2Gegen Qinlian decoction enhances immunity and protects intestinal barrier function in colorectal cancer patients via gut microbiota显示文摘BACKGROUND We previously showed,using the Traditional Chinese Medicine System Pharmacology Database,that Gegen Qinlian decoction(GQD)had a direct antitumor effect,and was combined with programmed cell death protein(PD)-1 inhibitors to treat microsatellite stable(MSS)tumor-bearing mice.However,the effect of GQD on patients with colorectal cancer(CRC)is not clear.AIM To determine the therapeutic mechanism of GQD in improving immune function,reducing inflammation and protecting intestinal barrier function.METHODS Seventy patients with CRC were included in this study:37 in the control group and 33 in the treatment group.The proportions of CD4+T,CD8+T,natural killer(NK),NKT and T regulatory cells were measured by flow cytometry.Levels of the cytokines tumor necrosis factor(TNF)-α,interferon(IFN)-γ,interleukin(IL)-2,IL-6,IL-10 and serotonin(5-hydroxytryptamine;5-HT)in serum were assessed by enzyme-linked immunosorbent assay(ELISA).The expression of zonula occludens(ZO)-1,occludin,nuclear factor(NF)-κB and TNF-αin tumor and normal tissues was measured by immunohistochemistry.The composition of gut microbiota from patients in the treatment group was assessed using 16S rDNA analysis.RESULTS There were no adverse events in the treatment group.The proportion of CD4+T cells and NKT cells in the post-treatment group was significantly higher than that in the pre-treatment and control groups(P<0.05).The level of TNF-αin the posttreatment group was significantly lower than that in the pre-treatment and control groups(P<0.05).The concentration of 5-HT in the post-treatment group was significantly lower than that in the pre-treatment group(P<0.05).The expression of ZO-1 and occludin in tumor tissues in the treatment group was significantly higher than that in the control group(P<0.05).The expression of ZO-1 in normal tissues of the treatment group was significantly higher than that in the control group(P=0.010).Compared with the control group,expression of NF-κB and TNF-αin tumor tissues of the treatment group was significantly decreased(P<0.05).Compared with the pre-treatment group,GQD decreased the relative abundance of Megamonas and Veillonella.In addition,GQD increased the relative abundance of Bacteroides,Akkermansia and Prevotella.CONCLUSION GQD enhances immunity and protects intestinal barrier function in patients with CRC by regulating the composition of gut microbiota.Yang Li Zhong-Xin Li Chen-Yang Xie Jing Fan Ji Lv Xin-Jian Xu Jian Lv Wen-Tao Kuai Yi-Tao Jia 2020World Journal of Gastroenterology2020,26,48:23
3Regulation of Leaf Senescence and Crop Genetic Improvement显示文摘Leaf senescence can impact crop production by either changing photosynthesis duration,or by modifying the nutrient remobilization efficiency and harvest index.The doubling of the grain yield in major cereals in the last 50 years was primarily achieved through the extension of photosynthesis duration and the increase in crop biomass partitioning,two things that are intrinsically coupled with leaf senescence.In this review,we consider the functionality of a leaf as a function of leaf age,and divide a leaf's life into three phases:the functionality increasing phase at the early growth stage,the full functionality phase,and the senescence and functionality decreasing phase.A genetic framework is proposed to describe gene actions at various checkpoints to regulate leaf development and senescence.Four categories of genes contribute to crop production:those which regulate (I) the speed and transition of early leaf growth,(II) photosynthesis rate,(III) the onset and (IV) the progression of leaf senescence.Current advances in isolating and characterizing senescence regulatory genes are discussed in the leaf aging and crop production context.We argue that the breeding of crops with leaf senescence ideotypes should be an essential part of further crop genetic improvement.Xiao-Yuan Wu Ben-Ke Kuai Ji-Zeng Jia Hai-Chun Jing 2012Journal of Integrative Plant Biology2012,54,12:21
4Interleukin-8 associates with adhesion,migration,invasion and chemosensitivity of human gastric cancer cells显示文摘AIM:To investigate the relationship between Interleukin-8 (IL-8) and proliferation,adhesion,migration,invasion and chemosensitivity of gastric cancer (GC) cells.METHODS:The IL-8 cDNA was stably transfected into human GC cell line MKN-45 and selected IL-8-secreting transfectants.The expression of IL-8 in human GC cell line KATO-Ⅲ was inhibited by RNA interference.The expressions of mRNA and protein of IL-8 in GC cells were detected by real-time reverse transcriptionpolymerase chain reaction or enzyme-linked immunosorbent assay (ELISA).RESULTS:The overexpression of IL-8 resulted in an increased cell adhesion,migration and invasion,and a significant resistance to oxaliplatin in MKN-45 cells.Inhibition of IL-8 expression with small interfering RNA decreased the adhesion,migration and invasion functions and oxaliplatin resistance in KATO-Ⅲ cells.IL-8 increased NF-κB and Akt activities and adhesion molecules ICAM-1,VCAM-1,and CD44 expression in GC cells.CONCLUSION:Overexpression of IL-8 promotes the adhesion,migration,invasion,and chemoresistance of GC cells,indicating that IL-8 is an important therapeutic target in GC.Wen-Xia Kuai Qiong wang Xiao-Zhong Yang Yao Zhao Ren Yu Xiao-Jun Tang 2012World Journal of Gastroenterology2012,18,9:21

8Defined tumor antigen-specific T cells potentiate personalized TCR-T cell therapy and prediction of immunotherapy response显示文摘Personalized immunotherapy targeting tumor-specific antigens(TSAs)could generate efficient and safe antitumor immune response without damaging normal tissues.Although neoantigen vaccines have shown therapeutic effect in clinic trials,precise prediction of neoantigens from tumor mutations is still challenging.The host antitumor immune response selects and activates T cells recognizing tumor antigens.Hence,T cells engineered with T-cell receptors(TCRs)from these naturally occurring tumor antigen-specific T(Tas)cells in a patient will target personal TSAs in his/her tumor.To establish such a personalized TCR-T cell therapy,we comprehensively characterized T cells in tumor and its adjacent tissues by single-cell mRNA sequencing(scRNA-seq),TCR sequencing(TCR-seq)and in vitro neoantigen stimulation.Compared to bystander T cells circulating among tissues,Tas cells were characterized by tumor enrichment,tumor-specific clonal expansion and neoantigen specificity.We found that CXCL13 is a unique marker for both CD4^(+)and CD8^(+)Tas cells.Importantly,TCR-T cells expressing TCRs from Tas cells showed significant therapeutic effects on autologous patient-derived xenograft(PDX)tumors.Intratumoral Tas cell levels measured by CXCL13 expression precisely predicted the response to immune checkpoint blockade,indicating a critical role of Tas cells in the antitumor immunity.We further identified CD200 and ENTPD1 as surface markers for CD4^(^(+))and CD8^(^(+))Tas cells respectively,which enabled the isolation of Tas cells from tumor by Fluorescence Activating Cell Sorter(FACS)sorting.Overall,our results suggest that TCR-T cells engineered with Tas TCRs are a promising agent for personalized immunotherapy,and intratumoral Tas cell levels determine the response to immunotherapy.Jingjing He Xinxin Xiong Han Yang Dandan Li Xuefei Liu Shuo Li Shuangye Liao Siyu Chen Xizhi Wen Kuai Yu Lingyi Fu Xingjun Dong Kaiyu Zhu Xiaojun Xia Tiebang Kang Chaochao Bian Xiang Li Haiping Liu Peirong Ding Xiaoshi Zhang Zhenjiang Liu Wende Li Zhixiang Zuo Penghui Zhou 2022Cell Research2022,32,6:10
9Mitochondrial uncoupling protein 2 expression in colon cancer and its clinical significance显示文摘AIM: To detect the expression of mitochondrial uncoupling protein 2 (UCP2) in colon cancer and analyze the relation between UCP2 expression and clinical pathological features of colon cancer.METHODS: Fifteen colon tissue samples and 15 its adjacent tissue samples were obtained from colon cancer patients during surgical interventions. UCP2 expression was detected with immunohistochemical method in 10 normal controls, 10 hyperplastic polyp patients, 20 tubular adenoma patients and 78 colon cancer patients. Patients with rectal cancer were excluded. Quantitative reverse transcription polymerase chain reaction and Western blotting were used to detect UCP2 expressions in colon cancer tissue samples and its adjacent tissue samples. Relation between UCP2 expression and clinical pathological features of colon cancer was also analyzed. RESULTS: The UCP2 mRNA expression level was fourfold higher in colon cancer tissue samples than in its adjacent tissue samples. The UCP2 protein expression level was three-fold higher in colon cancer tissue samples than in its adjacent normal tissue samples. The UCP2 was mainly expressed in cytoplasm. The UCP2 was not expressed in normal colon mucosa. Strong positive staining for UCP2 with a diffuse distribution pattern was identified throughout the mucosa in colon cancer tissue samples with a positive expression rate of 85.9%. The UCP2 expression level was higher in colon cancer tissue samples at clinical stages Ⅲ and Ⅳ than in those at stageⅠ+ Ⅱ. Univariate analysis showed that the high UCP2 expression level was significantly correlated to colon cancer metastasis (hazard ratio = 4.321, confidence interval = 0.035-0.682, P = 0.046). 10Spatial correlation analysis of sea-bottom backscattering显示文摘The principle of the spatial correlation characteristics of sea-bottom backscattering was expounded.The Kirchhoff model is introduced to simulate sea-bottom backscattering and the spatial correlation is calculated.Some sediment characteristic parameters,such as the ratio of sediment mass density to water mass density,the ratio of sediment sound speed to water sound speed,the attenuation coefficient of sound waves,the strength and exponent of seabottom interface roughness spectrum,are considered while simulating and their influences on spatial correlation are analyzed.By establishing the expressions of the sediment characteristic parameters with the logarithmic sediment grain size,the variation tendency of the spatial correlation versus the logarithmic sediment grain size is obtained.The width of the major lobe of the spatial correlation function narrows as the logarithmic sediment grain size increases.The comparisons between simulated data and in-situ data collected in October 2007 have proven that their spatial correlation function waveforms agree well under the same sediment characteristics.KUAI Duojie WANG Changhong FENG Lei WANG Yuling QIU Wei YI Huiqin CHEN Long 2012Chinese Journal of Acoustics2012,31,3:9
11Development of an automatic monitoring system for rice light-trap pests based on machine vision显示文摘Monitring pest populations in paddy fields is important to effectively implement integrated pest management.Light traps are widely used to monitor field pests all over the world.Most conventional light traps still involve manual identification of target pests from lots of trapped insects,which is time-consuming,labor-intensive and error-prone,especially in pest peak periods.In this paper,we developed an automatic monitoring system for rice light-trap pests based on machine vision.This system is composed of an itelligent light trap,a computer or mobile phone client platform and a cloud server.The light trap firstly traps,kills and disperses insects,then collects images of trapped insects and sends each image to the cloud server.Five target pests in images are automatically identifed and counted by pest identification models loaded in the server.To avoid light-trap insects piling up,a vibration plate and a moving rotation conveyor belt are adopted to disperse these trapped insects.There was a close correlation(r=0.92)between our automatic and manual identification methods based on the daily pest number of one-year images from one light trap.Field experiments demonstrated the effectiveness and accuracy of our automatic light trap monitoring system.YAO Qing FENG Jin TANG Jian XU Wei-gen ZHU Xu-hua YANG Bao-jun LU Jun XIE Yi-ze YAO Bo WU Shu-zhen KUAI Nai-yang WANG Li-jun 2020Journal of Integrative Agriculture2020,19,10:8
12Reverse Genetic Identification of CRN1 and its Distinctive Role in Chlorophyll Degradation in Arabidopsis显示文摘Recent identification of NYE1/SGR1 brought up a new era for the exploration of the regulatory mechanism of Chlorophyll (Chl) degradation.Cluster analysis of senescence associated genes with putative chloroplast targeting sequences revealed several genes sharing a similar expression pattern with NYE1.Further characterization of available T-DNA insertion lines led to the discovery of a novel stay-green gene CRN1 (Co-regulated with NYE1).Chl breakdown was significantly restrained in crn1-1 under diversified senescence scenarios,which is comparable with that in acd1-20,but much more severe than that in nye11.Notably,various Chl binding proteins,especially trimeric LHCP II,were markedly retained in crn1-1 four days after dark-treatment,possibly due to a lesion in disassociation of protein-pigment complex.Nevertheless,the photochemical efficiency of PSII in crn1-1 declined,even more rapidly,two days after dark-treatment,compared to those in Col-0 and nye1-1.Our results suggest that CRN1 plays a crucial role in Chl degradation,and that loss of its function produces various side-effects,including those on the breakdown of Ch-protein complex and the maintenance of the residual photosynthetic capability during leaf senescence.Guodong Ren Qian Zhou Shouxin Wu Yufan Zhang Lingang Zhang Jirong Huang Zhenfei Sun Benke Kuai 2010Journal of Integrative Plant Biology2010,52,5:8
13Carrier frequency offset and impulse noise estimation for underwater acoustic orthogonal frequency division multiplexing显示文摘The carrier frequency offset(CFO)and impulse noise always affect the performance of underwater acoustic communication_systems.The CFO and impulse noise could be estimated by using the null subcarriers to cancel the effects of the two types of interference.The null subcarriers estimation methods include optimal separate estimation and joint estimation.The separate estimation firstly estimates the CFO value and then estimates the impulse noise value.However,the CFO and impulse noise always affect each other when either of them is estimated separately.The performance could be improved by using the joint estimation.The results of simulations and experiments have showed that these two optimization methods have good performance and the joint estimation has better performance than the separate estimation method.There is 3 dB performance gain at the BER value of 10^(-2)when using the joint estimation method.Thus these methods could improve the system robustness by using the CFO compensation and impulse noise suppression.SUN Haixin XU Xiaoka MA Li KUAI Xiaoyan CHENG En CHEN En 2014Chinese Journal of Acoustics2014,33,3:8

17Protective effect of oxysophoridine on cerebral ischemia/reperfusion injury in mice显示文摘Oxysophoridine, a new alkaloid extracted from Sophora alopecuroides L., has been shown to have a protective effect against ischemic brain damage. In this study, a focal cerebral ischemia/reperfusion injury model was established using middle cerebral artery occlusion in mice. Both 62.5, 125, and 250 mg/kg oxysophoridine, via intraperitoneal injection, and 6 mg/kg nimodipine, via intragastric administration, were administered daily for 7 days before modeling. After 24 hours of reperfusion, mice were tested for neurological deficit, cerebral infarct size was assessed and brain tissue was collected. Results showed that oxysophoridine at 125, 250 mg/kg and 6 mg/kg nimodipine could reduce neurological deficit scores, cerebral infarct size and brain water content in mice. These results provided evidence that oxysophoridine plays a protective role in cerebral ischemia/reperfusion injury. In addition, oxysophoridine at 62.5, 125, and 250 mg/kg and 6 mg/kg nimodipine increased adenosine-triphosphate content, and decreased malondialdehyde and nitric oxide content. These compounds enhanced the activities of glutathione-peroxidase, superoxide dismutase, catalase, and lactate dehydrogenase, and decreased the activity of nitric oxide synthase. Protein and mRNA expression levels of N-methyl-D-aspartate receptor subunit NR1 were markedly inhibited in the presence of 250 mg/kg oxysophoridine and 6 mg/kg nimodipine. 18Iterative Block DFE for Underwater Acoustic Single Carrier System显示文摘In order to gain a better performance and reduce the computational complexity of the filter design in the underwater acoustic single carrier system, a new Iterative Block DFE (IBDFE) is proposed, which operates iteratively on blocks of the received signal, and fully implements its filtering operations by Discrete Fourier Transforms (DFTs). Two design methods are considered for IBDFE: one is HD-IBDFE, and the other is SD-IBDFE. In this paper, the first one is adopted. In this scheme, hard detected data are used as input of the feedback, and filters are designed according to the correlation between detected and transmitted data. In the simulations and experiments, the method of the HD-IBDFE is introduced, and for comparison, the performance of H-DFE is shown, as well as that of ZF, MMSE and Matched-Filter-Bound (MFB). From the results of the simulations on a Rayleigh fading channel, it is easily found that on average, the HD-IBDFE outperforms H-DFE. When the number of iteration is 4, HD-IBDFE outperforms ZF, MMSE and H-DFE by about 11dB, 5dB and 0.7dB respectively at BER=10-4. As for MFB, the HD-IBDFE still has a degradation of 1.6 dB. In addition, the experimental results in both experiment pool and shallow water also verify the achievement. The simulation and experiment results indicate that the IBDFE outperforms other equalization schemes.
