| 2 | Development of Hydro-Informatic Modelling System and its application显示文摘The understanding of hydrological cycle is the core of hydrology and the scientific base of water resources management. Meanwhile, simulation of hydrological cycle has long been regarded as an important tool for the assessment, utilization and protection of water resources. In this paper, a new tool named Hydro-Informatic Modelling System (HIMS) has been developed and introduced with case studies in the Yellow River Basin in China and 331 catchments in Australia. The case studies showed that HIMS can be employed as an integrated platform for hydrological simulation in different regions. HIMS is a modular based framework of hydrological model designed for different utilization such as flood forecasting, water resources planning and evaluating hydrological impacts of climate change and human activities. The unique of HIMS is its flexibility in providing alternative modules in the simulation of hydrological cycle, which successfully overcome the difficulties in the availability of input data, the uncertainty of parameters, and the difference of rainfall-runoff processes. | 3 | Cloning of fatty acid elongase1 gene and molecular identification of A and C genome in Brassica species显示文摘The fatty acid elongase 1 (FAE1) genes of Brassic napus were cloned from two cultivars, i.e. Zhong- shuan No. 9 with low erucic acid content, and Zhongyou 821 with high erucic acid content, using the degenerate PCR primers. The sequence analysis showed that there was no intron within the FAE1 genes. The FAE1 genes from Zhongyou 821 contained a coding sequence of 1521 nucleotides, and those cloned from Zhongshuan No. 9 contained a 1517 bp coding sequence. Alignment of the FAE1 sequences from Brassica rapa, B. oleracea and B. napus detected 31 single nucleotide polymorphic sites (2.03%), which resulted in 7 amino-acid substitutions. Further analysis indicated that 19 SNPs were genome-specific, of which, 95% were synonymous mutations. The nucleotide substitution at po- sition 1217 in the FAE1 genes led to a specific site of restricted cleavage. An AvrII cleavage site was present only in the C genome genes and absent in the A genome FAE1 genes. Digestion profile of the FAE1 sequences from B. rapa, B. oleracea and B. napus produced with AvrII confirmed that the FAE1 genes of B. oleracea origin was recognized and digested, while that of B. rapa origin could not. The results indicated that by AvrII cleavage it was possible to distinguish B. rapa from B. oleracea and be- tween the A and C genome of B. napus. | 5 | Long Chain Acyl-Coenzyme A Synthetase 4(BnLACS4) Gene from Brassica napus Enhances the Yeast Lipid Contents显示文摘Long-chain acyl-Coenzyme A(CoA) synthetases(LACSs) catalyze the formation of long-chain acyl-CoA,and play important roles in fatty acid metabolism including phospholipids,triacylglycerol(TAG) biosynthesis and fatty acid β-oxidation.Here,we report the characterization of a LACS gene from Brassica napus.It is highly homologous to Arabidopsis LACS4 and thus designated as BnLACS4.The cloned gene BnLACS4 could complement a LACS-deficient yeast strain YB525.It is mainly expressed in flowers and developing seeds where lipid biosynthesis is at high rate in Brassica napus.When transiently expressed in tobacco leaves,BnLACS4 is localized in endoplasmic reticulum(ER),the common site for eukaryotic pathway of lipid biosynthesis.Expression of BnLACS4 in the yeast strain pep4 increased its lipid content.Taken together,our results suggest that BnLACS4 may be involved in lipid biosynthesis in B.napus. | TAN Xiao-li ZHENG Xiang-feng ZHANG Zhi-yan WANG Zheng XIA Heng-chuan LU Changming GU Shou-lai | 2014 | Journal of Integrative Agriculture2014,13,1: | 4 |

| 10 | Reappraise role of lymph node status in patterns of recurrence following curative resection of gastric adenocarcinoma显示文摘Objective: To examine the association between lymph node status and recurrence patterns in completely resected gastric adenocarcinoma.Methods: We retrospectively assessed 1,694 patients who underwent curative gastrectomy from January 2010 to August 2014. Patients stratified according to lymph node status and recurrence patterns among different subgroups were compared.Results: Of all, 517(30.5%) patients developed recurrent disease, and complete data of recurrence could be obtained in 493(95.4%) patients. For p^(N0) patients, the patterns of recurrence were different according to p T stage: locoregional recurrence was most common in patients with p T1-2 disease(57.1%), distant recurrence was most common in patients with p T3 disease(57.1%), and peritoneal recurrence was most common in patients with p T4 a disease(66.7%). For p^(N+) patients, distant metastasis was most common pattern irrespective of p T stage. The site-specific trend of recurrence showed that locoregional recurrence increased within 5 years in patients with p^(N0)-2 disease but plateaued 3 years after surgery in patients with p N3 disease. Time to recurrence was significantly longer for the p^(N0) patients compared with the p^(N+) patients(median: 25 vs. 16 months, P=0.001).Moreover, post-recurrence survival was significantly better for the p^(N0) patients than for the p^(N+) patients(median:12 vs. 6 months, P<0.001), especially in patients with non-peritoneal recurrence, late recurrence, single recurrence,and receipt of potential curative treatment.Conclusions: Among clinicopathologic factors, lymph node status is the most important factor associated with recurrence patterns after curative gastrectomy. | 11 | Analysis on virus resistance and fruit quality for T4 generation of transgenic papaya显示文摘Molecular biological characterization,fruit char-acters,and nutrients were analyzed for T4 generation of transgenic papaya.All transgenic papaya plants with the mutated replicase(RP)gene from papaya ringspot virus(PRSV)showed high resistance or immunity against PRSV in the field.The RP transgene can be steadily inherited to,and expressed at RNA level,the progenies.The growth char-acteristics of transgenic papaya were much better than non-transgenic papaya in the field.The non-transgenic papaya seedlings began to show typical symptoms caused by PRSV after being inoculated with PRSV.They died quickly and never grew to produce fruit.The adult trees developed yellow leaves and produced smaller fruits and were doomed to a slow death after some time,while most of transgenic papaya plants(about 91.8%)did not show any symptoms caused by PRSV,and produced more,bigger,and high quality fruits.Compared with non-transgenic plants,the fresh fruit length of T4 gen-eration of transgenic papaya increased 2.6%-5%,and the diameter decreased 0.6%-1.5%.The flesh thickness of fresh fruit increased 12%-15%,which made it fitter for eating.Although the fresh fruit quality changed,there was no sig-nificant difference between transgenic and non-transgenic papaya.The quality characteristics of dry fruit including the contents of water,lipid,N,protein,reduced sugar,vitamin A,vitamin C,and carotene in the T4 generation of transgenic papaya were all the same as their non-transgenic parents.This means that transgenic plants and non-transgenic plants are substantially equivalent,and the transgene has no effect on dry fruit quality.In this study,we found that vitamin A and vitamin C in red-fleshed papaya were 1.4-1.8 and 1.78-2.07 times more than the yellow-fleshed ones,respectively,while N and protein were only 84.2%-92.1%and 82.1%-98.9%of the yellow-fleshed ones. | WEI Xiangdong LAN Congyu LU Zhijing YE Changming | 2007 | Frontiers in Biology2007,2,3: | 1 |
