2Preparation and characterization of monodisperse spherical particles of X-ray nano-phosphors based on Gd_2O_2S:Tb显示文摘Monodisperse spherical Gd2O2S:Tb nanoparticles have been prepared using an improved homogeneous precipitation method combined with solid-gas sulfuration technology. The effects of Tb3+-doped concentration on luminescent intensity and color purity of samples were investigated, and the optimal Tb3+-doped concentration was determined. 3Recombinant rabies virus expressing IL-15 enhances immunogenicity through promoting the activation of dendritic cells in mice显示文摘Rabies remains a public health threat that kills approximately 59,000 people worldwide each year,most of which are from the developing countries of Africa and Asia where dog rabies are endemic.Therefore, developing an affordable and efficacious vaccine is crucial for rabies control in these countries. Interleukin(IL)-15, an immunoregulatory cytokine, is a pluripotent molecule with therapeutic potential, which targets many cell types and links the innate and adaptive immune system. In this study, IL-15 gene was cloned and inserted into the genome of a recombinant rabies virus(RABV) strain LBNSE(designated as LBNSE-IL15), and the effect of over-expression of IL-15 on the immunogenicity of RABV was investigated. It was found that mice vaccinated with LBNSEIL15 could induce significantly higher level of virus-neutralizing antibody(VNA) than those immunized with LBNSE, resulting in the higher protection after challenge. Further investigation was performed to find out the possible role of IL-15 plays in the process of antibody induction, and it was found that LBNSE-IL15 could enhance the maturation of dendritic cells(DCs) in immunized mice. Furthermore, the mice immunized with LBNSE-IL15 could promote the T_(FH) cells differentiation and the generation of germinal center B cells and plasma cells. 4Crustal S-velocity structure and radial anisotropy beneath the southern part of central and western North China Craton and the adjacent Qilian Orogenic Belt from ambient noise tomography显示文摘The crustal S-velocity structure and radial anisotropy along a dense linear portable seismic array with 64 broadband seismic stations were investigated from ambient noise tomography with about one-year-long ambient noise recordings. The array transverses the southern part of the central North China Craton(CNCC) and western NCC(WNCC) from east to west and reaches the adjacent Qilian Orogenic Belt(QOB). The phase velocity structures of Rayleigh waves at 5–35 s and Love waves at 5–30 s were measured. The crustal S-velocity structures(Vsv and Vsh) were constructed from the dispersion data(Rayleigh and Love waves,respectively) from point-wise linear inversion with prior information of the Moho depth and average crustal Vp/Vs ratio. The radial anisotropy along the profile was calculated based on the discrepancies between Vsv and Vsh as 2×(Vsh.Vsv)/(Vsh+Vsv). The results show distinct structural variations in the three major tectonic units. The crustal architecture in the southern CNCC is complicated and featured with wide-distributed low-velocity zones(LVZs), which may be a reflection of crustal modification resulting from Mesozoic-Cenozoic tectonics and magmatic activities. The pronounced positive radial anisotropy in the lower-lowermost crust beneath the Shanxi-Shaanxi Rift and the neighboring areas could be attributed to the underplating of mantle mafic-ultramafic materials during the Mesozoic-Cenozoic tectonic activation. In southern Ordos, the overall weak lateral velocity variations, relative high velocity and large-scale positive radial anisotropy in mid-lower crust probably suggest that the current crustal structure has preserved its Precambrian tectonic characteristics. The low-velocity westward-dipping sedimentary strata in the Ordos Block could be attributed to the Phanerozoic whole-basin tilting and the uneven erosion since late Cretaceous. Integrated with previous studies, the systematic comparison of crustal architecture was made between the southern and northern part of CNCC-WNCC. The similarities and differences may have a relation with the tectonic events and deformation histories experienced before and after the Paleoproterozoic amalgamation of the NCC. 5A Recombinant Rabies Virus Expressing Fms-like Tyrosine Kinase 3 Ligand(Flt3L) Induces Enhanced Immunogenicity in Mice显示文摘Rabies is a zoonotic disease that still causes 59,000 human deaths each year,and rabies vaccine is the most effective way to control the disease.Our previous studies suggested that the maturation of DC plays an important role in enhancing the immunogenicity of rabies vaccine.Flt3L has been reported to own the ability to accelerate the DC maturation,therefore,in this study,a recombinant rabies virus expressing mouse Flt3L,designated as LBNSE-Flt3L,was constructed,and its immunogenicity was characterized.It was found that LBNSE-FU3L could enhance the maturation of DC both in vitro and in vivo,and significantly more TFH cells and Germinal Center B(GC B)cells were generated in mice immunized with LBNSE-FU3L than those immunized with the parent virus LBNSE.Consequently,expressing of Flt3L could elevate the level of virus-neutralizing antibodies(VNA)in immunized mice which provides a better protection from a lethal rabies virus challenge.Taken together,our study extends the potential of Flt3L as a good adjuvant to develop novel rabies vaccine by enhancing the VNA production through activating the DC—Tfh^GC B axis in immunized mice.Yachun Zhang Jie Yang Mingming Li Min Cui Zhen FFu Ling Zhao Ming Zhou 2019Virologica Sinica2019,34,6:3
6Rice FLOURY ENDOSPERM 18 encodes a pentatricopeptide repeat protein required for 5′ processing of mitochondrial nad5 messenger RNA and endosperm development显示文摘Pentatricopeptide repeat(PPR) proteins, composing one of the largest protein families in plants,are involved in RNA binding and regulation of organelle RNA metabolism at the posttranscriptional level. Although several PPR proteins have been implicated in endosperm development in rice(Oryza sativa), the molecular functions of many PPRs remain obscure. Here, we identified a rice endosperm mutant named floury endosperm 18(flo18) with pleiotropic defects in both reproductive and vegetative development.Map-based cloning and complementation tests showed that FLO18 encodes a mitochondriontargeted P-type PPR protein with 15 PPR motifs.Mitochondrial function was disrupted in the flo18 mutant, as evidenced by decreased assembly of Complex I in the mitochondrial electron transport chain and altered mitochondrial morphology. Loss of FLO18 function resulted in defective 5′-end processing of mitochondrial nad5 transcripts encoding subunit 5 of nicotinamide adenine dinucleotide hydrogenase. 8The impact of lipids on the cancer—immunity cycle and strategies for modulating lipid metabolism to improve cancer immunotherapy显示文摘Lipids have been found to modulate tumor biology,including proliferation,survival,and metastasis.With the new understanding of tumor immune escape that has developed in recent years,the influence of lipids on the cancer—immunity cycle has also been gradually discovered.First,regarding antigen presentation,cholesterol prevents tumor antigens from being identified by antigen presenting cells.Fatty acids reduce the expression of major histocompatibility complex class I and costimulatory factors in dendritic cells,impairing antigen presentation to T cells.Prostaglandin E2(PGE2)reduce the accumulation of tumor-infiltrating dendritic cells.Regarding T-cell priming and activation,cholesterol destroys the structure of the T-cell receptor and reduces immunodetection.In contrast,cholesterol also promotes T-cell receptor clustering and relative signal transduction.PGE2 represses T-cell proliferation.Finally,regarding T-cell killing of cancer cells,PGE2 and cholesterol weaken granule-dependent cytotoxicity.Moreover,fatty acids,cholesterol,and PGE2 can improve the activity of immunosuppressive cells,increase the expression of immune checkpoints and promote the secretion of immunosuppressive cytokines.Given the regulatory role of lipids in the cancer—immunity cycle,drugs that modulate fatty acids,cholesterol and PGE2 have been envisioned as effective way in restoring antitumor immunity and synergizing with immunotherapy.These strategies have been studied in both preclinical and clinical studies.Mingming Zheng Wenxin Zhang Xi Chen Hongjie Guo Honghai Wu Yanjun Xu Qiaojun He Ling Ding Bo Yang 2023Acta Pharmaceutica Sinica B2023,13,4:1
9Cellular antioxidant activity and cytotoxicity assay of canolol显示文摘To better understand canolol as an efficient intracellular antioxidant agent in food related fields, potential cellular antioxidant activity and cytotoxicity were evaluated using multiple assays, including cellular antioxidant activity(CAA), 3-(4,5)-dimethylthiahiazo(-z-y1)-3,5-di-phenytetrazoliumromide(MTT) and methylene blue assay. CAA assay was used to identify the half maximal effective concentration of canolol and other bioactive components, such as sinapic acid, resveratrol and sesamol in oil crops. Results showed that antioxidant capacity(without PBS wash) varies between different compounds. Capacity variation followed the order: sinapic acid > canolol > resveratrol > sesamol. While with PBS wash, the order changed to sinapic acid > resveratrol > canolol > sesamol. Half maximal inhibitory concentration of canolol measured by MTT was 1887.08±1.22 μmol/L, implying that cytotoxicity of canolol was greatly lower than that of resveratrol, sesamol and sinapic acid. Methylene blue assay showed that cell viability was still up to 90% exposed to canolol at a high concentration of 4444 μmol/L. 12Effect on the Performance of Drilling Fluids at Downhole Rock Surfaces at Low Temperatures显示文摘To maintain gas hydrate stability, low-temperature drilling fluids and high drilling speeds should be used while drilling in gas hydrate-bearing sediments. The effect of the drilling fluid on downhole rock surfaces at low temperatures is very important to increase the drilling rate. This paper analyzed the action mechanism of the drilling fluid on downhole rock surfaces and established a corresponding evaluation method. The softening effect of six simulated drilling fluids with 0.1 wt.% of four common surfactants and two common organic salts on the downhole rock surface strength was evaluated experimentally using the established method at low temperature. The experimental results showed that the surfactants and organic salts used in the drilling fluids aided in the reduction of the strength of the downhole rock surface, and the established evaluation method was able to quantitatively reveal the difference in the softening effect of the different drilling fluids through comparison with water. 17Puerarin inhibits NHE1 activity by interfering with the p38 pathway and attenuates mitochondrial damage induced by myocardial calcium overload in heart failure rats显示文摘Previous studies have shown that puerarin plays a key role in protecting humans and animals from cardiovascular diseases.The exact mechanism of the therapeutic effect of puerarin on various cardiovascular diseases(protective effect on cardiomyocytes)is still unclear.In the present study,we identify the role of puerarin in an animal model of experimental heart failure(HF)and explore its underlying mechanisms.The HF rat model is induced by in-traperitoneal injection of adriamycin(ADR),and puerarin is administered intragastrically at low,medium,and high concentrations.We demonstrate that puerarin significantly improves myocardial fibrosis and inflammatory in-filtration and,as a result,improves cardiac function in ADR-induced HF rats.Mechanistically,we find for the first time that puerarin inhibits overactivated Na+/H+exchange isoform 1(NHE1)in HF,which may improve HF by decreasing Na+and Ca2+ion concentrations and attenuating mitochondrial damage caused by calcium overload;on the other hand,puerarin inhibits the activation of the p38 pathway in HF,reduces the expressions of TGF-βand proinflammatory cytokines,and suppresses myocardial fibrosis.In conclusion,our results suggest that Puerarin is an effective drug against HF and may play a protective role in the myocardium by inhibiting the activation of p38 and its downstream NHE1.Guopin Pan Baoyue Cui Mingming Han Laibiao Lin Yinlan Li Ling Wang Shuang Guo Yaling Yin Heqin Zhan Peng Li 2024Acta Biochimica et Biophysica Sinica2024,56,2:0

19Melatonin restores normal Bax and Bcl-2 protein expression in the subgranular zone of the dentate gyrus in pinealectomized rats显示文摘In this study, we sought to elucidate the effects of melatonin on learning and memory as well as apoptosis and expression of the Bax or Bcl-2 proteins in the subgranular zone of the dentate gyrus in pinealectomized rats. Using the Morris water maze and the olfactory memory tests, we found that the average escape latency in pinealectomized rats was clearly increased compared with sham-operated rats. Moreover, the average escape latency in the melatonin-treated and pinealectomized rats was longer than that in the sham-operated rats and shorter than that in the pinealectomized and untreated rats. Immunohistochemistry and terminal-deoxynucleoitidyl transferase mediated nick end labeling (TUNEL) showed that there were fewer Bax immunoreactive cells and TUNEL-positive (apoptotic) cells but more Bcl-2 immunoreactive cells in the melatonin-treated rats compared with the pinealectomized rats. The sham-operated rats showed numbers of these cells similar to the melatonin-treated rats. These experimental findings demonstrate that melatonin treatment may reduce abnormal apoptosis by promoting gene expression of Bax and suppressing gene expression of Bcl-2 in the subgranular zone of the dentate gyrus in pinealectomized rats.
