1Sequencing of Cultivated Peanut, Arachis hypogaea, Yields Insights into Genome Evolution and Oil Improvement显示文摘Cultivated peanut (Arachis hypogaea) is an allotetraploid crop planted in Asia, Africa, and America for edible oil and protein. To explore the origins and consequences of tetraploidy, we sequenced the allotetraploid A. hypogaea genome and compared it with the related diploid Arachis duranensis and Arachis ipaensis genomes. We annotated 39 888 A-subgenome genes and 41 526 B-subgenome genes in allotetraploid peanut. The A. hypogaea subgenomes have evolved asymmetrically, with the B subgenome resembling the ancestral state and the A subgenome undergoing more gene disruption, loss, conversion, and transposable element proliferation, and having reduced gene expression during seed development despite lacking genome-wide expression dominance. Genomic and transcriptomic analyses identified more than 2 500 oil metabolism-related genes and revealed that most of them show altered expression early in seed development while their expression ceases during desiccation, presenting a comprehensive map of peanut lipid biosynthesis. 2Relationship between nutrients and plankton biomass in the turbidity maximum zone of the Pearl River Estuary显示文摘Nutrients,dissolved and particulate organic carbon and plankton(bacterio-,phyto-and zoo-)were compared in the turbidity maximum zone(TMZ) and adjacent areas(non-TMZ) in the Pearl River estuary.Our results showed that high levels of suspended substances had marked effect on dynamics of nutrients and plankton in the TMZ.Based on the cluster analysis of total suspended solids(TSS) concentrations,all stations were divided into two groups,TMZ with average TSS of 171 mg/L and non-TMZ of 45 mg/L.Suspended substances adsorbed PO^(3-)_4 and dissolved organic carbon,resulting in higher particulate phosphorus and organic carbon(POC)and lower PO^(3-)_4 and DOC in the TMZ,compared to the non-TMZ.However,suspended substances had limited effect on nitrogenous nutrients.Phytoplankton growth was light-limited due to high concentrations of suspended substances in the TMZ and a peak of phytoplankton abundance appeared in the non-TMZ.In contrast,the highest bacterial abundance occurred in the TMZ,which was likely partly responsible for low DOC levels.Two peaks of zooplankton abundance observed in the TMZ and non-TMZ in the Pearl River estuary were primarily supported by bacteria and phytoplankton,respectively.Our finding implied that high levels of suspended solids in the TMZ affect the trophic balance.Zhen Shi Jie Xu Xiaoping Huang Xia Zhang Zhijian Jiang Feng Ye Ximei Liang 2017Journal of Environmental Sciences2017,29,7:12
3Carbon pools and fluxes in the China Seas and adjacent oceans显示文摘The China Seas include the South China Sea, East China Sea, Yellow Sea, and Bohai Sea. Located off the Northwestern Pacific margin, covering 4700000 km^2 from tropical to northern temperate zones, and including a variety of continental margins/basins and depths, the China Seas provide typical cases for carbon budget studies. The South China Sea being a deep basin and part of the Western Pacific Warm Pool is characterized by oceanic features; the East China Sea with a wide continental shelf, enormous terrestrial discharges and open margins to the West Pacific, is featured by strong cross-shelf materials transport; the Yellow Sea is featured by the confluence of cold and warm waters; and the Bohai Sea is a shallow semiclosed gulf with strong impacts of human activities. Three large rivers, the Yangtze River, Yellow River, and Pearl River, flow into the East China Sea, the Bohai Sea, and the South China Sea, respectively. The Kuroshio Current at the outer margin of the Chinese continental shelf is one of the two major western boundary currents of the world oceans and its strength and position directly affect the regional climate of China. These characteristics make the China Seas a typical case of marginal seas to study carbon storage and fluxes. This paper systematically analyzes the literature data on the carbon pools and fluxes of the Bohai Sea,Yellow Sea, East China Sea, and South China Sea, including different interfaces(land-sea, sea-air, sediment-water, and marginal sea-open ocean) and different ecosystems(mangroves, wetland, seagrass beds, macroalgae mariculture, coral reefs, euphotic zones, and water column). Among the four seas, the Bohai Sea and South China Sea are acting as CO_2 sources, releasing about0.22 and 13.86–33.60 Tg C yr^(-1) into the atmosphere, respectively, whereas the Yellow Sea and East China Sea are acting as carbon sinks, absorbing about 1.15 and 6.92–23.30 Tg C yr^(-1) of atmospheric CO_2, respectively. Overall, if only the CO_2 exchange at the sea-air interface is considered, the Chinese marginal seas appear to be a source of atmospheric CO_2, with a net release of 6.01–9.33 Tg C yr^(-1), mainly from the inputs of rivers and adjacent oceans. The riverine dissolved inorganic carbon (DIC) input into the Bohai Sea and Yellow Sea, East China Sea, and South China Sea are 5.04, 14.60, and 40.14 Tg C yr^(-1),respectively. The DIC input from adjacent oceans is as high as 144.81 Tg C yr^(-1), significantly exceeding the carbon released from the seas to the atmosphere. In terms of output, the depositional fluxes of organic carbon in the Bohai Sea, Yellow Sea, East China Sea, and South China Sea are 2.00, 3.60, 7.40, and 5.92 Tg C yr^(-1), respectively. The fluxes of organic carbon from the East China Sea and South China Sea to the adjacent oceans are 15.25–36.70 and 43.93 Tg C yr^(-1), respectively. The annual carbon storage of mangroves, wetlands, and seagrass in Chinese coastal waters is 0.36–1.75 Tg C yr^(-1), with a dissolved organic carbon(DOC) output from seagrass beds of up to 0.59 Tg C yr^(-1). Removable organic carbon flux by Chinese macroalgae mariculture account for 0.68 Tg C yr^(-1) and the associated POC depositional and DOC releasing fluxes are 0.14 and 0.82 Tg C yr^(-1), respectively. Thus, in total, the annual output of organic carbon, which is mainly DOC, in the China Seas is 81.72–104.56 Tg C yr^(-1). The DOC efflux from the East China Sea to the adjacent oceans is 15.00–35.00 Tg C yr^(-1). The DOC efflux from the South China Sea is 31.39 Tg C yr^(-1). 4Holocene aeolian stratigraphic sequences in the eastern portion of the desert belt(sand seas and sandy lands) in northern China and their palaeoenvironmental implications显示文摘This paper presents the environmental history and its responses to palaeoclimatic changes since the start of the Holocene in the eastern portion of the desert belt(sand seas and sandy lands) in northern China by comparing the aeolian sandpalaeosol sequences and their palaeoclimatic proxies. The optically stimulated luminescence(OSL) ages of the aeolian sandpalaeosol sedimentary sequences and a series of palaeoenvironmental proxies show that:(1) The large-scale dune landscape currently in the Kubuqi Sand Sea was formed during the Holocene in general;and the palaeosol was generally developed during the period of 4–2 ka, indicating conditions favorable for vegetation growth, soil development, and organic carbon accumulation due to increased precipitation or effective moisture and weakened aeolian activities;the large-scale expansion of dunes in the recent 2 ka is closely linked to human activities. The variable discharge of the Yellow River with diversions for irrigation may have resulted in a more consistent supply of aeolian particles for dune field expansion.(2) The dune landscape of the Hunshandake Sandy Land was likely formed around 12 ka, and before this, the western part of the Hunshandake Sandy Land would have been covered by a single large lake;it was obviously wetter than today in the sandy land during the period of 9.6–3 ka and the palaeosols were developed at the same time. But the aeolian activities have not been completely dormant in this long-lasting wetter epoch;because the Holocene wetter period was likely time-transgressive across the region.(3) The palaeosol of the Hulunbuir Sandy Land began to develop as early as 14.5 ka, probably continuing until the last 2 ka. The palaeosol development of various dune fields in the eastern portion of the desert belt(sand seas and sandy lands) in northern China is spatially heterogeneous, and even the palaeosol development time in different locations within each sandy land is inconsistent. During the middle Holocene(especially the 7.5–3.5 ka), all the sandy lands were stabilized in general and the intensity of aeolian activities was significantly weakened. The number of palaeoenvironmental records in the eastern portion of the desert belt(sand seas and sandy lands) in northern China has increased rapidly in the past decade, but the amount of published data still does not match the vast extent of the dune fields. 5Investigation of influence of coal properties on dense-phase pneumatic conveying at high pressure显示文摘Experiments of dense-phase pneumatic conveying of pulverized coal using nitrogen were carried out in a test facility at pressures of up to 3.7 MPa to study the effects of coal type, particle size and moisture content on flow characteristics. The Jenike shear test and scanning electron microscopy (SEM) were employed to provide a better understanding of effects of the material properties on flow characteristics. Two kinds of pulverized coals, Yanzhou and Datong, with similar particle size, moisture content and density, were used in the test. Pressure drop increases with increasing the particle size at similar solid-gas ratio, superficial velocity and pressure in the receiving hopper, and pressure drops through different test sections decrease firstly and then rise with increasing the conveying velocity for the same particle size, mass flow rate and pressure in the receiving hopper. The flowability of pulverized coal decreases with increasing the moisture content in the range from 3.24% to 8.18%. Unconfined yield strength (UYS) increases and flow function (FF) decreases with increasing the moisture content. Results of the shearing tests are consistent with the results of the conveying study. 7Field trial of an HTS filter system on a CDMA base station显示文摘This paper presents the field trial results of a high temperature superconductor filter system (HTSFS) in a code division multiple access (CDMA) mobile communication base station (BTS) in Tangshan, China. The center frequency of the HTSFS is 830 MHz with 1.4% fractional bandwidth. The noise figure (NF) of the HTSFS, including connectors and cables, is 0.8 dB. The field trial results show that after the HTSFS was installed in the receiver front-end of a CDMA base station, the transmitting power of handset was decreased by 3.1 dB on average. 10Effects of Tillage Management on Infiltration and Preferential Flow in a Black Soil, Northeast China显示文摘The impacts of no-tillage (NT) and moldboard plough (MP) managements on infiltration rate and preferential flow were characterized using a combined technique of double-ring device and dye tracer on a black soil (Mollisols) in Northeast China. The objective of this study is to evaluate how tillage practices enhance soil water infiltration and preferential flow in favor of soil erosion control in the study area. The steady infiltration rates under NT management are 1.6 and 2.1 times as high as those under MP management in the 6th and 8th years of the tillage management in place, while the infiltrated water amounts under NT management are 1.4 and 2.0 times as high as those under MP management, respectively. The depth of methylene blue penetrated into NT soil increases from 43cm in the 6th year to 57cm in the 8th year, which are 16cm and 19cm deeper than those in MP soil, respectively. The results of morphologic image show that more biological macro-pores occur in NT soil than in MP soil. These macro-pores play a key role in enhancing preferential flow in NT soil, which in turn promotes water infiltration through preferential pathways in NT soil. 14Thrombolysis with alteplase 3-4.5 hours after acute ischaemic stroke:the first multicentre,phase III trial in China显示文摘background and purpose Data on the efficacy and safety of alteplase for acute ischaemic stroke(AIS)administered 3-4.5 hours after the onset of stroke symptoms in Chinese patients is limited.We sought to determine whether AIS patients would benefit from thrombolysis with alteplase between 3 and 4.5 hours after the onset of stroke symptoms in a prospective,multicentre,single-arm trial in China.Materials and methods Eligible AIS patients were given 0.9 mg/kg alteplase intravenously.The primary efficacy endpoint was a favourable outcome at 3 months,defined as a score of 0 or 1 on the modified Rankin Scale.Thresholds for the primary efficacy endpoint were determined to be 40%based on the literature review.The primary safety endpoint was symptomatic intracranial haemorrhage(sICH)according to the European Cooperative Acute Stroke Study III(ECASS III)trial definition.Post hoc analysis between this study and the ECASS III trial were compared using the propensity score matching(PSM)method.results A total of 120 eligible AIS patients from 11 sites in China received thrombolysis therapy in this study.The median time from onset of symptoms to needle was 3 hours 54 min.The percentage of patients with a favourable outcome was 63.3%(95%CI 54.4 to 71.4),significantly higher than the predefined threshold(p<0.0001).Three patients(2.5%,95%CI 0.5 to 7.1)had sICH,including two fatal sICH.Six patients died within 3 months after treatment.The post hoc PSM analysis showed a numerically higher rate of the primary efficacy endpoint in this study(63.3%)than the matched placebo arm(56.7%)in the ECASS III trial.Conclusions Intravenous alteplase with a standard dose administered between 3 and 4.5 hours after onset of symptoms is effective and safe for Chinese AIS patients.Huaguang Zheng Yi Yang Huisheng Chen Chuanling Li Yangkun Chen Fu-Dong Shi Li Yang Xiaoping Cui Zuneng Lu Yanling Liang Songbiao Cui Anding Xu Yiqing Wu Yaqing Sun Yongjun Wang 2020Stroke & Vascular Neurology2020,5,3:4
15Sub-micrometer-scale chemical analysis by nanosecond-laser-induced tip-enhanced ablation and ionization time-of-flight mass spectrometry显示文摘The tip-enhanced near-field technique has drawn great attention recently becauseit is a promising technique for nanoscale chemical analysis when combined withother spectroscopic methods. Furthermore, this integration can improve the spatialresolution of mass spectrometry. In this stud~ a nanosecond-laser (nsqaser)-inducedtip-enhanced ablation and ionization source was coupled to an in-house-builtlaser ionization time-of-flight mass spectrometer. Sub-micrometer-sized craters(with diameters of 200-300 nm) were observed on titanium (Ti) film coated ontoa gold (Au) substrate. The corresponding mass spectra were acquired, whichverified that sub-micrometer-sized ablation (and subsequently ionization) couldbe achieved by the ns-laser-induced tip-enhanced electromagnetic field. Inaddition, formation of the crater was studied with an increased number of laserpulses. 17Resection of Giant Hepatic Cavernous Hemangiomas after Dissection of the Third Porta Hepatis显示文摘Objective:To estabhsh a novel and safe operation technique for the resection of giant hepatic cavernous hemangiomas involving the retro-hepatic vena cava.Methods:After ligating the hepatic artery of affected lobe, the short hepatic veins at the third porta hepatis were dissected and ligated individually to separate the tumor from the retrohepatic vena cava, followed by the resection of the tumor under intermittent interruption of the porta hepatis.Results:A total of 62 giant hepatic cavernous hemangiomas were successfully resected without hepatic vascular exclusion. Right and caudate lobectomies were done in 27 cases, right hemihepatectornies in 5 cases, right upper segnentectomies in 7 cases, right posterior lobectomies in 7 cases, extended left and caudate lobectomies in 10 cases, and caudate lobectomies in 6 cases. The blood transfusion requirement during operation was 1 400 ml on average. All did well postoperatively during a follow up of 4 - 84 months.Conclusion:It is safe and feasible to resect giant hepatic cavernous hemangioma following dissection of the third porta hepatis. 19Modified methods for culturing myoblasts of rats: Combination of multi-enzymatic digestion and double purification显示文摘BACKGROUND: With developments of tissue engineering and genetic engineering, we aim to culture myoblasts, which are characterized by high purity, high quality and high production, for wide application in neural regeneration researches. OBJECTIVE: To modify traditional dissociation method in order to obtain myoblasts, which are characterized by high purity, high quality and high production, and explore the biological properties under in vitro culture. DESIGN: Observational study. SETTING: Basic Institute of Academy of Military Medical Sciences of Chinese PLA. MATERIALS: Four neonatal Wistar rats of 5 days old, both genders and mean body mass of 10 g were selected in this study. The main reagents and devices were detailed as follows: DMEM medium (Gibco Company), fetus bovine serum (FBS, Hycolne Company), collagenase Ⅱ(Sigma Company), trypsin (Sigma Company), dispase Ⅱ (Sigma Company), desmin antibody (Fuzhou Maixin Company), antibody Ⅱ and ABC kit (Wuhan Boster Biotechnology Company), desk centrifuge (KUBATO, Japan), and inverted phase contrast microscope (LEICA DMIRB, Germany). METHODS: The experiment was carried out in the Basic Institute of Academy of Military Medical Sciences of Chinese PLA from June to October 2006. Neonatal rats were sacrificed under sterile condition to obtain skeletal muscles of limbs, which were washed with cold PBS (containing benzylpenicillin and estreptomicina), and muscular tissue was sheared into pieces. Then, those muscular pieces were added with mixed digestive enzyme (containing 2 g/L collagenase Ⅱ + 5 g/L dispase Ⅱ + 0.28 g/L CaCl2) as twice volume as pieces, dealt with mechanical pipetting for 5 minutes and cultured in CO2 incubator for 10 minutes. The operation was done for three times and the muscular pieces were digested for 45 minutes in total. Moreover, cells were suspended again in order to obtain myoblasts from skeletal muscle of neonatal rats. In addition, myoblasts were purified with differential attachment technique and enzyme digestion so as to observe morphological characteristics and growth, draw growth curve, analyze surface structure under scanning electron microscope, and evaluate with Desmin immunohistochemical staining. MAIN OUTCOME MEASURES: Morphological characteristics and growth of myoblasts cultured in vitro. RESULTS: ① Growth of myoblasts of skeletal muscle: Primary cells had well growth, mature and differentiation. The positive rate of Desmin was 94% and purification of cells was ideal. Growth curve of cells demonstrated that myoblasts which were characterized by high purification started proliferation plentiful through transient growth lag phase (about at one or two days after inoculation). If myoblasts were not dealt with any interventions, they might become sarcotubule gradually at 3–5 days after proliferative phase. During this period, myoblasts maintained a monocaryon-bipolarity state under inverted phase contrast microscope. Furthermore, the growth of cells was the strongest and reproductive activity was the most powerful. This suggested that myotube started to form; in addition, muscle fiber of contractility might form under a well culturing condition. ② Immunocytochemical stain with desmin antibody: Interzonal fiber of desmin from myoblasts showed strongly positive reaction. Positive staining existed in cytoplasm had a high nucleus-cytoplasm ratio. However, myoblasts showed negative or mildly positive reaction. CONCLUSION: It is ideal for modified multi-enzymatic digestion and double purification method to dissociate and purify myoblasts of skeletal muscle; meanwhile, these two methods are both the effective ways to provide convenient conditions to obtain seed cells for neural regeneration researches.Li Zhang Wei Wang Ming Fan Xiaoping Chen Shuhong Liu Liang Sun 2007Neural Regeneration Research2007,2,1:3
20Effect of puerarin on myocardial perfusion and ventricular wall motion in patients with acute coronary syndrome显示文摘Objective To investigate the effects of puerarin (Pur) on myocardial perfusion and ventricular wall motion in patients with acute coronary syndrome (ACS).Methods Thirty-seven patients with ACS were randomly divided into two groups:conventional treatment group (n= 17,11 males,range of age:32-80 years,average age:60.9±4.9 years) and Pur treatment group (n=20,12 males,range of age:40-76 years,average age:62.7±3.5 years).Patients in the conventional treatment group received standard treatment according to the current guidelines,while patients in the Pur treatment group received intravenous administration of Pur (500 mg/day) for 10 days plus conventional treatment.Real-time myocardial contrast echocardiography (RT-MCE) was performed to evaluate the change in myocardial perfusion index (MPI) and ventricular wall motion index (VWMI) at admission and 10 days after treatment.Results At 10 days after treatment,MPI was significantly higher (P<0.01) and VWMI significantly lower (P<0.01) in the Pur group comparing with those in the conventional group.Conclusions Puerarin might improve myocardial microcirculation perfusion and ventricular wall motion in patients with ACS.Ling Niu Dongye Li Yong Xia Defeng Pan Xiaoping Wang Yan Yan Li Liang Tongda Xu Cardiovascular Institute of Xuzhou Medical College, Department of Cardiology, Affiliated Hospital, Xuzhou Medical College, Xuzhou, Jiangsu 221002, China 2008Journal of Geriatric Cardiology2008,5,3:3
