3Molecular dissection of plant height QTLs using recombinant inbred lines from hybrids between common wheat (Triticum aestivum L.) and spelt wheat (Triticum spelta L.)显示文摘In wheat, plant height is an important agronomic trait, and a number of quantitative trait loci (QTLs) controlling plant height have been located. In this study, using the conditional and unconditional QTL mapping methods, combined with data from five different growth stages over two years of field trials, the developmental behavior for plant height in wheat was dissected. Nine unconditional QTLs and 8 conditional QTLs were identified, of which 6 were detected by both methods. None of the 11 QTLs was detected at all of the 5 investigated developmental stages, but 7 QTLs were detected at certain stages in both years. Further analysis identified 9 unconditional QTLs at different stages, which could explain the phenotypic variation from 4.81% to 17.35%. It was noteworthy that one major QTL designated QHt-4B-2, which was located on chromosome 4B, was detected on May 18 and 25 in both years, and its genetic contributions to plant height ranged from 13.42% to 16.13%. Moreover, of the 8 conditional QTLs identified, six were detected in both years, in the order of QHt-3B→QHt-4B-1→QHt-4B-2→QHt-4D→QHt-5A and QHt-2B expressed at the same developmental stage. 4Genomic and genic sequence variation in synthetic hexaploid wheat (AABBDD) as compared to their parental species显示文摘In order to understand the genomic changes during the evolution of hexaploid wheat, two sets of synthetic hexaploid wheat from hybridization between maternal tetraploid wheat (AABB) and paternal diploid goat grass (DD) were used for DNA-AFLP and single strand conformation polymorphism (SSCP) analysis to determine the genomic and genic variation in the synthetic hexaploid wheat. Results indicated that more DNA sequences from paternal diploid species were eliminated in the synthetic hexaploid wheat than from maternal tetraploid wheat, suggesting that genome from parental species of lower ploidity tends to be eliminated preferentially. However, sequence variation detected by SSCP procedure was much lower than those detected by DNA-AFLP, which indicated that much less variation in the genic regions occurred in the synthetic hexaploid wheat, and sequence variations detected by DNA-AFLP could be derived mostly from non-coding regions and repetitive sequences. Our results also indicated that sequence variation in 4 genes can be detected in hybrid F1, which suggested that this type of sequence variation could be resulted from distant hybridization. 5A universal pipeline for mobile mRNA detection and insights into heterografting advantages under chilling stress显示文摘Heterografting has long been used to enhance the chilling tolerance of temperature-sensitive crops,including watermelon,whose mechanism is known to involve bidirectional long-distance mRNA movements.Despite several studies reporting on mobile mRNA(mb-mRNA)profiles in plants,accurate identification of mb-mRNAs is challenging owing to an array of technical problems.Here,we developed a bioinformatical pipeline that took most of the known technical concerns into consideration and is considered to be a universal tool for mb-mRNA detection in heterografts.By applying this pipeline to a commercial watermelon–bottle gourd heterografting system,we detected 130 and 1144 mb-mRNAs upwardly and 167 and 1051 mb-mRNAs downwardly transmitted under normal and chilling-stress conditions,respectively.Quantitative real-time PCR indicated a high accuracy rate(88.2%)of mb-mRNA prediction with our pipeline.We further revealed that the mobility of mRNAs was not associated with their abundance.Functional annotation and classification implied that scions may convey the stress signal to the rootstock,subsequently triggering energy metabolism reprogramming and abscisic acid-mediated stress responses by upward movement of effective mRNAs,ultimately leading to enhanced chilling tolerance.This study provides a universal tool for mb-mRNA detection in plant heterografting systems and novel insights into heterografting advantages under chilling stress.Ying Wang Lingping Wang Nailin Xing Xiaohua Wu Xinyi Wu Baogen Wang Zhongfu Lu Pei Xu Ye Tao Guojing Li Yuhong Wang 2020Horticulture Research2020,7,1:3

7Oxygen Permeation and Stability of Ce_(0.8)Gd_(0.2)O_(2-δ)-PrBaCo_(2-x)FexO_(5+δ) Dual-phase Composite Membranes显示文摘Dual-phase membranes of 60 wt% Ce_(0.8)Gd_(0.2)O_(2-δ)-40 wt% Pr Ba Co2exFexO3 d(0 x 2) were prepared by a combined citrate and ethylene diamine tetraacetic acid(EDTA) complexing method. X-ray diffraction(XRD)results revealed the good chemical compatibility between ion-conducting phase CGO and electron-conducting phases PBC2 xFxO after sintering in air. The Fe ionic dopant had a significant effect on the phase structure stability and oxygen permeability under CO2 atmosphere, which was confirmed by XRD, thermogravimetrye differential scanning calorimetry(TGeD SC), scanning electron microscopy(SEM) and oxygen permeation experiments. CGOeP BC0.5F1.5O dual-phase membrane demonstrated a stable oxygen permeation flux of2.71x10-7mol cm 2s 1with 50 mol% He/CO2 as the sweep gas at 925 C, and this value was much higher than that of perovskite-type membranes. The rate-limiting step in the oxygen permeation process changed from the bulk diffusion to the surface oxygen exchange when the CGOeP BC0.5F1.5O membrane thickness decreased to 0.8 mm or less. 10Development of Insertion and Deletion Markers for Bottle Gourd Based on Restriction Site-associated DNA Sequencing Data显示文摘Bottle gourd is an important cucurbit crop worldwide. To provide more available molecular markers for this crop, a bioinformatic approach was employed to develop insertion–deletions(In Dels) markers in bottle gourd based on restriction site-associated DNA sequencing(RAD-Seq)data. A total of 892 Indels were predicted, with the length varying from 1 bp to 167 bp. Single-nucleotide In Dels were the predominant types of In Dels. To validate these In Dels, PCR primers were designed from 162 loci where In Dels longer than 2 bp were predicated. A total of 112 In Dels were found to be polymorphic among 9 bottle gourd accessions under investigation. The rate of prediction accuracy was thus at a high level of72.7%.
