| 1 | Herbal decoctosome is a novel form of medicine显示文摘Traditionally, herbal medicine is consumed by drinking decoctions produced by boiling herbs with water. The functional components of the decoction are heat stable. Small RNAs(sRNAs) were reported as a new class of functional components in decoctions. However, the mechanisms by which sRNAs survive heat treatment of the decoction and enter cells are unclear.Previous studies showed that plant-derived exosome-like nanoparticles(ELNs), which we call botanosomes, could deliver therapeutic reagents in vivo. Here, we report that heat-stable decoctosomes(ELNs) from decoctions have more therapeutic effects than the decoctions in vitro and demonstrate therapeutic efficacy in vivo. Furthermore, sRNAs, such as HJT-sRNA-m7 and PGY-sRNA-6, in the decoctosome exhibit potent anti-fibrosis and anti-inflammatory effects, respectively. Decoctosome is comprised of lipids, chemical compounds, proteins, and s RNAs. A medical decoctosome mimic is called bencaosome. A single lipid sphinganine(d22:0) identified in the decoctosome was mixed and heated with the synthesized sRNAs to form the simplest bencaosome. This simple bencaosome structure was identified by critical micelle concentration(cmc) assay that sRNAs coassembled with sphinganine(d22:0) to form the lipid layers of vesicles. The heating process facilitates co-assembly of sRNAs and sphinganine(d22:0) until a steady state is reached. The artificially produced sphinganine-HJT-sRNA-m7 and sphinganinePGY-sRNA-6 bencaosomes could ameliorate bleomycin-induced lung fibrosis and poly(I:C)-induced lung inflammation, respectively, following oral administration in mice. | 2 | Plant-derived phosphocholine facilitates cellular uptake of anti-pulmonary fibrotic HJT-sRNA-m7显示文摘Pulmonary fibrosis, a progressive chronic disease with a high mortality rate, has limited treatment options. Currently, lung transplantation remains the only effective treatment. Here we report that a small RNA, HJT-sRNA-m7, from a Chinese herbal medicine Hong Jing Tian(HJT, RHODIOHAE CRENULATAE RADIX ET RHIZOMA, Rhodiola crenulata) can effectively reduce the expressions of fibrotic hallmark genes and proteins both in alveolar in vitro and in mouse lung tissues in vivo. We also discovered over one hundred oil-soluble chemicals from HJT decoctions, most of which are found in lipid extracts from other Chinese herbals decoctions, including Pu Gong Ying(PGY, TARAXACI HERBA, Taraxacum mongolicum), Chuan Xin Lian(CXL, changed to 'ANDROGRAPHIS HERBA, Andrographis paniculata'), and Jin Yin Hua(JYH, lonicera japonica or Honeysuckle). We identified the active component in these decoctions as two forms of phosphocholines, PC(18:0/18:2) and PC(16:0/18:2). These PCs potentially could form liposomes with small RNAs to enter human alveolar and gastric cells. | 3 | The PI3K/Akt/GSK-3β/ROS/eIF2B pathway promotes breast cancer growth and metastasis via suppression of NK cell cytotoxicity and tumor cell susceptibility显示文摘Objective: To examine the effect of pSer9-GSK-3β on breast cancer and to determine whether the underlying metabolic and immunological mechanism is associated with ROS/eIF2B and natural killer(NK) cells.Methods: We employed TWS119 to inactivate GSK-3β by phosphorylating Ser9 and explored its effect on breast cancer and NK cells. The expression of GSK-3β, natural killer group 2 member D(NKG2D) ligands, eIF2B was quantified by PCR and Western blot. We measured intracellular reactive oxygen species(ROS) and mitochondrial ROS using DCFH-DA and MitoSOX^(TM) probe,respectively, and conducted quantitative analysis of cellular respiration on 4T1 cells with mitochondrial respiratory chain complex Ⅰ/Ⅲ kits.Results: Our investigation revealed that TWS119 downregulated NKG2D ligands(H60 a and Rae1), suppressed the cytotoxicity of NK cells, and promoted the migration of 4T1 murine breast cancer cells. Nevertheless, LY290042, which attenuates p-GSK-3β formation by inhibiting the PI3K/Akt pathway, reversed these effects. We also found that higher expression of p Ser9-GSK-3β induced higher levels of ROS, and observed that abnormality of mitochondrial respiratory chain complex Ⅰ/Ⅲ function induced the dysfunction of GSK-3β-induced electron transport chain, naturally disturbing the ROS level. In addition, the expression of NOX3 and NOX4 was significantly up-regulated, which affected the generation of ROS and associated with the metastasis of breast cancer. Furthermore, we found that the expression of pSer535-eIF2B promoted the expression of NKG2D ligands(Mult-1 and Rae1) following by expression of pSer9-GSK-3β and generation of ROS.Conclusions: The PI3K/Akt/GSK-3β/ROS/eIF2B pathway could regulate NK cell activity and sensitivity of tumor cells to NK cells,which resulted in breast cancer growth and lung metastasis. Gut microbes are thought to be essential for the facilitation of infantile and childhood development and immunity by assisting in breaking down food substances to liberate nutrients, protecting against pathogens, stimulating or modulating the immune system, and exerting control over the hypothalamic–pituitary–adrenal axis.This review aims to summarize the current understanding of the colonization and development of the gut microbiota in early life, highlighting the recent findings regarding the role of intestinal microbes in pediatric diseases. | 6 | Increasing yield potential through manipulating of an ARE1 ortholog related to nitrogen use efficiency in wheat by CRISPR/Cas9显示文摘Wheat(Triticum aestivum L.)is a staple food crop consumed by more than 30%of world population.Nitrogen(N)fertilizer has been applied broadly in agriculture practice to improve wheat yield to meet the growing demands for food production.However,undue N fertilizer application and the low N use efficiency(NUE)of modern wheat varieties are aggravating environmental pollution and ecological deterioration.Under nitrogen-limiting conditions,the rice(Oryza sativa)abnormal cytokinin response1 repressor1(are1)mutant exhibits increased NUE,delayed senescence and consequently,increased grain yield.However,the function of ARE1 ortholog in wheat remains unknown.Here,we isolated and characterized three TaARE1 homoeologs from the elite Chinese winter wheat cultivar ZhengMai 7698.We then used CRISPR/Cas9-mediated targeted mutagenesis to generate a series of transgene-free mutant lines either with partial or triple-null taare1 alleles.All transgene-free mutant lines showed enhanced tolerance to N starvation,and showed delayed senescence and increased grain yield in field conditions.In particular,the AABBdd and aabbDD mutant lines exhibited delayed senescence and significantly increased grain yield without growth defects compared to the wild-type control.Together,our results underscore the potential to manipulate ARE1 orthologs through gene editing for breeding of high-yield wheat as well as other cereal crops with improved NUE. | Jiahui Zhang Huating Zhang Shaoya Li Jingying Li Lei Yan Lanqin Xia | 2021 | Journal of Integrative Plant Biology2021,63,9: | 13 |

A single nucleotide polymorphism in the NRT1.1 B gene between japonica and indica rice is responsible for the improved nitrogen use efficiency in indica rice. Herein, we precisely replaced the japonica NRT1.1 B allele with the indica allele, in just one generation, using CRISPR/Cas9 gene-editing technology.No additional selective pressure was needed to enrich the precise replacement events. | 9 | Sirt1-ROS-TRAF6 Signaling-Induced Pyroptosis Contributes to Early Injury in Ischemic Mice显示文摘Stroke is an acute cerebro-vascular disease with high incidence and poor prognosis,most commonly ischemic in nature.In recent years,increasing attention has been paid to inflammatory reactions as symptoms of a stroke.However,the role of inflammation in stroke and its underlying mechanisms require exploration.In this study,we evaluated the inflammatory reactions induced by acute ischemia and found that pyroptosis occurred after acute ischemia both in vivo and in vitro,as determined by interleukin-1β,apoptosis-associated speck-like protein,and caspase-1.The early inflammation resulted in irreversible ischemic injury,indicating that it deserves thorough investigation.Meanwhile,acute ischemia decreased the Sirtuin 1(Sirtl)protein levels,and increased the TRAF6(TNF receptor associated factor 6)protein and reactive oxygen species(ROS)levels.In further exploration,both Sirtl suppression and TRAF6 activation were found to contribute to this pyroptosis.Reduced Sirtl levels were responsible for the production of ROS and increased TRAF6 protein levels after ischemic exposure.Moreover,N-acetyl-L-cysteine,an ROS scavenger,suppressed the TRAF6 accumulation induced by oxygen-glucose deprivation via suppression of ROS bursts.These phenomena indicate that Sirtl is upstream of ROS,and ROS bursts result in increased TRAF6 levels.Further,the activation of Sirtl during the period of ischemia reduced ischemiainduced injury after 72 h of reperfusion in mice with middle cerebral artery occlusion.In sum,these results indicate that pyroptosis-dependent machinery contributes to the neural injury during acute ischemia via the Sirt1-ROS-TRAF6 signaling pathway.We propose that inflammatory reactions occur soon after oxidative stress and are detrimental to neuronal survival;this provides a promising therapeutic target against ischemic injuries such as a stroke. | Weijie Yan Wei Sun Jiahui Fan Haiqing Wang Song Han Junfa Li Yanling Yin | 2020 | Neuroscience Bulletin2020,36,8: | 9 |
| 10 | Visual Fixation Assessment in Patients with Disorders of Consciousness Based on Brain-Computer Interface显示文摘Visual fixation is an item in the visual function subscale of the Coma Recovery Scale-Revised(CRS-R).Sometimes clinicians using the behavioral scales find it difficult to detect because of the motor impairment in patients with disorders of consciousness(DOCs). Braincomputer interface(BCI) can be used to improve clinical assessment because it directly detects the brain response to an external stimulus in the absence of behavioral expression. In this study, we designed a BCI system to assist the visual fixation assessment of DOC patients. The results from 15 patients indicated that three showed visual fixation in both CRS-R and BCI assessments and one did not show such behavior in the CRS-R assessment but achieved significant online accuracy in the BCI assessment. The results revealed that electroencephalography-based BCI can detect the brain response for visual fixation. | 11 | Psychological impact of the COVID-19 pandemic on healthcare workers:a cross-sectional study in China显示文摘Background Healthcare workers fighting against the coronavirus disease 2019(COVID-19)pandemic are under tremendous pressure,which puts them at an increased risk of developing psychological problems.Aims This study aimed to investigate the prevalence of psychological problems in different healthcare workers(ie,physicians,medical residents,nurses,technicians and public health professionals)during the COVID-19 pandemic in China and explore factors that are associated with the onset of psychological problems in this population during this public health crisis.Methods A cross-sectional,web-based survey was conducted in February 2020 among healthcare workers during the COVID-19 pandemic.Psychological problems were assessed using the Generalized Anxiety Disorder Scale,Patient Health Questionnaire and Insomnia Severity Index.Logistic regression analyses were used to explore the factors that were associated with psychological problems.Results The prevalence of symptoms of anxiety,depression,insomnia and the overall psychological problems in healthcare workers during the COVID-19 pandemic in China was 46.04%,44.37%,28.75%and 56.59%,respectively.The prevalence of the overall psychological problems in physicians,medical residents,nurses,technicians and public health professionals was 60.35%,50.82%,62.02%,57.54%and 62.40%,respectively.Compared with healthcare workers who did not participate in front-line work,front-line healthcare workers had a higher risk of anxiety,insomnia and overall psychological problems.In addition,attention to negative or neutral information about the pandemic,receiving negative feedback from families and friends who joined front-line work,and unwillingness to join front-line work if given a free choice were three major factors for these psychological problems.Conclusions Psychological problems are pervasive among healthcare workers during the COVID-19 pandemic.Receiving negative information and participating in front-line work appear to be.important risk factors for psychological problems.The psychological health of different healthcare workers should be protected during the COVID-19 pandemic with timely interventions and proper information feedback. | Jianyu Que Le Shi Jiahui Deng Jiajia Liu Li Zhang Suying Wu Yimiao Gong Weizhen Huang Kai Yuan Wei Yan Yankun Sun Maosheng Ran Yanping Bao Lin Lu | 2020 | General Psychiatry2020,33,3: | 8 |

| 13 | Diclofop-methyl affects microbial rhizosphere community and induces systemic acquired resistance in rice显示文摘Diclofop-methyl(DM),a widely used herbicide in food crops,may partly contaminate the soil surface of natural ecosystems in agricultural area and exert toxic effects at low dose to nontarget plants.Even though rhizosphere microorganisms strongly interact with root cells,little is known regarding their potential modulating effect on herbicide toxicity in plants.Here we exposed rice seedlings(Xiushui 63) to 100 μg/L DM for 2 to 8 days and studied the effects of DM on rice rhizosphere microorganisms,rice systemic acquired resistance(SAR) and rice-microorganisms interactions.The results of metagenomic 16 S rDNA Illumina tags show that DM increases bacterial biomass and affects their community structure in the rice rhizosphere.After DM treatment,the relative abundance of the bacterium genera Massilia and Anderseniella increased the most relative to the control.In parallel,malate and oxalate exudation by rice roots increased,potentially acting as a carbon source for several rhizosphere bacteria.Transcriptomic analyses suggest that DM induced SAR in rice seedlings through the salicylic acid(but not the jasmonic acid) signal pathway.This response to DM stress conferred resistance to infection by a pathogenic bacterium,but was not influenced by the presence of bacteria in the rhizosphere since SAR transcripts did not change significantly in xenic and axenic plant roots exposed to DM.The present study provides new insights on the response of rice and its associated microorganisms to DM stress. | Si Chen Xingxing Li Michel Lavoie Yujian Jin Jiahui Xu Zhengwei Fu Haifeng Qian | 2017 | Journal of Environmental Sciences2017,29,1: | 8 |
| 14 | Cryogenic 3D printing of dual-delivery scaffolds for improved bone regeneration with enhanced vascularization显示文摘Three-dimensional(3D)printing has been increasingly employed to produce advanced bone tissue engineering scaffolds with biomimetic structures and matched mechanical strengths,in order to induce improved bone regeneration in defects with a critical size.Given that the successful bone regeneration requires both excellent osteogenesis and vascularization,endowing scaffolds with both strong bone forming ability and favorable angiogenic potential would be highly desirable to induce improved bone regeneration with required vascularization.In this investigation,customized bone tissue engineering scaffolds with balanced osteoconductivity/osteoinductivity were produced via cryogenic 3D printing ofβ-tricalcium phosphate and osteogenic peptide(OP)containing water/poly(lactic-co-glycolic acid)/dichloromethane emulsion inks.The fabricated scaffolds had a hierarchically porous structure and were mechanically comparable to human cancellous bone.Angiogenic peptide(AP)containing collagen I hydrogel was then coated on scaffold surface to further provide scaffolds with angiogenic capability.A sequential release with a quick AP release and a slow but sustained OP release was obtained for the scaffolds.Both rat endothelial cells(ECs)and rat bone marrow derived mesenchymal stem cells(MSCs)showed high viability on scaffolds.Improved in vitro migration and angiogenesis of ECs were obtained for scaffolds delivered with AP while enhanced osteogenic differentiation was observed in scaffolds containing OP.The in vivo results showed that,toward scaffolds containing both AP and OP,the quick release of AP induced obvious angiogenesis in vivo,while the sustained OP release significantly improved the new bone formation.This study provides a facile method to produce dual-delivery scaffolds to achieve multiple functions. | Chong Wang Jiahui Lai Kai Li Shaokui Zhu Bingheng Lu Jia Liu Yujin Tang Yen Wei | 2021 | Bioactive Materials2021,6,1: | 7 |
| 15 | Oxymatrine reduces neuroinflammation in rat brain A signaling pathway显示文摘Cerebral neuroinflammation models were established by injecting 10 μg lipopolysaccharide into the hippocampus of male Sprague-Dawley rats. The rats were treated with an intraperitoneal injection of 120, 90, or 60 mg/kg oxymatrine daily for three days prior to the lipopolysaccharide injection. Twenty-four hours after model induction, the hippocampus was analyzed by real-time quantitative PCR, and the cerebral cortex was analyzed by enzyme-linked immunosorbent assay and western blot assay. The results of the enzyme-linked immunosorbent assay and the real-time quantitative PCR showed that the secretion and mRNA expression of the pro-inflammatory cytokines interleukin-1β and tumor necrosis factor-α were significantly decreased in the hippocampus and cerebral cortex of model rats treated with oxymatrine. Western blot assay and real-time quantitative PCR analysis indicated that toll-like receptor 4 mRNA and protein expression were significantly decreased in the groups receiving different doses of oxymatrine. Additionally, 120 and 90 mg/kg oxymatrine were shown to reduce protein levels of nuclear factor-κB p65 in the nucleus and of phosphorylated IκBα in the cytoplasm of brain cells, as detected by western blot assay. | 17 | Development of ensemble learning models to evaluate the strength of coal-grout materials显示文摘In the loose and fractured coal seam with particularly low uniaxial compressive strength(UCS),driving a roadway is extremely difficult as roof falling and wall spalling occur frequently.To address this issue,the jet grouting(JG)technique(high-pressure grout mixed with coal particles)was first introduced in this study to improve the self-supporting ability of coal mass.To evaluate the strength of the jet-grouted coal-grout composite(JG composite),the UCS evolution patterns were analyzed by preparing 405 specimens combining the influential variables of grout types,curing time,and coal to grout(C/G)ratio.Furthermore,the relationships between UCS and these influencing variables were modeled using ensemble learning methods i.e.gradient boosted regression tree(GBRT)and random forest(RF)with their hyperparameters tuned by the particle swarm optimization(PSO).The results showed that the chemical grout composite has higher short-term strength,while the cement grout composite can achieve more stable strength in the long term.The PSO-GBRT and PSO-RF models can both achieve high prediction accuracy.Also,the variable importance analysis demonstrated that the grout type and curing time should be considered carefully.This study provides a robust intelligent model for predicting UCS of JG composites,which boosts JG design in the field. | Yuantian Sun Guichen Li Nong Zhang Qingliang Chang Jiahui Xu Junfei Zhang | 2021 | International Journal of Mining Science and Technology2021,31,2: | 6 |
| 18 | Strain hardening behavior of Mg-Y alloys after extrusion process显示文摘The strain hardening is an effective mode of enhancing mechanical properties in alloys.In this work,the strain hardening behaviors of Mg-xY(x=1,2,and 3 wt%)after extrusion process was investigated using uniaxial tensile tests.Results suggest that the Mg-xY alloys are composed ofα-Mg with a little amount of Mg24Y5 phase.The average grain size reduces from 19.8μm to 12.2μm as the Y content adds from 1 wt%to 2 wt%.Nevertheless,when Y content reaches 3 wt%,the grain size reaches to 12.9μm,which is close to that of Mg-2Y.The strain hardening rate decreases from 883 MPa to 798 MPa at(σ-σ0.2)=40 MPa,and Mg-2Y and Mg-3Y have the similar strain hardening response.Moreover,Mg-1Y shows an obvious ascending stage after the steep decreasing stage,which is mainly caused by the activation of twinning.The strain hardening behavior of Mg-xY is explained based on understanding the roles of the deformation mechanisms via deformation microstructure analysis and Visco-Plastic Self Consistent(VPSC)model.The variation of strain hardening characteristics with increasing Y content is related to the effects of grain size and texture. | Chaoyue Zhao Ziyan Li Jiahui Shi Xianhua Chen Teng Tu Zhu Luo Renju Cheng Andrej Atrens Fusheng Pan | 2019 | Journal of Magnesium and Alloys2019,7,4: | 6 |
| 19 | Characterization of particulate products for aging of ethylbenzene secondary organic aerosol in the presence of ammonium sulfate seed aerosol显示文摘Aging of secondary organic aerosol(SOA) particles formed from OH– initiated oxidation of ethylbenzene in the presence of high mass(100–300 μg/m^3) concentrations of(NH_4)_2SO_4seed aerosol was investigated in a home-made smog chamber in this study.The chemical composition of aged ethylbenzene SOA particles was measured using an aerosol laser time-of-flight mass spectrometer(ALTOFMS) coupled with a Fuzzy C-Means(FCM) clustering algorithm.Experimental results showed that nitrophenol,ethyl-nitrophenol,2,4-dinitrophenol,methyl glyoxylic acid,5-ethyl-6-oxo-2,4-hexadienoic acid,2-ethyl-2,4-hexadiendioic acid,2,3-dihydroxy-5-ethyl-6-oxo-4-hexenoic acid,1H-imidazole,hydrated N-glyoxal substituted1H-imidazole,hydrated glyoxal dimer substituted imidazole,1H-imidazole-2-carbaldehyde,N-glyoxal substituted hydrated 1H-imidazole-2-carbaldehyde and high-molecular-weight(HMW) components were the predominant products in the aged particles.Compared to the previous aromatic SOA aging studies,imidazole compounds,which can absorb solar radiation effectively,were newly detected in aged ethylbenzene SOA in the presence of high concentrations of(NH_4)_2SO_4seed aerosol.These findings provide new information for discussing aromatic SOA aging mechanisms. | Mingqiang Huang Jiahui Zhang Shunyou Cai Yingmin Liao Weixiong Zhao Changjin Hu Xuejun Gu Li Fang Weijun Zhang | 2016 | Journal of Environmental Sciences2016,28,9: | 6 |
