1Analysis of in vivo patterns of caspase 3 gene expression in primary hepatocellular carcinoma and its relationship to p21^(WAF1) expression and hepatic apoptosis显示文摘AIM To detect the expression of caspase 3gene in primary human hepatocellular carcinoma(HCC)and investigate its relationship to p21WAF1gene expression and HCC apoptosis.METHODS In situ hybridization was employedto determine caspase 3 and p21WAF1expression inHCC.In situ end-labeling was used to detecthepatocytic apoptosis in HCC.RESULTS Twenty-one of 39(53.8%)cases ofHCC were found to express caspase 3transcripts,while 45.2% of HCC failed toexpress caspase 3.Non-cancerous adjacent livertissues showed more positive caspase 3(87.5%,7/8)as compared with HCC(P<0.05).The expression of caspase 3 is correlated withHCC differentiation,72.2%(13/18)ofmoderately to highly differentiated HCC showedcaspase 3 transcripts positive,while only 38.1%of poorly differentiated HCC harbored caspase 3transcripts(P<0.05).No relationship wasfound between caspase 3 expression and tumorsize or grade or metastasis,although 52.5%(5/8)of HCC with metastasis were caspase 3positive and a little higher than that with nometastasis(51.6%,P>0.05).Expression of caspase 3 alone did not affect the apoptosisindex(AI)of HCC.The AI was 7.12%o in caspase3-positive tumors(n=21),while in caspase 3-negative cases(n=18)6.59%0(P>0.05).Expression of caspase 3 clearly segregated withp21WAF1positive tumors as compared withp21WAF1-negative cases(16 of 23,69.6% versus5 of 16,31.3%)with statistical significance(P=0.017).In the cases with positive caspase 3and negative p21WAF1,the Al was found slightlyhigher,but with no statistical significance,thanthat with expression of p21WAF1and caspase 3(7.21‰ vs 6.98‰,P>0.05).CONCLUSION Loss of caspase 3 expressionmay contribute to HCC carcinogenesis,althoughthe expression of caspase 3 does not correlatewell with cell apoptosis in HCC.p21WAF1may bemerely one of the inhibitors which can reducecaspase 3 mediated cell apoptosis in HCCs.Bao Hua Sun Jun Zhang Bao JǜWang Xi Ping Zhao You Kun Wang Zhi Qun Yu Dong Liang Yang Lian Jie Hao Department of Clinical Immunology,Tongji Hospital,Tongji Medical University,Wuhan 430030,Hubei Province,China 2000World Journal of Gastroenterology2000,6,3:65
22018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. Based on the globally recent development and the evidence-based data in China, 2018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents was jointly prepared by the Pediatric Cardiology Society, Chinese Pediatric Society, Chinese Medical Association(CMA)/Committee on Pediatric Syncope, Pediatricians Branch, Chinese Medical Doctor Association(CMDA)/Committee on Pediatric Cardiology, Chinese College of Cardiovascular Physicians, Chinese Medical Doctor Association(CMDA)/Pediatric Cardiology Society, Beijing Pediatric Society, Beijing Medical Association(BMA). 4Effect of Qingfei Mixture (清肺合剂) on Pediatric Mycoplasma Pneumoniae Pneumonia with Phlegm Heat Obstructing Fei (Lung) Syndrome显示文摘Objective: To explore the effect and mechanism of Qingfei Mixture(清肺合剂), a Chinese medicine, in treating mycoplasma pneumonia(MP) in MP patients and rat model. Methods: A total of 46 MP children with phlegm heat obstructing Fei(Lung) syndrome were randomly assigned to two groups by the method of random number table, with 23 children in each group. The control group was treated with intravenous infusion of azithromycin; the treatment group received intravenous infusion of azithromycin and oral administration of Qingfei Mixture. The treatment course was 7 days. Major symptoms and minor symptoms were observed and scored before and after treatments. A rat model of MP was also established. A total of 120 wistar rats were randomly divided into 5 groups: a normal group, infection group, Qingfei Mixture treatment group, azithromycin treatment group, and Qingfei Mixture + azithromycin treatment group. Each group contained 24 rats, from which every 6 were euthanatized 1, 3, 7 and 14 days after infection. MP DNA in pulmonary tissue homogenates was detected using real-time fluorescence quantitative polymerase chain reaction. Pathology was assessed after hematoxylin(HE) staining and lung tissue pathology scores were determined in pulmonary tissue. Transmission electron microscopic detection and electronic image analysis were performed on lung tissue 3 days after infection. Interleukin(IL)-17 was detected in serum using enzymelinked immunosorbent assay(ELISA) 7 days after infection. Results: In the clinical study, both control and the treatment group showed improved results on removing symptoms of phlegm heat syndrome compared to the control group(P<0.05). In animal experiments, On the 7th day after MP infection, as detected by electron microscopy, the pulmonary capillary basement membranes of the azithromycin + Qingfei Mixture treatment group were much thinner than those of the azithromycin or Qingfei mixture treatment groups(P<0.05). The level of serum IL-17 in the azithromycin + Qingfei Mixture treatment group was lower than that in the azithromycin or Qingfei Mixture groups(P<0.01). 5Magnolol additive as a replacer of antibiotic enhances the growth performance of Linwu ducks显示文摘Magnolol rich in Magnolia officinalis is a bioactive polyphenolic compound. The aim of this study was to examine the effects of magnolol additive(MA) on growth performance, expression levels of antioxidantrelated genes, and intestinal mucosal morphology of Linwu ducks aged from 49 to 70 days, comparing with that of an antibiotic additive(colistin sulfate [CS]). A total of 275,49-day-old ducks were assigned to5 groups with 5 cages of 11 ducks each and fed diets supplemented with 0,100, 200 and 300 mg of MA/kg and 300 mg of CS/kg for 3 weeks, respectively. The results showed that the average daily body weight gain(ADG) was increased significantly in MA-fed groups(200 and 300 mg/kg), compared with the basal diet(BD) group(P < 0.05). The mRNA levels of superoxide dismutase-1(SOD1), manganese superoxide dismutase-2(MnSOD2) and catalase(CAT) were also increased significantly in MA groups(P < 0.05). In addition, hematoxylin and eosin staining revealed that Linwu ducks fed the diets with MA had more intact intestinal mucosa than those fed the BD and CS diets. In addition, ileal villus height, ileal villus height/crypt depth ratio(V/C) and duodenal V/C were also improved significantly(P < 0.05). 7Effect of Integrated Chinese and Western Medicine Therapy on Severe Hand, Foot and Mouth Disease: A Prospective, Randomized, Controlled Trial显示文摘Objective: To evaluate the efficacy and safety of Chinese medicine(CM) plus Western medicine(WM) in the treatment of pediatric patients with severe hand, foot and mouth disease(HFMD) by conducting a prospective, controlled, and randomized trial. Methods: A total of 451 pediatric patients with severe HFMD were randomly assigned to receive WM therapy alone(224 cases, WM therapy group) or CM [Reduning Injection(热毒宁注射液) or Xiyanping Injection(喜炎平注射液)] plus WM therapy(227 cases, CM plus WM therapy group) for 7–10 days, according to a web-based randomization system. The primary outcome was fever clearance time, which was presented as temperature decreased half-life time. The secondary outcomes included the rate of rash/herpes disappearance within 120 h, as well as the rate for cough, runny nose, lethargy and weakness, agitation or irritability, and vomiting clearance within 120 h. The drug-related adverse events were also recorded. Results: The temperature decreased half-life time was 40.4 h in the WM therapy group, significantly longer than 27.2 h in the CM plus WM therapy group(P<0.01). Moreover, the rate for rash/herpes disappearance within 120 h was 43.6%(99/227) in the CM plus WM therapy group, significantly higher than 29.5%(66/224) in the WM therapy group(P<0.01). In addition, the rate for cough, lethargy and weakness, agitation or irritability disappearance within 120 h was 32.6%(74/227) in the CM plus WM therapy group, significantly higher than 19.2%(43/224) in the WM therapy group(P<0.01). No drug-related adverse events were observed during the course of the study. Conclusions: The combined CM and WM therapy achieved a better therapeutic efficacy in treating severe HFMD than the WM therapy alone. The World Health Organization's (WHO) Global Tuberculosis Report 2018[1] has reported that in 2017, 10.0 million people across the world had developed TB diseases that resulted in an estimated 1.6 million deaths, and 889, 000 people developed TB in China that led to 39, 000 TB-related deaths. 9Polymer assisted deposition of high-quality CsPbI2Br film with enhanced film thickness and stability显示文摘Inorganic halide perovskites such as cesium lead iodide(CsPbI3)have drawn tremendous attention,as their tunable band gaps are desirable for solar cells as well as light emitting diodes.However,due to their low Goldschmidt tolerance factor,the cubic phase of bulk CsPbX3-the variant with desirable band gap-is not stable in ambient,especially in humid air.Besides,the low solubility of CsX in precursor makes it difficult to control the film thickness and morphology of CsPbX3,which becomes another obstacle for the practical application of inorganic perovskite.Here,we report a polymer assisted deposition of high-quality CsPbI2Br film by spin-coating a polymer-blended CsPbI2Br precursor.The long-chained polymer increases the viscosity of the solution,which enables us to achieve a ca.700-nm thick film with a low solution concentration of CsPbI2Br.Moreover,the polymer network helps to regulate the crystallization process and provides more crystallization sites for perovskite film,reducing grain size and thus improving the film coverage.Perovskite solar cells with the polymer network exhibit improved efficiency and reproducibility(0.72%standard deviation).Moreover,the device demonstrates excellent robustness against moisture and oxygen,and maintains 90%of its initial power conversion efficiency(PCEs)after aging 4 months in ambient conditions.The conception of polymer incorporation into inorganic perovskite films paves a way to further increase the performance,stability and reproducibility of inorganic perovskite devices.Jing Wei Xi Wang Xiangyu Sun Zhaofeng Yang Iwan Moreels Kun Xu Hongbo Li 2020Nano Research2020,13,3:6

11Effects of Structural Changes in Subchondral Bone on Articular Cartilage in a Beagle Dog Model显示文摘Objective Using MR T2-mapping and histopathologic score for articular cartilage to evaluate the effect of structural changes in subchondral bone on articular cartilage. Methods Twenty-four male Beagle dogs were randomly divided into a subchondral bone defect group(n = 12) and a bone cement group(n = 12). Models of subchondral bone defectin the medial tibial plateau and subchondral bone filled with bone cement were constructed. In all dogs, the left knee joint was used as the experimental sideand the right knee as the sham side. The T2 value for articular cartilage at the medial tibial plateau was measured at postoperative weeks 4, 8, 16, and 24. The articular cartilage specimens were stained with hematoxylin and eosin, and evaluated using the Mankin score. Results There was a statistically significant difference(P < 0.05) in Mankin score between the bone defect group and the cement group at postoperative weeks 16 and 24. There was a statistically significant difference in the T2 values between the bone defect group and its sham group(P < 0.05) from week 8, and between the cement group and its sham group(P < 0.05) from week 16. There was significant difference in T2 values between the two experimental groups at postoperative week 24(P < 0.01). The T2 value for articular cartilage was positively correlated with the Mankin score(ρ = 0.758, P < 0.01). Conclusion Structural changes in subchondral bone can lead to degeneration of the adjacent articular cartilage. Defects in subchondral bone cause more severe degeneration of cartilage than subchondral bone filled with cement. The T2 value for articular cartilage increases with the extent of degeneration. 12Two-photon MINFLUX with doubled localization precision显示文摘Achieving localization with molecular precision has been of great interest for extending fluorescence microscopy to nanoscopy.MINFLUX pioneers this transition through point spread function(PSF)engineering,yet its performance is primarily limited by the signal-to-background ratio.Here we demonstrate theoretically that two-photon MINFLUX(2p-MINFLUX)could double its localization precision through PSF engineering by nonlinear effect.Cramér-Rao Bound(CRB)is studied as the maximum localization precision,and CRB of two-photon MINFLUX is halved compared to single-photon MINFLUX(1p-MINFLUX)in all three dimensions.Meanwhile,in order to achieve same localization precision with 1p-MINFLUX,2p-MINFLUX requires only 1/4 of fluorescence photons.Exploiting simultaneous two-photon excitation of multiple fluorophore species,2p-MINFLUX may have the potential for registration-free nanoscopy and multicolor tracking.Kun Zhao Xinzhu Xu Wei Ren Dayong Jin Peng Xi 2022eLight2022,2,1:4

14Effects of neural stem cell transplantation on the motor function of rats with contusion spinal cord injuries:a meta-analysis显示文摘Objective:To judge the efficacies of neural stem cell(NSC)transplantation on functional recovery following contusion spinal cord injuries(SCIs).Data sources:Studies in which NSCs were transplanted into a clinically relevant,standardized rat model of contusion SCI were identified by searching the PubMed,Embase and Cochrane databases,and the extracted data were analyzed by Stata 14.0.Data selection:Inclusion criteria were that NSCs were used in in vivo animal studies to treat contusion SCIs and that behavioral assessment of locomotor functional recovery was performed using the Basso,Beattie,and Bresnahan lo-comotor rating scale.Exclusion criteria included a follow-up of less than 4 weeks and the lack of control groups.Outcome measures:The restoration of motor function was assessed by the Basso,Beattie,and Bresnahan locomotor rating scale.Results:We identified 1756 non-duplicated papers by searching the aforementioned electronic databases,and 30 full-text articles met the inclusion criteria.A total of 37 studies reported in the 30 articles were included in the meta-analysis.The meta-analysis results showed that transplanted NSCs could improve the motor function recovery of rats following contusion SCIs,to a moderate extent(pooled standardized mean difference(SMD)=0.73;95%confidence interval(CI):0.47–1.00;P<0.001).NSCs obtained from different donor species(rat:SMD=0.74;95%CI:0.36–1.13;human:SMD=0.78;95%CI:0.31–1.25),at different donor ages(fetal:SMD=0.67;95%CI:0.43–0.92;adult:SMD=0.86;95%CI:0.50–1.22)and from different origins(brain-derived:SMD=0.59;95%CI:0.27–0.91;spinal cord-derived:SMD=0.51;95%CI:0.22–0.79)had similar efficacies on improved functional recovery;however,adult induced pluripotent stem cell-derived NSCs showed no significant efficacies.Furthermore,the use of higher doses of transplanted NSCs or the administration of immunosuppressive agents did not promote better locomotor function recovery(SMD=0.45;95%CI:0.21–0.70).However,shorter periods between the contusion induction and the NSC transplantation showed slightly higher efficacies(acute:SMD=1.22;95%CI:0.81–1.63;subacute:SMD=0.75;95%CI:0.42–1.09).For chronic injuries,NSC implantation did not significantly improve functional recovery(SMD=0.25;95%CI:–0.16 to 0.65).Conclusion:NSC transplantation alone appears to be a positive yet limited method for the treatment of contusion SCIs.Kai Qian Tuo-Ye Xu Xi Wang Tao Ma Kai-Xin Zhang Kun Yang Teng-Da Qian Jing Shi Li-Xin Li Zheng Wang 2020Neural Regeneration Research2020,15,4:4
15Electrochemical property of LiFePO4/C composite cathode with different carbon sources显示文摘To improve the performance of battery cathode materials that consist of carbonaceous organic material,carbon coatings on lithium iron phosphate (LiFePO_4/C)materials were synthesized by different carbon sources.LiFePO_4/C was synthesized by a combination method of sol-gel and gas-phase diffused permeation. LiFeO_4/C materials were prepared by coating different carbon contents. High-performance composite materials were prepared by combining carbon with element doped by two modified methods. The elements of Fe and C came from Fe3+and sucrose, glucose, citric acid. Thermogravimetrydifferential thermal analysis (TG-DTA), X-ray diffractometer (XRD), scanning electron microscope (SEM),cycle voltammetry (CV), and charge-discharge test were used to characterize and test the surface morphology,structure, and electrochemical performance. The results show that LiFePO_4/C synthesized with sucrose has higher specific discharge capacity than the other materials. The specific discharge capacity of this material is 84.27 mAh·g^(-1). 18Activation of Piezo1 contributes to matrix stiffness-induced angiogenesis in hepatocellular carcinoma显示文摘Background:Despite integrin being highlighted as a stiffness-sensor molecule in matrix stiffness-driven angiogenesis,other stiffness-sensor molecules and their mechanosensory pathways related to angiogenesis in hepatocellular carcinoma(HCC)remain obscure.Here,we explored the interplay between Piezo1 and integrinβ1 in the mechanosensory pathway and their effects on HCC angiogenesis to better understand matrix stiffness-induced angiogenesis.Methods:The role of Piezo1 in matrix stiffness-induced angiogenesis was investigated using orthotopic liver cancer SD rat models with high liver stiffness background,and its clinical significance was evaluated in human HCC tissues.Matrix stiffness-mediated Piezo1 upregulation and activation were assayed using an in vitro fibronectin(FN)-coated cell culture system with different stiffness,Western blotting and Ca^(2+)probe.The effects of shPiezo1-conditioned medium(CM)on angiogenesis were examined by tube formation assay,wound healing assay and angiogenesis array.The underlying mechanism by which Piezo1 participated in matrix stiffness-induced angiogenesis was analyzed by microRNA quantitative real-time polymerase chain reaction(qRT-PCR),matrix stiffness measurement,dual-luciferase reporter assay,ubiquitination assay and co-immunoprecipitation.Results:Increased matrix stiffness significantly upregulated Piezo1 expression at both cellular and tissue levels,and high expression of Piezo1 indicated an unfavorable prognosis.High matrix stiffness also noticeably enhanced the activation level of Piezo1,similar to its expression level.Piezo1 knockdown significantly suppressed tumor growth,angiogenesis,and lung metastasis of HCC rat models with high liver stiffness background.shPiezo1-CM from HCC cells attenuated tube formation and migration abilities of vascular endothelial cells remarkably,and analysis of differentially expressed pro-angiogenic factors revealed that Piezo1 promoted the expression and secretion of vascular endothelial growth factor(VEGF),CXC chemokine ligand 16(CXCL16)and insulin-like growth factor binding protein 2(IGFBP2).Matrix stiffness-caused Piezo1 upregulation/activation restrained hypoxia inducible factor-1α(HIF-1α)ubiquitination,subsequently enhanced the expression of downstream pro-angiogenic factors to accelerate HCC angiogenesis.Besides,collagen 1(COL1)-reinforced tissue stiffening resulted in more expression of Piezo1 via miR-625-5p.Conclusions:This study unravels a new mechanism by which the inte-grinβ1/Piezo1 activation/Ca2+influx/HIF-1αubiquitination/VEGF,CXCL16 and IGFBP2 pathway participates in matrix stiffness-driven HCC angiogenesis.Simultaneously,a positive feedback regulation loop as stiff matrix/integrinβ1/miR-625-5p/Piezo1 and COL1/stiffer matrix mediates matrix stiffness-caused Piezo1 upregulation.Miao Li Xi Zhang Mimi Wang Yaohui Wang Jiali Qian Xiaoxia Xing Zhiming Wang Yang You Kun Guo Jie Chen Dongmei Gao Yan Zhao Lan Zhang Rongxin Chen Jiefeng Cui Zhenggang Ren 2022Cancer Communications2022,42,11:3
19Elevated neutrophil extracellular traps by HBV-mediated S100A9-TLR4/RAGE-ROS cascade facilitate the growth and metastasis of hepatocellular carcinoma显示文摘Background:Neutrophil extracellular traps(NETs)are considered significant contributors to cancer progression,especially metastasis.However,it is still unclear whether NETs are involved in hepatitis B virus(HBV)-related hepatocarcinogenesis and have potential clinical significance during evaluation and management for hepatocellular carcinoma(HCC).In this study,we aimed to investigate the functional mechanism of NETs in HBV-related hepatocarcinogenesis and their clinical significance.Methods:A total of 175 HCC patients with and without HBV infection and 58 healthy controls were enrolled in this study.NETs weremeasured in tissue specimens,freshly isolated neutrophils and blood serum from these patients,and the correlation of circulating serum NETs levels with malignancy was evaluated.The mechanism by which HBV modulates NETs formation was explored using cell-based studies.In addition,in vitro and in vivo experiments were further performed to clarify the functional mechanism of NETs on the growth and metastasis of HCC.Results:We observed an elevated level of NETs in blood serum and tissue specimens from HCC patients,especially those infected with HBV.NETs facilitated the growth and metastasis of HCC both in vitro and in vivo,which were mainly dominated by increased angiogenesis,epithelial-mesenchymal transition(EMT)-related cell migration,matrix metalloproteinases(MMPs)-induced extracellular matrix(ECM)degradation and NETs-mediated cell trapping.Inhibition of NETs generation by DNase 1 effectively abrogated the NETs-aroused HCC growth and metastasis.In addition,HBV-induced S100A9 accelerated the generation of NETs,which was mediated by activation of toll-like receptor(TLR4)/receptor for advanced glycation end products(RAGE)-reactive oxygen species(ROS)signaling.Further,circulatory NETs were found to correlate with viral load,TNM stage and metastasis status in HBV-related HCC,and the identified NETs could predict extrahepatic metastasis,with an area under the ROC curve(AUC)of 0.83 and 90.3%sensitivity and 62.8%specificity at a cutoff value of 0.32.Conclusions:Our findings indicated that activation of RAGE/TLR4-ROS signaling by HBV-induced S100A9 resulted in abundant NETs formation,which subsequently facilitated the growth and metastasis of HCC cells.More importantly,the identified circulatory NETs exhibited potential as an alternative biomarker for predicting extrahepatic metastasis in HBV-related HCC.Xi Zhan RuiWu Xue-Hua Kong Yan You Kun He Xiao-Yu Sun Yong Huang Wei-Xian Chen Liang Duan 2023Cancer Communications2023,43,2:3
