| 2 | Effects of dietary Clostridium butyricum supplementation on growth performance,intestinal development,and immune response of weaned piglets challenged with lipopolysaccharide显示文摘Background: Weanling pigs, with immature immune system and physiological function, usually experience postweaning diarrhea. This study determined the effects of dietary Clostridium butyricum supplementation on growth performance, diarrhea, and immunity of weaned pigs challenged with lipopolysaccharide(LPS).Methods: In Experiment(Exp.) 1,144 weaned piglets were weaned at 21 d and randomly assigned to six groups,with six replicates per group and four pigs per replicate, receiving a control diet(CON) or diet supplemented with antibiotics(AB) or C. butyricum(CB)(0.1%, 0.2%, 0.4%, or 0.8%), respectively. All diets in Exp. 1 were a highly digestible basal diet, with 3,000 mg/kg zinc oxide supplied in the first 2 wk only. In Exp. 2, 180 piglets were weaned at 21 d and randomly assigned to five groups, with six replicates per group and six pigs per replicate, receiving CON, AB, or CB(0.2%, 0.4%, or 0.6%) diets. The digestibility of diets was lower than those in Exp. 1, and did not include zinc oxide. At 36 d of Exp. 2, 12 piglets were selected from each of the CON and 0.4% CB groups, six piglets were intraperitoneally injected with LPS(50 μg/kg body weight) and the other six piglets with normal saline;animals were killed at 4 h after injection to collect blood, intestine, and digesta samples for biochemical analysis.Results: In Exp. 1, CB and AB diets had no effect on growth performance of piglets. In Exp. 2, 0.4% CB decreased feed-gain ratio(P < 0.1), diarrhea score(P < 0.05), and increased duodenal, jejunal, and ileal villus height and jejunal villus height/crypt depth(P < 0.05). The 0.4% CB decreased the plasma tumor necrosis factor(TNF) α(P < 0.05) but increased ileal mucosa IL-10 and TLR2 mRNA expression(P < 0.05). Furthermore, 0.4% CB altered the microbial profile, with Bacillus and Ruminococcaceae UGG-003 at genus level and Lactobacillus casei and Parasutterella secunda at species level were higher than CON in colonic content(P < 0.05).Conclusions: Dietary C. butyricum supplementation had positive effects on growth of weaned piglets with less digestible diets. There was a tendency to reduce the feed-gain ratio, which could reduce feed costs in pig production. | 3 | Geochemical Signatures of Early Paleogene Source Rocks in the Sanshui Basin, South China显示文摘The Honggang member of the early Paleogene Buxin Formation is the main source rock in the Sanshui Basin, characterized by organic-rich black shales with the cyclic recurrence of organic- poor sediments. The geochemical characteristics of the Honggang member have been documented to determine the organic matter types and depositional environments in this paper. The organic matter of the black shales mainly consists of a mixture of land plant-derived and phytoplankton-derived organic matter. Total organic carbon content (TOC)–sulfur–iron (Fe) relationships suggest that the organic- rich black shales were deposited under dysoxic-to-euxinic water conditions. The time that iron minerals remained in contact with H2S in anoxic waters possibly influenced the formation of syngenetic pyrite, and organic carbon controlled the formation of diagenetic pyrite. Organic-poor intervals usually show pyrite sulfur enrichment and higher degree of pyritization values relative to low organic carbon contents. This resulted from HS– diffusing downward from overlying organic-rich sediments and formed Fe sulfides through reactions with sufficient Fe. Trace elements generally exhibit low concentrations and little TOC dependence, suggesting some degree of depletion in these elements in the early Paleogene sediments of the Sanshui Basin. | 4 | Body mass index does not affect the survival of pancreatic cancer patients显示文摘AIM to evaluate the association of body mass index(b MI) with the overall survival of pancreatic ductal adenocarcinoma(PDAC) patients.METHODS A retrospective analysis of PDAC patients diagnosed in the National Cancer Center of China between January 1999 and December 2014 was performed. these patients were categorized into four b MI groups(< 18.5, 18.5-22.9, 23-27.4 and ≥ 27.5 kg/m2). χ2 tests for comparison of the proportions of categorical variables, and Student's t-test or Mann-Whitney test for continuous variables were employed. Survival analysis was performed with the Kaplan-Meyer method. their HRs of mortality and 95%CIs were estimated using the Cox proportional hazards model.RESULTS With a median age of 59.6 years(range: 22.5-84.6 years), in total 1783 PDAC patients were enrolled in this study. their mean usual b MI was 24.19 ± 3.53 for the whole cohort. More than half of the patients(59.3%) experienced weight loss during the disease onset and progression. Compared with healthy-weight individuals, newly diagnosed patients who were overweight or obese had more severe weight loss during their disease onset and progression(P < 0.001). Individuals who were overweight or obese were associated with positive smoking history(P < 0.001). A significant difference in comorbidity of diabetes(P = 0.044) and coronary artery disease(P < 0.001) was identified between high b MI and normal-weight patients. After a median follow-up of 8 mo, the survival analysis showed no association between b MI and the overall survival(P = 0.90, n = 1783). | 8 | Suitability projection for Chinese ski areas under future natural andsocioeconomic scenarios显示文摘Ski tourism is extremely sensitive to climate change and is also heavily affected by socioeconomic conditions.Although some ski areas arestill profitable under current climate and socioeconomic conditions,they will become difficult to operate in the face of rising winter temper-atures,which will result in further economic losses,resource waste and environmental damage.This study projects variability in the suitability ofski area development across China in the coming decades.Natural suitability under three representative concentration pathway emission sce-narios(RCP2.6,RCP4.5 and RCP8.5),socioeconomic suitability under four shared socioeconomic pathways(SSP1,SSP2,SSP3,and SSP5)andintegrated suitability under four climatic-socioeconomic scenarios(RCP2.6-SSP1,RCP4.5-SSP2,RCP8.5-SSP3,and RCP8.5-SSP5)are re-ported.Furthermore,the suitability of 731 existing ski areas in China is assessed.The results show a substantial decline in integrated suitabilityfor most regions of China except for some very cold areas,where higher air temperatures will make visitors feel more comfortable and therelatively poor socioeconomic conditions will improve in the 2030s,2050s and 2090s.The average higher integrated suitability area(integratedsuitability values greater than 0.5)under four climatic-socioeconomic scenarios decreases from the current 29.9%e14.4%,5.0%and 4.5%bythe 2030s,2050s and 2090s,respectively.Under RCP2.6-SSP1,the higher integrated suitability area is projected to decrease from the current28.0%e5.2%by the 2050s and then increase to 5.3%by the 2090s.Under RCP4.5-SSP2,RCP8.5-SSP3,and RCP8.5-SSP5,the higher inte-grated suitability area is projected to continuously decrease from 30.3%,30.6%and 30.6%in the 2010s to 4.1%,4.4%and 4.4%in the 2090s,respectively.By the 2090s,41,138 and 277 existing ski areas are projected to be closed under RCP2.6-SSP1,RCP4.5-SSP2,and RCP8.5-SSP3/RCP8.5-SSP5,respectively.It is clear that emission pathways and climate change adaptation and mitigation strategies will greatly shape thedevelopment of China's regional ski tourism. | DENG Jie CHE Tao JIANG Tong DAI Li-Yun | 2021 | Advances in Climate Change Research2021,12,2: | 5 |

| 10 | Expression of S100B during the innate immune of corneal epithelium against fungi invasion显示文摘AIM: To explore the expression of S100 B in corneal epithelial cells under Aspergillus stimulation both in vivo and in vitro.·METHODS: Immortalized human corneal epithelial cells(HCECs) were exposed to inactive Aspergillus fumigatus(A. fumigatus) conidia at 0, 4, 8, 12, 16, and 24 h respectively. The corneas of Wistar rats were exposed to active A. fumigatus at 0, 12, 24, 48 h and the normal rat corneas were used for normal control. The m RNA level of S100 B was evaluated by real time quantitative reverse transcription-polymerase chain reaction(q RT-PCR).S100 B protein expression in cornea epithelium was detected by immunohistochemical/immunocytochemical staining(IHC/ICC).· RESULTS: Histopathology revealed a significant inflammatory cell infiltration in fungal keratitis human and rat cornea. Corneal epithelial cells didn't express or rarely express S100 B at baseline. A. fumigatus significantly induced S100 B m RNA expression in cultured corneal epithelial cells in a time depended manner in vitro, the m RNA began to rise significantly at8 h in vitro(P <0.05) and continue to rise as time prolonged(P <0.01). In vivo, S100 B m RNA level was low in the normal corneas. However, it was increased in keratitis corneas from 12 h after infection(P <0.05) and reached to a peak at 24h(P <0.001). | 13 | Expression of indoleamine 2,3-dioxygenase in a murine model of Aspergillus fumigatus keratitis显示文摘AIM: To observe the presence and expression of indoleamine 2,3-dioxygenase(IDO) during the corneal immunity to Aspergillus fumigatus(A. fumigatus) in the murine models.·METHODS: The murine model of fungal keratitis was established by smearing with colonies of A. fumigatus after scraping central epithelium of cornea and covering with contact lenses in C57BL/6 mice. The mice were randomly divided into control group, sham group and A.fumigatus keratitis group. The cornea was monitored daily using a slit lamp and recorded disease score after infection. Corneal lesion was detected by immunofluorescence staining. IDO m RNA and protein were also detected by quantitative reverse transcription-polymerase chain reaction(q RT-PCR) and Western blot.· RESULTS: The disease score and slit lamp photography indicated that disease severity was consistent with corneal inflammation in the murine models, and the disease scores in A. fumigatus keratitis group were obviously higher than those in the sham group. By immunofluorescence staining, IDO was mainly localized in corneal epithelium and stroma in the murine corneal tissues with A. fumigatus keratitis. Compared with the sham group, IDO m RNA expression was significantly enhanced in corneal epithelium infected by A. fumigatus. Furthermore, IDO protein expression detected by Western blot was in accord with transcript levels of IDO m RNA measured by q RT-PCR. | 15 | Hierarchical coupling effect in hollow Ni/NiFe2O4-CNTs microsphere via spray-drying for enhanced oxygen evolution electrocatalysis显示文摘Design and fabrication of cost-effective transition metal and their oxides-based nanocomposites are of paramount significance for metal-air batteries and water-splitting.However,the traditional optimized designs for nanostructure are complicated,low-efficient and underperform for wide-scale applications.Herein,a novel hierarchical framework of hollow Ni/NiFe2O4-CNTs compositemicrosphere forcibly-assembled by zero-dimensional(OD)Ni/NiFo204 nanoparticle(<16 nm)and one-dimensional(1D)self-supporting CNTs was fabricated successfully.Benefitted from the unique nanostructure,such monohybrids can achieve remarkable oxygen evolution reaction(OER)performance in alkaline media with a low overpotential and superior durability,which exceeds most of the commercial catalysts based on IrO/RuO2 or other non-noble metal nanomaterials.The enhanced OER performance of Ni/NiFe2OA-CNTs composite is mainly ascribed to the increased catalytic activity and the optimized conductivity induced by the effects of strong hierarchical coupling and charge transfers between CNTs and Ni/NiFe204 nanoparticles.These effects are greatly boosted by the polarized heterojunction interfaces confirmed by electron holography.The density functional theory(DFT)calculation indicates the epitaxial Ni further enriches the intrinsic electrons contents of NiFe204 and thus accelerates absorption/desorption kinetics of OER intermediates.This work hereby paves a facile route to construct the hollow composite microsphere with excellent OER electrocatalytic activity based on non-noble metal oxide/CNTs. | Xuefeng Yu Guanyu Chen Yizhe Wang Jiwei Liu Ke Pei Yunhao Zhao Wenbin You Lei Wang Jie Zhang Linshen Xing Jingjun Ding Guangzhou Ding Min Wang Renchao Che | 2020 | Nano Research2020,13,2: | 3 |

| 17 | ASB3 knockdown promotes mitochondrial apoptosis via activating the interdependent cleavage of Beclin1 and caspase-8 in hepatocellular carcinoma显示文摘Apoptosis and autophagy are distinct cellular processes that can be highly interconnected. The cross talk between the two processes is indispensable in determining the overall cell fate. Although the apoptosis-promoting effect of caspases has been demonstrated, the roles of autophagy-related proteins and even autophagy itself in regulating apoptosis remain poorly understood. In our present study, we found that downregulation of ubiquitin E3 ligase ASB3 led to enhanced mitochondrial apoptosis as well as autophagy, which synergistically promoted cell death in hepatocellular carcinoma(HCC). We observed the activation of caspase-8 and decrease of autophagy protein Beclin1 in apoptotic cells that were depleted of ASB3. Beclin1 was mainly cleaved by activated caspase-8 and active Beclin1 initiated mitochondrial apoptosis via locating its C-terminal fragment to mitochondria. In addition, knocking down of Beclin1 markedly blocked the apoptosis, indicating its essential role in the process.Notably, our study indicated that enhanced autophagy level might be involved in the activation of caspase-8 and promote the apoptosis. Taken together, our results demonstrated that ASB3 can regulate mitochondrial pathway of apoptosis by controlling caspase-8 mediated cleavage of Beclin1 in HCC. | 18 | Mincle in the innate immune response of mice fungal keratitis显示文摘AIM: To investigate how macrophage inducible C-type lectin(Mincle) influences inflammation in mice fungal keratitis induced by Aspergillus fumigatus(A. fumigatus).METHODS: C57 BL/6 mice were infected with A. fumigatus after pretreated with Mincle agonist TDB or Mincle neutralizing antibody(MincleAb), taking DMSO or IgG as control group respectively. The cornea lesions were monitored with slit-lamp microscope and evaluated by clinical score. Mincle expression was assessed using reverse transcriptionploymerase chain reaction(RT-PCR) and immunostaining. The expression of cytokines(IL-1β, TNF-α and IL-6) chemokines(CXCL-1 and MIP-2) was determined by RTPCR and ELISA. Neutrophil infiltration was observed by immunostaining. The levels of nitric oxide(NO) generated by corneas were tested by Griess reaction. RESULTS: Mincle mRNA and protein levels were higher in infected corneas than normal corneas of C57 BL/6 mice, saving clinical scores revealed differences. When pretreated with Mincle agonist TDB, the mRNA and protein levels of IL-1β, TNF-α and IL-6 in infected corneas were significantly increased compared with the control group(P<0.01). Results of the counterpart in corneas pretreated with Mincle neutralizing antibody was decreased consistently(P<0.01). Expression of CXCL1 and MIP-2 mRNA levels were upregulated in TDB group and down-regulated in Mincle Ab group(P<0.01), coincide with neutrophil aggregation degree in corneas showed by immunostaining. As for the concentration of NO, it was promoted in TDB group compared with DMSO control group, and decreased in Mincle Ab group compared with Ig G control group.CONCLUSION: Mincle plays a dual role in mice fungal keratitis. It participates in the innate immune system by enhancing inflammation. | 19 | Strain heterogeneity,cooccurrence network,taxonomic composition and functional profile of the healthy ocular surface microbiome显示文摘Background:There is growing evidence indicating that the microbial communities that dwell on the human ocular surface are crucially important for ocular surface health and disease.Little is known about interspecies interactions,functional profiles,and strain heterogeneity across individuals in healthy ocular surface microbiomes.Methods:To comprehensively characterize the strain heterogeneity,cooccurrence network,taxonomic composition and functional profile of the healthy ocular surface microbiome,we performed shotgun metagenomics sequencing on ocular surface mucosal membrane swabs of 17 healthy volunteers.Results:The healthy ocular surface microbiome was classified into 12 phyla,70 genera,and 140 species.The number of species in each healthy ocular surface microbiome ranged from 6 to 47,indicating differences in microbial diversity among individuals.The species with high relative abundances and high positivity rates were Streptococcus pyogenes,Staphylococcus epidermidis,Propionibacterium acnes,Corynebacterium accolens,and Enhydrobacter aerosaccus.A correlation network analysis revealed a competitive interaction of Staphylococcus epidermidis with Streptococcus pyogenes in ocular surface microbial ecosystems.Staphylococcus epidermidis and Streptococcus pyogenes revealed phylogenetic diversity among different individuals.At the functional level,the pathways related to transcription were the most abundant.We also found that there were abundant lipid and amino acid metabolism pathways in the healthy ocular surface microbiome.Conclusion:This study explored the strain heterogeneity,cooccurrence network,taxonomic composition,and functional profile of the healthy ocular surface microbiome.These findings have important significance for the future development of probiotic-based eye therapeutic drugs. | Yutong Kang Shudan Lin Xueli Ma Yanlin Che Yiju Chen Tian Wan Die Zhang Jiao Shao Jie Xu Yi Xu Yongliang Lou Meiqin Zheng | 2021 | Eye and Vision2021,8,1: | 3 |
