| 1 | Dendritic Cells Transduced with SOCS1 Gene Exhibit Regulatory DC Properties and Prolong Allograft Survival显示文摘SOCS1 is a key regulator of cytokine signaling and is important for maintaining balance in the immune system. It is thought to participate in negative feedback loops in cytokine signaling and may be an important signal for the regulation of dendritic cell (DC) maturation. However, it remains unclear whether DCs transduced with SOCS1 exhibit characteristics of regulatory DCs and induce allogeneic T-cell hyporesponsiveness. In this study, we constructed adenoviral vector coding SOCS1 (Ad-SOCS1) that can efficiently increase SOCS1 gene expression in bone marrow-derived dendritic cells. DCs transduced with Ad-SOCS1 (DC-SOCS1) expressed low levels of costimulatory and MHC molecules, were resistant to maturation and activation stimulation, induced allogeneic T-cell hyporesponsiveness, and promoted the generation of regulatory-like T cells in vitro. DC-SOCS1 pretreatment significantly prolonged the survival of allografts and led to a substantial increase in the generation of regulatory T cells. | 2 | Types,Characteristics and Significances of Migrating Pathways of Gas-bearing Fluids in the Shenhu Area,Northern Continental Slope of the South China Sea显示文摘The first marine gas hydrate expedition in China has been conducted by Guangzhou Marine Geological Survey in the Shenhu Area, northern continental slope of the South China Sea. Previous study has analyzed the P-T conditions, geophysical anomalies and saturation calculations of these gas hydrates, but has not documented in detail the migration of gas-bearing fluids in the study area. Based on the interpretations of 2D/3D seismic data, this work identified two types of migration pathways for gas-bearing fluids in the Shenhu area, i.e., vertical and lateral pathways. The vertical pathways(largescale faults, gas chimneys and mud diapirs) presented as steep seismic reflection anomalies, which could be traced downward to the Eocene source rocks and may penetrate into the Late Miocene strata. The deeper gases/fluids might be allowed migrating into the shallower strata through these vertical conduits. However, the distributions showed distinct differences between these pathways. Large-scale faults developed only in the north and northeast of the Shenhu area, while in the drilling area gas chimneys were the sole vertical migration pathways. Since the Pliocene, normal faults, detachment faults and favorable sediments have constituted the lateral pathways in the Shenhu gas hydrate drilling area. Although these lateral pathways were connected with gas chimneys, they exerted different effects on hydrate formation and accumulation. Gas-bearing fluids migrated upward along gas chimneys might further migrate laterally because of the normal faults, thereby enlarging the range of the chimneys. Linking gas chimneys with the seafloor, the detachment faults might act as conduits for escaping gases/fluids. Re-deposited sediments developed at the early stage of the Quaternary were located within the gas hydrate stability zone, so hydrates would be enriched in these favorable sediments. Compared with the migration pathways(large-scale faults and mud diapirs) in the LW3-1 deep-sea oil/gas field, the migration efficiency of the vertical pathways(composed of gas chimneys) in the gas hydrate drilling area might be relatively low. Description and qualitative discrimination of migration pathways in the Shenhu gas hydrate drilling area are helpful to further understand the relationship between good-quality deep source rocks and shallow, mainly biogenicallyproduced, hydrates. As the main source rocks of the Baiyun sag, lacustrine mudstones in the Wenchang and Enping Formations may provide thermogenic methane. Gas chimneys with relatively low migration efficiency created the vertical pathways. Caused by the Dongsha tectonic movement, the release of overpressured fluids might reduce the vertical migration rates of the thermogenic methane. The thick bathyal/abyssal fine-grained sediments since the Late Miocene provided migration media with low permeability. These preconditions may cause carbon isotopic fractionation ofthermogenic methane during long-distance vertical migrations. | 3 | Finite element investigation of the poroelastic effect on the Xinfengjiang Reservoir-triggered earthquake显示文摘Coulomb failure stress changes (ΔCFS) are used in the study of reservoir-induced seismicity (RIS) generation.The threshold value of ΔCFS that can trigger earthquakes is an important issue that deserves thorough research.The M s 6.1 earthquake in the Xinfengjiang Reservoir in 1962 is well acknowledged as the largest reservoir-induced earthquake in China.Therefore, it is a logical site for quantitative calculation of ΔCFS induced by the filling of the reservoir and for investigating the magnitude of CFS that can trigger reservoir seismic activities.To better understand the RIS mechanism, a three-dimensional poroelastic finite element model of the Xinfengjiang Reservoir is proposed here, taking into consideration of the precise topography and dynamic water level.We calculate the instant changes of stress and pore pressure induced by water load, and the time variation of effective stresses due to pore water diffusion.The CFS on the seismogenesis faults and the accumulation of strain energy in the reservoir region are also calculated.Primary results suggest that the reservoir impoundment increases both pore pressure and CFS on the fault at the focal depth.The diffusion of pore pressure was likely the main factor that triggered the main earthquake, whereas the elastic stress owing to water load was relatively small.The magnitude of CFS on seismogenesis fault can reach approximately 10 kPa, and the ΔCFS values at the hypocenter can be about 0.7-3.0 kPa, depending on the fault diffusion coefficient.The calculated maximum vertical subsidence caused by the water load in the Xinfengjiang Reservoir is 17.5 mm, which is in good agreement with the observed value of 15 mm.The accumulated strain energy owing to water load was only about 7.3×10 11 J, even less than 1% of the seismic wave energy released by the earthquake.The reservoir impoundment was the only factor that triggered the earthquake. | CHENG HuiHong ZHANG Huai ZHU BoJing SUN YuJun ZHENG Liang YANG ShaoHua SHI YaoLin | 2012 | Science China Earth Sciences2012,55,12: | 11 |

| 5 | Optimizing the CRISPR/Cas9 system for genome editing in grape by using grape promoters显示文摘The efficacy of the CRISPR/Cas9 system in grapevine(Vitis vinifera L.)has been documented,but the optimization of this system,as well as CRISPR/Cas9-mediated multiplex genome editing,has not been explored in this species.Herein,we identified four VvU3 and VvU6 promoters and two ubiquitin(UBQ)promoters in grapevine and demonstrated that the use of the identified VvU3/U6 and UBQ2 promoters could significantly increase the editing efficiency in grape by improving the expression of sgRNA and Cas9,respectively.Furthermore,we conducted multiplex genome editing using the optimized CRISPR/Cas9 vector that contained the conventional multiple sgRNA expression cassettes or the polycistronic tRNA-sgRNA cassette(PTG)by targeting the sugar-related tonoplastic monosaccharide transporter(TMT)family members TMT1 and TMT2,and the overall editing efficiencies were higher than 10%.The simultaneous editing of TMT1 and TMT2 resulted in reduced sugar levels,which indicated the role of these two genes in sugar accumulation in grapes.Moreover,the activities of the VvU3,VvU6,and UBQ2 promoters in tobacco genome editing were demonstrated by editing the phytoene desaturase(PDS)gene in Nicotiana benthamiana leaves.Our study provides materials for the optimization of the CRISPR/Cas9 system.To our knowledge,our simultaneous editing of the grape TMT family genes TMT1 and TMT2 constitutes the first example of multiplex genome editing in grape.The multiplex editing systems described in this manuscript expand the toolbox of grape genome editing,which would facilitate basic research and molecular breeding in grapevine. | Chong Ren Yanfei Liu Yuchen Guo Wei Duan Peige Fan Shaohua Li Zhenchang Liang | 2021 | Horticulture Research2021,8,1: | 6 |
| 6 | Preparation of DHA-and EPA-enriched Glycerides by Enzymatic Interesterification Using Tuna Oil and Capric Acid显示文摘This study focused on the preparation of docosahexaenoic acid(DHA) and eicosapentaenoic acid(EPA)enriched-triacylglycerols by enzymatic interesterification using tuna oil and capric acid. The content of DHA+EPA is 26.86% in the tuna oil used in this study. A response surface methodology(RSM) was used to optimize the reaction parameters(reaction temperature, substrate molar ratio, enzyme amount and reaction time), and the optimized conditions were determined to be: reaction temperature 58℃, substrate molar ratio(capric acid : tuna oil) 4:1, enzyme amount 4%, and reaction time 7.5 h. Under the optimized conditions, the content of DHA+EPA in the glycerides was 40.03%, which is 13.17% higher than that in raw tuna oil. | 17 | Multiscale analysis of Asian Monsoon over the past 640 ka显示文摘The empirical mode decomposition(EMD) method is used to re-analyse the high-resolution and precisely-dated stalagmite record from Chinese caves over the past 640 ka. Results show that(1) the variation in the Asian Monsoon can be completely decomposed into ten quasiperiod oscillations, among which the precession and semiprecession band oscillations are the most prominent periodicities, with contribution rates of 31.1% and 30.7%, respectively;(2) the cross-spectrum analysis of the semiprecession component and bi-hemisphere insolation(BHI) are strongly correlated, indicating an amplified response of precipitation and temperature variability to the interhemispheric insolation in the low-latitude regions, thus further affecting the intensity of the Asian Monsoon;(3) on millennial timescales, obvious oscillations at the 5 ka and 1–2 ka bands roughly correspond to the classical Bond and Dansgaard-Oeschger(DO) cycles. Additionally, a strong correlation is found between the detrended stalagmite δ18 O records and Ca/Sr sequence from the North Atlantic(especially at the 5 ka band).
