| 3 | Combination of mild therapeutic hypothermia and adipose-derived stem cells for ischemic brain injury显示文摘Mild therapeutic hypothermia has been shown to mitigate cerebral ischemia, reduce cerebral edema, and improve the prognosis of patients with cerebral ischemia. Adipose-derived stem cell-based therapy can decrease neuronal death and infiltration of inflammatory cells, exerting a neuroprotective effect. We hypothesized that the combination of mild therapeutic hypothermia and adipose-derived stem cells would be neuroprotective for treatment of stroke. A rat model of transient middle cerebral artery occlusion was established using the nylon monofilament method. Mild therapeutic hypothermia(33°C) was induced after 2 hours of ischemia. Adipose-derived stem cells were administered through the femoral vein during reperfusion. The severity of neurological dysfunction was measured by a modified Neurological Severity Score Scaling System. The area of the infarct lesion was determined by 2,3,5-triphenyltetrazolium chloride staining. Apoptotic neurons were detected by terminal deoxynucleotidyl transferase-mediated d UTP-biotin nick end labeling(TUNEL) staining. The regeneration of microvessels and changes in the glial scar were detected by immunofluorescence staining. The inflammatory responses after ischemic brain injury were evaluated by in situ staining using markers of inflammatory cells. The expression of inflammatory cytokines was measured by reverse transcription-polymerase chain reaction. Compared with mild therapeutic hypothermia or adipose-derived stem cell treatment alone, their combination substantially improved neurological deficits and decreased infarct size. They synergistically reduced the number of TUNEL-positive cells and glial fibrillary acidic protein expression, increased vascular endothelial growth factor levels, effectively reduced inflammatory cell infiltration and down-regulated the m RNA expression of the proinflammatory cytokines interleukin-1β, tumor necrosis factor-α and interleukin-6. | 6 | Concurrent topology optimization for minimization of total mass considering load-carrying capabilities and thermal insulation simultaneously显示文摘The present work introduces a novel concurrent optimization formulation to meet the requirements of lightweight design and various constraints simultaneously.Nodal displacement of macrostructure and effective thermal conductivity of microstructure are regarded as the constraint functions, which means taking into account both the loadcarrying capabilities and the thermal insulation properties.The effective properties of porous material derived from numerical homogenization are used for macrostructural analysis. Meanwhile, displacement vectors of macrostructures from original and adjoint load cases are used for sensitivity analysis of the microstructure. Design variables in the form of reciprocal functions of relative densities are introduced and used for linearization of the constraint function. The objective function of total mass is approximately expressed by the second order Taylor series expansion. Then, the proposed concurrent optimization problem is solved using a sequential quadratic programming algorithm, by splitting into a series of sub-problems in the form of the quadratic program. Finally, several numerical examples are presented to validate the effectiveness of the proposed optimization method. The various effects including initial designs, prescribed limits of nodal displacement, and effective thermal conductivity on optimized designs are also investigated. | 11 | Seismic Acquisition Parameters Analysis for Deep Weak Reflectors in the South Yellow Sea显示文摘The Mesozoic-Paleozoic marine residual basin in the South Yellow Sea(SYS) is a significant deep potential hydrocarbon reservoir. However, the imaging of the deep prospecting target is quite challenging due to the specific seismic-geological conditions. In the Central and Wunansha Uplifts, the penetration of the seismic wavefield is limited by the shallow high-velocity layers(HVLs) and the weak reflections in the deep carbonate rocks. With the conventional marine seismic acquisition technique, the deep weak reflection is difficult to image and identify. In this paper, we could confirm through numerical simulation that the combination of multi-level air-gun array and extended cable used in the seismic acquisition is crucial for improving the imaging quality. Based on the velocity model derived from the geological interpretation, we performed two-dimensional finite difference forward modeling. The numerical simulation results show that the use of the multi-level air-gun array can enhance low-frequency energy and that the wide-angle reflection received at far offsets of the extended cable has a higher signal-to-noise ratio(SNR) and higher energy. The driver uses external serial programmed pulses and internal clock to simplify brightness control. By embedding a 5-bit digital analog converter (DAC) into the driver, wide dimming range is achieved. Moreover, a new dynamic slope compensation circuit is presented and other key circuits of the driver are optimized to get higher efficiency and fast transition response. A practical circuit is implemented with 0.6 um bipolar complementary-metal-oxide-semiconductor double-diffused-metal-oxide-semiconductor (BCD) technology. Site-specific modification is achieved by a single guide RNA(usually about 20nucleotides) that is complementary to a target gene or locus and is anchored by a protospaceradjacent motif. Cas9 nuclease then cleaves the targeted DNA to generate double-strand breaks(DSBs), which are subsequently repaired by non-homologous end joining(NHEJ) or homology-directed repair(HDR) mechanisms. NHEJ may introduce indels that cause frame shift mutations and hence the disruption of gene functions. When combined with double or multiplex guide RNA design, NHEJ may also introduce targeted chromosome deletions,whereas HDR can be engineered for target gene correction, gene replacement, and gene knock-in. In this review, we briefly survey the history of the CRISPR/Cas9 system invention and its genome-editing mechanism. | 20 | The LRXs-RALFs-FER module controls plant growth and salt stress responses by modulating multiple plant hormones显示文摘Salt stress is a major environmental factor limiting plant growth and productivity.We recently discovered an important new salt tolerance pathway,where the cell wall leucine-rich repeat extensins LRX3/4/5,the RAPID ALKALINIZATION FACTOR(RALF)peptides RALF22/23 and receptor-like kinase FERONIA(FER)function as a module to simultaneously regulate plant growth and salt stress tolerance.However,the intracellular signaling pathways that are regulated by the extracellular LRX3/4/5-RALF22/23-FER module to coordinate growth,cell wall integrity and salt stress responses are still unknown.Here,we report that the LRX3/4/5-RALF22/23-FER module negatively regulates the levels of jasmonic acid(JA),salicylic acid(SA)and abscisic acid(ABA).Blocking JA pathway rescues the dwarf phenotype of the lrx345 and fer-4mutants,while disruption of ABA biosynthesis suppresses the salt-hypersensitivity of these mutants.Many salt stress-responsive genes display abnormal expression patterns in the lrx345 and fer-4 mutants,as well as in the wild type plants treated with epigallocatechin gallate(EGCG),an inhibitor of pectin methylesterases,suggesting cell wall integrity as a critical factor that determines the expression pattern of stress-responsive genes.Production of reactive oxygen species(ROS)is constitutively increased in the lrx345 and fer-4mutants,and inhibition of ROS accumulation suppresses the salt-hypersensitivity of these mutants.Together,our work provides strong evidence that the LRX3/4/5-RALF22/23-FER module controls plant growth and salt stress responses by regulating hormonal homeostasis and ROS accumulation. | Chunzhao Zhao Wei Jiang Omar Zayed Xin Liu Kai Tang Wenfeng Nie Yali Li Shaojun Xie Yuan Li Tiandan Long Linlin Liu Yingfang Zhu Yang Zhao Jian-Kang Zhu | 2021 | National Science Review2021,8,1: | 3 |
