| 1 | Myeloid-specific targeting of Notch ameliorates murine renal fibrosis via reduced infiltration and activation of bone marrowderived macrophage显示文摘Macrophages play critical roles in renal fibrosis.However,macrophages exhibit ontogenic and functional heterogeneities,and which population of macrophages contributes to renal fibrosis and the underlying mechanisms remain unclear.In this study,we genetically targeted Notch signaling by disrupting the transcription factor recombination signal binding protein-JK(RBP-J),to reveal its role in regulation of macrophages during the unilateral ureteral obstruction(UUO)-induced murine renal fibrosis.Myeloid-specific disruption of RBP-J attenuated renal fibrosis with reduced extracellular matrix deposition and myofibroblast activation,as well as attenuated epithelial-mesenchymal transition,likely owing to the reduced expression of TGF-β.Mean while,RBP-J deletion significantly hampered macrophage infiltration and activation in fibrotic kidney,although their proliferation appeared unaltered.By using macrophage clearance experiment,we found that kidney resident macrophages made negligible contribution,but bone marrow(BM)-derived macrophages played a major role in renal fibrogenesis.Further mechanistic analyses showed that Notch blockade reduced monocyte emigration from BM by down-regulating CCR2 expression.Finally,we found that myeloid-specific Notch activation aggravated renal fibrosis,which was mediated by CCR2^+macrophages infiltration.In summary,our data have unveiled that myeloid-specific targeting of Notch could ameliorate renal fibrosis by regulating BM-derived macrophages recruitment and activation,providing a novel strategy for intervention of this disease. | Yali Jiang Yuanyuan Wang Pengfei Ma Dongjie An Junlong Zhao Shiqian Liang Yuchen Ye Yingying Lu Peng Zhang Xiaowei Liu Hua Han Hongyan Qin | 2019 | Protein & Cell2019,10,3: | 10 |
| 2 | The radioactive abnormality characteristics of typical regions in Ordos Basin and its geological implications显示文摘There are many results of single mineral enrichment characteristic, such as oil, gas, coal and uranium, but little is known about the synergistic research of these important minerals and the study of uranium enrichment features in the deep basin. So, the study on the paragenesis regularity and coexisting relation of many minerals in the basin will promote the integrated forecast and cooperative exploitation of the basin. Based on the plentiful logging data and geological data, this paper studies the distributing feature of higher Gamma abnormality. The analysis on 33 core samples' test results indicates that the increasing of Gamma abnormality is due to the increasing of the uranium element, and the enrichment of uranium is a result of the activation and conglomeration of uranium. | 4 | Terrestrial input and nutrient change reflected by sediment records of the Changjiang River Estuary in recent 80 years显示文摘A variety of environmental problems have been observed in the Changjiang River Estuary and adjacent coastal area, including eutrophication, harmful algal blooms(HABs), and hypoxia in recent decades. Application of sedimentary biogenic element indicators on the study of paleoenvironment can reconstruct environmental evolution history of waters. Two 210Pb-dated cores were collected from the Changjiang River Estuary(S3) and adjacent coastal area(Z13), and total organic carbon(TOC), total nitrogen(TN), biogenic silicon(BSi), total phosphorus(TP) and phosphorus(P) species were analyzed. Three stages of environmental changes are deduced by the nutrient sedimentary records. First, nutrient concentration increased rapidly since the 1950 s, which attributed to agriculture development and overused chemical fertilizers. Second, nutrient concentration kept high and primary production began to promote during the 1960 s to 1980 s, while diatom abundance and proportion began to decline since the 1970 s, accompanied by reduced 23 Si O- concentration and flux from the river. Third, due to several dams and bridges constructed, river runoff and coastal hydrodynamic conditions reduced to a certain extent since the 1990 s, which aggravated the unbalance in nutrient structure. | 5 | Cloning, expression and protective immunity evalua-tion of the full-length cDNA encoding succinate dehydrogenase iron-sulfur protein of Schistosoma japonicum显示文摘1071-bp fragment was obtained from the Schistosoma japonicum (Chinese strain) adult cDNA library after the 3′ and 5′ ends of the incomplete expression sequence tag (EST) of succinate dehydrogenase iron-sulfur protein of Schistosoma japonicum (SjSDISP) were amplified by the anchored PCR with 2 pairs of primers designed according to the EST of SjSDISP and the sequence of multiclone sites of the library vector. Sequence analysis indicated that the fragment was a full-length cDNA with a complete open reading frame (ORF), encoding 278 amino acid residues. The fragment was cloned into prokary- otic expression vector pQE30, and subsequently sequenced and expressed in Escherichia coli. SDS-PAGE and Western-blot analyses showed that the recombinant protein was about 32 kD and could be recognized by the polyclonal antisera from rabbits immunized with Schistosoma japonicum adult worm antigen. Compared with the FCA controls, mice vaccinated with rSjSDISP (test) or rSjGST (posi- tive control) all revealed high levels of specific antibody and significant reduction in worm burden, liver eggs per gram (LEPG), fecal eggs per gram (FEPG) and intrauterine eggs. These results suggest that SjSDISP may be a novel and partially protective vaccine candidate against schistosomiasis. | 7 | Structural variation during dog domestication: insights from gray wolf and dhole genomes显示文摘Several processes like phenotypic evolution, disease susceptibility and environmental adaptations, which fashion the domestication of animals, are largely attributable to structural variations(SVs) in the genome.Here, we present high-quality draft genomes of the gray wolf(Canis lupus) and dhole(Cuon alpinus) with scaffold N50 of 6.04 Mb and 3.96 Mb, respectively. Sequence alignment comprising genomes of three canid species reveals SVs specific to the dog, particularly 16 315 insertions, 2565 deletions, 443 repeats, 16 inversions and 15 translocations. Functional annotation of the dog SVs associated with genes indicates their enrichments in energy metabolisms, neurological processes and immune systems. Interestingly, we identify and verify at population level an insertion fully covering a copy of the AKR1 B1(Aldo-Keto Reductase Family 1 Member B) transcript. Transcriptome analysis reveals a high level of expression of the new AKR1 B1 copy in the small intestine and liver, implying an increase in de novo fatty acid synthesis and antioxidant ability in dog compared to gray wolf, likely in response to dietary shifts during the agricultural revolution. For the first time, we report a comprehensive analysis of the evolutionary dynamics of SVs during the domestication step of dogs. | 8 | Classification of estuaries in China based on eutrophication susceptibility to nutrient load显示文摘Recently, environmental pressures along coasts have increased substantially. Classification of estuaries according to their susceptibility to eutrophication nutrient load is a useful method to determine priority management objects and to enforce control measures. Using historical monitoring data from 2007 to 2012, from 65 estuaries, including 101 estuarine monitoring sections and 260 coastal monitoring stations, a nutrient-driven phytoplankton dynamic model was developed based on the relationship among phytoplankton biomass, Total Nitrogen(TN) load and physical features of estuaries. The ecological filter effect of estuaries was quantified by introducing conversion efficiency parameter values into the model. Markov Chain Monte Carlo algorithm of Bayesian inference was then employed to estimate parameters in the model. The developed model fitted well to the observed chlorophyll, primary production, grazing, and sinking rates. The analysis suggests that an estuary with Q/V(the ratio of river flow to estuarine volume) greater than 2.0 per year and ε(conversion efficiency ratio) less than 1.0 g C/g N can be classified as less susceptible to TN load, Q/V between 0.7 to 2.0 per year and ε between 1.0 to 3.0 g C/g N as moderately susceptible, and ε greater than 3.0 g C/g N as very susceptible. The estuaries with Q/V less than 0.7 per year vary greatly in their susceptibility. The estuaries with high and moderate susceptibility accounted for 67% of all the analyzed estuaries. Significant progress has been attained in construction of the network units with zero-dimensional spherical shapes.However, owing to the limitations of synthetic methods, construction of porous building blocks in one dimension featuring high aspect ratios for porous nanonetwork structured polymer(PNSP) remains largely unexplored. Here we present the successful design and preparation of PNSP with a novel type of one-dimensional network unit, i.e., microporous heterogeneous nanowire. Well-defined core-shell polymer nanoobjects prepared from a gelable block copolymer, poly(3-(triethoxysilyl)propyl methacrylate)-block-polystyrene are employed as building blocks, and facilely transformed into PNSP via hypercrosslinking of polystyrene shell. The as-prepared PNSP exhibits unique three-dimensional hierarchical nanonetwork morphologies with large surface area.
