| 2 | The single nucleotide polymorphisms of the chicken myostatin gene are associated with skeletal muscle and adipose growth显示文摘Myostatin, a new member of the TGF-β superfamily, is predominantly expressed in skeletal muscle cells and functions as a negative regulator of skeletal muscle growth in animals. Recently, we have reported three single nucleotide polymorphisms (SNPs) in the chicken myostatin gene. Herein, we investigate the association of those SNPs with the production traits in a F2 chicken line derived from Broilers crossing to Silky with the least square analysis. The results show that the BB and AA genotypes are strongly associated with abdominal fat weight (AFW), abdominal fat percentage (AFP), and birth weight (BW) (P < 0.05). Breast muscle percentage (BMP) of the AA type is higher than that of the AB type. The breast muscle weight and breast muscle percentages of F2 individuals have significant difference between CC and DD genotypes (P < 0.05). Breast muscle weight (BMW) of EF birds is higher than that of EE birds (P < 0.05). | 5 | Application of Principal Component Analysis(PCA)to the Evaluation and Screening of Multiactivity Fungi显示文摘Continued innovation in screening methodologies remains important for the discovery of high-quality multiactive fungi,which have been of great significance to the development of new drugs.Mangrove-derived fungi,which are well recognized as prolific sources of natural products,are worth sustained attention and further study.In this study,118 fungi,which mainly included Aspergillus spp.(34.62%)and Penicillium spp.(15.38%),were isolated from the mangrove ecosystem of the Maowei Sea,and 83.1%of the cultured fungi showed at least one bioactivity in four antibacterial and three antioxidant assays.To accurately evaluate the fungal bioactivities,the fungi with multiple bioactivities were successfully evaluated and screened by principal component analysis(PCA),and this analysis provided a dataset for comparing and selecting multibioactive fungi.Among the 118 mangrove-derived fungi tested in this study,Aspergillus spp.showed the best comprehensive activity.Fungi such as A.clavatonanicus,A.flavipes and A.citrinoterreus,which exhibited high comprehensive bioactivity as determined by the PCA,have great potential in the exploitation of natural products and the development of new drugs.This study demonstrated the first use of PCA as a time-saving,scientific method with a strong ability to evaluate and screen multiactive fungi,which indicated that this method can affect the discovery and development of new drugs. | YANG Zonglin SHI Yaqi LI Pinglin PAN Kanghong LI Guoqiang LI Xianguo YAO Shuo ZHANG Dahai | 2022 | Journal of Ocean University of China2022,21,3: | 4 |

| 8 | Overexpression of miR-101 suppresses collagen synthesis by targeting EZH2 in hypertrophic scar fibroblasts显示文摘Background:MicroRNA-101(miR-101)is a tumor suppressor microRNA(miRNA)and its loss is associated with the occurrence and progression of various diseases.However,the biological function and target of miR-101 in the pathogenesis of hypertrophic scars(HS)remains unknown.Methods:We harvested HS and paired normal skin(NS)tissue samples from patients and cultured their fibroblasts(HSF and NSF,respectively).We used quantitative reverse transcriptase polymerase chain reaction(qRT-PCR),fluorescence in situ hybridization(FISH),enzyme-linked immunosorbent assays(ELISA)and Western blot analyses to measure mRNA levels and protein expression of miR-101,enhancer of zeste homolog 2(EZH2),collagen 1 and 3(Col1 and Col3)andα-smooth muscle actin(α-SMA)in different in vitro conditions.We also used RNA sequencing to evaluate the relevant signaling pathways and bioinformatics analysis and dual-luciferase reporter assays to predict miR-101 targets.We utilized a bleomycin-induced fibrosis mouse model in which we injected miR-101 mimics to evaluate collagen deposition in vivo.Results:We found low expression of miR-101 in HS and HSF compared to NS and NSF.Overexpressing miR-101 decreased Col1,Col3 andα-SMA expression in HSF.We detected high expression of EZH2 in HS and HSF.Knockdown of EZH2 decreased Col1,Col3 andα-SMA in HSF.Mechanistically,miR-101 targeted the 3-untranslated region(3UTR)of EZH2,as indicated by the decreased expression of EZH2.Overexpressing EZH2 rescued miR-101-induced collagen repression.MiR-101 mimics effectively suppressed collagen deposition in the bleomycin-induced fibrosis mouse model.Conclusions:Our data reveal that miR-101 targets EZH2 in HS collagen production,providing new insight into the pathological mechanisms underlying HS formation. | Jie Li Yan Li Yunchuan Wang Xiang He Jing Wang Weixia Cai Yanhui Jia Dan Xiao Jian Zhang Ming Zhao Kuo Shen Zichao Li Wenbin Jia Kejia Wang Yue Zhang Linlin Su Huayu Zhu Dahai Hu | 2021 | Burns & Trauma2021,9,1: | 3 |

| 10 | One-step synthesis of dimethyl ether from syngas on ordered mesoporous copper incorporated alumina显示文摘Ordered mesoporous copper incorporated Al_2O_3(Cu/Al_2O_3) with high Cu dispersion were prepared by a facile solution combustion synthesis method using aluminum nitrate and copper nitrate as oxidants and urea as fuel. It is a facile and green route to synthesize catalysts for dimethyl ether directly from syngas. Cu/Al_2O_3 catalysts were characterized by XRD, N_2 adsorption–desorption, SEM-EDS, and H_2-TPR.The results indicate that the catalysts obtain an ordered mesoporous structure and copper is homogenously dispersed. The mesoporous Cu/Al_2O_3 catalysts were utilized as bifunctional catalysts in syngas to dimethyl ether reaction(STD). The copper content affects the catalytic performance in STD reaction. The CO conversion and DME selectivity of Cu/Al_2O_3 with 15% copper molar ratio achieve 52.9% and 66.1%,respectively. | 14 | Functional interplay among the flavivirus NS3 protease, helicase, and cofactors显示文摘Flaviviruses are positive-sense RNA viruses, and many are important human pathogens. Nonstructural protein 2B and 3 of the flaviviruses(NS2BNS3) form an endoplasmic reticulum(ER) membrane-associated hetero-dimeric complex through the NS2B transmembrane region. The NS2BNS3 complex is multifunctional. The N-terminal region of NS3, and its cofactor NS2B fold into a protease that is responsible for viral polyprotein processing, and the C-terminal domain of NS3 possesses NTPase/RNA helicase activities and is involved in viral RNA replication and virus particle formation. In addition, NS2BNS3 complex has also been shown to modulate viral pathogenesis and the host immune response. Because of the essential functions that the NS2BNS3 complex plays in the flavivirus life cycle, it is an attractive target for antiviral development.
