1Auditory characteristics of noise-exposed memberscrossing age-related groups显示文摘Objective: To report audiological characteristics in a group of noise-exposed crew members on board ships.Methods and materials: Clinical and audiological measurements including pure-tone thresholds, acoustic immittance results and tinnitus questionnaires were collected from both the ship crew members(study subjects) and their land based colleagues(controls).Results: 1) Noise exposed crew members showed not only high frequency, but also low frequency hearing loss; 2) Hearing impairment increased with age, with 65.5% of crew members younger than 50 years showing normal hearing while only 14.9% of those older than 50 years had normal hearing; 3)hearing loss gradually increased with the extension of on board career time; and 4) Most study subjects reported high pitch tinnitus, significantly more than the control group although not significantly different among different age groups.Conclusion: Noise induced hearing impairment from working on board ships shows specific frequency and age characteristics. 2Recovery of yttrium from deep-sea mud显示文摘Deep-sea mud rich in rare earth yttrium has received lots of attention from the international community as a new resource for Y. A novel process, which mainly includes acid leaching, solvent extraction, and oxalic acid precipitation-roasting, is proposed for recovery of Y from deep-sea mud. A series of experiments were conducted to inspect the impacts of various factors during the process and the optimum conditions were determined. The results show that the Y of deep-sea mud totally exists in apatite minerals which can be decomposed by hydrochloric acid and sulfuric acid solution. The highest leaching efficiency of Y is 94.53% using hydrochloric acid and 84.38% using sulfuric acid under the conditions of H^+concentration 2.0 mol/L, leaching time 60 min, liquid-solid ratio 4:1 and room temperature 25 ℃(only in case of sulfuric acid, when using hydrochloric acid, the leaching temperature should be 60 ℃). Because of the much lower leaching temperature, sulfuric acid leaching is preferred. The counter current extraction and stripping tests were simulated by a cascade centrifugal extraction tank device. Using 10 vol% P204,15 vol% TBP and 75 vol% sulfonated kerosene as extractant, 98.79% Y^(3+) and 42.60% Fe^(3+) are extracted from sulfuric acid leaching liquor(adjusted to pH = 2.0) after seven-stage counter current extraction with O/A ratio of 1:1 at room temperature, while other metals ions such as Al^(3+), Ca^(2+), Mg^(2+)and Mn^(2+) are almost not extracted. The Y^(3+) in loaded organic can be selectively stripped using 50 g/L sulfuric acid solution and the stripping efficiency reaches 99.86% after seven-stage counter current stripping with O/A ratio of 10:1 at room temperature, while only 2.26% co-extracted Fe^(3+) is stripped. The Y^(3+) of loaded strip liquor can be precipitated by oxalic acid to further separate Y^(3+) and Fe^(3+). The precipitation efficiency of Y^(3+) in loaded strip liquor can be 98.56% while Fe^(3+) is not precipitated under the conditions of oxalic acid solution concentration 200 g/L, quality ratio of oxalic acid to Y of 2, and precipitation time 0.5 h. And the precipitate was roasted at 850 ℃ for 3 h to obtain the oxide of Y in which the purity of Y_2 O_3/REO is 79.02% and the contents of major non-rare earth impurities are less than 0.21%.Over the whole process included acid leaching, solvent extraction, and oxalic acid precipitation-roasting,the yttrium yield is 82.04%.Kuifang Zhang Zhiqiang Liu Changyong Sun Hongyang Cao Kechao Zhu Wei Zhu Wei Li 2018Journal of Rare Earths2018,36,8:2
3Proper animal experimental designs for preclinical research of biomaterials for intervertebral disc regeneration显示文摘Low back pain is a vital musculoskeletal disease that impairs life quality,leads to disability and imposes heavy economic burden on the society,while it is greatly attributed to intervertebral disc degeneration(IDD).However,the existing treatments,such as medicines,chiropractic adjustments and surgery,cannot achieve ideal disc regeneration.Therefore,advanced bioactive therapies are implemented,including stem cells delivery,bioreagents administration,and implantation of biomaterials etc.Among these researches,few reported unsatisfying regenerative outcomes.However,these advanced therapies have barely achieved successful clinical translation.The main reason for the inconsistency between satisfying preclinical results and poor clinical translation may largely rely on the animal models that cannot actually simulate the human disc degeneration.The inappropriate animal model also leads to difficulties in comparing the efficacies among biomaterials in different reaches.Therefore,animal models that better simulate the clinical charateristics of human IDD should be acknowledged.In addition,in vivo regenerative outcomes should be carefully evaluated to obtain robust results.Nevertheless,many researches neglect certain critical characteristics,such as adhesive properties for biomaterials blocking annulus fibrosus defects and hyperalgesia that is closely related to the clinical manifestations,e.g,low back pain.Herein,in this review,we summarized the animal models established for IDD,and highlighted the proper models and parameters that may result in acknowledged IDD models.Then,we discussed the existing biomaterials for disc regeneration and the characteristics that should be considered for regenerating different parts of discs.Finally,well-established assays and parameters for in vivo disc regeneration are explored.Yizhong Peng Xiangcheng Qing Hongyang Shu Shuo Tian Wenbo Yang Songfeng Chen Hui Lin Xiao Lv Lei Zhao Xi Chen Feifei Pu Donghua Huang Xu Cao Zengwu Shao 2021Biomaterials Translational2021,2,2:2

7Delaminated layered double hydroxide delivers DNA molecules as sandwich nanostructure into cells via a non-endocytic pathway显示文摘Layered double hydroxides(LDHs) are effective molecular carriers in cytological research, gene therapy,and transgenic applications. Herein, we investigated the internalization behavior of the LDH-DNA bioconjugates via a microscopic approach and analyzed the internalization pathway by dissipative particle dynamics(DPD) simulations. We experimentally found that LDH can efficiently carry DNA into the nucleus of cell in BY-2 suspension cells. Furthermore, atomic force microscopy and X-ray diffraction analysis demonstrated that the LDH-DNA bioconjugates mainly exist as a DNA-LDH-DNA sandwich complex,while the LDH-DNA-LDH sandwich complex and DNA-LDH complex cannot be excluded. The DPD simulations further indicated that only the DNA-LDH-DNA sandwich structure could penetrate the plasma membrane(PM), while PM is impermeable to the LDH-DNA-LDH sandwich complex and the DNA-LDH complex. This work provides novel perspective for understanding the membrane penetration mechanism of LDH nano-sheets and new insights into the design of novel molecular delivery systems.ó 2017 Science China Press. 10Aristolochic acids exposure was not the main cause of liver tumorigenesis in adulthood显示文摘Aristolochic acids(AAs) have long been considered as a potent carcinogen due to its nephrotoxicity. Aristolochic acid I(AAI) reacts with DNA to form covalent aristolactam(AL)-DNA adducts,leading to subsequent A to T transversion mutation, commonly referred as AA mutational signature. Previous research inferred that AAs were widely implicated in liver cancer throughout Asia. In this study, we explored whether AAs exposure was the main cause of liver cancer in the context of HBV infection in China's Mainland. Totally 1256 liver cancer samples were randomly retrieved from 3 medical centers and a refined bioanalytical method was used to detect AAI-DNA adducts. 5.10% of these samples could be identified as AAI positive exposure. Whole genome sequencing suggested 8.41% of 107 liver cancer patients exhibited the dominant AA mutational signature, indicating a relatively low overall AAI exposure rate. In animal models, long-term administration of AAI barely increased liver tumorigenesis in adult mice, opposite from its tumor-inducing role when subjected to infant mice. Furthermore, AAI induced dose-dependent accumulation of AA-DNA adduct in target organs in adult mice, with the most detected in kidney instead of liver. 12Nanogenerator-based bidirectional pressure sensor array and its demonstration in underwater invasive species detection显示文摘Lakes areas,which cause catastrophic damages in both commercial fishery and ecological systems.However,current assessment strategies may pose challenges for lake-wide abundance estimation and non-target anadromous species preservation.Here,we demonstrate an efficacious species-specific non-destructive sensing system based on porous ferroelectret nanogenerator for in-situ monitoring of lamprey spawning migration using their unique suction behavior.Simulations show that the porous structure enables a redistribution of surface charges under bidirectional deformations,which allows the detection of both positive and negative pressures.The quasi-piezoelectric effect is further validated by quantitative analysis in a wide pressure range of−50 to 60 kPa,providing detailed insights into transduction working principles.For reliable lamprey detection,a 4×4-pixel sensor array is developed and integrated with a complementary metal-oxide-semiconductor(CMOS)based signal processing array thus constituting a sensing panel capable of recording oral suction patterns in an underwater environment.Yunqi Cao Hongyang Shi Xiaobo Tan Nelson Sepúlveda 2023Nano Research2023,16,9:0
13A valine residue deletion in ZmSig2A, a sigma factor, accounts for a revertible leaf-color mutation in maize显示文摘A nuclear-encoded sigma(σ) factor is essential for the transcriptional regulation of plant chloroplastencoded genes. Five putative maize r factors have been identified by database searches, but their functions are unknown. We report a maize leaf color mutant etiolated/albino leaf 1(eal1) that was derived from space mutation breeding. The eal1 mutant displays etiolated or albino leaves that then gradually turn to normal green at the seedling stage. The changes in eal1 leaf color are associated with changes in photosynthetic pigment content and chloroplast development. Map-based cloning revealed that a single amino-acid deletion changing Val_(480)-Val_(481)-Val_(482) to Val_(480)-Val_(481), in the C-terminal domain σ_(4) of the putative σ factor ZmSig2A, is responsible for the eal1 mutation. In comparison with the expression level of the wild-type(WT) allele ZmSig2A^(+) in WT plants, much higher expression of the mutant allele ZmSig2A^(⊿V) in eal1 plants was detected before the eal1 plants turned to normal green. ZmSig2A shows the highest similarity to rice OsSig2A and Arabidopsis SIG2. Ectopic expression of ZmSig2A^(+) or ZmSig2A^(⊿V) driven by the cauliflower mosaic virus 35 S promoter rescued the pale green leaf of the sig2 mutant, but ectopic expression of ZmSig2A^(⊿V) driven by the SIG2 promoter did not. We propose that the Val deletion generated a new weak allele of ZmSig2A that cannot completely abolish the ZmSig2A function. Some genes involved in chloroplast development and photosynthesis-associated nuclear genes showed significant expression differences between eal1 and WT plants. We conclude that ZmSig2A encoding a r factor is essential for maize chloroplast development. 14LncRNA ZFAS1 regulates cardiomyocyte differentiation of human embryonic stem cells显示文摘Background:Cardiomyocytes derived from human embryonic stem cells(hESCs)are regulated by complex and stringent gene networks during differentiation.Long non-coding RNAs(lncRNAs)exert critical epigenetic regulatory functions in multiple differentiation processes.However,the involvement of lncRNAs in the differentiation of hESCs into cardiomyocytes has not yet been fully elucidated.Here,we identified the key roles of ZFAS1(lncRNA zinc finger antisense 1)in the differentiation of cardiomyocytes from hESCs.Methods:A model of cardiomyocyte differentiation from stem cells was established using the monolayer differentiation method,and the number of beating hESCs-derived cardiomyocytes was calculated.Gene expression was analyzed by quantitative real-time PCR(qRTPCR).Immunofluorescence assays were performed to assess the expression of cardiac troponin T(cTnT)andα-actinin protein in cardiomyocytes.Results:qRT-PCR showed that ZFAS1 expression in the mesoderm was significantly higher than that in embryonic stem cells,cardiac progenitor cells,and cardiomyocytes.Knockdown of ZFAS1 inhibited cardiomyocyte differentiation from hESCs,which was characterized by reduced expression of the cardiac-specific markers cTnT,α-actinin,myosin heavy chain 6(MYH6),and myosin heavy chain 7(MYH7).In contrast,ZFAS1 overexpression remarkably increased the percentage of spontaneously beating cardiomyocytes.In terms of the mechanism,we found that ZFAS1 is an antisense lncRNA at the 5′end of the protein-coding gene ZNFX1.Knockdown of ZFAS1 could increase the mRNA expression level of ZNFX1.Furthermore,qRT-PCR demonstrated that the silencing of ZNFX1 led to an increase in cardiac-specific markers that predicted the promotion of cardiomyocyte differentiation.Conclusion:Altogether,these data suggest that lncRNA-ZFAS1 is required for cardiac differentiation by functionally inhibiting the expression of ZNFX1,which may provide a reference for the treatment of heart disease to a certain extent.YANG CAO YINING LIU YANG YU XIAOFEI GUO XIUXIU WANG WENYA MA HANJING LI ZHONGYU REN XINLU GAO SIJIA LI HAOYU JI HONGYANG CHEN HONG YAN YANAN TIAN XIN WANG BENZHI CAI 2023BIOCELL2023,47,6:0
15ZmMS39 encodes a callose synthase essential for male fertility in maize(Zea mays L.)显示文摘Callose contributes to many biological processes of higher plants including pollen development,cell plate and vascular tissue formation,as well as regulating the transport function of plasmodesmata.The functions of callose synthase genes in maize have been little studied.We describe a maize male-sterile mutant 39(ms39)characterized by reduced plant height.In this study,we confirmed using CRISPR/Cas9 technology that a mutation in Zm00001d043909(ZmCals12),encoding a callose synthase,is responsible for the male sterility of the ms39 mutant.Compared with male-fertile plants,callose deposition around the dyads and tetrads in ms39 anthers was significantly reduced.Increased cell autophagy observed in ms39 anthers may have been due to the premature programmed cell death of tapetal cells,leading to collapse of the anther wall structure.Disordered glucose metabolism in ms39 may have intensified autophagy in anthers.Evaluation of the ms39 gene on maize heterosis by paired-crossed experiment with 11 maize inbred lines indicated that ms39 can be used for maize hybrid seed production.Qunkai Niu Ziwen Shi Peng Zhang Shuai Su Bin Jiang Xiaowei Liu Zhuofan Zhao Suzhi Zhang Qiang Huang Chuan Li Tao Yu Hongyang Yi Tingzhao Rong Moju Cao 2023The Crop Journal2023,11,2:0
16Construction of oxygen defective ZnO/ZnFe_(2)O_(4) yolk-shell composite with photothermal effect for tetracycline degradation:Performance and mechanism insight显示文摘Oxygen vacancy induced photothermal effect is of great significant but lack of adequate attentions for environmental remediation.In this paper,green recyclable ZnO/ZnFe_(2)O_(4) with oxygen vacancy was prepared by a solvothermal-calcination method.The UV-vis light capture ability of ZnO/ZnFe_(2)O_(4) microspheres is improved with the multiple light reflections due to the yolk-shell structure,and the oxygen vacancy expands the absorption range of photocatalyst and enhances photothermal conversion.The optimized photocatalyst can heat the solution from room temperature to 70°C within 60 min of visible light illumination,and the light to heat conversion efficiency is achieved by 61.3%.Compared with the degradation efficiency at 20°C,photothermal catalysis achieves a stable degradation in 80 min,and the degradation efficiency is increased by 41.5%.This can be attributed to the fact that light induced thermal energy accelerates the migration of electrons and holes,and promotes the diffusion of free radicals by heating active centers in situ.The active species contributing to the degradation,in order of importance,are the superoxide radical,hydroxyl radical,hole and electron.The light-to-thermal assisted photocatalysis with ZnO/ZnFe_(2)O_(4) provides a new sight for the pollution control in the future practical applications.Ke Zhang Hongyang Cao Afzal Dar Danqing Li Lina Zhou Chuanyi Wang 2023Chinese Chemical Letters2023,34,1:0
