2Epigallocatechin-3-gallate inhibits angiotensin Il-induced cardiomyocyte hypertrophy via regulating Hippo signaling pathway in H9c2 rat cardiomyocytes显示文摘AngiotensinⅡ(All) has been well known to induce cardiomyocyte hypertrophy. Epigallocatechin-3-gallate (EGCG) is the main active comp on ent of green tea and it has bee n show n to exhibit strong cardioprotective potential, although the underlying molecular mechanisms remain unclear. In this study, we investigated the role and mechanism of EGCG in preventing All-induced cardiomyocyte hypertrophy using rat H9c2 cardiomyocytes cells. Reactive oxygen species assay, cell size, and mRNA expression of cardiac hypertrophy markers ANP and BNP were assessed in response to All treatment. In addition, expression of proteins involved in Hippo signaling pathway were determined by western blot analysis. We found that All treatment resulted in significant upregulation of ANP and BNP expression levels and increase in H9c2 cell size, which were markedly attenuated by EGCG treatment. Furthermore, our results suggested that EGCG inhibited Allinduced cardiac hypertrophy via regulating the Hippo signaling pathway. 3Vertical structure and seasonal variation of tidal and residual currents in the northern Huanghai Sea显示文摘Analysis of tidal current and sea level has been made based on the observations from the summer of 2006 to the winter of 2007,respectively.The result indicates that a two-layer structure of residual current exists in summer,with its upper layer going northwestward and the lower layer southeastward.In addition,some strong residuals exist in the neighboring depth of the pycnocline during the current period of astronomical tide.In winter,except some individual layersthe residual currents mostly direct to the northwest,from which we can see the fluctuation of abnormal sea-level and the appearance of associated current because of the changes of the wind field.The analysis of tidal ellipse indicates that the direction of the maximum semidiurnal component is clockwise from summer to winter,with an angle of 16-18.Moreover,in summer the semidiurnal component rotates with depth clockwise while the diurnal component counterclockwise.However,the vertical structure is almost homogeneous in winter.SONG Jun GUO Junru QIAO Lulu MU Lin YAO Zhigang LIU Yang LI Huan BAO Xianwen SUN Xiaoyan GAO Jia 2011Acta Oceanologica Sinica2011,30,5:4
4Determining the optimal time for bortezomib-based induction chemotherapy followed by autologous hematopoietic stem cell transplant in the treatment of multiple myeloma显示文摘In our study, we determined the efficacy of bortezomib-based induction therapy followed by autologous stem cell transplant (ASCT) in newly diagnosed and relapsed/refractory (R/R) multiple myeloma (MM) patients and compared the advantages of early versus late transplant. We used a retrospective analysis to examine 62 patients, including 46 cases of newly diagnosed MM (early transplant group) and 16 cases of relapsed/refractory MM (late transplant group). All of these patients received bortezomib-based induction therapy followed by ASCT. The efficacy and side effects of the treatment regimen were analyzed. Patients' overall survival (OS) and progression-free survival (PFS) times were determined. The ratio of complete remission to near-complete remission (CR/nCR) was 69.5% versus 56.2% (P=0.361), respectively, for the early transplant group versus the late transplant group, respectively, after receiving bortezomib-based induction therapy; the overall response rates of the two group were 91.3% and 81.2%, respectively (P=0.369). After receiving ASCT, the CR/nCR of the two groups increased to 84.8% and 81.3%, respectively. The median time required for neutrophil engraftment of the early transplant group and the late transplant group was 11 and 14.5 days, respectively (P=0.003); the median time required for platelet engraftment was 13 and 21.5 days (P=0.031), respectively. There were no significant differences in the toxic side effects observed during induction therapy and ASCT between the two groups. The OS of the two groups was not statistically different (P=0.058). The PFS of the early transplant group and the late transplant group was 41.6 and 26.5 months, respectively (P=0.008). Multivariate analysis demonstrated that the time of receiving ASCT, the types of M protein, and the International Staging System (ISS) stage were all independent factors that influenced PFS. 6Definition of water exchange zone between the Bohai Sea and Yellow Sea and the effect of winter gale on it显示文摘The marine dynamic environment of the Bohai Sea and the Yellow Sea in the winter of 2006 is simulated by the Regional Ocean Modelling System(ROMS) marine numerical model. Using the simulated temperature and salinity, the water exchange zone between the Bohai Sea and Yellow Sea is defined through the Spectral Mixture Model(SMM). The influence of winter gales on the water exchange is also discussed. It is found that the Yellow Sea water masses in winter are distributed in a 'tongue' shape in the Bohai Strait region, the water exchange zone presents a zonal distribution along the margin of the 'tongue', with a tendency of running from northwest to southeast, and the water exchange is intensified at the tip of the 'tongue'. Besides, the coastal area in the northernmost Yellow Sea does not participate in the water exchange between the Bohai Sea and Yellow Sea. The result shows that the winter gale events play a role in enhancing the water exchange. It is specifically shown by the facts: the Yellow Sea warm current is enhanced to intrude the Bohai Sea by the gale process; the water exchange zone extends into the Bohai Sea; the water exchange belt in the southern part becomes wider; the mixture zone of river runoff with the Bohai Sea water upon its entry is enlarged and shifts northwards. 7Characterization of a Novel Weak Allele of RGA1/D1 and Its Potential Application in Rice Breeding显示文摘Semi-dwarfing improves the lodging resistance and yield of rice,and the vast majority of modern rice varieties harbor the sd1 allele to decrease plant height,resulting in reduced genetic diversity and negative agronomic traits.Thus,exploring alternative sources of dwarfism is imperative for rice breeding.Here,we identified a novel RGA1 allele,d1-w,from a local indica variety Xiaolixiang(XLX)using a map-based cloning approach.Compared with other rice varieties,RGA1 in XLX contained a unique single nucleotide polymorphism that resulted in an additional transcript and reduced functional RGA1 transcript level.The RGA1 from Nipponbare was introduced into XLX to estimate the value of d1-w in rice breeding.Compared with transgenic XLX plants(XLX^(D1)),XLX exhibited reduced plant height,increased stem strength,lower reactive oxygen species accumulation,delayed senescence,stronger photosynthesis,higher grain yield and quality(including external,milling and nutritional qualities),and enhanced resistance to drought and Rhizoctonia solani.Therefore,we proposed that the d1-w allele has potential as an excellent dwarfism resource for rice breeding.LIU Yantong LI Ting JIANG Zhishu ZENG Chuihai HE Rong QIU Jiao LIN Xiaoli PENG Limei SONG Yongping ZHOU Dahu CAI Yicong ZHU Changlan FU Junru HE Haohua XU Jie 2022Rice science2022,29,6:2

9Comparison of dominant frequency attenuation of blasting vibration for different charge structures显示文摘Dominant frequency attenuation is a significant concern for frequency-based criteria of blasting vibration control.It is necessary to develop a concise and practical prediction equation describing dominant frequency attenuation.In this paper,a prediction equation of dominant frequency that accounts for primary parameters influencing the dominant frequency was proposed based on theoretical and dimensional analyses.Three blasting experiments were carried out in the Chiwan parking lot for collecting blasting vibration data used to conduct regression analysis of the proposed prediction equation.The fitting equations were further adopted to compare the reliability of three different types of dominant frequencies in the proposed equation and to explore the effects of different charge structures on the dominant frequency attenuation.The apparent frequency proved to be more reliable to express the attenuation law of the dominant frequency.The reliability and superiority of the proposed equation employing the apparent frequency were verified by comparison with the other five prediction equations.The smaller blasthole diameter or decoupling ratio leads to the higher initial value and corresponding faster attenuation of the dominant frequency.The blasthole diameter has a greater influence on the dominant frequency attenuation than the decoupling ratio does.Among the charge structures applied in the experiments,the charge structure with decoupling ratio of 1.5 and blasthole diameter of 48 mm results in the greatest initial value and corresponding fastest attenuation of the dominant frequency.Pengchang Sun Wenbo Lu Junru Zhou Xincheng Huang Ming Chen Qi Li 2022Journal of Rock Mechanics and Geotechnical Engineering2022,14,2:2

11Upregulation of GLT25D1 in Hepatic Stellate Cells Promotes Liver Fibrosis via the TGF-β1/SMAD3 Pathway In Vivo and In vitro显示文摘Background and Aims:Collagenβ(1-O)galactosyltransferase 25 domain 1(GLT25D1)is associated with collagen production and glycosylation,and its knockout in mice results in embryonic death.However,its role in liver fibrosis remains elusive,particularly in hepatic stellate cells(HSCs),the primary collagen-producing cells associated with liver fibrogenesis.Herein,we aimed to elucidate the role of GLT25D1 in HSCs.Methods:Bile duct ligation(BDL)-induced mouse liver fibrosis models,primary mouse HSCs(mHSCs),and transforming growth factor beta 1(TGF-β1)-stimulated LX-2 human hepatic stellate cells were used in in vivo and in vitro studies.Stable LX-2 cell lines with either GLT25D1 overexpression or knockdown were established using lentiviral transfection.RNA-seq was performed to investigate the genomic differences.HPLCMS/MS were used to identify glycosylation sites.Scanning electronic microscopy(SEM)and second-harmonic generation/two-photon excited fluorescence(SHG/TPEF)were used to image collagen fibril morphology.Results:GLT25D1 expression was upregulated in nonparenchymal cells in human cirrhotic liver tissues.Meanwhile,its knockdown attenuated collagen deposition in BDL-induced mouse liver fibrosis and inhibited mHSC activation.GLT25D1 was overexpressed in activated versus quiescence LX-2 cells and regulated in vitro LX-2 cell activation,including proliferation,contraction,and migration.GLT25D1 also significantly increased liver fibrogenic gene and protein expression.GLT25D1 upregulation promoted HSC activation and enhanced collagen expression through the TGF-β1/SMAD signaling pathway.Mass spectrometry showed that GLT25D1 regulated the glycosylation of collagen in HSCs,affecting the diameter of collagen fibers.Conclusions:Collectively,the upregulation of GLT25D1 in HSCs promoted the progression of liver fibrosis by affecting HSCs activation and collagen stability.Shiwei Wang Lingling He Fan Xiao Meixin Gao Herui Wei Junru Yang Yang Shu Fuyang Zhang Xiaohui Ye Ping Li Xiaohua Hao Xingang Zhou Hongshan Wei 2023Journal of Clinical and Translational Hepatology2023,11,1:1

15Electrostatic Interaction Tailored Anion-Rich Solvation Sheath Stabilizing High-Voltage Lithium Metal Batteries显示文摘Through tailoring interfacial chemistry,electrolyte engineering is a facile yet effective strategy for highperformance lithium(Li)metal batteries,where the solvation structure is critical for interfacial chemistry.Herein,the effect of electrostatic interaction on regulating an anion-rich solvation is firstly proposed.The moderate electrostatic interaction between anion and solvent promotes anion to enter the solvation sheath,inducing stable solid electrolyte interphase with fast Li+transport kinetics on the anode.This asdesigned electrolyte exhibits excellent compatibility with Li metal anode(a Li deposition/stripping Coulombic efficiency of 99.3%)and high-voltage LiCoO_(2) cathode.Consequently,the 50μm-thin Li||high-loading LiCoO_(2) cells achieve significantly improved cycling performance under stringent conditions of high voltage over 4.5 V,lean electrolyte,and wide temperature range(-20 to 60℃).This work inspires a groundbreaking strategy to manipulate the solvation structure through regulating the interactions of solvent and anion for highperformance Li metal batteries.Junru Wu Ziyao Gao Yao Wang Xu Yang Qi Liu Dong Zhou Xianshu Wang Feiyu Kang Baohua Li 2022Nano-Micro Letters2022,14,9:1

17125I Radioactive Particles Drive Protective Autophagy in Hepatocellular Carcinoma by Upregulating ATG9B显示文摘Background and Aims:125I radioactive particles implantation have demonstrated efficacy in eradicating hepatocellular carcinoma(HCC).However,progressive resistance of HCC to 125I radioactive particles has limited its wide clinical application.Methods:We investigated the cellular responses to 125I radioactive particles treatment and autophagy-related 9B(ATG9B)silencing in HCC cell lines and Hep3B xenografted tumor model using Cell Counting Kit-8 reagent,western blotting,immunofluorescence,flow cytometry,transmission electron microscopy and immunohistochemistry.Results:In this study,we demonstrated that 125I radioactive particles induced cell apoptosis and protective autophagy of HCC in vitro and in vivo.Inhibition of autophagy enhanced the radiosensitivity of HCC to 125I radioactive particles.Moreover,125I radioactive particles induced autophagy by upregulating ATG9B,with increased expression level of LC3B and decreased expression level of p62.Furthermore,ATG9B silencing downregulated LC3B expression and upregulated p62 expression and enhanced radiosensitivity of HCC to 125I radioactive particles in vitro and in vivo.Conclusions:Inhibition of ATG9B enhanced the antitumor effects of 125I particle radiation against HCC in vitro and in vivo.Our findings suggest that 125I particle radiation plus chloroquine or/and the ATG9B inhibitor may be a novel therapeutic strategy for HCC.Yunhua Xiao Jing Yuan Chongshuang Yang Junru Xiong Liangyu Deng Qinghua Liang Chuang He Liangshan Li Fengtian He Xuequan Huang 2023Journal of Clinical and Translational Hepatology2023,11,2:1
