| 1 | Cloning,expression and binding specificity analysis of odorant binding protein 3 of the lucerne plant bug,Adelphocoris lineolatus(Goeze)显示文摘Credible evidence shows that odorant binding proteins(OBPs)are required for insect olfaction perception and play a key role in transporting hydrophobic odorants across the sensillum lymph to the olfactory receptors(ORs).In the present study,a novel OBP(AlinOBP3)gene from the lucerne plant bug,Adelphocoris lineolatus,was cloned and expressed.The expression pattern of AlinOBP3 was evaluated by qPCR,which indicated that AlinOBP3 was dominantly expressed in antennae.The binding properties of AlinOBP3 with 9 cotton volatiles and 5 sex pheromone analogs were measured by fluorescence competitive binding assays with the fluorescence probe 1-NPN.The results revealed that of 9 cotton volatiles,Myrcene,β-Ocimene and α-Phellandrene can bind with AlinOBP3.α-Phellandrene especially bound to AlinOBP3 with a high binding affinity,with a dissociation constant of 56.68 μmol/L.Of the 5 sex pheromone analogs,Hexyl butyrate had the strongest binding affinity with AlinOBP3,with a dissociation constant as 59.53 μmol/L.Butyl butyrate,trans-2-Hexenyl butyrate and Ethyl butyrate had medium binding affinities with AlinOBP3,with dissociation constants of 227.39,108.77 and 143.47 μmol/L,respectively.The results suggest that AlinOBP3 might be a pheromone binding protein(PBP)with a dual-function for the perception of sex pheromones and plant volatiles. | GU ShaoHua SUN Yang REN LiYan ZHANG XueYing ZHANG YongJun WU KongMing GUO YuYuan | 2010 | Chinese Science Bulletin2010,55,34: | 7 |
| 2 | Electron-Deficient Zn-N_(6) Configuration Enabling Polymeric Carbon Nitride for Visible-Light Photocatalytic Overall Water Splitting显示文摘Despite of suitable band structures for harvesting solar light and driving water redox reactions,polymeric carbon nitride(PCN)has suffered from poor charge transfer ability and sluggish surface reaction kinetics,which limit its photocatalytic activity for water splitting.Herein,atomically dispersed Zn-coordinated three-dimensional(3D)sponge-like PCN(Zn-PCN)is synthesized through a novel intermediate coordination strategy.Advanced characterizations and theoretical calculations well evidence that Zn single atoms are coordinated and stabilized on PCN in the form of Zn-N_(6) configura-tion featured with an electron-deficient state.Such an electronic configuration has been demonstrated contributive to promoted electron excitation,accelerated charge separation and transfer as well as reduced water redox barriers.Further benefited from the abundant surface active sites derived from the 3D porous structure,Zn-PCN realizes visible-light photocatalysis for overall water splitting with H_(2) and O_(2) simultaneously evolved at a stoichiometric ratio of 2:1.This work brings new insights into the design of novel single-atom photocatalysts by deepening the understanding of electronic configurations and reactive sites favorable to excellent photocatalysis for water splitting and related solar energy conversion reactions. | Daming Zhao Yiqing Wang Chung-Li Dong Fanqi Meng Yu-Cheng Huang Qinghua Zhang Lin Gu Lan Liu Shaohua Shen | 2022 | Nano-Micro Letters2022,14,12: | 2 |

| 4 | Preparation and characterization of some surface negatively charged residue mutants of cytochrome b_5显示文摘Site-directed mutagenesis was used to obtain seven variants of tryptic fragment of bovine liver cytochrome bs (cyt bs), in which the negatively charged residues around the heme exposed edge of cyt bs were replaced by hydropho-bic amino acid alanine. Double-site mutants, triple-site mutants and even quadruple-site mutants were obtained. DNA sequencing and molecular weight measurements of the mutant proteins both confirmed that these site-directed muta-genesises were successfully performed. Spectroelectrochem-istry of these mutant proteins revealed that the apparent redox potentials of these mutant proteins caused a positive shift of 2-10 mV. | 14 | Early Diagnosis of Bipolar Disorder Coming Soon:Application of an Oxidative Stress Injury Biomarker(BIOS)Model显示文摘Early distinction of bipolar disorder(BD)from major depressive disorder(MDD)is difficult since no tools are available to estimate the risk of BD.In this study,we aimed to develop and validate a model of oxidative stress injury for predicting BD.Data were collected from 1252 BD and 1359 MDD patients,including 64 MDD patients identified as converting to BD from 2009 through 2018.30 variables from a randomly-selected subsample of 1827(70%)patients were used to develop the model,including age,sex,oxidative stress markers(uric acid,bilirubin,albumin,and prealbumin),sex hormones,cytokines,thyroid and liver function,and glycolipid metabolism.Univariate analyses and the Least Absolute Shrinkage and Selection Operator were applied for data dimension reduction and variable selection.Multivariable logistic regression was used to construct a model for predicting bipolar disorder by oxidative stress biomarkers(BIOS)on a nomogram.Internal validation was assessed in the remaining 784 patients(30%),and independent external validation was done with data from 3797 matched patients from five other hospitals in China.10 predictors,mainly oxidative stress markers,were shown on the nomogram.The BIOS model showed good discrimination in the training sample,with an AUC of 75.1%(95%CI:72.9%–77.3%),sensitivity of 0.66,and specificity of 0.73.The discrimination was good both in internal validation(AUC 72.1%,68.6%–75.6%)and external validation(AUC 65.7%,63.9%–67.5%).In this study,we developed a nomogram centered on oxidative stress injury,which could help in the individualized prediction of BD.For better real-world practice,a set of measurements,especially on oxidative stress markers,should be emphasized using big data in psychiatry. | Zhiang Niu Xiaohui Wu Yuncheng Zhu Lu Yang Yifan Shi Yun Wang Hong Qiu Wenjie Gu Yina Wu Xiangyun Long Zheng Lu Shaohua Hu Zhijian Yao Haichen Yang Tiebang Liu Yong Xia Zhiyu Chen Jun Chen Yiru Fang | 2022 | Neuroscience Bulletin2022,38,9: | 1 |

| 16 | Construction of Ad-EGFP-BDNF vector and its expression in neural stem cells显示文摘BACKGROUND:Brain-derived neurotrophic factor(BDNF) provides nourishment to injured neurons.Neural stem cells can differentiate into neurons to repair neuronal injury in vivo.It has been hypothesized that continuous secretion of BDNF from neural stem cells could benefit brain injury repair.OBJECTIVE:To transfect BDNF and enhanced green fluorescent protein(EGFP) into neural stem cells with adenovirus vector and to observe expression of BDNF and EGFP in transfected neural stem cells.DESIGN,TIME AND SETTING:Observational,cellular,molecular study was performed at the Biochemistry Laboratory,Tongji University School of Medicine,China from July 2004 to September 2006.MATERIALS:Neural stem cells were provided by the Anatomy and Histoembryology Laboratory of Fudan University Medical School,China.METHODS:BDNF cDNA was extracted by reverse transcription polymerase chain reaction from the rat hippocampus.Following gene cloning and packaging by HEK293.BDNF,the EGFP gene was transfected into cultured neural stem cells with the Ad-EGFP-BDNF vector.BDNF-expressing neural stem cell clones were selected by G418 selection.MAIN OUTCOME MEASURES:EGFP expression and cell morphology were observed by fluorescent microscopy;neural stem cell expressing BDNF mRNA was examined by reverse transcription polymerase chain reaction;BDNF expression was detected by enzyme-linked immunosorbent assay from supernatant of infected neural stem cells.RESULTS:High transfection efficiency was obtained using 5 × 108 virus titers to transfect neural stem cells.G418-resistant neural stem cell clones integrated BDNF mRNA fragments.Enzyme-linked immunosorbent assay results showed that BDNF expression in the supernatant increased with increasing culture time and peaked at 72 hours.CONCLUSION:Adenovirus-mediated BDNF and EGFP genes were successfully transfected into neural stem cells and were expressed in neural stem cells for a long period of time. | Shaohua Li Zhenhua Li Huilong Wen Xin Gu Yinjiang Cui Yang Wang Lixia Lu Yanna Li Manli Na | 2010 | Neural Regeneration Research2010,5,13: | 1 |

| 19 | Deep-Learning-Based Production Decline Curve Analysis in the Gas Reservoir through Sequence Learning Models显示文摘Production performance prediction of tight gas reservoirs is crucial to the estimation of ultimate recovery,which has an important impact on gas field development planning and economic evaluation.Owing to the model’s simplicity,the decline curve analysis method has been widely used to predict production performance.The advancement of deep-learning methods provides an intelligent way of analyzing production performance in tight gas reservoirs.In this paper,a sequence learning method to improve the accuracy and efficiency of tight gas production forecasting is proposed.The sequence learning methods used in production performance analysis herein include the recurrent neural network(RNN),long short-term memory(LSTM)neural network,and gated recurrent unit(GRU)neural network,and their performance in the tight gas reservoir production prediction is investigated and compared.To further improve the performance of the sequence learning method,the hyperparameters in the sequence learning methods are optimized through a particle swarm optimization algorithm,which can greatly simplify the optimization process of the neural network model in an automated manner.Results show that the optimized GRU and RNN models have more compact neural network structures than the LSTM model and that the GRU is more efficiently trained.The predictive performance of LSTM and GRU is similar,and both are better than the RNN and the decline curve analysis model and thus can be used to predict tight gas production. | Shaohua Gu Jiabao Wang Liang Xue Bin Tu Mingjin Yang Yuetian Liu | 2022 | Computer Modeling in Engineering & Sciences2022,,6: | 0 |
