| 2 | Newborn screening with targeted sequencing:a multicenter investigation and a pilot clinical study in China显示文摘Different newborn screening(NBS) programs have been practiced in many countries since the 1960 s. It is of considerable interest whether next-generation sequencing is applicable in NBS. We have developed a panel of 465 causative genes for 596 early-onset, relatively high incidence, and potentially actionable severe inherited diseases in our Newborn Screening with Targeted Sequencing(NESTS) program to screen 11,484 babies in 8 Women and Children’s hospitals nationwide in China retrospectively. The positive rate from preliminary screening of NESTS was 7.85%(902/11,484). With 45.89%(414/902) follow-up of preliminary positive cases, the overall clinically confirmative diagnosis rate of monogenic disorders was 12.07%(50/414), estimating an average of 0.95%(7.85% × 12.07%) clinical diagnosis rate, suggesting that monogenic disorders account for a considerable proportion of birth defects. The disease/gene spectrum varied in different regions of China. NESTS was implemented in a hospital by screening 3923 newborns to evaluate its clinical application. The turn-around time of a primary report, including the sequencing period of < 7 days, was within 11 days by our automatic interpretation pipeline. | 3 | Improvement of Foaming and Emulsifying Properties of Gluten by Conjugation with Fructose through Maillard Reaction显示文摘Gluten has poor emulsifying and foaming ability due to its amino acid composition. In this study, Maillard reaction was used to improve the emulsifying and foaming properties of gluten. The processing conditions for the preparation of gluten-fructose conjugates were optimized by using Box-Behnken model to achieve optimum foaming and emulsifying activity, respectively.The results showed that glycated gluten exhibited enhanced emulsifying activity compared to native control. The processing conditions for the preparation of gluten-fructose conjugates with optimum emulsifying activity were as follows: the temperature was 48℃, reaction time was 72 h, and maltose/gluten(W/W) ratio was 125%. Under such condition, the average emulsifying activity was 66.54%, being improved by about 2.5 times compared with that of native control. The Foaming properties of gluten also increased significantly by glycation modification. The optimum conditions of response were as below: the temperature was 48℃, reaction time was 66 h, and maltose/gluten ratio(W/W) was 110%. | 8 | Influencing Factors and Improvement Suggestions for Agricultural Green Development Performance:Empirical Insights from China显示文摘Agricultural green development is an essential direction for global sustainable agriculture.The academic literature,however,needs to place greater emphasis on studying the factors influencing agricultural green development performance and how such performance can be improved.A theoretical framework for agricultural green development performance was constructed in this paper using the Super-SBM model,which considers undesirable outputs,to measure the agricultural green development performance of 330 cities at or above the prefecture level in China(excluding Tibet Autonomous Region,Hong Kong,Macao and Taiwan of China)from 2007 to2018.Furthermore,the influencing mechanism of agricultural green development performance was then analyzed using a spatial econometric model.The results show that:1)from 2007 to 2018,China’s agricultural green development performance experienced three stages of evolution:‘rise,decline and rise’.2)The regions with high performance agricultural green development are mainly distributed in eastern China,northeastern China,and southern Qinghai Province.3)The agricultural economic level,industrialization process,and labor quality play significant roles in promoting local agricultural green development performance,while such performance is obviously inhibited by the openness level and the government’s environmental regulations.Local agricultural green development performance is significant inhibited by the agricultural economic level and accelerated industrialization process in neighboring cities,while significantly promoted by the agricultural industrial structure in neighboring cities.Some suggestions for improving agricultural green development performance are proposed based on these research results,which can provide scientific references for promoting sustainable agriculture. | LI Erling ZHANG Mengzhen LI Ruolan DENG Qingqing | 2023 | Chinese Geographical Science2023,33,5: | 0 |
| 9 | Characterizing structure of cross-disciplinary impact of global disciplines:A perspective of the Hierarchy of Science显示文摘Purpose:Interdisciplinary fields have become the driving force of modern science and a significant source of scientific innovation.However,there is still a paucity of analysis about the essential characteristics of disciplines’cross-disciplinary impact.Design/methodology/approach:In this study,we define cross-disciplinary impact on one discipline as its impact to other disciplines,and refer to a three-dimensional framework of variety-balance-disparity to characterize the structure of cross-disciplinary impact.The variety of cross-disciplinary impact of the discipline was defined as the proportion of the high cross-disciplinary impact publications,and the balance and disparity of cross-disciplinary impact were measured as well.To demonstrate the cross-disciplinary impact of the disciplines in science,we chose Microsoft Academic Graph(MAG)as the data source,and investigated the relationship between disciplines’cross-disciplinary impact and their positions in the Hierarchy of Science(HOS).Findings:Analytical results show that there is a significant correlation between the ranking of cross-disciplinary impact and the HOS structure,and that the discipline exerts a greater cross-disciplinary impact on its neighboring disciplines.Several bibliometric features that measure the hardness of a discipline,including the number of references,the number of cited disciplines,the citation distribution,and the Price index have a significant positive effect on the variety of cross-disciplinary impact.The number of references,the number of cited disciplines,and the citation distribution have significant positive and negative effects on balance and disparity,respectively.It is concluded that the less hard the discipline,the greater the cross-disciplinary impact,the higher balance and the lower disparity of cross-disciplinary impact.Research limitations:In the empirical analysis of HOS,we only included five broad disciplines.This study also has some biases caused by the data source and applied regression models.Practical implications:This study contributes to the formulation of discipline-specific policies and promotes the growth of interdisciplinary research,as well as offering fresh insights for predicting the cross-disciplinary impact of disciplines.Originality/value:This study provides a new perspective to properly understand the mechanisms of cross-disciplinary impact and disciplinary integration. | Ruolan Liu Jin Mao Gang Li Yujie Cao | 2024 | Journal of Data and Information Science2024,9,1: | 0 |
| 10 | An UPLC-MS/MS method to monitor Estriol injection and comparison of pharmacokinetic characteristics after irradiation显示文摘Objective:To develop an UPLC-MS/MS method for the accurate quantification of Estriol(E3),a new radioprotective agent,and observe the variation in pharmacokinetic characteristics of E3 after irradiation.As a hormone drug,the gender differences of E3 metabolism were also concerned here.Methods:Various chromatographic and mass spectrometric conditions of E3 were optimized.The specificity,linearity,precision and accuracy of UPLC-MS/MS method were validated.Twenty SD rats,half male and half female,were administered with intramuscular(im)injection of 2.7 mg/kg E3,and then divided randomly into two groups,sham-irradiated group(Con)and irradiated group(IR).IR group was irradiated to 7 Gy ofγ-rays.Blood samples were collected at different times post-irradiation and E3 concentrations were detected.The changes of concentration-time variation and pharmacokinetic parameters for E3 after irradiation were investigated,together with metabolic differences between male and female rats.Results:The range of the calibration curve of UPLC-MS/MS for E3 was 1.00–200.0 ng/mL.Con group reached maximum concentration(Cmax)(77.57±18.71)ng/mL at(0.68±0.29)h(Tmax)after im injection.The drug concentration-time profiles and pharmacokinetic parameters of E3 were consistent before and after irradiation.The areas under time curve(AUC0-t)of E3 were(353±74)h⋅ng/mL for Con group,and(299±74)h⋅ng/mL for IR group(P>0.05).There were also no statistical differences in pharmacokinetic parameters between female and male rats.The elimination half-lifes(T1/2)of males and females were(3.02±0.68)h and(3.01±0.42)h in Con group,and(3.64±0.51)h and(3.38±0.60)h in IR group,respectively(P>0.05).Conclusion:A rapid and sensitive UPLC-MS/MS method for determination of E3 was established.The pharmacokinetic characteristics of E3 in rat were not affected by 7 Gy irradiation and gender differences.This study provided a theoretical basis for the development and application of new radiation injury treatment drug。 | Hanming Zhang Zhiyun Meng Tong Ye Taoyun Liu Jian Li Fei Ma Ruolan Gu Xiaoxia Zhu Zhuona Wu Guifang Dou Hui Gan | 2021 | Radiation Medicine and Protection2021,2,2: | 0 |

| 13 | SR-BI expression regulates the gastric cancer tumor immune microenvironment and is associated with poor prognosis显示文摘Aim:Scavenger receptor class B,type I(SR-BI)is an integral plasma membrane protein that has been reported to be overexpressed in various malignancies,such as renal cancer,breast cancer,and prostate cancer,and is an independent prognostic factor.However,the clinical value and expression of SR-BI in GC are unknown.Our research aimed to explore the role of SR-BI in combination with immune markers as a diagnostic and prognostic marker for gastric cancer(GC).Methods:GC tissues,paracancerous tissues,and clinicopathological data of 149 patients were collected.The expression level of SR-BI,Tumor-infiltrating lymphocytes(TILs),and PD-L1 were evaluated by immunohistochemistry(IHC).The associations of the SR-BI staining intensity with clinicopathological features and immune markers were determined by the chi-square test.Univariate and multivariate COX regression analyses were used to evaluate independent prognostic factors.Kaplan–Meier analyses were performed to plot the survival curve.Results:Our results indicated that SR-BI was expressed at higher levels in tumor tissues than in adjacent paracancerous tissues(p<0.001),and patients with high levels of SR-BI expression had a worse prognosis.Univariate and multivariate analyses revealed that high SR-BI expression was an independent factor for poor prognosis.The chi-square test determined that the expression of SR-BI was negatively correlated with CD4+T cells and CD8+T cells(CD4+T cells,p=0.013;CD8+T cells,p=0.021),and positively correlated with PD-L1(p=0.022).Finally,survival analysis revealed that CD4+T cells were associated with the prognosis of GC patients(p=0.019),and the combined survival analysis of SR-BI and CD4+T cells was also statistically significant(p=0.030).Conclusion:SR-BI is highly expressed in GC tissue and associated with poor prognosis.Moreover,SR-BI can also regulate the GC tumor immune microenvironment. | YI WANG SHANGQI CHEN PENGCHENG YU ZHEHAN BAO CAN HU YUHANG XIA RUOLAN ZHANG LI YUAN HUA RUAN JIANCHENG SUN | 2023 | BIOCELL2023,47,5: | 0 |

A gentle water-air interface method was adopted to spreadchromatin from interphase nucleus of chicken erythrocyte. The chromatin was ah-sorbed on APS-Inica surfaCe and studied with AFM. Beads-on-a-string were observedand many higher-order structures such as superbeads with dimensions 40-60urn indiameter and 4-7 urn in height were found to string together to make chromatin fibers.When sample spreading and absorbing time were shortened, higher-order chromatinfibers with 60-120 urn in width were observed in air as well as under water environment.
