1Expenditure and financial burden for the diagnosis and treatment of colorectal cancer in China:a hospital.based,multicenter,cross-sectional survey显示文摘Background: The increasing prevalence of colorectal cancer(CRC) in China and the paucity of information about relevant expenditure highlight the necessity of better understanding the financial burden and effect of CRC diagnosis and treatment. We performed a survey to quantify the direct medical and non-medical expenditure as well as the resulting financial burden of CRC patients in China.Methods: We conducted a multicenter, cross-sectional survey in 37 tertiary hospitals in 13 provinces across China between 2012 and 2014. Each enrolled patient was interviewed using a structured questionnaire. All expenditure data were inflated to the 2014 Chinese Yuan(CNY; 1 CNY = 0.163 USD). We quantified the overall expenditure and financial burden and by subgroup(hospital type, age at diagnosis, sex, education, occupation, insurance type, household income, clinical stage, pathologic type, and therapeutic regimen). We then performed generalized linear modeling to determine the factors associated with overall expenditure.Results: A total of 2356 patients with a mean age of 57.4 years were included, 57.1 % of whom were men; 13.9% of patients had stage I cancer; and the average previous-year household income was 54,525 CNY.The overall average direct expenditure per patient was estimated to be 67,408 CNY, and the expenditures for stage Ⅰ, Ⅱ, Ⅲ, and Ⅳ disease were 56,099 CNY, 59,952 CNY, 67,292 CNY, and 82,729 CNY, respectively. Non-medical expenditure accounted for 8.3%of the overall expenditure. The 1-year out-of-pocket expenditure of a newly diagnosed patient was 32,649 CNY, which accounted for 59.9% of their previous-year household income and caused 75.0% of families to suffer an unmanageable financial burden. Univariate analysis showed that financial burden and overall expenditure differed in almost all subgroups(P < 0.05), except for sex. Multivariate analysis showed that patients who were treated in specialized hospitals and those who were diagnosed with adenocarcinoma or diagnosed at a later stage were likely to spend more,whereas those with a lower household income and those who underwent surgery spent less(all P < 0.05).Conclusions: For patients in China, direct expenditure for the diagnosis and treatment of CRC seemed catastrophic,and non-medical expenditure was non-ignorable. 3The C_2H_2 -type Zinc Finger Protein ZFP182 is Involved in Abscisic Acid-Induced Antioxidant Defense in Rice显示文摘C2H2 -type zinc finger proteins (ZFPs) are thought to play important roles in modulating the responses of plants to drought, salinity and oxidative stress. However, direct evidence is lacking for the involvement of these ZFPs in abscisic acid (ABA)-induced antioxidant defense in plants. In this study, the role of the rice (Oryza sativa L. sub. japonica cv. Nipponbare) C 2 H 2 -type ZFP ZFP182 in ABA-induced antioxidant defense and the relationship between ZFP182 and two rice MAPKs, OsMPK1 and OsMPK5 in ABA signaling were investigated. ABA treatment induced the increases in the expression of ZFP182, OsMPK1 and OsMPK5, and the activities of superoxide dismutase (SOD) and ascorbate peroxidase (APX) in rice leaves. The transient gene expression analysis and the transient RNA interference (RNAi) analysis in protoplasts showed that ZFP182, OsMPK1 and OsMPK5 are involved in ABA-induced up-regulation in the activities of SOD and APX. Besides, OsMPK1 and OsMPK5 were shown to be required for the up-regulation in the expression of ZFP182 in ABA signaling, but ZFP182 did not mediate the ABA-induced up-regulation in the expression of OsMPK1 and OsMPK5. 4Tracing the first hematopoietic stem cell generation in human embryo by single-cell RNA sequencing显示文摘Tracing the emergence of the first hematopoietic stem cells(HSCs)in human embryos,particularly the scarce and transient precursors thereof,is so far challenging,largely due to the technical limitations and the material rarity.Here,using single-cell RNA sequencing,we constructed the first genome-scale gene expression landscape covering the entire course of endothelial-to-HSC transition during human embryogenesis.The transcriptomically defined HSC-primed hemogenic endothelial cells(HECs)were captured at Carnegie stage(CS)12-14 in an unbiased way,showing an unambiguous feature of arterial endothelial cells(ECs)with the up-regulation of RUNX1,MYB and ANGPTL Importantly,subcategorizing CD34+CD45-ECs into a CD44+population strikingly enriched HECs by over 10-fold.We further mapped the developmental path from arterial ECs via HSC-primed HECs to hematopoietic stem progenitor cells,and revealed a distinct expression pattern of genes that were transiently over-represented upon the hemogenic fate choice of arterial ECs#including EMCN,PROCR and RUNX1T1.We also uncovered another temporally and molecularly distinct intra-embryonic HEC population,which was detected mainly at earlier CS 10 and lacked the arterial feature.Finally,we revealed the cellular components of the putative aortic niche and potential cellular interactions acting on the HSCprimed HECs.The cellular and molecular programs that underlie the generation of the first HSCs from HECs in human embryos,together with the ability to distinguish the HSC-primed HECs from others,will shed light on the strategies for the production of clinically useful HSCs from pluripotent stem cells.Yang Zeng Jian He Zhijie Bai Zongcheng Li Yandong Gong Chen Liu Yanli Ni Junjie Du Chunyu Ma Lihong Bian Yu Lan Bing Liu 2019Cell Research2019,29,11:14

6Apatinib, a selective VEGFR2 inhibitor, improves the delivery of chemotherapeutic agents to tumors by normalizing tumor vessels in LoVo colon cancer xenograft mice显示文摘Tumor vascular normalization has been proposed as a therapeutic strategy for malignant neoplasms, which can also interpret the synergistic effect of anti-angiogenesis agents combined with chemotherapy. Apatinib (Apa), a highly selective VEGFR2 inhibitor, attracts much attentions due to its encouraging anticancer activity, especially in the clinical trials of combined treatment. In this study, we investigated whether Apa could promote vascular normalization in tumor in a certain time window. Mice bearing LoVo colon cancer xenograft were orally administrated Apa (150 mg kg^-1 per day) for 5, 7, 10, or 12 days. Apa significantly inhibited tumor growth and decreased the microvessel density. Using multi-photon microscopy and electron microscopy, we found that Apa improved tumor vessel morphology by pruning distorted vessel branches and decreased the gap between endothelial cells after a 7-day treatment. Furthermore, Apa decreased vessel leakage and in creased pericyte coverage on vascular en dothelial cells, suggesting that tumor vessels were more mature and integrated. The intratumoral distribution of adriamycin (ADR) in Apa group was improved from day 7 to 10 without change in plasma drug concen tration. Tumor blood perfusion was also in creased in this window, and the expression of hypoxia induced factor 1a was downregulated, suggesting the effect of Apa on alleviating tumor hypoxic micro-e nvironme nt. In conclusi on, Apa may improve the effective perfusi on of tumor vessels and in crease the intratumoral distribution of ADR in a certain time window via normalizing tumor vessels. 8miR-21 promotes the differentiation of hair folliclederived neural crest stem cells into Schwann cells显示文摘Hair follicle-derived neural crest stem cells can be induced to differentiate into Schwann cells in vivo and in vitro. However, the underlying regulatory mechanism during cell differentiation remains poorly understood. This study isolated neural crest stem cells from human hair follicles and induced them to differentiate into Schwann cells. Quantitative RT-PCR showed that microRNA(miR)-21 expression was gradually increased during the differentiation of neural crest stem cells into Schwann cells. After transfection with the miR-21 agonist(agomir-21), the differentiation capacity of neural crest stem cells was enhanced. By contrast, after transfection with the miR-21 antagonist(antagomir-21), the differentiation capacity was attenuated. Further study results showed that SOX-2 was an effective target of miR-21. Without compromising SOX2 mRNA expression, miR-21 can down-regulate SOX protein expression by binding to the 3′-UTR of miR-21 mRNA. Knocking out the SOX2 gene from the neural crest stem cells significantly reversed the antagomir-21 inhibition of neural crest stem cells differentiating into Schwann cells. 10Targeting redox-altered plasticity to reactivate synaptic function: A novel therapeutic strategy for cognitive disorder显示文摘Redox-altered plasticity refers to redox-dependent reversible changes in synaptic plasticity via altering functions of key proteins, such as N-methyl-D-aspartate receptor(NMDAR). Age-related cognitive disorders includes Alzheimer’s disease(AD), vascular dementia(VD), and age-associated memory impairment(AAMI). Based on the critical role of NMDAR-dependent long-term potentiation(LTP) in memory, the increase of reactive oxygen species in cognitive disorders, and the sensitivity of NMDAR to the redox status, converging lines have suggested the redox-altered NMDAR-dependent plasticity might underlie the synaptic dysfunctions associated with cognitive disorders. In this review, we summarize the involvement of redox-altered plasticity in cognitive disorders by presenting the available evidence. According to reports from our laboratory and other groups, this 'redox-altered plasticity' is Hydrogen sulfidemore similar to functional changes rather than organic injuries, and strategies targeting redox-altered plasticity using pharmacological agents might reverse synaptic dysfunctions and memory abnormalities in the early stage of cognitive disorders. 11Embryonic endothelial evolution towards first hematopoietic stem cells revealed by single-cell transcriptomic and functional analyses显示文摘Hematopoietic stem cells(HSCs)in adults are believed to be born from hemogenic endothelial cells(HECs)in mid-gestational embryos.Due to the rare and transient nature,the HSC-competent HECs have never been stringently identified and accurately captured,let alone their genuine vascular precursors.Here,we first used high-precision single-cell transcriptomics to unbiasedly examine the relevant EC populations at continuous developmental stages with intervals of 0.5 days from embryonic day(E)9.5 to E11.0.As a consequence,we transcriptomically identified two molecularly different arterial EC populations and putative HSCprimed HECs,whose number peaked at El 0.0 and sharply decreased thereafter,in the dorsal aorta of the aorta-gonadmesonephros(AGM)region.Combining computational prediction and in vivo functional validation,we precisely captured HSCcompetent HECs by the newly constructed Neurl3-EGFP reporter mouse model,and realized the enrichment further by a combination of surface markers(Procr+Kit+CD44+,PK44).Surprisingly,the endothelial-hematopoietic dual potential was rarely but reliably witnessed in the cultures of single HECs.Noteworthy,primitive vascular ECs from E8.0 experienced two-step fate choices to become HSC-primed HECs,namely an Initial arterial fate choice followed by a hemogenic fate conversion.This finding resolves several previously observed contradictions.Taken together,comprehensive understanding of endothelial evolutions and molecular programs underlying HSC-primed HEC specification in vivo will facilitate future investigations directing HSC production in vitro.Siyuan Hou Zongcheng Li Xiaona Zheng Yun Gao Ji Dong Yanli Ni Xiaobo Wang Yunqiao Li Xiaochen Ding Zhilin Chang Shuaili Li Yuqiong Hu Xiaoying Fan Yu Hou Lu Wen Bing Liu Fuchou Tang Yu Lan 2020Cell Research2020,30,5:8

13Dissecting human embryonic skeletal stem cell ontogeny by single-cell transcriptomic and functional analyses显示文摘Human skeletal stem cells(SSCs)have been discovered in fetal and adult long bones.However,the spatiotemporal ontogeny of human embryonic SSCs during early skeletogenesis remains elusive.Here we map the transcriptional landscape of human limb buds and embryonic long bones at single-cell resolution to address this fundamental question.We found remarkable heterogeneity within human limb bud mesenchyme and epithelium,and aligned them along the proximal-distal and anterior-posterior axes using known marker genes.Osteo-chondrogenic progenitors first appeared in the core limb bud mesenchyme,which give rise to multiple populations of stem/progenitor cells in embryonic long bones undergoing endochondral ossification.Importantly,a perichondrial embryonic skeletal stem/progenitor cell(eSSPC)subset was identified,which could self-renew and generate the osteochondral lineage cells,but not adipocytes or hematopoietic stroma.eSSPCs are marked by the adhesion molecule CADM1 and highly enriched with FOXP1/2 transcriptional network.Interestingly,neural crest-derived cells with similar phenotypic markers and transcriptional networks were also found in the sagittal suture of human embryonic calvaria.Taken together,this study revealed the cellular heterogeneity and lineage hierarchy during human embryonic skeletogenesis,and identified distinct skeletal stem/progenitor cells that orchestrate endochondral and intramembranous ossification.Jian He Jing Yan Jianfang Wang Liangyu Zhao Qian Xin Yang Zeng Yuxi Sun Han Zhang Zhijie Bai Zongcheng Li Yanli Ni Yandong Gong Yunqiao Li Han He Zhilei Bian Yu Lan Chunyu Ma Lihong Bian Heng Zhu Bing Liu Rui Yue 2021Cell Research2021,31,7:5

17Targeting a novel inducible GPX4 alternative isoform to alleviate ferroptosis and treat metabolic-associated fatty liver disease显示文摘Metabolic-associated fatty liver disease(MAFLD),which is previously known as non-alcoholic fatty liver disease(NAFLD),represents a major health concern worldwide with limited therapy.Here,we provide evidence that ferroptosis,a novel form of regulated cell death characterized by iron-driven lipid peroxidation,was comprehensively activated in liver tissues from MAFLD patients.The canonical-GPX4(cGPX4),which is the most important negative controller of ferroptosis,is downregulated at protein but not mRNA level.Interestingly,a non-canonical GPX4 transcript-variant is induced(inducible-GPX4,iGPX4)in MAFLD condition.The high fat-fructose/sucrose diet(HFFD)and methionine/choline-deficient diet(MCD)-induced MAFLD pathologies,including hepatocellular ballooning,steatohepatitis andfibrosis,were attenuated and aggravated,respectively,in cGPX4-and iGPX4-knockin mice.cGPX4 and iGPX4 isoforms also displayed opposing effects on oxidative stress and ferroptosis in hepatocytes.Knockdown of iGPX4 by siRNA alleviated lipid stress,ferroptosis and cell injury.Mechanistically,the triggered iGPX4 interacts with cGPX4 to facilitate the transformation of cGPX4 from enzymatic-active monomer to enzymatic-inactive oligomers upon lipid stress,and thus promotes ferroptosis.Co-immunoprecipitation and nano LC–MS/MS analyses confirmed the interaction between iGPX4 and cGPX4.Our results reveal a detrimental role of non-canonical GPX4 isoform in ferroptosis,and indicate selectively targeting iGPX4 may be a promising therapeutic strategy for MAFLD.Jie Tong Dongjie Li Hongbo Meng Diyang Sun Xiuting Lan Min Ni Jiawei Ma Feiyan Zeng Sijia Sun Jiangtao Fu Guoqiang Li Qingxin Ji Guoyan Zhang Qirui Shen Yuanyuan Wang Jiahui Zhu Yi Zhao Xujie Wang Yi Liu Shenxi Ouyang Chunquan Sheng Fuming Shen Pei Wang 2022Acta Pharmaceutica Sinica B2022,12,9:3
18Adriamycin Thermotherapy through the Hepatic Artery Using VX2 Carcinoma in Rabbit Liver as a Model显示文摘OBJECTIVE It has been reported that heating can enhance sensitivity of rabbit VX2 cells to adriamycin and increase the intracellular concentration of adriamycin. This study was designed to evaluate the anti-tumor effect of interventional hyperthermia and interventional thermochemotherapy on VX2 carcinoma in rabbit liver. METHODS VX2 carcinoma cells were surgically implanted into the right liver lobe of 60 male New Zealand white rabbits, which were randomly divided into 4 groups (15 per group). The 4 groups (designated as 1, 2, 3, 4 respectively) were injected with 10 ml of the following via the hepatic artery: physiological saline (37℃); adriamycin (37℃); physiological saline (60℃); adriamycin (60℃). One week later, the tumor volume, serum level of aspartate transaminase (AST) and the survival of the rabbits bearing VX2 were observed and compared among the different treated groups. RESULTS The tumor growth rate in group 4 (ADM 60℃) (0.53±0.21)% was significantly lower than that in group 1 (3.48±1.17)%, in group 2 (1.09±0.26)% and group 3 (3.32±1.28)% (P<0.05, P<0.05, P<0.01, respectively). The days of survival days for group 4 (87.0±2.0) were significantly more than that in group 1 (40.0±3.0). Group 4 showed a significantly higher increase in serum AST compared to group 1 (P<0.05), but without significant differences compared to the other groups (P>0.05). 20Delineating spatiotemporal and hierarchical development of human fetal innate lymphoid cells显示文摘Whereas the critical roles of innate lymphoid cells(ILCs)in adult are increasingly appreciated,their developmental hierarchy in early human fetus remains largely elusive.In this study,we sorted human hematopoietic stem/progenitor cells,lymphoid progenitors,putative ILC progenitor/precursors and mature ILCs in the fetal hematopoietic,lymphoid and non-lymphoid tissues,from 8 to 12 post-conception weeks,for single-cell RNA-sequencing,followed by computational analysis and functional validation at bulk and single-cell levels.We delineated the early phase of ILC lineage commitment from hematopoietic stem/progenitor cells,which mainly occurred in fetal liver and intestine.We further unveiled interleukin-3 receptor as a surface marker for the lymphoid progenitors in fetal liver with T,B,ILC and myeloid potentials,while IL-3RA–lymphoid progenitors were predominantly B-lineage committed.Notably,we determined the heterogeneity and tissue distribution of each ILC subpopulation,revealing the proliferating characteristics shared by the precursors of each ILC subtype.Additionally,a novel unconventional ILC2 subpopulation(CRTH2–CCR9+ILC2)was identified in fetal thymus.Taken together,our study illuminates the precise cellular and molecular features underlying the stepwise formation of human fetal ILC hierarchy with remarkable spatiotemporal heterogeneity.Chen Liu Yandong Gong Han Zhang Hua Yang Yang Zeng Zhilei Bian Qian Xin Zhijie Bai Man Zhang Jian He Jing Yan Jie Zhou Zongcheng Li Yanli Ni Aiqing Wen Yu Lan Hongbo Hu Bing Liu 2021Cell Research2021,31,10:2
