conduction mechanism. The temperature dependent I-V measurement indicates that the carrier transport can be ascribed to the hopping conduction rather than the metallic conductive filament. | Da Chen Shihua Huang Lü He | 2017 | Journal of Semiconductors2017,38,4: | 0
Simulation approach for optimization of ZnO/c-WSe2 heterojunction solar cells显示文摘Taking into account defect density in WSe_2,interface recombination between ZnO and WSe_2,we presented a simulation study of ZnO/crystalline WSe_2 heterojunction(HJ) solar cell using wxAMPS simulation software. The optimal conversion efficiency 39.07%for n-ZnO/p-c-WSe_2 HJ solar cell can be realized without considering the impact of defects.High defect density(> 1.0×10^(11) cm^(-2)) in c-WSe_2 and large trap cross-section(> 1.0×10^(-10) cm^2) have serious impact on solar cell efficiency.A thin p-WSe_2 layer is intentionally inserted between ZnO layer and c-WSe_2 to investigate the effect of the interface recombination. The interface properties are very crucial to the performance of ZnO/c-WSe_2 HJ solar cell. The affinity of ZnO value range between 3.7-4.5 eV gives the best conversion efficiency. | Shihua Huang Qiannan Li Dan Chi Xiuqing Meng Lü He | 2017 | Journal of Semiconductors2017,38,4: | 0
Relationship between Ham Quality and Body Weight of Fragrance Pig显示文摘[Objective] The paper was to determine the weight range of fragrance pigs suitable for processing small ham. [Method]In terms of differences in sensory state and physical and chemical indicators of hams produced by fragrance pigs with different weights,29 fragrance pigs at five weight grades were chosen and processed in accordance with Chinese ham processing methods. The products were evaluated by sense, and their salt and moisture contents were also measured.[Result]The fragrance pigs less than 30 kg were not suitable for making Chinese ham, while the fragrance pigs weighed 32-45 kg could be made into the ham with prominent flavor, beautiful appearance, good color, moderate taste, and suitable moisture content,of which the pigs weighed 40- 45 kg were the best materials for processing small ham. [Conclusion] The study provided scientific basis for industrial development of fragrance pig. | Shen Xuelin Duan Yongbang Zhu Qiujin He Chengyuan Yang Yuanpiao Pan Shihua | 2014 | Animal Husbandry and Feed Science2014,6,3: | 0
A single-nucleus transcriptomic atlas of the dog hippocampus reveals the potential relationship between specific cell types and domestication显示文摘The process of domestication has led to dramatic differences in behavioral traits between domestic dogs and gray wolves.Whole-genome research found that a class of putative positively selected genes were related to various aspects of learning and memory, such as long-term potentiation and long-term depression. In this study,we constructed a single-nucleus transcriptomic atlas of the dog hippocampus to illustrate its cell types, cell lineage and molecular features.Using the transcriptomes of 105057 nuclei from the hippocampus of a Beagle dog,we identified 26 cell clusters and a putative trajectory of oligodendrocyte development.Comparative analysis revealed a significant convergence between dog differentially expressed genes(DEGs)and putative positively selected genes(PSGs).Forty putative PSGs were DEGs in glutamatergic neurons, especially in Cluster 14,which is related to the regulation of nervous system development. In summary, this study provides a blueprint to understand the cellular mechanism of dog domestication. | Qi-Jun Zhou Xingyan Liu Longlong Zhang Rong Wang Tingting Yin Xiaolu Li Guimei Li Yuqi He Zhaoli Ding Pengcheng Ma Shi-Zhi Wang Bingyu Mao Shihua Zhang Guo-Dong Wang | 2022 | National Science Review2022,9,11: | 0
Virus inoculation and treatment regimens for evaluating anti-filovirus monoclonal antibody efficacy in vivo显示文摘The development of monoclonal antibodies to treat disease caused by filoviruses, particularly Ebola virus, has risen steeply in recent years thanks to several key studies demonstrating their remarkable therapeutic potential. The increased drive to develop new and better monoclonal antibodies has necessarily seen an increase in animal model efficacy testing, which is critical to the pre-clinical development of any novel countermeasure.Primary and secondary efficacy testing against filoviruses typically makes use of one or more rodent models(mice, guinea pigs, and occasionally hamsters)or the more recently described ferret model, although the exact choice of model depends on the specific filovirus being evaluated.Indeed,no single small animal model exists for all filoviruses, and the use of any given model must consider the nature of that model as well as the nature of the therapeutic and the experimental objectives.Confirmatory evaluation,on the other hand,is performed in nonhuman primates(rhesus or cynomolgus macaques)regardless of the filovirus. In light of the number of different animal models that are currently used in monoclonal antibody efficacy testing,we sought to better understand how these efficacy tests are being performed by numerous different laboratories around the world. To this end,we review the animal models that are being used for antibody efficacy testing against filoviruses, and we highlight the challenge doses and routes of infection that are used.We also describe the various antibody treatment regimens, including antibody dose, route, and schedule of administration, that are used in these model systems. We do not identify any single best model or treatment regimen, and we do not advocate for field-wide protocol standardization.Instead,we hope to provide a comprehensive resource that will facilitate and enhance the continued pre-clinical development of novel monoclonal antibody therapeutics. | Logan Banadyga Zachary Schiffman Shihua He Xiangguo Qiu | 2019 | Biosafety and Health2019,1,1: | 0
Formulation of Classical Swine Fever Immunization Procedures in Large scale Pig Farms显示文摘[Objective] The paper was to formulate classical swine fever( CSF) immunization procedures in large-scale pig farms. [Method] Antibody maintenance time and dynamics of material antibody was detected by enzyme-linked immunosorbent assay( ELISA),and scientific primary immunization time of piglets was explored. [Result] The positive rate of sows reached the highest level of 93% at 4 weeks post immunization. Piglets were immunized at 9 weeks of age,the antibody level did not increase at 3 weeks post immunization, but increased rapidly after secondary immunization; the antibody level in late fattening period was high, and pigs could be protected completely. [Conclusion]General prevention of CSF is suitable for scale farming. It takes about 4 weeks to produce CSF antibody. Therefore, pig farms should conduct immunization timely at one month prior to susceptible stage according to their infection pressure. | He Junchang Zhang Jianyuan Xie Youfu Fan Lizhe Gao Shihua | 2017 | Animal Husbandry and Feed Science2017,9,5: | 0
Characteristics of Hydrogen and Oxygen Isotopes and Noble Gas Isotopes in the Groundwater of Weishan, Wudalianchi, Northeast China显示文摘According to the hydrochemical characteristics, hydrogen and oxygen isotope characteristics and the ratio of noble gas isotopes of the sandstone aquifer and basalt aquifer, this study calculated the recharge temperature and residence time of groundwater in the Weishan area of Wudalianchi, also calculating the contribution of noble gas components from different sources to the samples. Based on the characteristics of hydrogen and oxygen isotopes and noble gases Xe and Ne, the recharge altitude and recharge temperature of the two aquifers were estimated, and the recharge temperature fitting with the NGT model as verified, the results showing that the main recharge altitude of groundwater in the region was 500–600 m, the recharge temperature being 2–7°C. He_(eq) and He_(ea) of the samples have been simulated using the OD model, the content of radioactive ~4He in the crust being obtained, the groundwater ages under the two conditions(closed condition and open condition) both being simulated. The results show that groundwater from the sandstone layer water is older than groundwater from the basalt layer. Hydrochemical characteristics and noble gas isotope ratios indicate that in the basalt aquifer and sandstone aquifer in the Weishan area, in addition to atmospheric and crustal helium, there is also an input of mantle-derived helium. The fault constitutes the uplift channel for groundwater containings mantle components, which results in the mantle source composition in water samples near the fault being much higher than those form non-fault areas. | WANG Shuai HUANG Xuelian QI Shihua HAN Yongjie KUANG Jian WANG Siqi WANG He XIAO Zhicai | 2022 | Acta Geologica Sinica(English Edition)2022,96,5: | 0
