1Time range of Mesozoic tectonic regime inversion in eastern North China Block显示文摘An important tectonic inversion took place in eastern North China Block(NCB) during Mesozoic, which caused a great lithosphere thinning, reconstruction of basin-range series, powerful interaction between mantle and crust, a vast granitic intrusion and volcanism, and large-scale metallogenic explosion. The time range of the Mesozoic tectonic regime inversion in the eastern North China Block is one of the key issues to understand mechanism of tectonic regime inversion. Our updated results for recognizing the time range are mainly obtained from the following aspects: structural analyses along northern and southern margins of the NCB and within the NCB for revealing tectonic inversion from compression to extension and structural striking from ~EW to NNE; geothermic analyses of the eastern sedimental basins for a great change of thermal history and regime; basin analysis for basin inversion from compression to extension and basin migration from ~EW to NNE; petrological and geochemical studies of volcanic rocks and lowermost crust xenoliths for recognizing peak period of mantle upwelling and intense interaction between mantle and crust, and main metallogenic epoch. 2Clinical and biochemical indexes from 2019-nCoV infected patients linked to viral loads and lung injury显示文摘The outbreak of the 2019-nCoV infection began in December 2019 in Wuhan,Hubei province,and rapidly spread to many provinces in China as well as other countries.Here we report the epidemiological,clinical,laboratory,and radiological characteristics,as well as potential biomarkers for predicting disease severity in 2019-nCoV-infected patients in Shenzhen,China.All 12 cases of the 2019-nCoV-infected patients developed pneumonia and half of them developed acute respiratory distress syndrome (ARDS).The most common laboratory abnormalities were hypoalbuminemia,lymphopenia,decreased percentage of lymphocytes (LYM) and neutrophils (NEU),elevated C-reactive protein (CRP) and lactate dehydrogenase (LDH),and decreased CD8 count.The viral load of 2019-nCoV detected from patient respiratory tracts was positively linked to lung disease severity.ALB,LYM,LYM (%),LDH,NEU (%),and CRP were highly correlated to the acute lung injury.Age,viral load,lung injury score,and blood biochemistry indexes,albumin (ALB),CRP,LDH,LYM (%),LYM,and NEU (%),may be predictors of disease severity.Moreover,the Angiotensin Ⅱlevel in the plasma sample from 2019-nCoV infected patients was markedly elevated and linearly associated to viral load and lung injury.Our results suggest a number of potential diagnosis biomarkers and angiotensin receptor blocker (ARB) drugs for potential repurposing treatment of 2019-nCoV infection.Yingxia Liu Yang Yang Cong Zhang Fengming Huang Fuxiang Wang Jing Yuan Zhaoqin Wang Jinxiu Li Jianming Li Cheng Feng Zheng Zhang Lifei Wang Ling Peng Li Chen Yuhao Qin Dandan Zhao Shuguang Tan Lu Yin Jun Xu Congzhao Zhou Chengyu Jiang Lei Liu 2020Science China(Life Sciences)2020,63,3:157

5Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29
6Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 8Synthesis,characterization,and adsorption performance of Pb(Ⅱ)-imprinted polymer in nano-TiO_2 matrix显示文摘Surface ion-imprinted in combination with sol-gel process was applied to synthesis a new Pb(Ⅱ)-imprinted polymer for selective separation and enrichment of trace Pb(II) from aqueous solution. The prepared material was characterized by using the infrared spectra, X-ray diffractometer, and scanning electron microscopy. The batch experiments were conducted to study the optimal adsorption condition of adsorption trace Pb(Ⅱ) from aqueous solutions on Pb(Ⅱ)-imprinted polymer. The equilibrium was achieved in approximately 4.0 h, and the experimental kinetic data were fitted the pseudo second-order model better. The maximum adsorption capacity was 22.7 mg/g, and the Langmuir equation fitted the adsorption isotherm data. The results of selectivity experiment showed that selectively adsorbed rate of Pb(Ⅱ) on Pb(Ⅱ)-imprinted polymer was higher than all other studied ions. Desorption conditions of the adsorbed Pb(Ⅱ) from the Pb(Ⅱ)-imprinted polymer were also studied in batch experiments. The prepared Pb(Ⅱ)-imprinted polymer was shown to be promising for the separation and enrichment of trace Pb(Ⅱ) from water samples. 9Menthol-modified casein nanoparticles loading 10-hydroxycamptothecin for glioma targeting therapy显示文摘Chemotherapy outcomes for the treatment of glioma remains unsatisfactory due to the inefficient drug transport across the blood–brain barrier(BBB) and insufficient drug accumulation in the tumor region. Although many approaches, including various nanosystems, have been developed to promote the distribution of chemotherapeutics in the brain tumor, the delivery efficiency and the possible damage to the normal brain function still greatly restrict the clinical application of the nanocarriers.Therefore, it is urgent and necessary to discover more safe and effective BBB penetration and gliomatargeting strategies. In the present study, menthol, one of the strongest BBB penetration enhancers screened from traditional Chinese medicine, was conjugated to casein, a natural food protein with brain targeting capability. Then the conjugate self-assembled into the nanoparticles to load anti-cancer drugs.The nanoparticles were characterized to have appropriate size, spheroid shape and high loading drug capacity. Tumor spheroid penetration experiments demonstrated that penetration ability of mentholmodified casein nanoparticles(M-CA-NP) into the tumor were much deeper than that of unmodified nanoparticles. In vivo imaging further verified that M-CA-NPs exhibited higher brain tumor distribution than unmodified nanoparticles. The median survival time of glioma-bearing mice treated with HCPT-MCA-NPs was significantly prolonged than those treated with free HCPT or HCPT-CA-NPs. HE staining ofthe organs indicated the safety of the nanoparticles. 10Association of biochar properties with changes in soil bacterial,fungal and fauna communities and nutrient cycling processes显示文摘Soil microorganisms play crucial roles in soil nutrient cycling,carbon sequestration,fertility maintenance and crop health and production.To date,the responses of microorganisms,such as microbial activity,diversity,community structure and nutrient cycling processes,to biochar addition have been widely reported.However,the relationships between soil microbial groups(bacteria,fungi and microscopic fauna)and biochar physicochemical properties have not been summarized.In this review,we conclude that biochar affects soil microbial growth,diversity and community compositions by directly provid-ing growth promoters for soil biota or indirectly changing soil basic properties.The porous structure,labile C,high pH and electrochemical properties of biochar play an important role in determining soil microbial abundance and communities,and their mediated N and P cycling processes,while the effects and underlying mechanisms vary with biochar types that are affected by pyrolysis temperature and feedstock type.Finally,we highlight some issues related to research methodology and subjects that are still poorly understood or controversial,and the perspectives for further research in microbial responses to biochar addition.Zhongmin Dai Xinquan Xiong Hang Zhu Haojie Xu Peng Leng Jihui Li CTang Jianming Xu 2021Biochar2021,3,3:12

12Application of bioactive hydrogels combined with dental pulp stem cells for the repair of large gap peripheral nerve injuries显示文摘Due to the limitations in autogenous nerve grafting or Schwann cell transplantation,large gap peripheral nerve injuries require a bridging strategy supported by nerve conduit.Cell based therapies provide a novel treatment for peripheral nerve injuries.In this study,we first experimented an optimal scaffold material synthesis protocol,from where we selected the 10%GFD formula(10%GelMA hydrogel,recombinant human basic fibroblast growth factor and dental pulp stem cells(DPSCs))to fill a cellulose/soy protein isolate composite membrane(CSM)tube to construct a third generation of nerve regeneration conduit,CSM-GFD.Then this CSM-GFD conduit was applied to repair a 15-mm long defect of sciatic nerve in a rat model.After 12 week post implant surgery,at histologic level,we found CSM-GFD conduit could regenerate nerve tissue like neuron and Schwann like nerve cells and myelinated nerve fibers.At physical level,CSM-GFD achieved functional recovery assessed by a sciatic functional index study.In both levels,CSM-GFD performed like what gold standard,the nerve autograft,could do.Further,we unveiled that almost all newly formed nerve tissue at defect site was originated from the direct differentiation of exogeneous DPSCs in CSM-GFD.In conclusion,we claimed that this third-generation nerve regeneration conduit,CSM-GFD,could be a promising tissue engineering approach to replace the conventional nerve autograft to treat the large gap defect in peripheral nerve injuries.Lihua Luo Yan He Ling Jin Yanni Zhang Fernando P.Guastaldi Abdullkhaleg A.Albashari Fengting Hu Xiaoyan Wang Lei Wang Jian Xiao Lingli Li Jianming Wang Akon Higuchi Qingsong Ye 2021Bioactive Materials2021,6,3:11

15Drug targeting through platelet membrane-coated nanoparticles for the treatment of rheumatoid arthritis显示文摘The effective drug treatment of rheumatoid arthritis (RA) is hindered by poordelivery efficiency to the diseased site and the serious side effects caused bywide-spread drug distribution. Traditional drug-targeting strategies, such asligand modification, are complex, laborious, and inefficient. Inspired by theintrinsic relationship between platelets and RA, platelet-mimetic nanoparticles(PNPs) were developed for targeted drug delivery in RA. Through plateletreceptor-mediated adhesion, an intact platelet membrane was coated onto poly(lactic-co-glycolic add) nanopartides, endowing the resulting PNPs with variousfunctional receptors. By coating with platelet membranes, the nanoparticleswere stabilized and had a better circulation profile, providing a benefit forpassive targeting. In vitro binding of PNPs to inflamed endothelium, and in vivoaccumulation in joints of a collagen-induced arthritis (CIA) mouse model of RAwere significantly improved via P-selectin and GVPI recognition, indicating thatthe PNPs could effectively target to RA tissues through multiple mechanisms,similar to natural platelets. Moreover, FK506, a model drug, was loaded into thePNPs and used to treat RA. Pharmacodynamic studies demonstrated that theFK506-PNPs had a notable anti-arthritic effect in CIA mice. 16Detection of ALK translocation in non-small cell lung carcinoma(NSCLC) and its clinicopathological significance using the Ventana immunohistochemical staining method: a single-center large-scale investigation of 1,504 Chinese Han patients显示文摘Objective: The novel fully automated immunohistochemistry(IHC) assay-Ventana anaplastic lymphoma kinase(ALK)-D5F3 for screening ALK rearrangements has been approved by China's Food and Drug Administration in 2013, our previous study disclosed a highly specificity and sensitivity nearly 100%, and its efficacy needs to be evaluated in a large cohort of primary lung adenocarcinoma patients, and to compare clinicopathological features with ALK(+) and ALK(-) lung adenocarcinoma.Methods: A total of 1,504 consecutive surgical lung adenocarcinoma cases of Chinese Han population were collected and re-diagnosed according to the 2011 multidisciplinary classification of lung adenocarcinoma.Fully automated Ventana ALK-D5F3 IHC staining with a binary scoring was adopted to evaluate staining and correlated with clinicopathological characters, including age, sex, differentiation degree, histological subtype, lymph node metastasis, and clinical staging.ALK(+) patients were followed-up, and targeted therapy of ALKinhibitors was adopted and observed in patients with stage Ⅳ according to the NCCN guideline.Results: ALK positive adenocarcinomas were identified in 6.6% of the surgically resected 1,504 NSCLCs, and significantly younger than the negative group(P<0.05).Mucinous adenocarcinoma(28.2%) was determined to be predominant in ALK(+) cases, followed by the solid type(11.7%), specific type(6.8%), papillary type(5.6%), acinar type(5.5%), and lepidic type(3.1%), and the differences were statistically significant(χ~2=42.011, P<0.05).ALK(+) adenocarcinoma with lymph node metastasis(10.8%) were significantly higher than that without lymph node metastasis(4.5%)(χ~2=19.809, P<0.05); and ALK(+) in phase Ⅳ(20%) was significantly higher than phase Ⅲ(12.9%), phase Ⅱ(4.2%), phase Ⅰ(4.5%), and phase 0(0)(χ~2=36.068, P<0.05).Multivariate logistic regression disclosed that patient age, AJCC staging, and histological mucinous subtype were correlated with ALK positive staining(OR=0.959, 1.578, 5.036, respectively).Sixty eight patients had followed-up results, five patients out of which primarily diagnosed or progressed into Stage Ⅳ benefited well from targeted therapy with Crizotinib.Conclusions: The ALK fusion protein was seen in 6.6% Chinese NSCLC patients, and mostly seen in younger, clinically higher staging, mucinous and solid predominant adenocarcinoma.Clinical trials in patients of Stage Ⅳ confirmed that ALK-D5F3 Ventana IHC is serviceable in screening ALK-positive candidates for molecular targeted therapy.Lin Yang Yun Ling Lei Guo Di Ma Xuemin Xue Bingning Wang Junling Li Jianming Ying 2016Chinese Journal of Cancer Research2016,28,5:8
17Impending HRT wave precursors to the Wenchuan M_s8.0 earthquake and methods of earthquake impending prediction by using HRT wave显示文摘We deployed four geo-electric monitoring stations in Sichuan and Yunnan provinces from 2004, using the new generation of equipment (PS-100) and technologies to capture the HRT wave earthquake precursor. Before the Wenchuan Ms8.0 earthquake, we recorded the HRT wave precursor at the only operating station in Hongge (HG, Δ=465 km) and found that significant impending signal had been recorded at the station in the early morning ( 0―5 am) of 12th of May, 2008. The precursor for this earthquake is consistent with precursors recorded for other strong earthquakes. The measured physical properties (geo-resistivity and telluric-current) show tidal wave period oscillations from several days to several months before the earthquakes and the amplitude of such HT oscillation increases significantly towards the occurrence of an earthquake. These HT and RT waves from the epicenter have a causal relationship with the earthquakes that happened several days later. The arrival time of two RT waves is proportional to the distance from the station to the epicenter. The estimated natural decay of the amplitude is correlated with the natural period (T0) of the earthquake fault, which is proportional to the fault length. From this relationship, we can predict the earthquake magnitude. For magnitude 6―9 earthquakes, the natural period is about 1―6 hours. Such oscillation comes from the epicenter area and they can propagate several thousand kilometers in the Earth's crust. Before a strong earthquake in the shallow crust, the conductive pore fluid will experience major changes before the fault rapture. Such fluid change will emit an oscillation in the pore fluid pressure. This is the mechanism for the HRT wave generation. Since the China Earthquake Administration funded the HRT wave short-term earth-quake prediction project in 2003, the first record of HRT precursor wave has been recorded from the 2004-12-26 Sumatra Mw9.0 earthquake with the largest epicentre distance Δ=2900 km. Thereafter, we have captured HRT waves from more than twenty strong earthquakes, which are well-matched and show repeatability, consistency and regularity. All our observation with the HRT waves demonstrate that HRT wave precursors to earthquakes indeed exist. Strong earthquakes can be predicted and short-term and impending earthquake prediction is achievable in the very near future. 18Reinforcement effect of fiber and deoiled asphalt on high viscosity rubber/SBS modified asphalt mortar显示文摘In this paper we investigate the reinforcement mechanism of high viscosity rubber/SBS modified asphalt mortar mixed with fiber(mineral, lignin or carbon fiber) and deoiled asphalt(DOA). The softening point, penetration and viscosity tests were conducted to characterize the engineering properties of asphalt-fiber mortar. The microstructure of fiber was observed using scanning electron microscopy(SEM), and the results indicated that fiber can effectively improve the toughness of asphalt matrix through forming a spatial network structure, and then adhesion and stabilization of asphalt binder. The cone penetration test was designed to study the rheological property of fiber modified asphalt. The results indicated that the reinforcement effect increased with fibers and DOA fraction increasing to a certain threshold, and the optimal fiber content was dependent on the fiber type and its length. 19Measuring the resilience of an airport network显示文摘Resilience is the ability of a system to withstand and stay operational in the face of an unexpected disturbance or unpredicted changes. Recent studies on air transport system resilience focus on topology characteristics after the disturbance and measure the robustness of the network with respect to connectivity. The dynamic processes occurring at the node and link levels are often ignored. Here we analyze airport network resilience by considering both structural and dynamical aspects. We develop a simulation model to study the operational performance of the air transport system when airports operate at degraded capacity rather than completely shutting down. Our analyses show that the system deteriorates soon after disruptive events occur but returns to an acceptable level after a period of time. Static resilience of the airport network is captured by a phase transition in which a small change to airport capacity will result in a sharp change in system punctuality. After the phase transition point, decreasing airport capacity has little impact on system performance. Critical airports which have significant influence on the performance of whole system are identified, and we find that some of these cannot be detected based on the analysis of network structural indicators alone. 20One Stone Four Birds:A Novel Liposomal Delivery System Multi-functionalized with Ginsenoside Rh2 for Tumor Targeting Therapy显示文摘Liposomes hold great potential in anti-cancer drug delivery and the targeting treatment of tumors.However,the clinical therapeutic efficacy of liposomes is still limited by the complexity of tumor microenvironment(TME)and the insufficient accumulation in tumor sites.Meanwhile,the application of cholesterol and polyethylene glycol(PEG),which are usually used to prolong the blood circulation and stabilize the structure of liposomes respectively,has been questioned due to various disadvantages.Herein,we developed a ginsenoside Rh2-based multifunctional liposome system(Rh2-lipo)to effectively address these challenges once for all.Different with the conventional’wooden’liposomes,Rh2-lipo is a much more brilliant carrier with multiple functions.In Rh2-lipo,both cholesterol and PEG were substituted by Rh2,which works as membrane stabilizer,long-circulating stealther,active targeting ligand,and chemotherapy adjuvant at the same time.Firstly,Rh2 could keep the stability of liposomes and avoid the shortcomings caused by cholesterol.Secondly,Rh2-lipo showed a specifically prolonged circulation behavior in the blood.Thirdly,the accumulation of the liposomes in the tumor was significantly enhanced by the interaction of glucose transporter of tumor cells with Rh2.Fourth,Rh2-lipo could remodel the structure and reverse the immunosuppressive environment in TME.When tested in a 4T1 breast carcinoma xenograft model,the paclitaxel-loaded Rh2-lipo realized high efficient tumor growth suppression.Therefore,Rh2-lipo not only innovatively challenges the position of cholesterol as a liposome component,but also provides another innovative potential system with multiple functions for anti-cancer drug delivery.Chao Hong Jianming Liang Jiaxuan Xia Ying Zhu Yizhen Guo Anni Wang Chunyi Lu Hongwei Ren Chen Chen Shiyi Li Dan Wang Huaxing Zhan Jianxin Wang 2020Nano-Micro Letters2020,12,10:8
