A genome-wide landscape of mRNAs,lncRNAs,and circRNAs during subcutaneous adipogenesis in pigs显示文摘Background: Preadipocyte differentiation plays a critical role in subcutaneous fat deposition in pigs. However, the roles of different RNAs,such as messenger RNAs(mRNAs),long non-coding RNAs(lncRNAs),and circular RNAs(circRNAs) in the differentiation process of subcutaneous preadipocytes, are still largely unclear. In the present study,a transcriptome analysis, including the analysis of mRNAs,lncRNAs,and circRNAs,during different differentiation stages, namely, day 0(D0),day 2(D2),day 4(D4),and day 8(D8),of subcutaneous preadipocytes from Chinese Erhualian pigs was performed. Results: A total of 1554,470,1344,1777,and 676 differentially expressed(DE) mRNAs,112,58,95,136,and 93 DE lncRNAs,and 902,787,710,932,and 850 DE circRNAs were identified between D2 and D0,between D4 and D2,between D8 and D4,between D4 and D0,and between D8 and D0,respectively.Furthermore,functional enrichment analysis showed that the common DE mRNAs during the entire differentiation process were mainly involved in lipid metabolic and cell differentiation processes.Additionally,co-expression network analysis identified the potential lncRNAs related to adipogenesis,e.g.,MSTRG.131380 and MSTRG.62128.Conclusions: Our study provides new insights of the expression changes of RNAs during adipogenic differentiation, which might contribute to the phenotype of subcutaneous adipogenesis. These results greatly improve our understanding of the molecular mechanisms regulating subcutaneous fat deposition in pigs. | Xin Liu Kaiqing Liu Baosen Shan Shengjuan Wei Dongfeng Li Haiyin Han Wei Wei Jie Chen Honglin Liu Lifan Zhang | 2019 | Journal of Animal Science and Biotechnology2019,10,1: | 5

Decentralized multi-agent reinforcement learning with networked agents: recent advances显示文摘Multi-agent reinforcement learning(MARL) has long been a significant research topic in both machine learning and control systems. Recent development of(single-agent) deep reinforcement learning has created a resurgence of interest in developing new MARL algorithms, especially those founded on theoretical analysis. In this paper, we review recent advances on a sub-area of this topic: decentralized MARL with networked agents. In this scenario, multiple agents perform sequential decision-making in a common environment, and without the coordination of any central controller, while being allowed to exchange information with their neighbors over a communication network. Such a setting finds broad applications in the control and operation of robots, unmanned vehicles, mobile sensor networks, and the smart grid. This review covers several of our research endeavors in this direction, as well as progress made by other researchers along the line. We hope that this review promotes additional research efforts in this exciting yet challenging area. | Kaiqing ZHANG Zhuoran YANG Tamer BAŞAR | 2021 | Frontiers of Information Technology & Electronic Engineering2021,22,6: | 1

An endolysin gene from Candidatus Liberibacter asiaticus confers dual resistance to huanglongbing and citrus canker显示文摘The most damaging citrus diseases are Huanglongbing(HLB)and citrus canker, which are caused by Candidatus Liberibacter asiaticus(CaLas)and Xanthomonas citri pv.citri(Xcc),respectively.Endolysins from bacteriophages are a possible option for disease resistance in plant breeding. Here,we report improvement of citrus resistance to HLB and citrus canker using the LasLYS1 and LasLYS2 endolysins from CaLas.LasLYS2 demonstrated bactericidal efficacy against several Rhizobiaceae bacteria and Xcc,according to inhibition zone analyses. The two genes, driven by a strong promoter from Cauliflower mosaic virus,35S,were integrated into Carrizo citrange via Agrobacterium-mediated transformation.More than 2 years of greenhouse testing indicated that LasLYS2 provided substantial and long-lasting resistance to HLB,allowing transgenic plants to retain low CaLas titers and no obvious symptoms while also clearing CaLas from infected plants in the long term.LasLYS2 transgenic plants with improved HLB resistance also showed resistance to Xcc,indicating that LasLYS2 had dual resistance to HLB and citrus canker.A microbiome study of transgenic plants revealed that the endolysins repressed Xanthomonadaceae and Rhizobiaceae populations in roots while increasing Burkholderiaceae and Rhodanobacteraceae populations, which might boost the citrus defense response, according to transcriptome analysis. We also found that Lyz domain 2 is the key bactericidal motif of LasLYS1 and LasLYS2.Four endolysins with potential resistance to HLB and citrus canker were found based on the structures of LasLYS1 and LasLYS2.Overall,the work shed light on the mechanisms of resistance of CaLas-derived endolysins, providing insights for designing endolysins to develop broad-spectrum disease resistance in citrus. | Lanzhen Xu Kaiqing Mo Danlu Ran Juanjuan Ma Lehuan Zhang Yijia Sun Qin Long Guojin Jiang Xiaochun Zhao Xiuping Zou | 2023 | Horticulture Research2023,10,9: | 0
Crosstalk between lung cancer cells and macrophages contributes to traditional Chinese medicine Feiyanninginduced anti-tumor activities by suppressing M2 macrophage polarization显示文摘Lung cancer is one of the leading cancers with high morbidity and fatality worldwide[1].There are two major types of lung cancer:small cell lung cancer(SCLC)and non-small cell lung cancer(NSCLC).NSCLC accounts for nearly 90%of all lung cancer cases[2].Although surgery, radiation therapy, chemotherapy, and immunotherapy[3]have been clinically used, the overall survival rate for lung cancer patients remains at approximately 19%,and the 5-year survival rate is only 5%[4].Therefore,alternative strategies are urgently needed to improve lung cancer treatment. | Kaiqing Zhang Yonglin Peng Qingting Wang Sheng Tan Bin Wang Huaqin Tian Xiaodong Zhao Ming Zhang | 2023 | Acta Biochimica et Biophysica Sinica2023,55,10: | 0
Matching-based two-color X-ray free-electron laser generation utilizing laser-plasma accelerated electron beam显示文摘Laser-plasma accelerators(LPAs)have great potential to realize a compact X-ray free-electron laser(FEL),which is limited by the beam properties currently.Two-color high-intensity X-ray FEL provides a powerful tool for probing ultrafast dynamic systems. In this paper,we present a simple and feasible method to generate a two-color X-ray FEL pulse based on an LPA beam.In this scheme,time-dependent mismatch along the bunch is generated and manipulated by the designed lattice system, enabling FEL lasing at different wavelength within two undulator sections. The time separation between the two pulses can be precisely adjusted by varying the time-delay chicane.Numerical simulations show that two-color soft X-ray FELs with gigawatt-level peak power and femtosecond duration can be generated, which confirm the validity and feasibility of the scheme. | Tao Liu Kaiqing Zhang Zheng Qi Si Chen Chao Feng Haixiao Deng Bo Liu Dong Wang | 2022 | High Power Laser Science and Engineering2022,10,1: | 0
Structural topology optimization subject to overhang angle constraint with overhang length relaxation in additive manufacturing显示文摘This paper studies additive manufacturing(AM) oriented structural topology optimization(TO).The minimum compliance design subject to overhang angle constraint with overhang length relaxation and horizontal minimum length control is considered.Although the overhang length relaxation allows additional flexibility for AM product design, there have been very limited studies on it.This paper elucidates that the overhang angle constraint we proposed can identify the lower boundary element that violates the overhang angle constraint.Taking advantage of this fact,we achieve the overhang length relaxation by specifying that the volume fraction of the elements that violate the overhang angle constraint in each local area of the design domain is less than a specified upper bound.A formula for estimating the maximum allowable overhang length of this method is proposed and verified. The horizontal minimum length constraint is also employed in this paper. While controlling the horizontal length size of the structural member, this constraint together with the overhang angle constraint with overhang length relaxation suppresses the hanging feature. The gradient-based optimization algorithm method of moving asymptotic(MMA) is used to solve the TO formulation.Numerical examples show the effectiveness of this method. It is observed that the new constraint alleviates the main issues of traditional overhang angle constraints,i.e.,gray element issue, stress concentration issue, and shattered structure issue.Compared with the strict traditional overhang angle constraint, the new formulation reduces structural compliance. | ZHANG KaiQing CHENG GengDong WANG Yu | 2022 | Science China(Technological Sciences)2022,65,6: | 0
Morphology modulation of artificial muscles by thermodynamic-twist coupling显示文摘Human muscles can grow and change their length with body development;therefore, artificial muscles that modulate their morphology according to changing needs are needed. In this paper,we report a strategy to transform an artificial muscle into a new muscle with a different morphology by thermodynamic-twist coupling, and illustrate its structural evolution during actuation. The muscle length can be continuously modulated over a large temperature range, and actuation occurs by continuously changing the temperature. This strategy is applicable to different actuation modes, including tensile elongation, tensile contraction and torsional rotation. This is realized by twist insertion into a fibre to produce torsional stress.Fibre anne aling causes partial thermodynamic relaxation of the spiral molecular chains, which serves as internal tethering and inhibits fibre twist release, thus producing a self-supporting artificial muscle that actuates under heating. At a sufficiently high temperature, further relaxation of the spiral molecular chains occurs, resulting in a new mus cle with a different length.A structural study provides an understanding of the thermo dynamic-twist coupling. This work provides a new design strategy for intelligent materials. | Xiaoyu Hu Jiatian Li Sitong Li Guanghao Zhang Run Wang Zhongsheng Liu Mengmeng Chen Wenqian He Kaiqing Yu Wenzhong Zhai Weiqiang Zhao Abdul Qadeer Khan Shaoli Fang Ray HBaughman Xiang Zhou Zunfeng Liu | 2023 | National Science Review2023,10,1: | 0
Online single-shot characterization of ultrafast pulses from high-gain free-electron lasers显示文摘X-ray free-electron lasers(FELs)provide cutting-edge tools for fundamental researches to study nature down to the atomic level at a time-scale that fits this resolution.A precise knowledge of temporal information of FEL pulses is the central issue for its applications. Here we proposed and demonstrated a novel method to determine the FEL temporal profiles online. This robust method, designed for ultrafast FELs,allows researchers to acquire real-time longitudinal profiles of FEL pulses as well as their arrive times with respect to the external optical laser with a resolution better than 6 fs.Based on this method,we can also directly measure various properties of FEL pulses and correlations between them online. This helps us to further understand the FEL lasing processes and realize the generation of stable, nearly fully coherent soft X-ray laser pulses at the Shanghai Soft X-ray FEL facility. This method will enhance the experimental opportunities for ultrafast science in various areas. | Li Zeng Chao Feng Duan Gu Xiaofan Wang Kaiqing Zhang Bo Liu Zhentang Zhao | 2022 | Fundamental Research2022,2,6: | 0
Spider Silk Supercontraction‑Inspired Cotton‑Hydrogel Self‑Adapting Textiles显示文摘Smart textiles are able to self-adapt to an irregular surface. So,they found new applications in intelligent clothes and equipments, where the properties and functionality of traditional polymeric fibers are insufficient, and hard to be realized.Inspired by the supercontraction behavior of the spider silk,we prepared a spinnable hydrogel to form a sheath-core-like composite yarn, after being coated on cotton yarn. The strong hydrogen bonding between the cotton yarn and the polar groups of the hydrogel provides an outstanding mechanical stability, and the twists insertion forms a spiral-like architecture, which exhibited moisture-responsive super contraction behavior.By structural tailoring the chirality of the fiber twists and coiling extends into homo-chiral and heterochiral architectures,as displays contraction and expansion when is exposed to the moisture.Once the relative humidity is increased from 60 to 90%,a homochiral yarn exhibits 90%contraction,while a heterochiral yarn shows 450% expansion, and the maximum work capacity reached up to 6.1 J/Kg.The super contracted yarn can be re-stretched to its original length manifesting cyclability, which can be exploited to build a smart textile, selfadaptive to irregular surfaces.Such a strategy may be further extended to a wide variety of materials to achieve intelligent textiles from common fiber or yarns. | Abdul Qadeer Khan Kaiqing Yu Jiatian Li Xueqi Leng Meilin Wang Xuesong Zhang Baigang An Bin Fei Wei Wei Huichuan Zhuang Muhammad Shafiq Lili Bao Zunfeng Liu Xiang Zhou | 2022 | Advanced Fiber Materials2022,4,6: | 0
