2A novel gene delivery system targeting cells expressing VEGF receptors显示文摘Two ligand oligopeptides GV1 and GV2 were designed according to the putative binding region of VEGF to its receptors. GV1, GV2 and endosome releasing oligopeptide HA20 were conjugated with poly-L-lysine or protamine and the resulting conjugates could interact with DNA in a noncovalent bond to form a complex. Using pSV2-β-galactosidase as a reporter gene, it has been demonstrated that exogenous gene was transferred into bovine aortic arch-derived endothelial cells (ABAE) andhuman malignant melanoma cell lines (A375) in vitro. In vivo experiments, exogenous gene was transferred into tumor vascular endothelial cells and tumor cells of subcutaneously transplanted human colon cancer LOVO, human malignant melanoma A375 and human hepatoma graft in nude mice. This system could also target gene to intrahepatically transplanted human hepatoma injected via portal vein in nude mice. 3Surface settlement caused by twin-parallel shield tunnelling in sandy cobble strata显示文摘City metro tunnels are usually constructed as twin-parallel tunnels and their adjacent construction may lead to surface deformation,affecting the surface environment and the safety of the tunnels.Due to its strong dispersion,sandy cobble strata can be easily disturbed by shield tunneling.Based on the project of the Chengdu Metro Line 1,field and model tests were carried out to study the surface settlement caused by shield tunneling in sandy cobble strata by measuring surface settlement curves,ground loss ratios and construction influence zones.The discrete element method(DEM) was used to study the factors affecting the formation of ground arches in sandy cobble strata at the microscopic level.Results show that the shape of the surface settlement curve in sandy cobble strata is different from that in soft soil.The buried depth and clear spacing of the two tunnels had a significant impact on the formation of ground arches.Chuan HE Kun FENG Yong FANG Ying-chao JIANG 2012Journal of Zhejiang University-Science A(Applied Physics & Engineering)2012,13,11:21

7Application of ANSYS 3D FEM in Studies of Surface Deformation Caused by Pipe Jacking显示文摘By using site observation data and establishing 3D model using ANSYS software,this paper has discussed the strain change of stratum stress during process of jacking-in and the im-pact of machine head on ground surface under different frontal resistances. Analysis of the two cases shows that soil pressure reaches its maximum point when the soil is right above machine head,and soil stress will gradually decline when machine head passes over it. It also shows that impact brought by pipe-jacking construction on stress change of the surrounding soil is limited. 8Long-term outcomes after hepatectomy of huge hepatocellular carcinoma: A single-center experience in China显示文摘Background: Currently, hepatectomy remains the first-line therapy for hepatocellular carcinoma(HCC). However, surgery for patients with huge( > 10 cm) HCCs is controversial. This retrospective study aimed to explore long-term survival after hepatectomy for patients with huge HCC. Methods: The records of 188 patients with pathologically confirmed HCC who underwent curative hepatectomy between 2007 and 2017 were reviewed;patients were divided into three groups according to tumor size: huge( > 10 cm;n = 84), large(5–10 cm;n = 51) and small( < 5 cm;n = 53) HCC. Kaplan-Meier analysis was used to assess overall survival(OS) and disease-free survival(DFS), and log-rank analysis was performed for pairwise comparisons among the three groups. Risk factors for survival and recurrence were analyzed using the Cox proportional hazard model. Results: The median follow-up period was 20 months. Although the prognosis of small HCC was better than that of huge and large HCC, OS and DFS were not significantly different between huge and large HCC( P = 0.099 and P = 0.831, respectively). A family history of HCC, poor Child-Pugh class, vascular invasion, diolame, pathologically positive margins, and operative time ≥240 min were identified as independent risk factors for OS and DFS in a multivariate model. Tumor size( > 10 cm) had significant effect on OS, and postoperative antiviral therapy and postoperative complications also had significant effects on DFS. Conclusions: Huge HCC is not a contraindication of hepatectomy. 9Temporal Variations of the Frontal and Monsoon Storm Rainfall during the First Rainy Season in South China显示文摘The temporal variations in storm rainfall during the first rainy season (FRS) in South China (SC) are investigated in this study. The results show that the inter-annual variations in storm rainfall during the FRS in SC seem to be mainly influenced by the frequency of storm rainfall, while both frequency and intensity affect the inter-decadal variations in the total storm rainfall. Using the definitions for the beginning and ending dates of the FRS, and the onset dates of the summer monsoon in SC, the FRS is further divided into two sub-periods, i.e., the frontal and monsoon rainfall periods. The inter-annual and inter-decadal variations in storm rainfall during these two periods are investigated here. 10The positive function of selenium supplementation on reducing nitrate accumulation in hydroponic lettuce(Lactuca sativa L.)显示文摘High nitrate(NO_3^- ) in vegetables, especially in leaf vegetables poses threaten to human health. Selenium(Se) is an important element for maintaining human health, and exogenous Se application during vegetable and crop production is an effective way to prevent Se deficiency in human bodies. Exogenous Se shows positive function on plant growth and nutrition uptake under abiotic and/or biotic stresses. However, the influence of exogenous Se on NO_3^- accumulation in hydroponic vegetables is still not clear. In the present study, hydroponic lettuce plants were subjected to six different concentrations(0, 0.1, 0.5, 5, 10 and 50 μmol L–1) of Se as Na2 Se O3. The effects of Se on NO_3^- content, plant growth, and photosynthetic capacity of lettuce(Lactuca sativa L.) were investigated. The results showed that exogenous Se positively decreased NO_3^- content and this effect was concentration-dependent. The lowest NO_3^- content was obtained under 0.5 μmol L–1 Se treatment. The application of Se enhanced photosynthetic capacity by increasing the photosynthesis rate(Pn), stomatal conductance(Cs) and the transpiration efficiency(Tr) of lettuce. The transportation and assimilation of NO_3^- and activities of nitrogen metabolism enzymes in lettuce were also analysed. The NO_3^- efflux in the lettuce roots was markedly increased, but the efflux of NO_3^- from the root to the shoot was decreased after treated with exogenous Se. Moreover, Se application stimulated NO_3^- assimilation by enhancing nitrate reductase(NR), nitrite reductase(Ni R), glutamine synthetase(GS) and glutamate synthase enzyme(GOGAT) activities. These results provide direct evidence that exogenous Se shows positive function on decreasing NO_3^- accumulation via regulating the transport and enhancing activities of nitrogen metabolism enzyme in lettuce. 11Alpl prevents bone ageing sensitivity by specifically regulating senescence and differentiation in mesenchymal stem cells显示文摘Mutations in the liver/bone/kidney alkaline phosphatase(Alpl) gene cause hypophosphatasia(HPP) and early-onset bone dysplasia,suggesting that this gene is a key factor in human bone development. However, how and where Alpl acts in bone ageing is largely unknown. Here, we determined that ablation of Alpl induces prototypical premature bone ageing characteristics, including bone mass loss and marrow fat gain coupled with elevated expression of p16^(INK4A)(p16) and p53 due to senescence and impaired differentiation in mesenchymal stem cells(MSCs). Mechanistically, Alpl deficiency in MSCs enhances ATP release and reduces ATP hydrolysis. Then, the excessive extracellular ATP is, in turn, internalized by MSCs and causes an elevation in the intracellular ATP level, which consequently inactivates the AMPKα pathway and contributes to the cell fate switch of MSCs. 12Application of video-assisted thoracic surgery in the standard operation for thoracic tumors显示文摘Objective: To evaluate the short-term outcomes of video-assisted thoracic surgery (VATS) for thoracic tumors. Methods: The data of 1,790 consecutive patients were retrospectively reviewed. These patients underwent VATS pulmonary resections, VATS esophagectomies, and VATS resections of mediastinal tumors or biopsies at the Cancer Institute & Hospital, Chinese Academy of Medical Sciences between January 2009 and January 2012. Results: There were 33 patients converted to open thoracotomy (OT, 1.84%). The overall morbidity and mortality rate was 2.79% (50/1790) and 0.28% (5/1790), respectively. The overall hospitalization and chest tube duration were shorter in the VATS lobectomy group (n=949) than in the open thoracotomy (OT) lobectomy group (n=753). There were no significant differences in morbidity rate, mortality rate and operation time between the two groups. In the esophageal cancer patients, no significant difference was found in the number of nodal dissection, chest tube duration, morbidity rate, mortality rate, and hospital length of stay between the VATS esophagectomy group (n=81) and open esophagectomy group (n=81). However, the operation time was longer in the VATS esophagectomy group. In the thymoma patients, there was no significant difference in the chest tube duration, morbidity rate, mortality rate, and hospital length of stay between the VATS thymectomy group (n=41) and open thymectomy group (n=41). However, the operation time was longer in the VATS group. The median tumor size in the VATS thymectomy group was comparable with that in the OT group. Conclusions: In early-stage (I/II) non-small cell lung cancer patients who underwent lobectomies, VATS is comparable with the OT approach with similar short-term outcomes. In patients with resectable esophageal cancer, VATS esophagectomy is comparable with OT esophagectomy with similar morbidity and mortality. VATS thymectomy for Masaoka stage I and II thymoma is feasible and safe, and tumor size is not contraindicated. 13Extrinsic calibration of a laser displacement sensor in a non-contact coordinate measuring machine显示文摘In order to implement 3D scanning of those complicated parts such as blades in the aviation field,a non-contact optical measuring system is established in the paper,which integrates a laser displacement sensor,a probe head,the frame of a coordinate measuring machine(CMM),etc.As the output of the laser sensor directly obtained possesses the 1D length of the laser beam,it needs to determine the unit direction vector of the laser beam denoted as(l,m,n)by calibration so as to convert the 1D values into 3D coordinates of target points.Therefore,an extrinsic calibration method based on a standard sphere is proposed to accomplish this task in the paper.During the calibration procedure,the laser sensor moves along with the motion of the CMM and gathers the required data on the spherical surface.Then,both the output of the laser sensor and the grating readings of the CMM are substituted into the constraint equation of the spherical surface,in which an over-determined nonlinear equation group containing unknown parameters is established.For the purpose of solving the equation group,a method based on non-linear least squares optimization is put forward.Finally,the system after calibration is utilized to measure the diameter of a metallic sphere 10 times from different orientations to verify the calibration accuracy.In the experiment,the errors between the measured results and the true values are all smaller than 0.03 mm,which manifests the validity and practicality of the extrinsic calibration method presented in the paper.Chao BI Jianguo FANG Kun LI Zhijun GUO 2017Chinese Journal of Aeronautics2017,30,4:8

15Hydrodynamics of a Newly China-Made Jiuling Bileaflet Heart Valve Prostheses显示文摘Jiuling bileaflet prosthetic heart valve is manufactured in compliance with the national standard of P.R.C. GB12279 90. This mechanical heart valve substitute is constructed entirely of solid low temperature pyrolytic carbon. There is no graphite substrate within the leaflets or orifice. This prosthesis presents with satisfactory histocompatibility and induces little hemocoagulation around it. Objective: This study aimed to assess the hydrodynamic function of the newly China made Jiuling bileaflet heart valve prostheses. Methods: Jiuling valve has been tested in a pulsatile flow simulator in the aortic position. The testing condition was set at the pulsatile frequency of 55, 75, and 100 beats/min and a constant cardiac output of 4 L/min. The mean pressure differences, leakage volumes and closing volumes across each valve, and the effective orifice areas have been analyzed. Results: The Jiuling heart valves prosthesis had the lowest pressure difference at any given tissue annulus diameter. The mean pressure difference of all the Jiuling valves decreases with increasing the tissue annulus diameter or pulse frequency, and is in the range of below 10 mmHg. CarboMedics (CarboMedics, Inc., Austin, Tex.) is similar to that of the 21 mm and 23 mm Jiuling heart valves at the given pulse rate under 4 L/min. C L tilting disc valve (Lanzhou CarboMedics Inc., China) is similar to that of the 23 mm Jiuling heart valve. The effective orifice area of the Jiuling mechanical valve increases with increasing the tissue annulus diameter or pulse frequency. CarboMedics and C L tilting disc valve is similar to that of the 21 mm and 23 mm Jiuling heart valves. The Closing volume of the Jiuling prosthetic cardiac valves increases with increasing the tissue annulus diameter, but decreases with increasing the pulse frequency, and is in the range of below 11% (normalized by stroke volume). CarboMedics is below 16.5% of stroke volume; and C L tilting disc valve is below 7.5%. The leakage volume of the Jiuling prosthetic cardiac valves increases with increasing the tissue annulus diameter, but decreases with increasing the pulse frequency, and are in the range of below 28.9% (normalized by stroke volume). The leakage volume of CarboMedics is higher than that of the same sized 25 mm Jiuling valve, and the C L tilting disc valve is also higher than that of Jiuling valve at 4 L/min. Conclusion: For a given tissue diameter, the Jiuling valve showed significantly better hemodynamic characteristics. The material used in the construction is biocompatible, durable, and thromboresistant. 17Review of Chinese atmospheric science research over the past 70 years: Synoptic meteorology显示文摘Synoptic meteorology is a branch of meteorology that uses synoptic weather observations and charts for the diagnosis,study,and forecasting of weather.Weather refers to the specific state of the atmosphere near the Earth’s surface during a short period of time.The spatial distribution of meteorological elements in the atmosphere can be represented by a variety of transient weather phenomena,which are caused by weather systems of different spatial and temporal scales.Weather is closely related to people’s life,and its development and evolution have always been the focus of atmospheric scientific research and operation.The development of synoptic meteorology is closely related to the development of observation systems,dynamical theories and numerical models.In China,observation networks have been built since the early 1950 s.Up to now,a comprehensive meteorological observation systembased on ground,air and space has been established.In particular,the development of a new generation of dense radar networks,the development of the Fengyun satellite series and the implementation of a series of large field experiments have brought our understanding of weather from large-scale environment to thermal dynamics,cloud microphysical structure and evolution characteristics of meso and micro-scale weather systems.The development of observation has also promoted the development of theory,numerical model and simulation.In the early days,China mainly used foreign numerical models.Lately,China has developed numerical model systems with independent intellectual property rights.Based on the results of high-resolution numerical simulations,in-depth understanding of the initiation and evolution mechanism and predictability of weather at different scales has been obtained.Synoptic meteorology has gradually changed from an initially independent development to a multidisciplinary approach,and the interaction between weather and the change of climate and environment has become a hot and frontier topic in atmospheric science.This paper reviews the important scientific and technological achievements made in China over the past 70 years in the fields of synoptic meteorology based on the literatures in China and abroad,from six aspects respectively including atmospheric dynamics,synoptic-scale weather,typhoon and tropical weather,severe convective weather,numerical weather prediction and data assimilation,weather and climate,atmospheric physics and atmospheric environment.Zhiyong MENG Fuqing ZHANG Dehai LUO Zhemin TAN Juan FANG Jianhua SUN Xueshun SHEN Yunji ZHANG Shuguang WANG Wei HAN Kun ZHAO Lei ZHU Yongyun HU Huiwen XUE Yaping MA Lijuan ZHANG Ji NIE Ruilin ZHOU Sa LI Hongjun LIU Yuning ZHU 2019Science China Earth Sciences2019,62,12:7
18Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Lung Cancer Patients显示文摘The prevalence of malnutrition is high among oncology patients in China. Although the Patient- Generated Subjective Global Assessment (PG-SGA) is widely recognized as an important nutritional assessment tool, it has not been validated for Chinese cancer patients. The purpose of this study was to conduct an analysis of nutrition screening and assessment data to validate the Chinese version of the PG-SGA in Chinese patients with lung cancer. This was an observational, cross-sectional study of 2,000 consecutive adult patients with lung cancer treated at several tertiary hospitals in China. Anthropometric and patient descriptive data were collected. The PG-SGA generated a score for the nutritional risk and a global rating for the nutritional status. The internal consistency, external consistency, and construct validity were evaluated, and a known-groups comparison and principal component analyses were conducted to evaluate the reliability and validity of the PG-SGA. The known-groups comparison demonstrated the ability of the PG-SGA to differentiate lung cancer patients of different nutritional and body mass index (BMI) groups. A confirmatory factor analysis supported the original four-factor structure of the PG-SGA. In terms of the internal and external consistency, the PG-SGA demonstrated intra-class correlation coefficients (ICC) up to 0.987. Alternative form validity was also demonstrated between Box 4 of the PG-SGA and the Karnofsky Performance Status score, with excellent external consistency (r = 0.972, P < 0.001). The construct validity was supported, and selected questionnaire dimensions were evident in the principal component analysis. Significant Spearman correlations (P < 0.001) were demonstrated. The Chinese version of the PG-SGA demonstrated significant reliability and sufficient exploratory properties to support its validity. It seems to be a valid tool that can be used to access the nutritional status of Chinese lung cancer patients. 20Kiwifruit Genome Database(KGD):a comprehensive resource for kiwifruit genomics显示文摘Kiwifruit(Actinidia spp.)plants produce economically important fruits containing abundant,balanced phytonutrients with extraordinarily high vitamin C contents.Since the release of the first kiwifruit reference genome sequence in 2013,large volumes of genome and transcriptome data have been rapidly accumulated for a handful of kiwifruit species.To efficiently store,analyze,integrate,and disseminate these large-scale datasets to the research community,we constructed the Kiwifruit Genome Database(KGD;http://gffzz6fc08c45f9754a68h0puoubfupuwu69kn.ffgz.tsg.suse.edu.cn/).The database currently contains all publicly available genome and gene sequences,gene annotations,biochemical pathways,transcriptome profiles derived from public RNA-Seq datasets,and comparative genomic analysis results such as syntenic blocks and homologous gene pairs between different kiwifruit genome assemblies.A set of user-friendly query interfaces,analysis tools and visualization modules have been implemented in KGD to facilitate translational and applied research in kiwifruit,which include JBrowse,a popular genome browser,and the NCBI BLAST sequence search tool.Other notable tools developed within KGD include a genome synteny viewer and tools for differential gene expression analysis as well as gene ontology(GO)term and pathway enrichment analysis.Junyang Yue Jiacheng Liu Wei Tang Ya Qing Wu Xiaofeng Tang Wei Li Ying Yang Lihuan Wang Shengxiong Huang Congbing Fang Kun Zhao Zhangjun Fei Yongsheng Liu Yi Zheng 2020Horticulture Research2020,7,1:6
