| 1 | Microstructure and properties of Al_2O_3-13%TiO_2 coatings sprayed using nanostructured powders显示文摘The microstructure and wear performance of Al2O3-13% TiO2 coatings prepared by plasma spraying of agglom- erated nanoparticle powders were investigated. SEM analysis showed that the as-sprayed Al2O3-TiO2 coatings comprise of two kinds of typical region: fully melted region and unmelted/partially melted nanostructured region, which is different than the conventional coating with lamellar structure. It is shown that the microhardness of the nanostructured coatings was about 15%-30% higher than that of the conventional coating and the wear resistance is significantly improved, especially under a high wear load. | 3 | Molecular diversity of arbuscular mycorrhizal fungi at a large-scale antimony mining area in southern China显示文摘Arbuscular mycorrhizal fungi(AMF) have great potential for assisting heavy metal hyperaccumulators in the remediation of contaminated soils. However, little information is available about the community composition of AMF under natural conditions in soils contaminated by antimony(Sb). The objective of this study was to investigate the characteristics of AMF molecular diversity, and to explore the effects of Sb content and soil properties on the AMF community structure in an Sb mining area. Four Sb mine spoils and one adjacent reference area were selected from around the Xikuangshan mine in southern China. The association of AMF molecular diversity and community composition with the rhizosphere soils of the dominant plant species was studied by Polymerase Chain Reaction-Denaturing Gradient Gel Electrophoresis(PCR-DGGE). Results from all five studied sites showed that the diversity of AMF decreased with increasing Sb concentration. Principal component analysis(PCA) indicated that the AMF community structure was markedly different among these groups. Further redundancy analysis(RDA) showed that Sb contamination was the dominating factor influencing the AMF community structure in the Sb mine area. However, the multivariate analysis showed that, apart from the soil Sb content, extractable nitrogen content and organic matter content also attributed to AMF sequence distribution type. Some AMF sequences were only found in the highly contaminated area and these might be ideal candidates for improving phytoremediation efficiency in Sb mining regions. | 8 | Mining of a streptothricin gene cluster from Streptomyces sp.TP-A0356 genome via heterologous expression显示文摘Streptothricins (STs) are used commercially to treat bacterial and fungal diseases in agriculture. Mining of the sequenced microbial genomes uncovered two cryptic ST clusters from Streptomyces sp. C and Streptomyces sp. TP-A0356. The ST cluster from S. sp. TP-A0356 was verified by successful heterologous expression in Streptomyces coelicolor M145. Two new ST analogs were produced together with streptothricin F and streptothricin D in the heterologous host. The ST cluster was further confirmed by inactivation of gene stnO, which was proposed encoding an aminomutase supplying -lysines for the poly-β-Lys chain formation. | 11 | Identifying EGFR-Expressed Cells and Detecting EGFR Multi-Mutations at Single-Cell Level by Microfluidic Chip显示文摘EGFR mutations companion diagnostics have been proved to be crucial for the efficacy of tyrosine kinase inhibitor targeted cancer therapies. To uncover multiple mutations occurred in minority of EGFR-mutated cells,which may be covered by the noises from majority of unmutated cells, is currently becoming an urgent clinical requirement. Here we present the validation of a microfluidic-chip-based method for detecting EGFR multimutations at single-cell level. By trapping and immunofluorescently imaging single cells in specifically designed silicon microwells, the EGFR-expressed cellswere easily identified. By in situ lysing single cells, the cell lysates of EGFR-expressed cells were retrieved without cross-contamination. Benefited from excluding the noise from cells without EGFR expression, the simple and cost-effective Sanger's sequencing, but not the expensive deep sequencing of the whole cell population, was used to discover multi-mutations. We verified the new method with precisely discovering three most important EGFR drugrelated mutations from a sample in which EGFR-mutated cells only account for a small percentage of whole cell population. The microfluidic chip is capable of discovering not only the existence of specific EGFR multi-mutations,but also other valuable single-cell-level information: on which specific cells the mutations occurred, or whether different mutations coexist on the same cells. | 12 | Development and analysis of a novel AF11-2 aptamer capable of enhancing the fluorescence of aflatoxin B1显示文摘Aflatoxin B1(AFB1)is one of the most common mycotoxins that threatens human health.As singlestranded oligonucleotides with high affinity and specificity,aptamers have incomparable effect on the targeted detection of AFB1.Herein,after 11 rounds of selection and analysis using a modified affinity chromatography-based SELEX strategy,the truncated 37 nt aptamer AF11-2 was successfully obtained.The aptamer shows good detection performance for AFB1,and can sensitively detect AFB1 in the range of 100-1000 nmol/L,with a detection limit of 42 nmol/L.In the detection of pretreated edible peanut oil samples,AF11-2 aptamer also showed a high recovery rate and good stability for AFB1,and achieved satisfactory results.In addition,AF11-2 aptamer can significantly enhance the fluorescence ability of AFB1,which is not available in traditional Afla17-2-3 aptamer.After molecular docking analysis,it was found that AF11-2 and Afla17-2-3 had different nucleotide binding sites for AFB1.Afla17-2-3 binds to the carbonyl O of AFB1,while AF11-2 binds to the pyrrolic O of AFB1,which may be the main reason that AF11-2 can enhance the fluorescence of AFB1. | Wenjing Li Yian Pei Jine Wang | 2022 | Chinese Chemical Letters2022,33,8: | 2 |

| 19 | Critical involvement of lysyl oxidase in seizure-induced neuronal damage through ERK-Alox5-dependent ferroptosis and its therapeutic implications显示文摘Recent insights collectively suggest the important roles of lysyl oxidase(LysOX)in the pathological processes of several acute and chronic neurological diseases,but the molecular regulatory mechanisms remain elusive.Herein,we explore the regulatory role of LysOX in the seizure-induced ferroptotic cell death of neurons.Mechanistically,LysOX promotes ferroptosis-associated lipid peroxidation in neurons via activating extracellular regulated protein kinase(ERK)-dependent 5-lipoxygenase(Alox5)signaling.In addition,overexpression of LysOX via adeno-associated viral vector(AAV)-based gene transfer enhances ferroptosis sensitivity and aggravates seizure-induced hippocampal damage.Our studies show that pharmacological inhibition of LysOX withβ-aminopropionitrile(BAPN)significantly blocks seizure-induced ferroptosis and thereby alleviates neuronal damage,while the BAPN-associated cardiotoxicity and neurotoxicity could further be reduced through encapsulation with bioresponsive amorphous calcium carbonate-based nanocarriers.These findings unveil a previously unrecognized LysOX-ERK-Alox5 pathway for ferroptosis regulation during seizure-induced neuronal damage.Suppressing this pathway may yield therapeutic implications for restoring seizure-induced neuronal injury. | Xiaoyuan Mao Xuan Wang Mingzhu Jin Qin Li Jining Jia Menghuan Li Honghao Zhou Zhaoqian Liu Weilin Jin Yanli Zhao Zhong Luo | 2022 | Acta Pharmaceutica Sinica B2022,12,9: | 1 |
| 20 | Molecular mechanism of mureidomycin biosynthesis activated by introduction of an exogenous regulatory gene ssa A into Streptomyces roseosporus显示文摘Mureidomycins(MRDs), a group of unique uridyl-peptide antibiotics, exhibit antibacterial activity against the highly refractory pathogen Pseudomonas aeruginosa. Our previous study showed that the cryptic MRD biosynthetic gene cluster(BGC) mrd in Streptomyces roseosporus NRRL 15998 could not be activated by its endogenous regulator 02995 but activated by an exogenous activator Ssa A from sansanmycin’s BGC ssa of Streptomyces sp. strain SS. Here we report the molecular mechanism for this inexplicable regulation. EMSAs and footprinting experiments revealed that Ssa A could directly bind to a 14-nt palindrome sequence of 5′-CTGRCNNNNGTCAG-3′ within six promoter regions of mrd. Disruption of three representative target genes(SSGG-02981, SSGG-02987 and SSGG-02994) showed that the target genes directly controlled by Ssa Awere essential for MRD production. The regulatory function was further investigated by replacing six regions of SSGG-02995 with those of ssa A.Surprisingly, only the replacement of 343–450 nt fragment encoding the 115–150 amino acids(AA) of Ssa A could activate MRD biosynthesis.
