2The altered PD-1/PD-L1 pathway delivers the ‘one-two punch’ effects to promote the Treg/Th17 imbalance in pre-eclampsia显示文摘The programmed cell death-1(PD-1)/PD-ligand 1(PD-L1)pathway is critical for normal pregnancy by promoting regulatory T(Treg)cell development and inhibiting the Th17 response.However,the relationship between the PD-1/PD-L1 pathway and the Treg/Th17 imbalance in pre-eclampsia(PE)is an enigma.In this study,decreased PD-1 and PD-L1 expression and a Treg/Th17 imbalance were observed at the maternal-fetal interface in PE.The regulatory effects of the PD-1/PD-L1 pathway on the Treg and Th17 cell quantities were determined in vitro by targeting T-cell proliferation,differentiation and transdifferentiation.First,decreased PD-1 expression might contribute to a higher Th17 cell frequency by promoting proliferation in PE.Second,the percentages of Treg but not Th17 cells differentiated from peripheral naive CD4+T cells were increased by PD-L1 Fc administration.This effect was accompanied by decreased PI3K/AKT/m-TOR and increased PTEN mRNA expression and was completely reversed by PD-1 blockade.Finally,the percentage of IL-17-producing Treg cells increased and was positively associated with the Th17 cell frequency in PE.Increased RORγt and IL-17 but not Foxp3 and IL-10 mRNA expression by Treg cells was observed with PD-1 blockade.Similar findings occurred when Treg cells were exposed to IL-6/IL-23/IL-1βand were reversed by PD-L1 Fc.Taken together,our findings indicate that the PD-1/PD-L1 pathway contributes to the Treg/Th17 imbalance via‘one-two punch’approaches:(i)promoting Th17 cell proliferation,(ii)inhibiting Treg cell differentiation and(iii)enhancing Treg cell plasticity into Th17 cells in PE.The therapeutic value of PD-L1 Fc for PE treatment will be explored in the future.Yonghong Zhang Zhaozhao Liu Mei Tian Xiaohui Hu Liling Wang Jinlu Ji Aihua Liao 2018Cellular & Molecular Immunology2018,15,7:21
3The Zoulang Nanshan Caledonian Subduct ion Complex in the Northern Qilian Mountains and Its Dynamics显示文摘The front of the Zoulang Nanshan Caledonian volcanic island arc zone in the northern Qilian Mountains is a forearc accretionary terrane, composed of multiple accretionary volcanic island arcs, flysch accretionary wedges,high-pressure metamorphosed detachment zones and remnants of ophiolites. It resulted from the northeastward subduction of the Early Palaeozoic Qilan oceanic crust beneath the Alxa block. High-pressure metamorphism, which occurred during the subduction, progressed through three stages: the initial stage of medium T-high P,the main stage of temperature decrease and pressure increase, and the lag stage of pressure decrease and temperature increase. 4Phylogeny and Classifcation of Prunus sensu lato (Rosaceae)显示文摘The classifcation of the economically important genus Prunus L.sensu lato(s.l.)is controversial due to the high levels of convergent or the parallel evolution of morphological characters.In the present study,phylogenetic analyses of ffteen main segregates of Prunus s.l.represented by eighty-four species were conducted with maximum parsimony and Bayesian approaches using twelve chloroplast regions(atpB-rbcL,matK,ndhF,psbA-trnH,rbcL,rpL16,rpoC1,rps16,trnS-G,trnL,trnL-F and ycf1)and three nuclear genes(ITS,s6pdh and SbeI)to explore their infrageneric relationships.The results of these analyses were used to develop a new,phylogeny-based classifcation of Prunus s.l.Our phylogenetic reconstructions resolved three main clades of Prunus s.l.with strong supports.We adopted a broad-sensed genus,Prunus,and recognised three subgenera corresponding to the three main clades:subgenus Padus,subgenus Cerasus and subgenus Prunus.Seven sections of subgenus Prunus were recognised.The dwarf cherries,which were previously assigned to subgenus Cerasus,were included in this subgenus Prunus.One new section name,Prunus L.subgenus Prunus section Persicae(T.T.Yü&L.T.Lu)S.L.Zhou and one new species name,Prunus tianshanica(Pojarkov)S.Shi,were proposed.Shuo Shi Jinlu Li Jiahui Sun Jing Yu Shiliang Zhou 2013Journal of Integrative Plant Biology2013,55,11:18
5Down.regulation of E.cadherin enhances prostate cancer chemoresistance via Notch signaling显示文摘Background:The chemoresistance of prostate cancer(PCa)is invariably associated with the aggressiveness and metastasis of this disease.New emerging evidence indicates that the epithelial-to-mesenchymal transition(EMT)may play pivotal roles in the development of chemoresistance and metastasis.As a hallmark of EMT,E-cadherin is suggested to be a key marker in the development of chemoresistance.However,the molecular mechanisms underlying PCa chemoresistance remain unclear.The current study aimed to explore the association between EMT and chemoresistance in PCa as well as whether changing the expression of E-cadherin would affect PCa chemoresistance.Methods:Parental PC3 and DU145 cells and their chemoresistant PC3-Tx R and DU145-Tx R cells were analyzed.PC3-Tx R and DU145-Tx R cells were transfected with E-cadherin-expressing lentivirus to overexpress E-cadherin;PC3 and DU145 cells were transfected with small interfering RNA to silence E-cadherin.Changes of EMT phenotype-related markers and signaling pathways were assessed by Western blotting and quantitative real-time polymerase chain reaction.Tumor cell migration,invasion,and colony formation were then evaluated by wound healing,transwell,and colony formation assays,respectively.The drug sensitivity was evaluated using MTS assay.Results:Chemoresistant PC3-Tx R and DU145-Tx R cells exhibited an invasive and metastatic phenotype that associated with EMT,including the down-regulation of E-cadherin and up-regulation of Vimentin,Snail,and N-cadherin,comparing with that of parental PC3 and DU145 cells.When E-cadherin was overexpressed in PC3-Tx R and DU145-Tx R cells,the expression of Vimentin and Claudin-1 was down-regulated,and tumor cell migration and invasion were inhibited.In particular,the sensitivity to paclitaxel was reactivated in E-cadherin-overexpressing PC3-Tx R and DU145-Tx R cells.When E-cadherin expression was silenced in parental PC3 and DU145 cells,the expression of Vimentin and Snail was up-regulated,and,particularly,the sensitivity to paclitaxel was decreased.Interestingly,Notch-1 expression was up-regulated in PC3-Tx R and DU145-Tx R cells,whereas the E-cadherin expression was down-regulated in these cells comparing with their parental cells.The use ofγ-secretase inhibitor,a Notch signaling pathway inhibitor,significantly increased the sensitivity of chemoresistant cells to paclitaxel.Conclusion:The down-regulation of E-cadherin enhances PCa chemoresistance via Notch signaling,and inhibiting the Notch signaling pathway may reverse PCa chemoresistance.Wenchu Wang Lihui Wang Atsushi Mizokami Junlin Shi Chunlin Zou Jinlu Dai Evan T. 6Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 7The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 8Immune mediators in the tumor microenvironment of prostate cancer显示文摘Prostate cancer tissue is composed of both cancer cells and host cells.The milieu of host components that compose the tumor is termed the tumor microenvironment(TME).Host cells can be those derived from the tissue in which the tumor originates(e.g.,fibroblasts and endothelial cells)or those recruited,through chemotactic or other factors,to the tumor(e.g.,circulating immune cells).Some immune cells are key players in the TME and represent a large proportion of non-tumor cells found within the tumor.Immune cells can have both anti-tumor and pro-tumor activity.In addition,crosstalk between prostate cancer cells and immune cells affects immune cell functions.In this review,we focus on immune cells and cytokines that contribute to tumor progression.We discuss T-regulatory and T helper17 cells and macrophages as key modulators in prostate cancer progression.In addition,we discuss the roles of interleukin-6 and receptor activator of nuclear factor kappa-B ligand in modulating prostate cancer progression.This review highlights the concept that immune cells and cytokines offer a potentially promising target for prostate cancer therapy.Jinlu Dai Yi Lu Hernan Roca Jill M. 9A Robust Control Strategy to Improve Transient Stability for AC-DC Interconnected Power System with Wind Farms显示文摘In view of the variable parameters that affect the transient stability of electromagnetic torque and mechanical torque balance in AC-DC system,and the uncertainty of wind power in large-scale interconnection of wind farm.This paper proposes a linear parameter varying(LPV)robust feedback control method for transient stability of interconnected systems.The proposed LPV robust feedback control method uses the DC channel power control and the mechanical power in the interconnected system as the control target to improve the transient stability of the interconnected system with wind farm channel.Firstly,aiming at the strong nonlinear characteristics of the interconnected system,the power balance and the wind power output uncertainty in the transient process,the transient process is designed as a linear model of variable parameters.Then,the H∞robust output feedback controller is designed according to the LPV model.The transient stability control strategy topology and transfer function of the interconnected system are proposed.Finally,the proposed scheme is verified by an interconnected system formed by four equal-value grids through AC and DC lines in a digital simulation platform.The results show that the LPV robust feedback control model proposed in this paper has better response characteristics and transient stability control effects for interconnected systems with wind power weak sendingend system.Qian Hui Jinlu Yang Xia Yang Zhe Chen Yan Li Yun Teng 2019CSEE Journal of Power and Energy Systems2019,5,2:6
10The Medium Energy X-ray telescope (ME) onboard the Insight-HXMT astronomy satellite显示文摘The Medium Energy X-ray telescope(ME) is one of the three main telescopes on board the Insight hard X-ray modulation telescope(Insight-HXMT) astronomy satellite. ME contains 1728 pixels of Si-PIN detectors sensitive in 5-30 ke V with a total geometrical area of 952 cm^2. The application specific integrated circuit(ASIC) chip, VA32TA6, is used to achieve low power consumption and low readout noise. The collimators define three kinds of field of views(FOVs) for the telescope, 1°×4°, 4°×4°,and blocked ones. Combination of such FOVs can be used to estimate the in-orbit X-ray and particle background components.The energy resolution of ME is ~3 ke V at 17.8 ke V(FWHM) and the time resolution is 255 μs. 12Effect of chiropractic manipulation on disrupted epithelium barrier and its mechanism of specialized pro-resolving mediators in a spleen-deficiency murine model显示文摘OBJECTIVE: To investigate the influence of spleen deficiency on the epithelial barrier of jejunum and lungs in a rat model of spleen-deficiency and the effect and potential specialized pro-resolving mediators (SPMs) mechanism of chiropractic manipulation. METHODS: Three-week-old male Sprague-Dawley rats were divided randomly into normal control group (n = 6), spleen-deficiency group (n = 5) and chiropractic group (n = 6). Spleen-deficiency model was induced in spleen-deficiency group and chiropractic group. Moreover, chiropractic manipulation was performed in chiropractic group. Four weeks later, systemic Th1/Th2 balance was evaluated by the ratio of plasma interferon (IFN)-γ/interleukin (IL)-4 levels by enzyme-linked immunosorbent assay (ELISA). Epithelial barrier integrity were assessed by the observation of morphological changes by hematoxylin-eosin staining and zonula occludens (ZO)-1 gene expressions by quantitative real time polymerase chain reaction in jejunum and lungs. Plasma resolvin D1 (RvD1) and lipoxin A4 (LXA4) levels were measures by ELISA for endogenous SPMs production. The levels of docosahexaenoic acid (DHA) and arachidonic acid (AA) in jejunum and lungs were also measured by HPLC-MS/ MS. RESULTS: Comparing with normal control group, spleen-deficiency group showed disrupted mucosa in jejunum, inflammatory condition in lungs, significantly decreased ratio of plasma IFN-γ/IL-4 levels and lower expressions of ZO-1 mRNA in both jejunum and lung tissues. Comparing with spleen-deficiency group, chiropractic group had less disrupted mucosa in jejunum and inflammatory condition in lungs, significantly increased systemic ratio of IFN-γ/IL-4 and expressions of ZO-1 mRNA in both jejunum and lung tissues. Chiropractic group had significantly enhanced plasma levels of RvD1 and LXA4, but had no significantly higher levels of DHA and AA in jejunum and lungs when comparing with spleen-deficiency group. 13Contrasting patterns of river runoff and sea-ice melted water in the Canada Basin显示文摘The fractions of river runoff and sea-ice melted water in the Canada Basin in summer 2003 were determined by the salinity-18O system. The fraction of river runoff(fR) was high in the upper 50 m of the water column and decreased with depth and latitude. The signals of the river runoff were confined to water depths above 200 m. The total amount of river runoff in the Canada Basin was higher than that in other arctic seas, indicating that the Canada Basin is a main storage region for river runoff. The penetration depth of the sea-ice melted water was less than 50 m to the south of 78°N, while it was about 150 m to the north of 78°N. The total amount of sea-ice melted water was much higher to the north of 78°N than to the south of 78°N, indicating the sea-ice melted waters accumulated on the ice edge. 14Influence of particle composition on thorium scavenging in the marginal China seas显示文摘Thorium-234 and particle composition (organic matter, biogenic silica, carbonate and lithogenic component) were examined in the East China Sea (ECS) and the northern South China Sea (NSCS) in order to constrain the particle types scavenging thorium isotopes. Good positive correlations be-tween particulate organic matter (POM) or carbonate and thorium-234 in suspended particulate matter (SPM) indicates that POM and carbonate are efficient to scavenge thorium-234. No relation-ship between biogenic silica and thorium-234 suggests that geochemical behavior of thorium-234 may be not influenced by biogenic silica. A simple model was used to evaluate the affinity of thorium-234 to different particle components. The results show that POM is the most efficient scavenger for thorium-234 in the ECS and the NSCS, followed by carbonate. The authors’ re-sults lend support to the utility of thorium-234 as a proxy of POC and carbonate in the upper layer. 15Effect of the Heat-treatment Temperature on the Mechanical Properties and Microstructural Evolution of Cold-rolled Twinning-induced Plasticity Steel显示文摘TWIP steels with 70% cold-rolled reduction were heated at 500, 600, 700, 800, 900, 1000, and 1100 ℃. Then, the properties before and after heating were examined through tensile and hardness experiments. The microstructures were also analyzed by optical microscopy and transmission electron microscopy. The relationship between the properties and microstructure was examined as well. Finally, the evolution process of cold-rolled microstructures during heating was discussed in detail. 16Adult islets cultured in collagen gel transdifferentiate into duct-like cells显示文摘AIM: To establish a model of islet-ductal cell bansdifferentiation to identify the transdifferentiated cells. METHODS: Collagen was extracted from rat tail at first. Purified rat islets were divided into three groups, embedded in collagen gel and incubated respectively in DMEM/F12 alone (control group), DMEM/F12 plus epidermal growth factor (EGF), DMEM/F12 plus EGF and cholera toxin (CT). Transdifferentiation was proved by microscopy, RT-PCR, immunohistochemistry and RIA.RESULTS: Islets embedded in collagen gel plus EGF and CT were cystically transformed and could express new gene cytokeratin 19 while still maintaining the expression of insulin and Pdx-1 genes. Immunohistochemistry demonstrated that the protein of cytokeratin 19 was only expressed in the third group. 18Variation of freshwater components in the Canada Basin during 1967–2010显示文摘As a conservative tracer, oxygen isotopes in seawater are widely used for water mass analysis, along with temperature and salinity. In this study, seawater oxygen-18 datasets in the Canada Basin during 1967–2010 were obtained from the four cruises of the Chinese National Arctic Research Expedition(1999, 2003, 2008, and 2010) and the NASA database. Fractions of sea ice meltwater and river runoff were determined from the salinity-18O system. Our results showed that the river runoff decreased from the south to the north in the Canada Basin. The enhanced amount of river runoff observed in the southern Canada Basin may originate from the Mackenzie River, transported by the Beaufort Gyre. The river runoff component showed maximum fractions during 1967–1969, 1978–1979, 1984–1985, 1993–1994, and 2008–2010, indicating the refresh time of the river runoff was 5.0–16.0 a in the Canada Basin. 19A spatial attentive and temporal dilated(SATD)GCN for skeleton-based action recognition显示文摘Current studies have shown that the spatial-temporal graph convolutional network(STGCN)is effective for skeleton-based action recognition.However,for the existing STGCN-based methods,their temporal kernel size is usually fixed over all layers,which makes them cannot fully exploit the temporal dependency between discontinuous frames and different sequence lengths.Besides,most of these methods use average pooling to obtain global graph feature from vertex features,resulting in losing much fine-grained information for action classification.To address these issues,in this work,the authors propose a novel spatial attentive and temporal dilated graph convolutional network(SATD-GCN).It contains two important components,that is,a spatial attention pooling module(SAP)and a temporal dilated graph convolution module(TDGC).Specifically,the SAP module can select the human body joints which are beneficial for action recognition by a self-attention mechanism and alleviates the influence of data redundancy and noise.The TDGC module can effectively extract the temporal features at different time scales,which is useful to improve the temporal perception field and enhance the robustness of the model to different motion speed and sequence length.Importantly,both the SAP module and the TDGC module can be easily integrated into the ST-GCN-based models,and significantly improve their performance.Extensive experiments on two large-scale benchmark datasets,that is,NTU-RGB+D and Kinetics-Skeleton,demonstrate that the authors’method achieves the state-of-the-art performance for skeleton-based action recognition.Jiaxu Zhang Gaoxiang Ye Zhigang Tu Yongtao Qin Qianqing Qin Jinlu Zhang Jun Liu 2022CAAI Transactions on Intelligence Technology2022,7,1:3
20BHMT Gene Polymorphisms as Risk Factors for Cleft Lip and Cleft Palate in a Chinese Population显示文摘Objective Convincing evidence suggests a link between increased risk of nonsyndromic cleft lip with or without cleft palate (NSCL/P) and low intake of folic acid by the mother during pregnancy. The present study was designed to explore if genetic variation in the betaine‐homocysteine methyltransferase (BHMT) gene contributes to NSCL/P. Methods DNA was obtained from 166 individuals with NSCL/P and 285 healthy subjects. Three known single nucleotide polymorphisms (SNPs) present in the BHMT gene (rs651852, rs3797546, and rs3733890) were investigated by real‐time PCR‐based TaqMan genotyping. Results Neither allelic nor genotypic association was found between NSCL/P and SNPs rs651852 and rs3733890. SNP rs3797546 did not show allelic association with NSCL/P; however, a higher proportion of NSCL/P patients carry the CC genotype compared with the TT+CT genotype (P=0.020, OR=2.10, 95% CI=1.11‐3.95).
