1Clinical and biochemical indexes from 2019-nCoV infected patients linked to viral loads and lung injury显示文摘The outbreak of the 2019-nCoV infection began in December 2019 in Wuhan,Hubei province,and rapidly spread to many provinces in China as well as other countries.Here we report the epidemiological,clinical,laboratory,and radiological characteristics,as well as potential biomarkers for predicting disease severity in 2019-nCoV-infected patients in Shenzhen,China.All 12 cases of the 2019-nCoV-infected patients developed pneumonia and half of them developed acute respiratory distress syndrome (ARDS).The most common laboratory abnormalities were hypoalbuminemia,lymphopenia,decreased percentage of lymphocytes (LYM) and neutrophils (NEU),elevated C-reactive protein (CRP) and lactate dehydrogenase (LDH),and decreased CD8 count.The viral load of 2019-nCoV detected from patient respiratory tracts was positively linked to lung disease severity.ALB,LYM,LYM (%),LDH,NEU (%),and CRP were highly correlated to the acute lung injury.Age,viral load,lung injury score,and blood biochemistry indexes,albumin (ALB),CRP,LDH,LYM (%),LYM,and NEU (%),may be predictors of disease severity.Moreover,the Angiotensin Ⅱlevel in the plasma sample from 2019-nCoV infected patients was markedly elevated and linearly associated to viral load and lung injury.Our results suggest a number of potential diagnosis biomarkers and angiotensin receptor blocker (ARB) drugs for potential repurposing treatment of 2019-nCoV infection.Yingxia Liu Yang Yang Cong Zhang Fengming Huang Fuxiang Wang Jing Yuan Zhaoqin Wang Jinxiu Li Jianming Li Cheng Feng Zheng Zhang Lifei Wang Ling Peng Li Chen Yuhao Qin Dandan Zhao Shuguang Tan Lu Yin Jun Xu Congzhao Zhou Chengyu Jiang Lei Liu 2020Science China(Life Sciences)2020,63,3:157
2Elevated plasma levels of selective cytokines in COVID-19 patients reflect viral load and lung injury显示文摘A recent outbreak of pneumonia in Wuhan,China was found to be caused by a 2019 novel coronavirus(2019-nCoV or SARS-CoV-2 or HCoV-19).We previously reported the clinical features of 12 patients with2019-n Co V infections in Shenzhen,China.To further understand the pathogenesis of COVID-19 and find better ways to monitor and treat the disease caused by 2019-n Co V,we measured the levels of 48 cytokines in the blood plasma of those 12 COVID-19 patients.Thirty-eight out of the 48 measured cytokines in the plasma of 2019-n Co V-infected patients were significantly elevated compared to healthy individuals.Seventeen cytokines were linked to 2019-nCoV loads.Fifteen cytokines,namely M-CSF,IL-10,IFN-α2,IL-17,IL-4,IP-10,IL-7,IL-1 ra,G-CSF,IL-12,IFN-γ,IL-1α,IL-2,HGF and PDGF-BB,were strongly associated with the lung-injury Murray score and could be used to predict the disease severity of2019-n Co V infections by calculating the area under the curve of the receiver-operating characteristics.Our results suggest that 2019-nCoV infections trigger extensive changes in a wide array of cytokines,some of which could be potential biomarkers of disease severity of 2019-n Co V infections.These findings will likely improve our understanding of the immunopathologic mechanisms of this emerging disease.Our results also suggest that modulators of cytokine responses may play a therapeutic role in combating the disease once the functions of these elevated cytokines have been characterized.Yingxia Liu Cong Zhang Fengming Huang Yang Yang Fuxiang Wang Jing Yuan Zheng Zhang Yuhao Qin Xiaoyun Li Dandan Zhao Shunwang Li Shuguang Tan Zhaoqin Wang Jinxiu Li Chenguang Shen Jianming Li Ling Peng Weibo Wu Mengli Cao Li Xing Zhixiang Xu Li Chen Congzhao Zhou William J Liu Lei Liu Chengyu Jiang 2020National Science Review2020,7,6:6
3A nanocomposite competent to overcome cascade drug resistance in ovarian cancer via mitochondria dysfunction and NO gas synergistic therapy显示文摘Ovarian cancer(OC)is one of the most common and recurring malignancies in gynecology.Patients with relapsed OC always develop'cascade drug resistance'(CDR)under repeated chemotherapy,leading to subsequent failure of chemotherapy.To overcome this challenge,amphiphiles(P1)carrying a nitric oxide(NO)donor(Isosorbide 5-mononitrate,ISMN)and high-density disulfide are synthesized for encapsulatingmitochondria-targeted tetravalent platinum prodrug(TPt)to construct a nanocomposite(INP@TPt).Mechanism studies indicated that INP@TPt significantly inhibited drug-resistant cells by increasing cellular uptake and mitochondrial accumulation of platinum,depleting glutathione,and preventing apoptosis escape through generating highly toxic peroxynitrite anion(ONOO−).To better replicate the microenvironmental and histological characteristics of the drug resistant primary tumor,an OC patient-derived tumor xenograft(PDXOC)model in BALB/c nude mice was established.INP@TPt showed the best therapeutic effects in the PDXOC model.The corresponding tumor tissues contained high ONOO−levels,which were attributed to the simultaneous release of O_(2)^(·−)and NO in tumor tissues.Taken together,INP@TPtbased systematic strategy showed considerable potential and satisfactory biocompatibility in overcoming platinum CDR,providing practical applications for ovarian therapy.Min Zhong Peiqin Liang Zhenzhen Feng Xin Yang Guang Li Rui Sun Lijuan He Jinxiu Tan Yangpengcheng Xiao Zhiqiang Yu Muhua Yi Xuefeng Wang 2023Asian Journal of Pharmaceutical Sciences2023,18,6:0
4Research of 1,3-Dihydroxyacetone Production by Overexpressing Glycerol Transporter and Glycerol Dehydrogenase显示文摘1,3-Dihydroxyacetone (DHA), a natural ketose, is widely used in the chemical, cosmetic, and pharmaceutical industries. The current method for DHA production is Gluconobacter oxydans ( G. oxydans ) fermentation, but the high concentration of glycerol in the fermentation broth inhibits cells growth. To overcome this obstacle, in this study, we overexpressed the glycerol transporter (GlpFp) by the use of promoters P tufB , P gmr , P glp1 , and P glp2 in G. oxydans 621H. The results show that the glycerol tolerances of strains overexpressing G lpF were all much better than that of the control strain. The glycerol dehydrogenase gene (G dh) was overexpressed by the promoters P tufB and P gdh , which increased the DHA titer by 12.7% compared with that of the control group. When G lpF and Gdh genes were co-overexpressed in G. oxydans 621H, the OD600 value of the engineered strains all increased, but the DHA titers decreased in di erent degrees, as compared with strains that overexpressed only G dh .
