| 1 | Construction of a regulable gene therapy vector targeting for hepatocellular carcinoma显示文摘AIM: To construct a gene modified hepatocellular carcinoma (HCC) specific EGFP expression vector regulated by abbreviated cis-acting element of AFP gene.METHODS: The minimal essential DNA segments of AFP gene enhancer and promoter were synthesized through PCR from Genome DNA of HepG2 cells. Gene fragments were then cloned into the multiple cloning site of non-promoter EGFP vector pEGFP-t. Recombinant plasmid was transferred into positive or negative AFP cell lines by means of lipofectamine. The expression of EGFP was tested by fluorescence microscope and flow cytometry. The effect of all-trans retinoic acid (ATRA) on the expression of EGFP was tested in different concentrations.RESULTS: By the methods of restriction digestion and sequence analyses we confirmed that the length, position and orientation of inserted genes of cis-acting element of AFP were all correct. The transcription of EGFP was under the control of AFP cis-acting element. The expressing EGFP can only been detected in AFP producing hepatoma cells.The expression rate of EGFP in G418 screened cell line was 34.9±4.1%. 48 h after adding 1×10-7M retinoic acid, EGFP expression rate was 14.7±3.5%. The activity of AFP gene promoter was significantly suppressed by addition of 1×10-7M retinoic acid (P<0.05, P=0.003, t=6.488).CONCLUSION: This recombinant expression vector can be used as a gene therapy vector for HCC. | 3 | Inhibitory effect of human telomerase antisense oligodeoxyribonucleotides on the growth of gastric cancer cell lines in variant tumor pathological subtype显示文摘AIM: To investigate the inhibitory effect of specialized human telomerase antisense oligodeoxyribonucleotides on the growth of well (MKN-28), moderately (SGC-7901)and poorly (MKN-45) differentiated gastric cancer cell lines under specific conditions and its inhibition mechanism,and to observe the correlation between the growth inhibition ratio and the tumor pathologic subtype of gastric cancer cells.METHODS: Telomerase activity in three gastric cancer cell lines of variant tumor pathologic subtype was determined by modified TRAP assay before and after the specialized human telomerase antisense oligodeoxyribonucleotides were dealt with under specific conditions. Effect of antisense oligomer under specific conditions of the growth and viability of gastric cancer cell lines was explored by using trypan blue dye exclusion assay, and cell apoptosis was detected by cell morphology observation, flow cytometry and TUNEL assay.RESULTS: Telomerase activity was detected in well,moderately and poorly differentiated gastric cancer cell lines (the quantification expression of telomerase activity was 43.7TPG, 56.5TPG, 76.7TPG, respectively).Telomerase activity was controlled to 30.2TPG, 36.3TPG and 35.2TPG for MKN-28, SGC-7901 and MKN-45 cell lines respectively after treatment with human telomerase antisense oligomers at the concentration of 5 μmol/L, and was entirely inhibited at 10 μmol/L, against the template region of telomerase RNA component, whereas no inhibition effect was detected in missense oligomers (P<0.05). After treatment with antisense oligomers at different concentrations under specific conditions for 96 h, significant growth inhibition effects were found in MKN-45 and SGC-7901gastric cancer cell lines (the inhibition ratio was 40.89%and 71.28%), but not in MKN-28 cell lines (15.86%). The ratio of inactive SGC-7901 cells increased according to the prolongation of treatment from 48 to 96 h. Missense oligomers could not lead to the same effect (P<0.05).Apoptosis of SGC-7901 and MKN-45 cells was detected not only by morphology and TUNEL assay but also by flow cytometry. The apoptotic rate reached 33.56% for SGC-7901 cells and 44.75% for MKN-45 cells.CONCLUSION: The viability and proliferation of gastric cancer cells can be inhibited by antisense telomerase oligomers. The growth inhibition of gastric cancer cells is correlated with concentration, time and sequence specialty of antisense oligomers. | 5 | Clinical significance of serum IGF-I,IGF-II and IGFBP-3 in liver cirrhosis显示文摘AIM: To investigate the relationship between insulin-like growth factor-Ⅰ, -Ⅱ (IGF-Ⅰ and IGF-Ⅱ), IGF-binding protein 3(IGFBP-3) and Child-Pugh score in patients with liver cirrhosis,and to search for potential clinical markers of liver function.METHODS: Forty-four patients with advanced liver cirrhosis of viral origin were divided into 3 groups according to severity of cirrhosis (Child-Pugh score) and 38 healthy subjects served as controls. Serum levels of IGF-Ⅰ, IGF-Ⅱ and IGFBP-3 were measured by immunoradiometric assay.RESULTS: Serum IGF-Ⅰ, IGF-Ⅱ and IGFBP-3 levels were significantly lower in patients with cirrhosis than in controls,and serum concentrations of IGF-Ⅰ, IGF-Ⅱ and IGFBP-3 were associated with the severity of liver dysfunction, and dropped sharply during the progression of liver failure. Among these 3 parameters, serum IGF-Ⅱ was the most sensitive and effective indicator for liver dysfunction. Concentrations of IGF-Ⅰ <30 ng/mL, IGF-Ⅱ <200 ng/mL and IGFBP-3 <6 ng/mL implied a negative prognosis for patients with liver cirrhosis.CONCLUSION: Serum IGF-Ⅰ, IGF-Ⅱ and IGFBP-3 may provide a new dimension in the assessment of liver dysfunction.Combined detection of serum IGF-Ⅰ, IGF-Ⅱ and IGFBP-3 with Child-Pugh score is more effective in predicting prognosis than Child-Pugh score alone. | Yun-LinWu JingYe ShuZhang JieZhong Rong-PingXi | 2004 | World Journal of Gastroenterology2004,10,18: | 7 |
| 6 | Radiation induced graft polymerization of multi-walled carbon nanotubes for superhydrophobic composite membrane preparation显示文摘Highly soluble multi-walled carbon nanotubes(MWNTs) were prepared by radiation-induced free radical graft polymerization of vinyl acetate(VAc) onto pristine MWNT surfaces. High resolution transmission electron microscopy(HR-TEM), Fourier transform infrared(FTIR) spectroscopy, and micro-Raman spectroscopy were used to confirm that poly(vinyl acetate)(PVAc) had been successfully grafted onto the surface of the MWNTs. The effects of experimental parameters on the degree of grafting(DG) of PVAc were also investigated, including adsorbed dose, dose rate, initial monomer concentration, and solvents. The grafted MWNTs(MWNTs-g-PVAc) exhibited good solubility in common organic solvents at high mass fraction. | 7 | Imaging algorithm for synthetic aperture interferometric radiometer in near field显示文摘With the benefits of digital IC technology development, the synthetic aperture interferometric radiometer (SAIR) technique is growing fast and expanding to more and more application areas. The near field imaging detection is a potential application which has received increasing demand recently. Because the Fourier imaging theory of the traditional SAIR is based on far-field approximation, it will be invalid for near-field condition. This paper is devoted to establishing a new accurate imaging algorithm for near-field SAIR imaging. Firstly, the visibility function in near field is deduced and the relationship of which to far-field visibility function is analyzed. Then, a numerical method based on pseudo inverse and focal plane approximation is developed. The effectivity of this method is tested with imaging simulation of point source and extended source, and the superiority is also demonstrated by comparing with the existing phase-modified Fourier transform method. The selective binding of OTA with mercury ions is confirmed by fluorescence in situ hybridization (FISH). | 13 | Molecular architecture of mouse and human pancreatic zymogen granules: protein components and their copy numbers显示文摘A molecular model of pancreatic zymogen granule (ZG) is critical for understanding its functions. We haveextensively characterized the composition and membrane topology of rat ZG proteins. In this study, wereport the development of targeted proteomics approaches to quantify representative mouse and humanZG proteins using LC-SRM and heavy isotope-labeled synthetic peptides. The absolute quantities of mouseRab3D and VAMP8 were determined as 1242 ± 218 and 2039±151 (mean ± SEM) copies per ZG. Thesize distribution and the averaged diameter of ZGs 750 ±23 nm (mean ± SEM) were determined byatomic force microscopy. The absolute quantification of Rab3 D was then validated using semi-quantitativeWestern blotting with purified GST-Rab3D proteins as an internal standard. To extend our proteomicsanalysis to human pancreas, ZGs were purified using human acini obtained from pancreatic islet trans-plantation center. One hundred and eighty human ZG proteins were identified for the first time includingboth the membrane and the content proteins. Furthermore, the copy number per ZG of human Rab3 D andVAMP8 were determined to be 1182 ± 45 and 485 ± 15 (mean ± SEM). The comprehensive proteomicanalyses of mouse and human pancreatic ZGs have the potential to identify species-specific ZG proteins.The determination of protein copy numbers on pancreatic ZGs represents a significant advance towardsbuilding a quantitative molecular model of a prototypical secretory vesicle using targeted proteomicsapproaches. | 18 | Comparative study on traumatic effect and L-selectin change made by two blooddrawing modes in rat显示文摘Objective: To compare carotid catheters blooddrawing and tail-cut blooddrawing in traumatizing rats and changing their internal L-selectin. Methods: Two ways of bloodletting were selected to produce hemorrhagic shock models. Monoclonal antibody sign and flow cytometer were used to test neutrophil L-selectin dynamic expression in rats. Results: No remarkable differences were shown among different time points in neutrophil L-selectin expressing amount (average fluorescence index)(P>0.05). Compared with the normal control group, all cell surface expressions in empirical control group increased. Cell surface expressions reached the summit at 3 hours and kept the high level at 4-5 hours. A progressive increase of the mRNA expression peaked at 5 hours; the expressions in both groups of carotid catheters blooddrawing increased compared with normal control group, and remained stable after 3 hours compared with the Tail-cut Blooddrawing Group. Conclusion: L-selectin expressions changed little in tail-cut blooddrawing rats, while carotid catheters resulted moderate or sever trauma. Therefore, it was suggested that it should not be set as hemorrhagic shock control model. As neutrophil L-selection expressions increased after trauma, the adhesion of leucocytes to the venule wall may be beneficial to the healing of wound and enhance the ability of anti-infection.
