1Enhancing the antihepatitis B virus immune response by adefovir dipivoxil and entecavir therapies显示文摘Chronicity of hepatitis B(CHB)infection is characterized by a weak immune response to the virus.Entecavir(ETV)and adefovir dipivoxil(ADV)are effective in suppressing hepatitis B virus(HBV)replication.However,the underlying immune mechanism in the antiviral response of patients treated with nucleoside or nucleotide analogs is not clearly understood.In this study,regulatory T cells(Tregs)and intracellular cytokines,including IL-2,interferon(IFN)-γ,tumor-necrosis factor(TNF)-α and IL-4,were measured prior to and at 12,24,36 and 48 weeks after treatment with ETV or ADV.The cytokines were increased from 24 to 48 weeks after treatment.Higher levels of Th1 cytokines were observed with ETV(n=29)versus ADV(n=28)treatment.By contrast,the numbers of Tregs in both groups were decreased.The altered cytokine profile and cellular component was accompanied by a decrease in HBV DNA levels in both groups,which may contribute to their therapeutic effect in CHB infection.Our findings suggest that the antiviral effect of the drugs may be attributed not only to their direct effect on virus suppression but also to their immunoregulatory capabilities.Yanfang Jiang Wanyu Li Lei Yu Jingjing Liu Guijie Xin Hongqing Yan Pinghui Sun Hong Zhang Damo Xu Junqi Niu 2011Cellular & Molecular Immunology2011,8,1:16

4Allele-aware chromosome-level genome assembly of Artemisia annua reveals the correlation between ADS expansion and artemisinin yield显示文摘Artemisia annua is the major natural source of artemisinin,an anti-malarial medicine commonly used worldwide.Here,we present chromosome-level haploid maps for two A.annua strains with different artemisinin contents to explore the relationships between genomic organization and artemisinin production.High-fidelity sequencing,optical mapping,and chromatin conformation capture sequencing were used to assemble the heterogeneous and repetitive genome and resolve the haplotypes of A.annua.Approximately 5o,ooo genes were annotated for each haplotype genome,and a triplication event that occurred approximately 58.12million years ago was examined for the first time in this species.A total of 3,903,467-5,193,414 variants(SNPs,indels,and structural variants)were identified in the 1.5-Gb genome during pairwise comparison between haplotypes,consistent with the high heterozygosity of this species.Genomic analyses revealed a correlation between artemisinin concents and the copy number of amorpha-4,11-dienes ynthasegenes.This correlation was further confirmed by resequencing of 36A.annua samples with varied artemisinin contents.Circular consensus sequencing of transcripts facilitated the detection of paralog expression.Collectively,our study provides chromosome-level allele-aware genome assemblies for two A.annua strains and new insights into the biosynthesis of artemisinin and its regulation,which will contribute to conquering malaria worldwide.Baosheng Liao Xiaofeng Shen Li Xiang Shuai Guo Shiyu Chen Ying Meng Yu Liang Dandan Ding Junqi Bai Dong Zhang Tomasz Czechowski Yi Li Hui Yao Tingyu Mai Caroline Howard Chao Sun Haitao Liu Jiushi Liu Jin Pei Jihai Gao Jigang Wang Xiaohui Qiu Zhihai Huang Hongyi Li Ling Yuan Jianhe Wei lan Graham Jiang Xu Boli Zhang Shilin Chen 2022Molecular Plant2022,15,8:9

696-Week Treatment of Tenofovir Amibufenamide and Tenofovir Disoproxil Fumarate in Chronic Hepatitis B Patients显示文摘Background and Aims:Tenofovir amibufenamide(TMF)is a novel phosphoramidated prodrug of tenofovir with nonin-ferior efficacy and better bone and renal safety to tenofovir disoproxil fumarate(TDF)in 48 weeks of treatment.Here,we update 96-week comparison results.Methods:Patients with chronic hepatitis B were assigned(2:1)to receive either 25 mg TMF or 300 mg TDF with matching placebo for 96 weeks.The virological suppression was defined as HBV DNA levels<20 IU/mL at week 96.Safety was evaluated thoroughly with focusing on bone,renal,and metabolic pa-rameters.Results:Virological suppression rates at week 96 were similar between TMF and TDF group in both HBeAg-positive and HBeAg-negative populations.Noninferior efficacy was maintained in the pooled population,while it was first achieved in patients with HBV DNA≥7 or 8 log10 IU/mL at baseline.Non-indexed estimated glomerular filtration rate for renal safety assessment was adopted,while a smaller decline of which was seen in the TMF group than in the TDF group(p=0.01).For bone mineral density,patients receiv-ing TMF displayed significantly lower reduction levels in the densities of spine,hip,and femur neck at week 96 than those receiving TDF.In addition,the lipid parameters were stable after week 48 in all groups while weight change still showed the opposite trend.Conclusions:TMF maintained similar efficacy at week 96 compared with TDF with continued superior bone and renal safety profiles(NCT03903796).Zhihong Liu Qinglong Jin Yuexin Zhang Guozhong Gong Guicheng Wu Lvfeng Yao Xiaofeng Wen Zhiliang Gao Yan Huang Daokun Yang Enqiang Chen Qing Mao Shide Lin Jia Shang Huanyu Gong Lihua Zhong Huafa Yin Fengmei Wang Peng Hu Qiong Wu Chao Pan Wen Jia Chuan Li Chang’an Sun Junqi Niu Jinlin Hou TMF Study Group 2023Journal of Clinical and Translational Hepatology2023,11,3:3

9Polymeric Complex Nanoparticles Enable the Fabrication of Mechanically Superstrong and Recyclable Poly(aryl ether sulfone)-based Polymer Composites显示文摘It is a long-term pursuit,and also,a challenge to significantly improve the mechanical strength of thermoplastic polymers using readily dispersed polymers as nanofillers.In this study,we demonstrated that in situ formed carboxylic acid-functionalized poly(aryl ether sulfone)(PAES-COOH)/polyvinylpyrrolidone(PVPON)complex nanoparticles can significantly enhance the mechanical strength of PAES-COOH by mixing PAES-COOH with a small fraction of PVPON.The PAES-COOH/PVPON10%composite,which contained∼10 wt%PVPON,exhibited a tensile strength of∼104.8 MPa and Young’s modulus of∼932.2 MPa,which were∼2.0 and∼1.7 times higher than those of the PAES-COOH,respectively.The PAES-COOH/PVPON nanoparticles which were uniformly dispersed in PAES-COOH matrices and had strong hydrogen-bonding interactions with PAES-COOH,served as nanofillers to reinforce the mechanical strength of the PAES-COOH.The PAES-COOH/PVPON_(10%)composites possessed excellent solvent-assisted healability,and the fractured composites could be healed to restore their original mechanical strength.Meanwhile,the PAES-COOH/PVPON_(10%)composites could be recycled multiple times,and yet retained their shape integration and their original mechanical strength.Xingyuan Lu Yuchao Luo Yixuan Li Chunyang Bao Xiaohan Wang Ni An Guibin Wang Junqi Sun 2020CCS Chemistry2020,2,1:1

17Fast-response fluorescent probe with favorable water solubility for highly sensitive imaging of endogenous tyrosinase in living cells and zebrafish model显示文摘Tyrosinase(TYR) is an important polyphenolic oxidase enzyme and usually regards as a biomarker of melano ma cancer.Highly effective tracking TYR activity in vivo will help to study the mechanism of TYR in living organisms and forecasts related diseases.In this study,we present a novel TYR-activatable fluo rescent probe(CHMC-DOPA) for tracking TYR activity in vitro and in vivo.CHMC-DOPA is constructed by incorporating dopamine(DOPA) moiety into a fluorescent chloro-hydroxyl-merocyanine(CHMC)scaffold.Upon exposure to TYR,the dopamine unit in CHMC-DOPA is oxidized to a dopaquinone derivative,and an intramolecular photo-induced electron transfer(PET) process between CHMC fluorophore and o-dopaquinone will take place,the fluorescence of CHMC-DOPA is quenched rapidly.Therefore,the evaluation of TYR activity is established in terms of the relationship between fluorescence quenching efficiency and TYR activity.In our experiments,CHMC-DOPA shows various advantages,such as fast response(8 min),low concentration of TYR activation(0.5 U/mL),good water-solubility,as well as the lowest detection limit(0.003 U/mL) compared with previously reported works.Furthermore,CHMCDO PA also exhibits excellent cell membrane permeability and low cytotoxicity,which is successfully used to monitor endogenous TYR activity in living cancer cells and zebrafish models.CHMC-DOPA performs well,and we anticipate that this newly designed novel platform will provide an alternative for high effective monitoring TYR activity in biosystems.Zheng Li Xiaofeng Xia Yu You Cuifen Lu Guichun Yang Chao Ma Junqi Nie Qi Sun Shuilin Wu Jun Ren Feiyi Wang 2021Chinese Chemical Letters2021,32,5:1
