1Interaction between the gut microbiome and mucosal immune system显示文摘The gut microbiota, the largest symbiotic ecosystem with the host, has been shown to play important roles in maintaining intestinal homeostasis. Dysbiosis of the gut microbiome is caused by the imbalance between the commensal and pathogenic microbiomes. The commensal microbiome regulates the maturation of the mucosal immune system, while the pathogenic microbiome causes immunity dysfunction, resulting in disease development.The gut mucosal immune system, which consists of lymph nodes, lamina propria and epithelial cells, constitutes a protective barrier for the integrity of the intestinal tract. The composition of the gut microbiota is under the surveillance of the normal mucosal immune system. Inflammation, which is caused by abnormal immune responses,influences the balance of the gut microbiome, resulting in intestinal diseases. 3Experiment and simulation study on construction of a three-dimensional network model显示文摘The construction of a network model is one of the key techniques in organic com-bination of microscopic flow experiment and simulation. The construction method of a three-dimensional network model is presented on the basis of CT scanning images in this paper. A series of CT slice images describing microscopic pore structure and fluid distribution of actual rock is obtained with the help of the in-dustrial microfocus CT system. Based on the extraction of pore space skeleton, pore and throat information, the corresponding network model is established, and the conversion from three-dimensional CT image information to pore-throat size distribution and topological information is also achieved. The feature of this me-thod lies in the fact that complicated pore space of rock may be characterized by pores and throats with a simple shape while keeping the geometry and flow char-acteristics. It is indicated that the calculated results of porosity, permeability, rela-tive permeability curve and microscopic remaining oil distribution match very well the experimental results of water flooding and polymer flooding. 5Effects of Shear Fracture on In-depth Profile Modification of Weak Gels显示文摘Two sand packs were filled with fine glass beads and quartz sand respectively.The characteristics of crosslinked polymer flowing through the sand packs as well as the influence of shear fracture of porous media on the in-depth profile modification of the weak gel generated from the crosslinked polymer were investigated.The results indicated that under the dynamic condition crosslinking reaction happened in both sand packs,and the weak gels in these two cases became small gel particles after water flooding.The differences were:the dynamic gelation time in the quartz sand pack was longer than that in the glass bead pack.Residual resistance factor(FRR)caused by the weak gel in the quartz sand pack was smaller than that in the glass bead pack.The weak gel became gel particles after being scoured by subsequent flood water.A weak gel with uniform apparent viscosity and sealing characteristics was generated in every part of the glass bead pack,which could not only move deeply into the sand pack but also seal the high capacity channels again when it reached the deep part.The weak gel performed in-depth profile modification in the glass bead pack,while in the quartz sand pack,the weak gel was concentrated with 100 cm from the entrance of the sand pack.When propelled by the subsequent flood water,the weak gel could move towards the deep part of the sand pack but then became tiny gel particles and could not effectively seal the high capacity channels there.The in-depth profile modification of the weak gel was very weak in the quartz sand pack.It was the shear fracture of porous media that mainly affected the properties and weakened the in-depth profile modification of the weak gel.Li Xianjie Song Xinwang Yue Xiang'an Hou Jirui Fang Lichun Zhang Huazhen 2007Petroleum Science2007,4,1:2

7Heterogeneity-diversity Relationships in Natural Areas of Yunnan,China显示文摘Understanding regional environmental heterogeneity(EH)and biodiversity relationships(heterogeneity-diversity relationships:HDRs)is the first step toward coupling environmental variables with biodiversity surrogates into regional systematic conservation planning.However,there is no universal method for determining regional HDRs that considers various environmental variables and biodiversity in different regions.This study selected 32 nature reserves as natural areas in Yunnan,China,to examine regional HDRs in Yunnan.We calculated 17 EH parameters(of soil,topography,and climate)and three(ecosystem,plant,and animal)biodiversity indices in the nature reserves.By examining the explanatory power of each EH parameter and area of the nature reserve,we identified the primary parameters and constructed an optimal model for each biodiversity index.The explanatory powers of these parameters varied for each biodiversity index,and those of climatic parameters were generally higher than soil and topographic heterogeneity ones.Heterogeneity of the temperature annual range,followed by area and heterogeneity of soil type,were important parameters for ecosystem diversity of Yunnan and the optimal model explained 56.9%.Plant diversity was explained 54.5%by its optimal model,consisting of heterogeneity of precipitation of the coldest quarter and annual precipitation.Heterogeneity of temperature annual range was important for animal diversity in Yunnan and explained 29.6%of its optimal model.This study suggests that EH parameters can be an effective surrogate for biodiversity,therefore,we suggested that the significance and role of climatically heterogeneous regions for the conservation of biodiversity in Yunnan should be further studied in the future.LIU Feng HU Jinming YANG Feiling LI Xinwang 2021Chinese Geographical Science2021,31,3:1
8Glucocorticoid receptor–IRS-1 axis controls EMT and the metastasis of breast cancers显示文摘Glucocorticoid receptor (GR) is involved in the transcriptional regulation of genes that are important for various biological functions, including tumor growth and metastatic progression. However, the cellular and biological effects of GR remain poorly understood. Here, we investigated the role of GR and its underlying mechanism in mediating breast cancer cell survival and metastasis. We observed that the GR levels were increased in drug-resistant breast cancer cells and in metastatic breast cancer samples. GR promoted tumor cell invasion and lung metastasis in vivo. The GR expression levels were negatively correlated with the survival rates of breast cancer patients. Both ectopic expression and knockdown of GR revealed that GR is a strong inducer of epithelial-to-mesenchymal transition (EMT), which is consistent with its effects on cell survival and metastasis. GR suppressed the expression of insulin receptor substrate 1 (IRS-1) by acting as an IRS-1 transcriptional repressor. In addition, GR has an opposite effect on the expression levels of IRS-2, indicating that GR is able to differentially regulate the IRS-1 and IRS-2 expression. The cellular and biological effects elicited by GR were consistent with the reduced levels of IRS-1 observed in cancer cells, and GR-mediated IRS-1 suppression activated the ERK2 MAP kinase pathway, which is required for GR-mediated EMT. 12Chinese registry of rheumatoid arthritis(CREDIT)V:sex impacts rheumatoid arthritis in Chinese patients显示文摘Background: The impact of sex on the clinical manifestations of rheumatoid arthritis (RA) were diversely reported in the literature. The Chinese Registry of rhEumatoiD arthrITis provides a platform for the investigation of this issue in Chinese patients.Methods: Demographic and clinical parameters were collected from all enrolled patients with RA and from patients with early RA (disease duration ≤6 months). The differences in data regarding disease activity, comorbidities, and medications for RA were compared between men and women. The proportions of patients who achieved remission and low disease activity were compared at enrollment and during 3-, 6-, and 12-month follow-up visits.Results: A total of 11,564 patients were enrolled, 83.6% of whom were female. In all the enrolled patients and patients with early RA, C-reactive protein (CRP, 12.0vs. 6.7 mg/L), pain visual analogue scale (4.8vs. 4.5), patient’s and physician’s global assessment (4.9vs. 4.5 and 4.9vs. 4.5), 28-joint disease activity score using DAS28-CRP (4.3vs. 4.0) simplified disease activity index (21.9vs. 19.9), and clinical disease activity index (19.3vs. 18.0) were significantly higher in men than in women. Additionally, the swollen joint count/tender joint count and DAS28 using erythrocyte sedimentation rate were higher in male patients than in female patients with early RA. More female patients with early RA reached the treatment target at baseline than male patients (23.4%vs. 18.2%, assessed by CDAI). At 3 months, 6 months, and 12 months, the proportion of remission and treatment target achievement was similar in both sexes. Coronary artery disease (CAD) and stroke were more frequent in men than in women.Conclusions: In Chinese patients with RA, men were found to have more active disease, as well as more cases of CAD and stroke. 14Sex differences in systemic lupus erythematosus(SLE):an inception cohort of the Chinese SLE Treatment and Research Group(CSTAR)registry XVII显示文摘Background:The onset and clinical presentation of systemic lupus erythematosus(SLE)are sex-related.Few studies have investigated the distinctions in clinical characteristics and treatment preferences in male and female SLE patients in the initial cohort.This study aimed to improve the understanding of Chinese SLE patients by characterizing the different sexes of SLE patients in the inception cohort.Methods:Based on the initial patient cohort established by the Chinese SLE Treatment and Research Group,a total of 8713 patients(795 men and 7918 women)with newly diagnosed SLE were enrolled between April 2009 and March 2021.Of these,2900 patients(347 men and 2553 women)were eligible for lupus nephritis(LN).A cross-sectional analysis of the baseline demographic characteristics,clinical manifestations,laboratory parameters,organ damage,initial treatment regimens,and renal pathology classification was performed according to sex.Results:In the SLE group,as compared to female patients,male patients had a later age of onset(male vs.female:37.0±15.8 years vs.35.1±13.7 years,P=0.006)and a higher SLE International Collaborative Clinic/American College of Rheumatology damage index score(male vs.female:0.47±1.13 vs.0.34±0.81,P=0.015),LN(male vs.female:43.6%vs.32.2%,P<0.001),fever(male vs.female:18.0%vs.14.6%,P=0.010),thrombocytopenia(male vs.female:21.4%vs.18.5%,P=0.050),serositis(male vs.female:14.7%vs.11.7%,P=0.013),renal damage(male vs.female:11.1%vs.7.4%,P<0.001),and treatment with cyclophosphamide(CYC)(P<0.001).The frequency of leukopenia(male vs.female:20.5%vs.25.4%,P=0.002)and arthritis(male vs.female:22.0%vs.29.9%,P<0.001)was less in male patients with SLE.In LN,no differences were observed in disease duration,SLE Disease Activity Index score,renal biopsy pathological typing,or 24-h urine protein quantification among the sexes.In comparisons with female patients with LN,male patients had later onset ages(P=0.026),high serum creatinine(P<0.001),higher end-stage renal failure rates(P=0.002),musculoskeletal damage(P=0.023),cardiovascular impairment(P=0.009),and CYC use(P=0.001);while leukopenia(P=0.017),arthritis(P=0.014),and mycophenolate usage(P=0.013)rates were lower.Conclusions:Male SLE patients had more severe organ damage and a higher LN incidence compared with female SLE patients;therefore,they may require more aggressive initial treatment compared to female patients.Yinli Gui Wei Bai Jian Xu Xinwang Duan Feng Zhan Chen Zhao Zhenyu Jiang Zhijun Li Lijun Wu Shengyun Liu Min Yang Wei Wei Ziqian Wang Jiuliang Zhao Qian Wang Xiaomei Leng Xinping Tian Mengtao Li Yan Zhao Xiaofeng Zeng CSTAR co-authors 2022Chinese Medical Journal2022,135,18:0
15Effect of Three Feed Antibiotics Substitutes on Growth Performance and Apparent Nutrient Digesti- bility of Guangxi Jinling Partridge显示文摘Totally 150 individuals of 1-day-old Guangxi Jinling partridges were randomly divided into five groups,30 chickens each group. Chickens in control group were fed with basal diet,while chickens in experimental groups were fed with the diet containing antibiotics,Chinese herbal medicine,probiotics and acidifier. The entire trial lasted 50 d,and growth performance and apparent nutrient digestibility of tested chickens were observed and measured. Compared with control group,the feed gain ratio in experimental groups significantly decreased( P < 0. 05),while that in probiotics group was the lowest. The average daily gain in probiotics group was significantly higher than that in control group or other experimental groups( P < 0. 05). The apparent digestibility of crude protein,crude fat and energy in probiotics group were significantly higher than that in control group or other experimental groups( P < 0. 05). The apparent digestibility of phosphorus,crude fat,crude fiber,calcium and crude protein in antibiotics group were significantly higher than that in control group( P < 0. 05). The apparent digestibility of crude protein,crude fiber and phosphorus in Chinese herbal medicine group were significantly higher than that in control group( P < 0. 05). The apparent digestibility of nutrient in acidifier group were significantly lower than that in control group or other experimental groups( P < 0. 05). 16Amphibian-derived peptide homodimer OA-GL17d promotes skin wound regeneration through the miR-663a/TGF-β1/Smad axis显示文摘Background:Amphibian-derived peptides exhibit considerable potential in the discovery and development of new therapeutic interventions for clinically challenging chronic skin wounds.MicroRNAs(miRNAs)are also considered promising targets for the development of effective therapies against skin wounds.However,further research in this field is anticipated.This study aims to identify and provide a new peptide drug candidate,as well as to explore the underlying miRNA mechanisms and possible miRNA drug target for skin wound healing.Methods:A combination of Edman degradation,mass spectrometry and cDNA cloning were adopted to determine the amino acid sequence of a peptide thatwas fractionated from the secretion of Odorrana andersonii frog skin using gel-filtration and reversed-phase high-performance liquid chromatography.The toxicity of the peptide was evaluated by Calcein-AM/propidium iodide(PI)double staining against human keratinocytes(HaCaT cells),hemolytic activity against mice blood cells and acute toxicity against mice.The stability of the peptide in plasma was also evaluated.The prohealing potency of the peptide was determined by MTS,scratch healing and a Transwell experiment against HaCaT cells,full-thickness injury wounds and scald wounds in the dorsal skin of mice.miRNA transcriptome sequencing analysis,enzyme-linked immunosorbent assay,real-time polymerase chain reaction and western blotting were performed to explore the molecular mechanisms.Results:A novel peptide homodimer(named OA-GL17d)that contains a disulfide bond between the 16th cysteine residue of the peptide monomer and the sequence‘GLFKWHPRCGEEQSMWT’was identified.Analysis showed that OA-GL17d exhibited no hemolytic activity or acute toxicity,but effectively promoted keratinocyte proliferation and migration and strongly stimulated the repair of full-thickness injury wounds and scald wounds in the dorsal skin of mice.Mechanistically,OA-GL17d decreased the level of miR-663a to increase the level of transforming growth factor-β1(TGF-β1)and activate the subsequent TGF-β1/Smad signaling pathway,thereby resulting in accelerated skin wound re-epithelialization and granular tissue formation.Conclusions:Our results suggest that OA-GL17d is a new peptide drug candidate for skin wound repair.This study emphasizes the importance of exogenous peptides as molecular probes for exploring competing endogenous RNA mechanisms and indicates that miR-663a may be an effective target for promoting skin repair.Yue Zhang Ying Wang Lin Zeng Yixiang Liu Huiling Sun Shanshan Li Siyu Wang Longjun Shu Naixin Liu Saige Yin Junsong Wang Dan Ni Yutong Wu Ying Yang Li He Buliang Meng Xinwang Yang 2022Burns & Trauma2022,10,1:0
