Analysis of status and countermeasures of cancer incidence and mortality in China显示文摘Cancer is the leading cause of human deaths in the world and produces serious economic burdens. On September 12, 2018, the academic journal A Cancer Journal for Clinicians published an article about the latest statistics of cancers worldwide, which provided a status report on the global burden of 36 cancers in 185 countries worldwide. Cancer has also become a serious public health problem in China and caused more and more attention of the government and people in recent years. This review analyzes the incidence, mortality and prevalent trend of cancers in China, discusses the reasons behind this status, and reviews the potential countermeasures for cancer prevention and control in China.Chunchun Wu Mengna Li Hanbing Meng Yukun Liu Weihong Niu Yao Zhou Ran Zhao Yumei Duan Zhaoyang Zeng Xiaoling Li Guiyuan Li Wei Xiong Ming Zhou 2019Science China(Life Sciences)2019,62,5:175
Comparing the Diagnostic Criteria for Gestational Diabetes Mellitus of World Health Organization 2013 with 1999 in Chinese Population显示文摘INTRODUCTION Gestational diabetes mellitus (GDM) was earlier defined as 'hyperglycemia first recognized during pregnancy' and has more recently been described by American Diabetes Association (ADA) (2012) as diabetes diagnosed during pregnancy that is not clearly overt diabetes.[1] The hyperglycemia and adverse pregnancy outcomes study (HAPO) demonstrated that the risk of adverse maternal, fetal, and neonatal outcomes continuously increase as a function of maternal glycemia at 24-28 weeks, even within ranges previously considered normal for pregnancy.[2] After reviewing the results of the HAPO study, many international diabetes study groups, including the International Association of Diabetes and Pregnancy Study Groups and ADA have adopted the 75 g oral glucose tolerance test (OGTT) at 24-28 weeks as a screening and diagnostic test and defined new cut-off values for GDM diagnosis. The Ministry of Health of China published the criteria for GDM,that is the same as the ADA criteria on July 1,2011.World Health Organization (WHO) has recommended the 75 g OGTT with different cut-offvalues as a diagnostic test since 1999,changed its recommendation on 2013.In Hongkong and Yunnan province in China, most hospital adopted the WHO 1999 criteria, nevertheless, there is no evidence of multicenter and large sample to show that the GDM population within the same ethnic group diagnosed by the new WHO criteria matches those diagnosed by the old one and consistency between the two criteria has not been reported. Thus,we conducted this study to analyze the two criteria in Chinese pregnant women.Weiwei Zhu Huixia Yang Yumei Wei Zilian Wang Xuelan Li Hairong Wu Nan Li Meihua Zhang Xinghui Liu Hua Zhang Yunhui Wang Jianmin Niu Yujie Gan Liruo Zhong Yunfeng Wang Anil Kapur 2015Chinese Medical Journal2015,,1:42
Characteristics of change in water quality along reclaimed water intake area of the Chaobai River in Beijing, China显示文摘To reveal the basic characteristics and controlling factors of water quality change in the project Wenyu to Chaobai reclaimed water diversion, the water quality in the study area was monitored for one year at seven monitoring sites. Inverse geochemical models of the statistical groups were developed using PHREEQC to elucidate the hydrochemistry characteristics of reclaimed water and the factors. The monitoring results indicated that nitrogen and phosphorus contents were significantly reduced along the river mainly caused by seasonal and location variation. The pH ranged from 7.44 to 9.81. Photosynthesis of algae and denitrification in anaerobic microenvironment ultimately led to a sudden p H increase after the Jian River and the Chaobai River confluence. Mg^(2+)and SO_4^(2-) levels dropped obviously in the summer and increased in winter seasons after intersection. Na+and Cl-are relatively stable, and marked drop in the concentration only after the two rivers meet. And there is a decrease of Ca^(2+) and HCO^(3-) and increase in CO_3^(2-) during monitoring period. As a whole, the primary ions and nutrient components, including nitrogen and phosphorus, had high levels in winter. Algae's photosynthesis and respiration were observed to have an impact on the river water quality; there was precipitation–dissolution of minerals and denitrification from upstream to downstream. Inverse geochemical PHREEQC modeling confirmed that there was precipitation of aragonite or calcite, and gypsum or anhydrite in summer, and dissolution in winter; as well as precipitation of dolomite in winter, and cationic exchange and denitrification along the river.Lei Yang Jiangtao He Yumei Liu Jian Wang Lie Jiang Guangcai Wang 2016Journal of Environmental Sciences2016,28,12:14
Heterotic Group Classification of 63 Inbred Lines and Hybrid Purity Identification by Using SSR Markers in Winter Cabbage(Brassica Oleracea L. var. capitata)显示文摘Winter cabbage is an important crop cultivated through winter in the region near the Yangtze River, enabling the supply of fresh cabbage there at that time of year. However, a problem has emerged regarding the newly developed parents of winter cabbage, which is completely different from spring and autumn cabbage, namely, how to combine these parents to breed an elite hybrid. To classify the heterotic groups and improve the efficiency of parent selection in winter cabbage breeding, 20 polymorphic SSR markers were selected to screen 63 winter cabbage inbred lines. Seventeen pairs among the 20 SSR markers amplified polymorphic bands. These primers amplified two to six bands, with an average of 2.8 bands per primer, and a total of 47 polymorphic bands were generated in the 63 inbred lines. These lines included flatheaded morphotype and round-headed morphotype, thus they were separately classified into heterotic groups based on the SSR markers. The flat-headed morphotype contained 21 inbred lines and was classified into three heterotic groups, named Hanchun 4, Jiali, and Dongsheng, in accordance with the representative germplasm contained in each group. The round-headed morphotype contained 42 inbred lines and was classified into five heterotic groups, named Parte, Bejo1039, YK-143, SCA002, and Golden B90. Meanwhile, parent analysis of 20 developed elite combinations showed that their parents were all distributed in different heterotic groups, indicating that the group classification was reasonable, which can provide a basis for further parent selection in winter cabbage breeding. Furthermore, polymorphic SSR primers were successfully used to identify the hybrid purity of three elite varieties.LI Xing YU Hailong LI Zhiyuan LIU Xiaoping FANG Zhiyuan LIU Yumei YANG Limei ZHUANG Mu LV Honghao ZHANG Yangyong 2018Horticultural Plant Journal2018,4,4:11
Identification of a senescence-related protease in coriander leaves显示文摘Senescence-related protease may play an important role in leaf senescence. By improved SDS-Gela-tin-PAGE assay, a 63 ku senescence-related protease (63 SRP) in coriander leaves was identified. Activity of 63 SRP was increased in parallel to the advance of coriander leaf senescence, and inhibited by treating the leaf with gibberellic acid, and enhanced by ethylene treatment. The 63 SRP was suggested to be a serine protease based on the fact that its activity was inhibited by the protease inhibitor PMSF. The optimal temperature for the activity of the 70 ku protease was 50°C. The maximal activity was observed at pH 6-9, some activity could be observed on the gel slices incubated at pH 5 or 11. The 63 SRP was partly purified by the way of ammonium sulfate precipitation and then gel slicing after gel elec-trophoresis.Weibo Jiang Xiangjuan Zhou Yumei Zhao Pengcheng Liu 2002Chinese Science Bulletin2002,47,13:10
Reduced EGFR signaling enhances cartilage destruction in a mouse osteoarthritis model显示文摘Osteoarthritis(OA) is a degenerative joint disease and a major cause of pain and disability in older adults. We have previously identified epidermal growth factor receptor(EGFR) signaling as an important regulator of cartilage matrix degradation during epiphyseal cartilage development. To study its function in OA progression, we performed surgical destabilization of the medial meniscus(DMM) to induce OA in two mouse models with reduced EGFR activity, one with genetic modification(EgfrWa5/1mice) and the other one with pharmacological inhibition(gefitinib treatment). Histological analyses and scoring at 3 months post-surgery revealed increased cartilage destruction and accelerated OA progression in both mouse models. TUNEL staining demonstrated that EGFR signaling protects chondrocytes from OA-induced apoptosis, which was further confirmed in primary chondrocyte culture. Immunohistochemistry showed increased aggrecan degradation in these mouse models, which coincides with elevated amounts of ADAMTS5 and matrix metalloproteinase 13(MMP13), the principle proteinases responsible for aggrecan degradation, in the articular cartilage after DMM surgery. Furthermore, hypoxia-inducible factor 2a(HIF2a), a critical catabolic transcription factor stimulating MMP13 expression during OA, was also upregulated in mice with reduced EGFR signaling. Taken together, our findings demonstrate a primarily protective role of EGFR during OA progression by regulating chondrocyte survival and cartilage degradation.Xianrong Zhang Ji Zhu Fei Liu Yumei Li Abhishek Chandra L Scott Levin Frank Beier Motomi Enomoto-Iwamoto Ling Qin 2014Bone Research2014,2,3:9
Optimized clinical practice for superaged patients with hip fracture:significance of damage control and enhanced recovery program显示文摘With the advent of global aging, the incidence, mortality, and medical costs of hip fracture among aged patients are increasing annually. The number of controlled clinical studies and health economics analyses that conform to evidence-based medicine principles is growing day by day. However, unfortunately,no specific recommendations regarding the procedures for the treatment of hip fracture are available.Meanwhile, the existence of both traditional treatment systems and new treatment theories means that most doctors confront difficult choices in their daily practice. These factors make the therapeutic approach for aged patients, especially among superaged patients with hip fracture, extremely challenging. This study focuses on superaged patients(>80 years as defined by the World Health Organization)with hip fracture and includes their preoperative pathological condition;therapeutic decision-making in terms of the benefit and risk ratio, damage control theory, and enhanced recovery after surgery were also investigated. These patients were discussed specifically by combining the current treatment strategies from several experts and the results of a meta-analysis published recently. The study presents some new ideas and approaches currently recognized in the field, such as preoperative assessment, surgical planning, safety consideration, complication intervention, and enhanced recovery implementation, and further presents some clear interpretations regarding misunderstandings in clinical practice. Finally, optimized treatment according to damage control principles and enhanced recovery after surgery during the perioperative period among superaged hip fracture patients is defined.Zaiyang Liu Jun Zhang Kaiqi He Yumei Zhang Yuan Zhang 2019Burns & Trauma2019,7,1:9
Interferon-inducible cytoplasmic lncLrrc55-AS promotes antiviral innate responses by strengthening IRF3 phosphorylation显示文摘Type I interferon (IFN-I) production is efficiently induced to ensure a potent innate immune response to viral infection. How this response can be enhanced, however, remains to be explored. Here, we identify a new cytoplasmic long non-coding RNA (lncRNA), lncLrrc55-AS, that drives a positive feedback loop to promote interferon regulatory factor 3 (IRF3) signaling and IFN-I production. We show that lncLrrc55-AS is virus-induced in multiple cell types via the IFN-JAK-STAT pathway. LncLrrc55-AS-deficient mice display a weakened antiviral immune response and are more susceptible to viral challenge. Mechanistically, lncLrrc55-AS binds phosphatase methylesterase 1 (PME-1), and promotes the interaction between PME-1 and the phosphatase PP2A, an inhibitor of IRF3 signaling. LncLrrc55-AS supports PME-1-mediated demethylation and inactivation of PP2A, thereby enhancing IRF3 phosphorylation and signaling. Loss of PME-1 phenocopies lncLrrc55-AS deficiency, leading to diminished IRF3 phosphorylation and IFN-I production. We have identified an IFN-induced lncRNA as a positive regulator of IFN-I production, adding mechanistic insight into lncRNA-mediated regulation of signaling in innate immunity and inflammation.Yumei Zhou Mengxuan Li Yiquan Xue Zhiqing Li Weitao Wen Xingguang Liu Yuanwu Ma Lianfeng Zhang Zhongyang Shen Xuetao Cao 2019Cell Research2019,29,8:8
Focal adhesion protein Kindlin-2 regulates bone homeostasis in mice显示文摘Our recent studies demonstrate that the focal adhesion protein Kindlin-2 is critical for chondrogenesis and early skeletal development. Here, we show that deleting Kindlin-2 from osteoblasts using the 2.3-kb mouse Col1 a1-Cre transgene minimally impacts bone mass in mice, but deleting Kindlin-2 using the 10-kb mouse Dmp1-Cre transgene, which targets osteocytes and mature osteoblasts, results in striking osteopenia in mice. Kindlin-2 loss reduces the osteoblastic population but increases the osteoclastic and adipocytic populations in the bone microenvironment. Kindlin-2 loss upregulates sclerostin in osteocytes, downregulates β-catenin in osteoblasts, and inhibits osteoblast formation and differentiation in vitro and in vivo. Upregulation ofβ-catenin in the mutant cells reverses the osteopenia induced by Kindlin-2 deficiency. Kindlin-2 loss additionally increases the expression of RANKL in osteocytes and increases osteoclast formation and bone resorption. Kindlin-2 deletion in osteocytes promotes osteoclast formation in osteocyte/bone marrow monocyte cocultures, which is significantly blocked by an anti-RANKLneutralizing antibody. Finally, Kindlin-2 loss increases osteocyte apoptosis and impairs osteocyte spreading and dendrite formation. Thus, we demonstrate an important role of Kindlin-2 in the regulation of bone homeostasis and provide a potential target for the treatment of metabolic bone diseases.Huiling Cao Qinnan Yan Dong Wang Yumei Lai Bo Zhou Qi Zhang Wenfei Jin Simin Lin Yiming Lei Liting Ma Yuxi Guo Yishu Wang Yilin Wang Xiaochun Bai Chuanju Liu Jian QFeng Chuanyue Wu Di Chen Xu Cao Guozhi Xiao 2020Bone Research2020,8,1:8
Expert consensus on dental caries management显示文摘Dental Caries is a kind of chronic oral disease that greatly threaten human being’s health.Though dentists and researchers struggled for decades to combat this oral disease, the incidence and prevalence of dental caries remain quite high. Therefore, improving the disease management is a key issue for the whole population and life cycle management of dental caries. So clinical difficulty assessment system of caries prevention and management is established based on dental caries diagnosis and classification.Dentists should perform oral examination and establish dental records at each visit. When treatment plan is made on the base of caries risk assessment and carious lesion activity,we need to work out patient-centered and personalized treatment planning to regain oral microecological balance,to control caries progression and to restore the structure and function of the carious teeth. And the follow-up visits are made based on personalized caries management. This expert consensus mainly discusses caries risk assessment, caries treatment difficulty assessment and dental caries treatment plan, which are the most important parts of caries management in the whole life cycle.Lei Cheng Lu Zhang Lin Yue Junqi Ling Mingwen Fan Deqin Yang Zhengwei Huang Yumei Niu Jianguo Liu Jin Zhao Yanhong Li Bin Guo Zhi Chen Xuedong Zhou 2022International Journal of Oral Science2022,14,2:8
Water quality characteristics and assessment of Yongding New River by improved comprehensive water quality identification index based on game theory显示文摘The Yongding New River is essential for the water supplies of Tianjin. To date, there is no comprehensive report that assesses the year-round water quality of the Yongding New River Main stream. Moreover, little attention has been given to determining a combined weight for improving the traditional comprehensive water quality identification index(ICWQII)by the game theory.Seven water quality parameters were investigated monthly along the main stream of the Yongding New River from May 2018 to April 2019.Organic contaminants and nitrogen pollution were mainly caused by point sources pollution, and the total phosphorus mainly by non-point source pollution.Dramatic spatio-temporal variations of water quality parameters were jointly caused by different pollutant sources and hydrometeorological factors. In terms of this study,an improved comprehensive water quality identification index(ICWQII)based on entropy weight or variation coefficient and traditional CWQII underestimated the water qualities, and an ICWQII based on the superstandard multiple method overvalued the assessments.By contrast, water qualities assessments done with an ICWQII based on the game theory matched perfectly with the practical situation. The ICWQII based on game theory proposed in this study takes into account not only the degree of disorder and variation of water quality data, but also the influence of standard-exceeded pollution indicators, whose results are relatively reasonable. All findings and the ICWQII based on game theory can provide scientific support for decisions related to the water environment management of the Yongding New River and other waters.Yu Liu Yucheng Hu Yumei Hu Yuqi Gao Zhenying Liu 2021Journal of Environmental Sciences2021,33,6:8
Utilization of Ogura CMS germplasm with the clubroot resistance gene by fertility restoration and cytoplasm replacement in Brassica oleracea L显示文摘Clubroot disease,a major plant root disease caused by Plasmodiophora brassicae, has become one of the most destructive diseases among cultivated cruciferous vegetables.However,clubroot-resistant Brassica oleracea materials are rare.A few clubroot-resistant cabbage varieties are available on the market, but all are Ogura cytoplasmic male sterile(CMS)types. Therefore,in this study,to reutilize the clubroot-resistant Ogura CMS germplasm of cabbage,a new fertility-restored Ogura CMS material,16Q2-11,was used as a bridge to transfer the clubroot resistance(CR)gene from the Ogura CMS cytoplasm to the normal cytoplasm by a two-step method(a fertility restoration and cytoplasm replacement method). In the first cross for fertility restoration of Ogura CMS clubroot-resistant cabbage(FRCRC),16Q2-11 was used as a restorer to cross with Ogura CMS materials containing the CR gene CRb2.Eleven Rfo-positive progenies were generated,of which four contained CRb2:F8-514,F8-620,F8-732 and F8-839.After inoculation with race 4 of P.brassicae, these four CRb2-positive individuals showed resistance.Furthermore,F8-514 and F8-839 were then used as male parents in the second cross of FRCRC to cross with cabbage inbred lines, resulting in the successful introgression of the CRb2 gene into the inbred lines. All offspring produced from this step of cross, which had a normal cytoplasm, showed a high resistance to race 4 of P.brassicae and could be utilized for the breeding of clubrootresistant cabbage varieties in the future. This is the first time that the Ogura CMS restorer has been used to restore the fertility of Ogura CMS clubroot-resistant cabbages, which could improve germplasm diversity in cabbage and provide a reference method for using CMS germplasm in Brassica crops.Wenjing Ren Zhiyuan Li Fengqing Han Bin Zhang Xing Li Zhiyuan Fang Limei Yang Mu Zhuang Honghao Lv Yumei Liu Yong Wang Hailong Yu Yangyong Zhang 2020Horticulture Research2020,7,1:7
Research progress of in-situ gelling ophthalmic drug delivery system显示文摘Blindness and vision impairment are the most devastating global health problems resulting in a substantial economic and social burden.Delivery of drug to particular parts of the anterior or posterior segment has been a major challenge due to various protective barriers and elimination mechanisms associated with the unique anatomical and physiological nature of the ocular system.Drug administration to the eye by conventional delivery systems results in poor ocular bioavailability(<5%).The designing of a novel approach for a safe, simple, and effective ocular drug delivery is a major concern and requires innovative strategies to combat the problem.Over the past decades, several novel approaches involving different strategies have been developed to improve the ocular delivery system.Among these, the ophthalmic in-situ gel has attained a great attention over the past few years. This review discussed and summarized the recent and the promising research progress of in-situ gelling in ocular drug delivery system.Yumei Wu Yuanyuan Liu Xinyue Li Dereje Kebebe Bing Zhang Jing Ren Jun Lu Jiawei Li Shouying Du Zhidong Liu 2019Asian Journal of Pharmaceutical Sciences2019,14,1:7
Facile synthesis of high electrical conductive CoP via solid-state synthetic routes for supercapacitors显示文摘Co-P precursor was prepared by a mechanical alloying method and then is controlled to synthesis of Co P phase through an annealing method. The optimal conditions of ball milling and annealing temperature are investigated. The Co P exhibits higher electrical conductivity than graphite and cobalt oxide, showing excellent pseudocapacitive properties due its high electrical conductivity which can result in a fast electron transfer in high rate charge–discharge possess. The as-obtained Co P electrode achieves a high specific capacitance of 447.5 F/g at 1 A/g, and displays an excellent rate capability as well as good cycling stability. Besides, the asymmetric supercapacitor(ASC) based on the Co P as the positive electrode and activated carbon(AC) as the negative electrode was assembled and displayed a high rate capability(60%of the capacitance is retained when the current density increased from 1 A/g to 12 A/g), excellent cycling stability(96.7% of the initial capacitance is retained after 5000 cycles), and a superior specific energy of19 Wh/kg at a power density of 350.8 W/kg. The results suggest that the Co P electrode materials have a great potential for developing high-performance electrochemical energy storage devices.Yumei Hu Maocheng Liu Qingqing Yang Lingbin Kong Long Kang 2017Journal of Energy Chemistry2017,26,1:7
Kindlin-2 inhibits Nlrp3 inflammasome activation in nucleus pulposus to maintain homeostasis of the intervertebral disc显示文摘Intervertebral disc(IVD) degeneration(IVDD) is the main cause of low back pain with major social and economic burdens;however, its underlying molecular mechanisms remain poorly defined. Here we show that the focal adhesion protein Kindlin-2 is highly expressed in the nucleus pulposus(NP), but not in the anulus fibrosus and the cartilaginous endplates, in the IVD tissues. Expression of Kindlin-2 is drastically decreased in NP cells in aged mice and severe IVDD patients. Inducible deletion of Kindlin-2 in NP cells in adult mice causes spontaneous and striking IVDD-like phenotypes in lumbar IVDs and largely accelerates progression of coccygeal IVDD in the presence of abnormal mechanical stress. Kindlin-2 loss activates Nlrp3 inflammasome and stimulates expression of IL-1β in NP cells, which in turn downregulates Kindlin-2. This vicious cycle promotes extracellular matrix(ECM) catabolism and NP cell apoptosis. Furthermore, abnormal mechanical stress reduces expression of Kindlin-2, which exacerbates Nlrp3 inflammasome activation, cell apoptosis, and ECM catabolism in NP cells caused by Kindlin-2 deficiency. In vivo blocking Nlrp3 inflammasome activation prevents IVDD progression induced by Kindlin-2 loss and abnormal mechanical stress. Of translational significance, adeno-associated virus-mediated overexpression of Kindlin-2 inhibits ECM catabolism and cell apoptosis in primary human NP cells in vitro and alleviates coccygeal IVDD progression caused by mechanical stress in rat. Collectively, we establish critical roles of Kindlin-2 in inhibiting Nlrp3 inflammasome activation and maintaining integrity of the IVD homeostasis and define a novel target for the prevention and treatment of IVDD.Sheng Chen Xiaohao Wu Yumei Lai Di Chen Xiaochun Bai Sheng Liu Yongchao Wu Mingjue Chen Yuxiao Lai Huiling Cao Zengwu Shao Guozhi Xiao 2022Bone Research2022,10,1:6
Construction and initial analysis of five Fosmid libraries of mitochondrial genomes of cotton(Gossypium)显示文摘Cotton(Gossypium)is an important crop providing textile fiber and edible oil.To gain the insights into mechanism of the cytoplasmic male sterility(CMS)inheritance,we constructed five fosmid libraries of mitochondrial genomes from mitotype of G.harknessii Brandegee.(one CMS line and its restorer),mitotype of G.hirsutum L.(one CMS line and its maintainer),and G.barbadense L.The numbers of the clones in these libraries ranged from 1152 to 2016 with an average insert size of 36.2 to 38.4 kb,equivalent to 70–119.3 mitogenomes. The libraries were screened with 28 markers derived from the conservative sequences and yielded 22,19,26,21,and 23 positive clones, respectively. These positive clones were used to construct the physical map of G.harknessii Brandegee.CMS line and G.barbadense L.mitogenomes that shared six syntenis regions.A total of 30 genes in nine clusters showed conservative and had high similarity with those in the mitochondrial genomes of cotton,Carica papaya,Cucurbita pepo and Nicotiana tabacum.Further investigation indicated that gene rrn26 had two copies in all five cotton mitogenomes, while genes atp1,rrn5 and rrn18 had two copies only in G.barbadense L.The positive clones and physical map are considered being useful resources in cotton genomics research.LI ShuangShuang LIU GuoZheng CHEN ZhiWen WANG YuMei LI PengBo HUA JinPing 2013Chinese Science Bulletin2013,58,36:5
Neuroprotective effects of combined lead and cadmium,as well as N-acetylcysteine,on cerebral cortical neurons following lipid peroxidation injury显示文摘BACKGROUND:Studies have reported the antioxidant effects of lead and cadmium in the central nervous system, but very few have addressed the combined toxicity of lead and cadmium. The mechanisms by which these combined heavy metals are toxic,as well as how to protect cells from these agents, remains poorly understood. OBJECTIVE:Primary cultured rat cortical neurons were used to determine the effects of combined lead and cadmium on levels of glutathione peroxidase(GSH-Px),superoxide dismutase(SOD), catalase(CAT),and acetylcholinesterase(AChE),as well as malondialdehyde(MDA),and to evaluate the neuroprotective effects of N-acetylcysteine(NAC). DESIGN,TIME AND SETTING:An in vitro toxicological observation was performed at the Comparative Medicine Center of Yangzhou University from August 2007 to April 2008. MATERIALS:Lead acetate, cadmium acetate, and NAC were purchased from Sigma-Aldrich(St. Louis,USA).Commercial kits of GSH-Px,SOD,CAT,AChE,and MDA were purchased from Nanjing Jiancheng Bioengineering Institute,Nanjing,China. METHODS:The cerebral cortical neurons were isolated from newborn Sprague dawley rats at 24 hours after birth and primary cultured for 6 days.Thereafter, the cells were treated with a range of cadmium doses(0,5.0,and 10.0μmol/L),lead doses(0,1.0,and 2.0μmol/L),or a combination of the two for 12 hours at 37℃in a 5%CO_2 incubator, respectively. In addition, the cells were incubated with different doses of cadmium and/or lead and(0 and 50μmol/L) NAC for 12 hours to assess the protective effects on cell survival. MAIN OUTCOME MEASURES:The activity of SOD,GSH-Px,CAT,and AChE,as well as MDA content,in the cell lysates was detected using commercial kits. RESULTS:At 12 hours after treatment, compared to the control group, activity of GSH-Px,SOD, and AChE in the lead, cadmium,or combined treated cells was significantly decreased with increasing doses of cadmium/or lead(P<0.05),but CAT activity and MDA levels were significantly increased(P<0.05).The combination of cadmium and lead led to higher levels of toxicity than individual exposure. CONCLUSION:The degree of oxidative damage increased when the two heavy metals were combined.NAC protected neonatal cortical neurons by increasing activity of anti-oxidative enzymes, and reducing lipid peroxidation, but the reduction was not statistically significant.Yumei Zhang Hao Lu Yan Yuan Xuezhong Liu Zongping Liu 2009Neural Regeneration Research2009,4,11:4

Growing vertical aligned mesoporous silica thin film on nanoporous substrate for enhanced degradation, drug delivery and bioactivity显示文摘Mesoporous silica thin film has been widely used in various fields, particularly the medical implant coating for drug delivery. However, some drawbacks remain with the films produced by traditional method(evaporation-induced self-assembly,EISA),such as the poor permeability caused by their horizontal aligned mesochannels. In this study, the vertical aligned mesoporous silica thin film(VMSTF)is uniformly grown alongside the walls of titania nanotubes array via a biphase stratification growth method, resulting in a hierarchical two-layered nanotubular structure.Due to the exposure of opened mesopores,VMSTF exhibits more appealing performances, including rapid degradation, efficient small-molecular drug(dexamethasone)loading and release, enhanced early adhesion and osteogenic differentiation of MC3T3-E1 cells. This is the first time successfully depositing VMSTF on nanoporous substrate and our findings suggest that the VMSTF may be a promising candidate for bone implant surface coating to obtain bioactive performances.Zhe Li Yide He Lasse Hyldgaard Klausen Ning Yan Jing Liu Fanghao Chen Wen Song Mingdong Dong Yumei Zhang 2021Bioactive Materials2021,6,5:4
