| 3 | Daclatasvir plus asunaprevir in treatment-na?ve patients with hepatitis C virus genotype 1b infection显示文摘AIM To assess daclatasvir plus asunaprevir(d UAL) in treatment-na?ve patients from China's Mainland, Russia and South Korea with hepatitis C virus(HCV) genotype 1 b infection. METHODS Patients were randomly assigned(3:1) to receive 24 wk of treatment with d UAL(daclatasvir 60 mg once daily and asunaprevir 100 mg twice daily) beginning on day 1 of the treatment period(immediate treatment arm) or following 12 wk of matching placebo(placebodeferred treatment arm). The primary endpoint was a comparison of sustained virologic response at posttreatment week 12(SVR12) compared with the historical SVR rate for peg-interferon plus ribavirin(70%) among patients in the immediate treatment arm. The first 12 wk of the study were blinded. Safety was assessed in d UAL-treated patients compared with placebo patients during the first 12 wk(doubleblind phase), and during 24 wk of d UAL in both arms combined.RESULTS In total, 207 patients were randomly assigned to immediate(n = 155) or placebo-deferred(n = 52) treatment. Most patients were Asian(86%), female(59%) and aged < 65 years(90%). Among them, 13% had cirrhosis, 32% had IL28 B non-CC genotypes and 53% had baseline HCV RNA levels of ≥ 6 million IU/m L. Among patients in the immediate treatment arm, SVR12 was achieved by 92%(95% confidence interval: 87.2-96.0), which was significantly higher than the historical comparator rate(70%). SVR12 was largely unaffected by cirrhosis(89%), age ≥ 65 years(92%), male sex(90%), baseline HCV RNA ≥ 6 million(89%) or IL28 B non-CC genotypes(96%), although SVR12 was higher among patients without(96%) than among those with(53%) baseline NS5 A resistanceassociated polymorphisms(at L31 or Y93 H). during the double-blind phase, aminotransferase elevations were more common among placebo recipients than among patients receiving d UAL. during 24 wk of d UAL therapy(combined arms), the most common adverse events(≥ 10%) were elevated alanine aminotransferase and upper respiratory tract infection; emergent grade 3-4 laboratory abnormalities were infrequently observed, and all grade 3-4 aminotransferase abnormalities(alanine aminotransferase, n = 9; aspartate transaminase, n = 6) reversed within 8-11 d. Two patients discontinued d UAL treatment; one due to aminotransferase elevations, nausea, and jaundice and the other due to a fatal adverse event unrelated to treatment. | 6 | A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. | 7 | Fluid Inclusions and Metallization of the Kendekeke Polymetallic Deposit in Qinghai Province, China显示文摘The Kendekeke polymetallic deposit, located in the middle part of the magmatic arc belt of Qimantag on the southwestern margin of the Qaidam Basin, is a polygenetic compound deposit in the Qimantag metallogenic belt of Qinghai Province. Multi-periodic ore-forming processes occurred in this deposit, including early-stage iron mineralization and lead-zinc-gold-polymetallic mineralization which was controlled by later hydrothermal process. The characteristics of the ore-forming fluids and mineralization were discussed by using the fluid inclusion petrography, Laser Raman Spectrum and micro-thermometry methods. Three stages, namely, S1-stage(copper-iron-sulfide stage), S2-stage(lead-zinc-sulfide stage) and C-stage(carbonate stage) were included in the hydrothermal process as indicated by the results of this study. The fluid inclusions are in three types: aqueous inclusion(type I), CO2-aqueous inclusion(type II) and pure CO2 inclusion(type III). Type I inclusions were observed in the S1-stage, having homogenization temperature at 240–320oC, and salinities ranging from 19.8% to 25.0%(wt % NaCl equiv.). All three types of inclusions, existing as immiscible inclusion assemblages, were presented in the S2-stage, with the lowest homogenization temperature ranging from 175 oC to 295oC, which represents the metallogenic temperature of the S2-stage. The salinities of these inclusions are in the range of 1.5% to 16%. The fluid inclusions in the C-stage belong to types I, II and III, having homogenization temperatures at 120–210oC, and salinities ranging from 0.9% to 14.5%. These observations indicate that the ore-forming fluids evolved from high-temperature to lowtemperature, from high-salinity to low-salinity, from homogenization to immiscible separation. Results of Laser Raman Spectroscopy show that high density of CO2 and CH4 were found as gas compositions in the inclusions. CO2, worked as the pH buffer of ore-forming fluids, together with reduction of organic gases(i.e. CH4, etc), affected the transport and sediment of the minerals. The fluid system alternated between open and close systems, namely, between lithostatic pressure and hydrostatic pressure systems. The calculated metallogenic pressures are in the range of 30 to 87 Mpa corresponding to 3 km mineralization depth. Under the influence of tectonic movements, immiscible separation occurred in the original ore-forming fluids, which were derived from the previous highsalinity, high-temperature magmatic fluids. | 12 | Black phosphorus inverter devices enabled by in-situ aluminum surface modification显示文摘Two-dimensional black phosphorus (BP) generally exhibits a hole-dominated transport characteristic when configured as field-effect transistor devices. The effective control of charge carrier type and concentration is very crucial for the application of BP in complementary electronics. Herein, we report a facile and effective electron doping methodology on BP, through in situ surface modification with aluminum (Al). The electron mobility of few-layer BP is found to be largely enhanced to ~ 10.6 cm^2·V^–1·s^–1 by over 6 times after aluminum modification. In situ photoelectron spectroscopy characterization reveals the formation of Al–P covalent bond at the interface, which can also serve as local gate to tune the transport properties in BP layers. | 16 | Early–Middle Triassic Intrusions in Western Inner Mongolia, China: Implications for the Final Orogenic Evolution in Southwestern Xing-Meng Orogenic Belt显示文摘The end-Permian to Early–Middle Triassic magmatic rocks in Inner Mongolia can provide valuable insights into the relationships between the collisional processes and the magmatic responses during the final orogenic evolution of Xing-Meng orogenic belt(XMOB). This paper presents zircon U-Pb ages and Hf isotopes, whole rock geochemical and Sr-Nd-Pb isotopic data for the Early–Middle Triassic diabases and monzogranites from the Langshan area, southwestern XMOB. Our results suggest that the studied diabases and monzogranites were respectively formed during Early Triassic and Middle Triassic. The Early Triassic diabases are characterized by 'arc-like' geochemical signatures, including enrichment in Rb, U and K, and depletion in Nb, Ta, P and Ti. They have negative to weak positive εNd(t) values(-3.1 to +1.5) and relatively high initial ratios of 208 Pb/204 Pb(35.968–37.346), 207 Pb/204 Pb(15.448–15.508) and 206 Pb/204 Pb(16.280–17.492), indicating a subduction-metasomatized enriched lithospheric mantle source. Their low Ba/Rb(2.72–6.56), Ce/Y(0.97–1.39) and(Tb/Yb)N ratios(1.31–1.45) suggest that the parental magma was likely originated from low degree partial melting of the phlogopite-bearing lherzolite in a spinel-stability field. The Middle Triassic monzogranites show high Sr/Y ratios, low Mg O, Cr and Ni contents, high Zr/Sm ratios(40–64), negative zircon εHf(t) values(-25.8 to-8.8), as well as relatively flat heavy rare earth element patterns. They were likely derived from low degree partial melting of a moderately thickened ancient lower crust. The diabases and the slightly postdated high Sr/Y granites in this study represent the magmatic responses to the final orogenic evolution in the southwestern XMOB. However, only one MDR gene have been cloned in maize, and molecular mechanisms of MDR are not clear. By map-based cloning, we have cloned a teosinte-derived allele of a resistance gene, Mexicana lesion mimic 1 (ZmMM1), which has a lesion mimic phenotype and confers resistance to northern leaf blight (NLB), gray leaf spot (GLS) and southern corn rust (SCR). Strong MDR conferred by the teosinte allele is linked with the polymorphisms in the 3' untranslated region of the ZmMM1 gene that cause increased accumulation of ZmMM1 protein. ZmMM1 acts as a transcription repressor and negatively regulates transcription of specific target genes including ZmMM1-target gene 3 (ZmMT3), which functions as a negative regulator of plant immunity and associated cell death. | 20 | Homologous fault monitoring technology of redundant INS in airborne avionics systems显示文摘Redundant technology plays an important role in improving the reliability and fault-tolerance of the airborne avionics systems.A Markov state transition model is introduced to the reliability analysis of the redundant inertial navigation system (RINS) in airborne navigation systems.An information processing mechanism based on difference filtering is put forward to strengthen the consistency between the outputs of the equal-precision inertia navigation system (INS).On this basis,the homologous fault monitoring algorithm is designed to realize the homologous fault monitoring of RINS.The simulation is carried out based on the above algorithms,and the results verify the effectiveness of the proposed fault monitoring algorithm based on difference filtering.Research results have good reference value for the configuration and design of RINS in airborne integrated avionics systems. | Xiuzhi Wu Jizhou Lai Min Liu Pin Lv | 2013 | Journal of Systems Engineering and Electronics2013,24,6: | 4 |
