Adenovirus-mediated gene delivery:Potential applications for gene and cell-based therapies in the new era of personalized medicine显示文摘With rapid advances in understanding molecular pathogenesis of human diseases in the era of genome sciences and systems biology,it is anticipated that increasing numbers of therapeutic genes or targets will become available for targeted therapies.Despite numerous setbacks, efficacious gene and/or cell-based therapies still hold the great promise to revolutionize the clinical management of human diseases. It is wildly recognized that poor gene delivery is the limiting factor for most in vivo gene therapies. There has been a long-lasting interest in using viral vectors, especially adenoviral vectors,to deliver therapeutic genes for the past two decades.Among all currently available viral vectors, adenovirus is the most efficient gene delivery system in a broad range of cell and tissue types. The applications of adenoviral vectors in gene delivery have greatly increased in number and efficiency since their initial development. In fact, among over 2000 gene therapy clinical trials approved worldwide since 1989,a significant portion of the trials have utilized adenoviral vectors. This review aims to provide a comprehensive overview on the characteristics of adenoviral vectors, including adenoviral biology, approaches to engineering adenoviral vectors, and their applications in clinical and preclinical studies with an emphasis in the areas of cancer treatment, vaccination and regenerative medicine.Current challenges and future directions regarding the use of adenoviral vectors are also discussed. It is expected that the continued improvements in adenoviral vectors should provide great opportunities for cell and gene therapies to live up to its enormous potential in personalized medicine.Cody S.Lee Elliot S.Bishop Ruyi Zhang Xinyi Yu Evan M.Farina Shujuan Yan Chen Zhao Zongyue Zeng Yi Shu Xingye Wu Jiayan Lei Yasha Li Wenwen Zhang Chao Yang Ke Wu Ying Wu Sherwin Ho Aravind Athiviraham Michael J.Lee Jennifer Moriatis Wolf Russell R.Reid Tong-Chuan He 2017Genes & Diseases2017,4,2:19
Influence of physical properties of carrier on the performance of dry powder inhalers显示文摘Dry powder inhalers(DPIs) offer distinct advantages as a means of pulmonary drug delivery and have attracted much attention in the field of pharmaceutical science. DPIs commonly contain micronized drug particles which, because of their cohesiveness and strong propensity to aggregate, have poor aerosolization performance. Thus carriers with a larger particle size are added to address this problem. However, the performance of DPIs is profoundly influenced by the physical properties of the carrier, particularly their particle size, morphology/shape and surface roughness. Because these factors are interdependent, it is difficult to completely understand how they individually influence DPI performance. The purpose of this review is to summarize and illuminate how these factors affect drug–carrier interaction and influence the performance of DPIs.Tingting Peng Shiqi Lin Boyi Niu Xinyi Wang Ying Huang Xuejuan Zhang Ge Li Xin Pan Chuanbin Wu 2016Acta Pharmaceutica Sinica B2016,6,4:8
Spatial-temporal Evolution and Determinants of the Belt and Road Initiative: A Maximum Entropy Gravity Model Approach显示文摘The spatial interaction model is an effective way to explore the geographical disparities inherent in the Belt and Road Initiative(BRI) by simulating spatial flows. The traditional gravity model implies the hypothesis of equilibrium points without any reference to when or how to achieve it. In this paper, a dynamic gravity model was established based on the Maximum Entropy(MaxEnt) theory to estimate and monitor the interconnection intensity and dynamic characters of bilateral relations. In order to detect the determinants of interconnection intensity, a Geodetector method was applied to identify and evaluate the determinants of spatial networks in five dimensions. The empirical study clearly demonstrates a heterogeneous and non-circular spatial structure. The main driving forces of spatial-temporal evolution are foreign direct investment, tourism and railway infrastructure construction, while determinants in different sub-regions show obvious spatial differentiation. Southeast Asian countries are typically multi-island area where aviation infrastructure plays a more important role. North and Central Asian countries regard oil as a pillar industry where power and port facilities have a greater impact on the interconnection. While Western Asian countries are mostly influenced by the railway infrastructure, Eastern European countries already have relatively robust infrastructure where tariff policies provide a greater impetus.HUANG Qinshi ZHU Xigang LIU Chunhui WU Wei LIU Fengbao ZHANG Xinyi 2020Chinese Geographical Science2020,30,5:7
Regulation of non-classical immune parameters in immune thrombocytopenic purpura mice by a spleen-invigorating,qi-replenishing and blood-containing formula显示文摘Objective:This study investigated the regulatory effect of non-classical immune parameters on immune thrombocytopenic purpura(ITP)mice by a spleen-invigorating,qi-replenishing and blood-containing formula(SQBF).Method:A total of 80 BALB/c mice were randomly divided into four equal groups(20 mice each):control group, model group, prednisone group and spleen-invigorating,qi-replenishing and blood-containing(SQBF)group.Mice in the model group, prednisone group, and SQBF group were administered anti-platelet serum to induce ITP.The dynamic variations of platelet counts in ITP mice were measured with an automatic blood analyzer before modeling and 48 h,and 8,12 and 15 days following APS injection.Levels of b-endorphin(b-EP),vasoactive intestinal peptide(VIP)and salivary IgA(SIgA)were detected by enzyme-linked immunosorbent assay(ELISA)on 15th day of experiment. Results:SQBF enhanced peripheral blood platelet counts in ITP mice similar to that of prednisone, and both groups showed a statistically significant response compared with the model group(P<.01).The SQBF significantly decreased b-EP levels compared with the model and prednisone intervention groups(P<.05),significantly increased the levels of VIP and SIgA in ITP mice compared with the model group(P<.05)and had significant protective effects on the thymus of ITP mice compared with the model group(P<.01).Conclusions:The SQBF had a similar effect to prednisone with regards to enhancing peripheral blood platelet counts in ITP mice.Furthermore,it decreased b-EP levels and increased VIP and SIgA,and protected the thymus. This shows that,on base of the brain-gut axis functions, some non-classical immune vascular active factors or neurotransmitters are also involved in immune responses, and also have relationship with the onset of ITP and bleeding and/or hemostasis. It needs further study to determine whether a change in these active factors is related to immediate hemostasis.Tiantian Li Hao He Li Hou Yue Xu Xiaoyong Wu Yanping Sun Lei Zheng Zhixiong Chen Xinyi Chen Bei Qin 2015Journal of Traditional Chinese Medical Sciences2015,2,2:6
Establishment and functional characterization of the reversibly immortalized mouse glomerular podocytes(imPODs)显示文摘Glomerular podocytes are highly specialized epithelial cells and play an essential role in establishing the selective permeability of the glomerular filtration barrier of kidney.Maintaining the viability and structural integrity of podocytes is critical to the clinical management of glomerular diseases, which requires a thorough understanding of podocyte cell biology. As mature podocytes lose proliferative capacity,a conditionally SV40 mutant tsA58-immortalized mouse podocyte line(designated as tsPC)was established from the Immortomouse over 20 years ago. However, the utility of the tsPC cells is hampered by the practical inconvenience of culturing these cells. In this study,we establish a user-friendly and reversibly-immortalized mouse podocyte line(designated as imPOD),on the basis of the tsPC cells by stably expressing the wildtype SV40 T-antigen,which is flanked with FRT sites. We show the imPOD cells exhibit long-term high proliferative activity, which can be effectively reversed by FLP recombinase. The imPOD cells express most podocyte-related markers, including WT-1,Nephrin,Tubulin and Vinculin, but not differentiation marker Synaptopodin. The imPOD cells do not form tumor-like masses in vivo.We further demonstrate that TGFb1 induces a podocyte injury-like response in the FLP-reverted imPOD cells by suppressing the expression of slit diaphragm-associated proteins P-Cadherin and ZO-1 and upregulating the expression of mesenchymal markers,a-SMA,Vimentin and Nestin,as well as fibrogenic factors CTGF and Col1a1.Collectively,our results strongly demonstrate that the newly engineered im-POD cells should be a valuable tool to study podocyte biology both under normal and under pathological conditions.Xinyi Yu Liqun Chen Ke Wu Shujuan Yan Ruyi Zhang Chen Zhao Zongyue Zeng Yi Shu Shifeng Huang Jiayan Lei Xiaojuan Ji Chengfu Yuan Linghuan Zhang Yixiao Feng Wei Liu Bo Huang Bo Zhang Wenping Luo Xi Wang Bo Liu Rex C.Haydon Hue H.Luu Tong-Chuan He Hua Gan 2018Genes & Diseases2018,5,2:5
TPGS/hyaluronic acid dual-functionalized PLGA nanoparticles delivered through dissolving microneedles for markedly improved chemo-photothermal combined therapy of superficial tumor显示文摘Nanoparticles(NPs)have shown potential in cancer therapy, while a single administration conferring a satisfactory outcome is still unavailable. To address this issue, the dissolving microneedles(DMNs)were developed to locally deliver functionalized NPs with combined chemotherapy and photothermal therapy(PTT).α-Tocopheryl polyethylene glycol succinate(TPGS)/hyaluronic acid(HA)dualfunctionalized PLGA NPs(HD10 NPs)were fabricated to co-load paclitaxel and indocyanine green.HD10 NPs significantly enhanced the cytotoxicity of low-dose paclitaxel because of active and mitochondrial targeting by HA and TPGS,respectively.PTT could further sensitize tumor cells toward chemotherapy by promoting apoptosis into the advanced period, highly activating caspase 3 enzyme, and significantly reducing the expression of survivin and MMP-9 proteins.Further, the anti-tumor effects of HD10 NPs delivered through different administration routes were conducted on the 4 T1 tumorbearing mice. After a single administration,HD10 NPs delivered with DMNs showed the best antitumor effect when giving chemotherapy alone. As expected, the anti-tumor effect was profoundly enhanced after combined therapy, and complete tumor ablation was achieved in the mice treated with DMNs and intra-tumor injection. Moreover,DMNs showed better safety due to moderate hyperthermia. Therefore, the DMNs along with combined chemo-photothermal therapy provide a viable treatment option for superficial tumors.Tingting Peng Yao Huang Xiaoqian Feng Chune Zhu Shi Yin Xinyi Wang Xuequn Bai Xin Pan Chuanbin Wu 2021Acta Pharmaceutica Sinica B2021,11,10:4
The development of a sensitive fluorescent protein-based transcript reporter for high throughput screening of negative modulators of lncRNAs显示文摘While the human genome is pervasively transcribed,<2%of the human genome is transcribed into protein-coding mRNAs,leaving most of the transcripts as noncoding RNAs,such as microRNAs and long-noncoding RNAs(lncRNAs),which are critical components of epigenetic regulation.lncRNAs are emerging as critical regulators of gene expression and genomic stability. However,it remains largely unknown about how lncRNAs are regulated. Here,we develop a highly sensitive and dynamic reporter that allows us to identify and/or monitor negative modulators of lncRNA transcript levels in a high throughput fashion.Specifically,we engineer a fluorescent fusion protein by fusing three copies of the PEST destruction domain of mouse ornithine decarboxylase(MODC)to the C-terminal end of the codon-optimized bilirubin-inducible fluorescent protein, designated as dBiFP,and show that the dBiFP protein is highly destabilized, compared with the commonly-used eGFP protein. We further demonstrate that the dBiFP signal is effectively down-regulated when the dBiFP and mouse lncRNA H19 chimeric transcript is silenced by mouse H19-specific siRNAs.Therefore, our results strongly suggest that the dBiFP fusion protein may serve as a sensitive and dynamic transcript reporter to monitor the inhibition of lncRNAs by microRNAs,synthetic regulatory RNA molecules,RNA binding proteins,and/or small molecule inhibitors so that novel and efficacious inhibitors targeting the epigenetic circuit can be discovered to treat human diseases such as cancer and other chronic disorders.Zongyue Zeng Bo Huang Shifeng Huang Ruyi Zhang Shujuan Yan Xinyi Yu Yi Shu Chen Zhao Jiayan Lei Wenwen Zhang Chao Yang Ke Wu Ying Wu Liping An Xiaojuan Ji Cheng Gong Chengfu Yuan Linghuan Zhang Wei Liu Yixiao Feng Bo Zhang Zhengyu Dai Yi Shen Xi Wang Wenping Luo Rex C.Haydon Hue H.Luu Lan Zhou Russell R.Reid Tong-Chuan He Xingye Wu 2018Genes & Diseases2018,5,1:4
PK-tailored tertiary prophylaxis in patients with severe hemophilia A at beijing Children’s Hospital显示文摘Importance:Tertiary prophylaxis using a low-dose regimen is usually insufficient to prevent recurrent joint bleeding and deterioration in joint diseases in children with severe hemophilia A.Pharmacokinetic(PK)dosing is a useful approach to increase the precision and efficiency of prophylaxis. Objective:To explore the efficacy of PK-tailored tertiary prophylaxis in children with severe hemophilia A.Methods:We implemented a PK-tailored tertiary prophylaxis program for 15 boys with severe hemophilia A aged 5-16 years at Beijing Children’s Hospital.Following PK testing and a 6-month evaluation period(phase I),15 patients were divided in two groups according to individual PK data and actual bleeding:(1)a PK-tailored group[modified prophylaxis regimen according to PK data for the next 6 months(phase II);n=8]and(2)a maintenance group(continued the original regimen for the next 6 months;n=7).We compared the bleeding rate, infusion frequency, and factor VIII(FVIII)consumption between the two groups.results:In the PK-tailored group, the median annual joint bleeding rate was reduced from 7.8 in phase I to 1.4 in phase II,mean annual total factor consumption increased from 1619.0 IU/kg in phase I to 2401.9 IU/kg in phase II,and median infusion frequency for prophylaxis increased from 104 times/year in phase I to 156 times/year in phase II(P<0.05).Although the FVIII consumption increased,it remained at approximately half of the standard method.Interpretation:PK-tailored prophylaxis may represent a more efficient approach to individual prophylaxis in China, but further studies are required to verify this.Peijing Li Zhenping Chen Xiaoling Cheng Yan Wang Ningning Zhang Yingzi Zhen Xinyi Wu Runhui Wu 2019Pediatric Investigation2019,3,1:3
A universal pipeline for mobile mRNA detection and insights into heterografting advantages under chilling stress显示文摘Heterografting has long been used to enhance the chilling tolerance of temperature-sensitive crops, including watermelon, whose mechanism is known to involve bidirectional long-distance mRNA movements.Despite several studies reporting on mobile mRNA(mb-mRNA)profiles in plants, accurate identification of mb-mRNAs is challenging owing to an array of technical problems. Here,we developed a bioinformatical pipeline that took most of the known technical concerns into consideration and is considered to be a universal tool for mb-mRNA detection in heterografts.By applying this pipeline to a commercial watermelon–bottle gourd heterografting system,we detected 130 and 1144 mb-mRNAs upwardly and 167 and 1051 mb-mRNAs downwardly transmitted under normal and chilling-stress conditions, respectively.Quantitative real-time PCR indicated a high accuracy rate(88.2%)of mb-mRNA prediction with our pipeline. We further revealed that the mobility of mRNAs was not associated with their abundance.Functional annotation and classification implied that scions may convey the stress signal to the rootstock, subsequently triggering energy metabolism reprogramming and abscisic acid-mediated stress responses by upward movement of effective mRNAs,ultimately leading to enhanced chilling tolerance. This study provides a universal tool for mb-mRNA detection in plant heterografting systems and novel insights into heterografting advantages under chilling stress.Ying Wang Lingping Wang Nailin Xing Xiaohua Wu Xinyi Wu Baogen Wang Zhongfu Lu Pei Xu Ye Tao Guojing Li Yuhong Wang 2020Horticulture Research2020,7,1:3
Adaptive cavity-enhanced dual-comb spectroscopy显示文摘Resolution and bandwidth are critical for cavity-enhanced dual-comb spectroscopy(CE-DCS). Here, we pioneer an adaptive approach in CE-DCS to improve the broadband as well as the resolution. Postcorrections to dualcomb interferograms adaptively compensate the relative phase jitters of the optical frequency combs and result in both a mode-resolved spectral resolution and a signal-to-noise ratio of 440:1 in 1 s. Meanwhile, an adaptive combcavity locking scheme exploits more than 90% of the comb modes, covering 340 cm-1(10 THz) at 6450 cm-1. For a single dual-comb interferogram, more than 40,000 comb teeth spaced by 250 MHz are measured in less than7.5 ms, contributing to a noise equivalent absorption per spectral element of 2 × 10-10 cm-1·Hz-1∕2. This adaptive cavity-enhanced dual-comb spectroscopy technique provides an attractive spectroscopic tool that may be utilized in trace-gas sensing, breath and cancer analysis, and engine combustion diagnosis.WEIPENG ZHANG XINYI CHEN XUEJIAN WU YAN LI HAOYUN WEI 2019Photonics Research2019,7,8:3
Rapid, visual, and equipment-free point-of-care testing for Staphylococcus aureus by direct recombinase polymerase amplification with SYBR GreenΙ显示文摘Food-borne diseases mainly caused by bacterial pathogens and the safety of genetically modified(GM)food have become global concerns[1].As a leading bacterial pathogen,Staphylococcus aureus can not only cause a variety of diseases[2]but also contaminate different kinds of animal food products[3].In addition,GM foods whose impact on human health and the natural environment is still a matter of controversy have gradually entered the commercial markets[4].Therefore,many countries have decided to implement labeling regulations for authorized genetically modified organisms(GMOs)to protect the consumers’right to know[5].Hence,it is critical to develop a rapid,simple,equipment-free,and on-site testing method to detect the GM contents in food samples and pathogen to ensure food safety.Xiaorui Fan Fangti Han Binan Zhao Yan Xu Xiao Zhao Xinyi Pu Yanan Du Qi Zhang Xiaoxia Zhang Wanjing Zhang Wenjing Wu Zhiwei Chen Kai Zhao 2021Acta Biochimica et Biophysica Sinica2021,53,9:3
Surface Oxygen Injection in Tin Disulfide Nanosheets for Efficient CO_(2) Electroreduction to Formate and Syngas显示文摘Surface chemistry modification represents a promising strategy to tailor the adsorption and activation of reaction intermediates for enhancing activity.Herein,we designed a surface oxygen-injection strategy to tune the electronic structure of SnS_(2) nanosheets, which showed effectively enhanced electrocatalytic activity and selectivity of CO_(2) reduction to formate and syngas(CO and H_(2)).The oxygen-injection SnS_(2) nanosheets exhibit a remarkable Faradaic efficiency of 91.6%for carbonaceous products with a current density of 24.1 mA cm^(−2) at−0.9 V vs RHE,including 83.2%for formate production and 16.5%for syngas with the CO/H_(2) ratio of 1:1.By operando X-ray absorption spectroscopy,we unravel the in situ surface oxygen doping into the matrix during reaction, thereby optimizing the Sn local electronic states.Operando synchrotron radiation infrared spectroscopy along with theoretical calculations further reveals that the surface oxygen doping facilitated the CO_(2) activation and enhanced the affinity for HCOO*species. This result demonstrates the potential strategy of surface oxygen injection for the rational design of advanced catalysts for CO_(2) electroreduction.Tao Chen Tong Liu Tao Ding Beibei Pang Lan Wang Xiaokang Liu Xinyi Shen Sicong Wang Dan Wu Dong Liu Linlin Cao Qiquan Luo Wei Zhang Wenkun Zhu Tao Yao 2021Nano-Micro Letters2021,13,11:3
Dietary Enteromorpha polysaccharide-Zn supplementation regulates amino acid and fatty acid metabolism by improving the antioxidant activity in chicken显示文摘Background:Enteromorpha prolifera(E.prolifera)polysaccharide has become a promising feed additive with a variety of physiological activities, such as anti-oxidant,anti-cancer,anti-diabetic,immunomodulatory,hypolipidemic,and cation chelating ability. However, whether Enteromorpha polysaccharide-trace element complex supplementation regulates amino acid and fatty acid metabolism in chicken is largely unknown. This study was conducted to investigate the effects of E.prolifera polysaccharide(EP)-Zn supplementation on growth performance, amino acid, and fatty acid metabolism in chicken. Methods:A total of 184 one-day-old Ross-308 broiler chickens were randomly divided into two treatment groups with 8 replicates,12 chickens per replicate, and fed either the basal diet(control group)or basal diet plus E.prolifera polysaccharide-Zinc(400 mg EP-Zn/kg diet). Results:Dietary EP-Zn supplementation significantly increased(P<0.05)the body weight, average daily gain, muscle antioxidant activity, serum HDL level, and reduced serum TG and LDL concentration. In addition, dietary EPZn supplementation could modulate ileal amino acid digestibility and upregulate the mRNA expression of amino acid transporter genes in the jejunum, ileum, breast muscle, and liver tissues(P<0.05).Compared with the control group, breast meat from chickens fed EP-Zn had higher(P<0.05)Pro and Asp content, and lower(P<0.05)Val,Phe,Gly,and Cys free amino acid content.Furthermore,EP-Zn supplementation upregulated(P<0.05)the mRNA expressions of mTOR and anti-oxidant related genes, while down-regulated protein degradation related genes in the breast muscle.Breast meat from EP-Zn supplemented group had significantly lower(P<0.05)proportions ofΣn-3 PUFA,and a higher percentage ofΣn-6 PUFA and the ratio of n-6/n-3 PUFA.Besides,EP-Zn supplementation regulated lipid metabolism by inhibiting the gene expression of key enzymes involved in the fatty acid synthesis and activating genes that participated in fatty acid oxidation in the liver tissue. Conclusions:It is concluded that EP-Zn complex supplementation regulates apparent ileal amino acid digestibility, enhances amino acid metabolism, and decreases oxidative stress-associated protein breakdown, thereby improving the growth performance.Furthermore,it promotes fatty acid oxidation and restrains fat synthesis through modulating lipid metabolism-related gene expression.Teketay Wassie Xinyi Duan Chunyan Xie Ruxia Wang Xin Wu 2022Journal of Animal Science and Biotechnology2022,13,3:3
Two-step Optimal Allocation of Stationary and Mobile Energy Storage Systems in Resilient Distribution Networks显示文摘Energy storage systems (ESSs) are acknowledged to be a promising option to cope with issues in high penetration of renewable energy and guarantee a highly reliable power supply. In this paper, a two-step optimal allocation model is proposed to obtain the optimal allocation (location and size) of stationary ESSs (SESSs) and mobile ESSs (MESSs) in the resilient distribution networks (DNs). In the first step, a mixed-integer linear programming (MILP) problem is formulated to obtain the preselected location of ESSs with consideration of different scenarios under normal operation conditions. In the second step, a two-stage robust optimization model is established to get the optimal allocation results of ESSs under failure operation conditions which are solved by column-and-constraint generation (C&CG) algorithm. A hybrid ESS allocation strategy based on the subjective and objective weight analysis is proposed to give the final allocation scheme of SESSs and MESSs. Finally, the proposed two-step optimal allocation model is demonstrated on a modified IEEE 33-bus system to show its effectiveness and merits.Xinyi Jiang Jian Chen Qiuwei Wu Wen Zhang Yicheng Zhang Jie Liu 2021Journal of Modern Power Systems and Clean Energy2021,9,4:2
Efficient and secure image authentication with robustness and versatility显示文摘Image authentication is the technology of verifying image origin, integrity and authenticity.A rich stream of research on image authentication has shown various trade-off among four favorable features, namely robustness, security, versatility and efficiency.Image data authentication has the highest level of security but provides no robustness/versatility.Image content authentication from robust hashing keeps robust to limited types of operations, and as a result its versatility is not satisfactory.Existing designs of image content authentication from advanced cryptographic primitives achieve robustness, security and versatility,at the cost of low efficiency. In this paper,we present a new design of image authentication with an improved trade-off among the aforementioned features. Our versatile design is robust to a number of predefined image processing operations. Its security can be reduced to q-strong Diffie-Hellman(q-SDH),a complexity problem used by existing cryptographic algorithms. From the aspect of efficiency, the new design has a constant-size authentication overhead(≤2 kB)and a constant verification time(around 0.05 s).While the time of generating authentication overhead increases linearly with the number of permissible editing operations,it only takes around 0.33 s for 1000 types of permissible operations. We believe the new design will facilitate image applications where trustworthy image editing is required.Haixia CHEN Xinyi HUANG Wei WU Yi MU 2020Science China(Information Sciences)2020,63,12:2
Blue-to-green manipulation of carbon dots from fluorescence to ultralong room-temperature phosphorescence for high-level anti-counterfeiting显示文摘Carbon dots(CDs) with fluorescence(FL) and room-temperature phosphorescence(RTP) optical properties have attracted dramatically growing interest in anti-counterfeiting application. Herein, color-tunable and stable FL and ultralong RTP(to naked eyes ~14 s) are successfully achieved in CDs system. Encoding information and patterns fabricated by directly screen-printing method are invisible to eyes under natural light. Interestingly, clear and multicolor patterns with tunable FL and RTP emissions are identified under the 365 nm, 395 nm and 465 nm excitation and removal of them, indicating potential application of carbon dots with different FL and RTP outputs in the high-level photonic anti-counterfeiting field.Youfusheng Wu Xinyi Fang Jiaqi Shi Weijing Yao Wei Wu 2021Chinese Chemical Letters2021,32,12:2
EMC10 governs male fertility via maintaining sperm ion balance显示文摘Infertility is a severe public health problem worldwide that prevails up to 15% in reproductive-age couples, and male infertility accounts for half of total infertility.Studies on genetically modified animal models have identifed lots of genes involved in the pathogenesis of male infertility. The underlying causes, however, remain largely unclear. In this study,we provide evidence that EMCIO,one subunit of endoplasmic reticulum (ER)membrane protein complex (EMC),is required for male fertility.EMC10 is significantly decreased in spermatozoa from patients with asthenozoospermia and positively associated with human sperm motility. Male mice lacking Emc10 gene are completely sterile.Emc10-null spermatozoa exhibit multiple defects including abnormal morphology, decreased motility, impaired capacitation, and impotency of acrosome reaction, thereby which are incapable of fertilizing intact or ZP-free oocytes. However, intracytoplasmic sperm injection could rescue this defect caused by EMC10 deletion. Mechanistically,EMC10 deficiency leads to inactivation of Na/K-ATPase,in turn giving rise to an increased level of intraceltutar Na^+ in spermatozoa, which contributes to decreased sperm motility and abnormal morphology.Other mechanistic investigations demonstrate that the absence of EMC10 results in a reduction of HCO3^- entry and subsequent decreases of both cAMP-dependent protein kinase A substrate phosphorylation and protein tyrosine phosphorytation. These data demonstrate that EMC10 is indispensable to mate fertility via maintaining sperm ion balance of Na^+ and HCO3^-,and also suggest that EMC10 is a promising biomarker for male fertility and a potential pharmaceutical target to treat male infertility.Yuchuan Zhou Fei Wu Man Zhang Zuquan Xiong Qianqian Yin Yanfei Ru Huijuan Shi Jinsong Li Shanhua Mao Yanliang Li Xinyi Cao Renming Hu Chong Wee Liew Qiang Ding Xuanchun Wang Yonglian Zhang 2018Journal of Molecular Cell Biology2018,10,6:2
Hematological Parameters in the First Trimester and the Risk of Gestational Diabetes Mellitus—Beijing,China,2017–2020显示文摘Summary What is already known about this topic?Hematological parameters may indicate the presence of chronic low-grade inflammation and increasing viscosity, which are involved in the pathological processes of gestational diabetes mellitus(GDM).However, the association between several hematological parameters in early pregnancy and GDM has yet to be elucidated. What is added by this report?Hematological parameters in the first trimester, particularly red blood cell(RBC)count and systematic immune index, have a significant impact on GDM incidence. The neutrophils(NEU)count in the first trimester was particularly pronounced for GDM.The upward trend of RBC,white blood cell(WBC),and NEU counts was consistent across all GDM subtypes. What are the implications for public health practice?Early pregnancy hematological parameters are associated with the risk of GDM.Xinyi Lyu Jiajing Jia Haishan Yang Yuzhi Deng Hanbin Wu Shuo Wang Chuanyu Zhao Jueming Lei Xiaoxuan Zou Ying Yang 2023China CDC weekly2023,5,9:1
Discovery of the mechanisms of acupuncture in the treatment of migraine based on functional magnetic resonance imaging and omics显示文摘Migraine is one of the most prevalent and disabling neurological disease, but the current pharmacotherapies show limited efficacy and often accompanied by adverse effects.Acupuncture is a promising complementary therapy, but further clinical evidence is needed. The influence of acupuncture on migraine is not an immediate effect, and its mechanism remains unclear. This study aims to provide further clinical evidence for the anti-migraine effects of acupuncture and explore the mechanism involved.A randomized controlled trial was performed among 10 normal controls and 38 migraineurs. The migraineurs were divided into blank control, sham acupuncture, and acupuncture groups.Patients were subjected to two courses of treatment, and each treatment lasted for 5 days, with an interval of 1 day between the two courses. The effectiveness of treatment was evaluated using pain questionnaire. The functional magnetic resonance imaging(fMRI)data were analyzed for investigating brain changes induced by treatments.Blood plasma was collected for metabolomics and proteomics studies.Correlation and mediation analyses were performed to investigate the interaction between clinical,fMRI and omics changes. Results showed that acupuncture effectively relieved migraine symptoms in a way different from sham acupuncture in terms of curative effect, affected brain regions, and signaling pathways. The anti-migraine mechanism involves a complex network related to the regulation of the response to hypoxic stress, reversal of brain energy imbalance, and regulation of inflammation. The brain regions of migraineurs affected by acupuncture include the lingual gyrus, default mode network, and cerebellum. The effect of acupuncture on patients’metabolites/proteins may precede that of the brain.Chong Li Xinyi Li Ke He Yang Wu Xiaoming Xie Jiju Yang Fan Zhang Yang Yue Huifeng Hao Shaokun Zhao Xin Li Guihua Tian 2023Frontiers of Medicine2023,17,5:1
In-situ TEM observation of the evolution of helium bubbles & dislocation loops and their interaction in Pd during He^(+) irradiation显示文摘The microstructural evolution of purity Pd under 30 keV He^(+)irradiation at 573 K was investigated by in-situ transmission electron microscopy. The nucleation, growth, merging, annihilation, size change, number density variation, and types of dislocation loops were analyzed under the influence of irradiation fluence and sample thickness. Both perfect dislocation loops with b=1/2<110>and faulted dislocation loops with b=1/3<111>were formed. However,at low irradiation fluence, most of the loops were 1/3<111>loops.The thickness of TEM foil obviously affected the ratio of 1/3<111>loop variants, the size and number density of dislocation loops, and the characteristics of bubble-loop complexes. With the increase of irradiation fluence, the size of dislocation loops increased, but loop volume number density remained almost constant until dislocation loops merged and evolved into dislocation network. There was an obvious interaction between dislocation loops and bubbles, indicating that 1/3<111>loop was first formed at the initial stage of irradiation, and when the loop grew to a certain size, obvious helium bubbles appeared inside its region.Qing Han Yipeng Li Guang Ran Xinyi Liu Lu Wu Yang Chen Piheng Chen Xiaoqiu Ye Yifan Ding Xiaoyong Wu 2021Journal of Materials Science & Technology2021,,28:1
