| 5 | Lymphangiogenesis contributes to exercise-induced physiological cardiac growth显示文摘Background:Promoting cardiac lymphangiogenesis exerts beneficial effects for the heart.Exercise can induce physiological cardiac growth with cardiomyocyte hypertrophy and increased proliferation markers in cardiomyocytes.However,it remains unclear whether and how lymphangiogenesis contributes to exercise-induced physiological cardiac growth.We aimed to investigate the role and mechanism of lymphangiogenesis in exercise-induced physiological cardiac growth.Methods:Adult C57 BL6/J mice were subjected to 3 weeks of swimming exercise to induce physiological cardiac growth.Oral treatment with vascular endothelial growth factor receptor 3(VEGFR3) inhibitor SAR1 3 1 675 was used to investigate whether cardiac lymphangiogenesis was required for exercise-induced physiological cardiac growth by VEGFR3 activation.Furthermore,human dermal lymphatic endothelial cell(LEC)-conditioned medium was collected to culture isolated neonatal rat cardiomyocytes to determine whether and how LECs could influence cardiomyocyte proliferation and hypertrophy.Results:Swimming exercise induced physiological cardiac growth accompanied by a remarkable increase of cardiac lymphangiogenesis as evidenced by increased density of lymphatic vessel endothelial hyaluronic acid receptor 1-positive lymphatic vessels in the heart and upregulated LYVE-1 and Podoplanin expressions levels.VEGFR3 was upregulated in the exercised heart,while VEGFR3 inhibitor SAR131675 attenuated exercise-induced physiological cardiac growth as evidenced by blunted myocardial hypertrophy and reduced proliferation marker Ki67 in cardiomyocytes,which was correlated with reduced lymphatic vessel density and downregulated LYVE-1 and Podoplanin in the heart upon exercise.Furthermore,LEC-conditioned medium promoted both hypertrophy and proliferation of cardiomyocytes and contained higher levels of insulinlike growth factor-1 and the extracellular protein Reelin,while LEC-conditioned medium from LECs treated with SAR131675 blocked these effects.Functional rescue assays further demonstrated that protein kinase B(AKT) activation,as well as reduced CCAAT enhancer-binding protein beta(C/EBPβ) and increased CBP/p300-interacting transactivators with E(glutamic acid)/D(aspartic acid)-rich-carboxylterminal domain 4(CITED4),contributed to the promotive effect of LEC-conditioned medium on cardiomyocyte hypertrophy and proliferation.Conclusion:Our findings reveal that cardiac lymphangiogenesis is required for exercise-induced physiological cardiac growth by VEGFR3 activation,and they indicate that LEC-conditioned medium promotes both physiological hypertrophy and proliferation of cardiomyocytes through AKT activation and the C/EBPβ-CITED4 axis.These results highlight the essential roles of cardiac lymphangiogenesis in exercise-induced physiological cardiac growth. | Yihua Bei Zhenzhen Huang Xing Feng Lin Li Meng Wei Yujiao Zhu Shuqin Liu Chen Chen Mingming Yin Huimin Jiang Junjie Xiao | 2022 | Journal of Sport and Health Science2022,11,4: | 2 |
| 6 | Rice FLOURY ENDOSPERM 18 encodes a pentatricopeptide repeat protein required for 5′ processing of mitochondrial nad5 messenger RNA and endosperm development显示文摘Pentatricopeptide repeat(PPR) proteins, composing one of the largest protein families in plants,are involved in RNA binding and regulation of organelle RNA metabolism at the posttranscriptional level. Although several PPR proteins have been implicated in endosperm development in rice(Oryza sativa), the molecular functions of many PPRs remain obscure. Here, we identified a rice endosperm mutant named floury endosperm 18(flo18) with pleiotropic defects in both reproductive and vegetative development.Map-based cloning and complementation tests showed that FLO18 encodes a mitochondriontargeted P-type PPR protein with 15 PPR motifs.Mitochondrial function was disrupted in the flo18 mutant, as evidenced by decreased assembly of Complex I in the mitochondrial electron transport chain and altered mitochondrial morphology. Loss of FLO18 function resulted in defective 5′-end processing of mitochondrial nad5 transcripts encoding subunit 5 of nicotinamide adenine dinucleotide hydrogenase. After calcinations, finer particles with narrow distribution and large surface area were obtained. After dry pressing, samples were sintered at 1500℃―1700℃ for 4 h in vacuum to produce transparent polycrystalline ceramics with uniform grains. Samples with 9 mol% lanthanum were transparent in visible light after being sintered at 1500℃ for 4 h. The grain sizes increased with lanthanum doping compared with those of pure yttria transparent ceramic sintered at the same conditions. Relative density of the transparent ceramic was 99.7%. Here, we provide a novel micro/nano-assembling method, which is accurate and efficient, especially suitable for the fabrication of micro/nano-scale electronic devices. Using this method, a self-powered ZnO/Sb-doped ZnO nanowire p–n homojunction ultraviolet detector(UVD) was fabricated, and the detailed photoelectric properties were tested. At a reverse bias of -0.1 V under UV light illumination, the photoresponse sensitivity of the UVD was 26.5 and the rise/decay time of the UVD was as short as 30 ms. However, P-type PPRs involved in both RNA splicing and editing have rarely been reported, and their underlying mechanism remains largely unknown. Here, we report a rice floury endosperm22(flo22) mutant with delayed amyloplast development in endosperm cells. Map-based cloning and complementation tests demonstrated that FLO22 encodes a mitochondrion-localized P-type PPR protein.Mutation of FLO22 resulting in defective transsplicing of mitochondrial nad1 intron 1 and perhaps causing instability of mature transcripts affected assembly and activity of complex Ⅰ, and mitochondrial morphology and function. RNA-seq analysis showed that expression levels of many genes involved in starch and sucrose metabolism were significantly down-regulated in the flo22mutant compared with the wild type, whereas genes related to oxidative phosphorylation and the tricarboxylic acid cycle were significantly upregulated. In addition to involvement in splicing as a P-type PPR protein, we found that FLO22 interacted with DYW3, a DYW-type PPR protein, and they may function synergistically in mitochondrial RNA editing. | 19 | Redirecting dynamic structural evolution of nickel-contained RuO_(2) catalyst during electrochemical oxygen evolution reaction显示文摘Electrochemical oxygen evolution reaction (OER) is a main efficiency bottleneck of water electrolysis.Commercial ruthenium oxide (RuO_(2)) catalyst displays remarkable activities but poor stability for OER.The instability stems from lattice oxygen oxidation,resulting in the oxidation of Ru^(4+) to soluble Ru^(4+)(x>4) species.Herein,we redirect dynamic structural evolution of Ru-based catalysts through introducing oxidized nickel (Ni) components.By virtue of comprehensive structural characterizations,such as high-angle annular dark-field scanning transmission electron microscopy (HAADF-STEM),X-ray photoelectron spectroscopy (XPS),operando Raman and so forth,it is demonstrated that when the atomic content of Ni exceeds that of ruthenium (Ru),the Ni components can efficiently inhibit the Ru^(4+) oxidation and structural collapse.Density functional theory (DFT) calculations suggest that the introduction of Ni component hinders the formation of oxygen vacancies,and makes lattice oxygen mediated mechanism turn to adsorbate evolution mechanism,which eventually improves the stability.The optimized nickel-contained RuO_(2) catalyst delivers an effective reactivity with an overpotential of less than 215 m V to attain 10 m A cm^(-2) and remarkable stability with only 5 mV increment after 5000 potential cycles.This work provides insights into the origin of dynamic structural evolution of transition-metalmodified RuO_(2) electrocatalysts. | Yuhan Zhao Menghua Xi Yanbin Qi Xuedi Sheng Pengfei Tian Yihua Zhu Xiaoling Yang Chunzhong Li Hongliang Jiang | 2022 | Journal of Energy Chemistry2022,31,6: | 1 |
