| 2 | Enhanced Expression of miR-425 Promotes Esophageal Squamous Cell Carcinoma Tumorigenesis by Targeting SMAD2显示文摘Esophageal squamous cell carcinoma(ESCC) is one of the most common and deadly cancers in the world. Currently, clinical therapy of ESCC remains limited and the five-year survival rate is poor. The function of miR-425 has been reported in multiple human cancers.However, the tumorigenic role and clinical significance of miR-425 in ESCC remains unclear. We found that enhanced expression of miR-425 in ESCC cell lines not only promoted cell proliferation and colony formation, but also increased cellular metastasis. Furthermore, we revealed the mechanism that miR-425 inhibited the expression of SMAD2 by targeting the second binding site in the 30-untranslated region(30-UTR) in ESCC. This mode of action influenced not only SMAD2 mRNA expression but also protein expression. In addition, we detected the expression of miR-425 in ESCC tissues and plasma. Moreover, we analyzed the relationship between miR-425 expression and SMAD2 m RNA expression. We found that miR-425 was overexpressed in ESCC tissues and the plasma relative to adjacent normal tissues and plasma of healthy individuals. Furthermore, there was a negative correlation between miR-425 expression and SMAD2. | 3 | TRAP1 Shows Clinical Significance and Promotes Cellular Migration and Invasion through STAT3/MMP2 Pathway in Human Esophageal Squamous Cell Cancer显示文摘Tumor necrosis factor receptor-associated protein 1(TRAP1), an important member of mitochondrial heat shock protein 90 family, is involved in multiple biological processes in several types of tumors. However, its pathological role in esophageal squamous cell cancer(ESCC) remains unknown. Herein, we demonstrated the clinical value of TRAP1, and its role in apoptosis and motility in ESCC. The clinical potential of TRAP1 was investigated through immunohistochemical analysis in 328 ESCC samples, which revealed that strong TRAP1 expression was associated with increased risk of lymph node metastasis, while high TRAP1 expression correlated with poor prognosis. Expression of TRAP1 was found to be an independent prognostic factor for patients with ESCC. Additionally, the upregulation of TRAP1 antagonized cisplatin-induced apoptosis while its downregulation sensitized cells to cisplatin-induced apoptosis. As revealed by the transwell assay, TRAP1 overexpression promoted cellular migration and invasion as compared to the control groups. In contrast,silencing of endogenous TRAP1 expression attenuated the ability of migration and invasion. Finally, the molecular mechanism investigated in the present study demonstrated that TRAP1-mediated migration and invasion occurred through STAT3/MMP2 signaling pathway. Eventually, a solid shell of Ag is capped onto the Ag-modified Fe3O4 nanoparticles by reducing aliquots of AgNO3 solution using NaBH4 as a reducing agent. The as-prepared Fe3O4 -core@Ag-shell nanoeggs, which are uniform in size, highly monodisperse, stable against aggregation and have high magnetization, were depended on thin shell of the attached nano-Ag, the size of Fe3O4 nanoparticles, the volume of thiol-terminated silane as well as the ultrasonication. | 5 | Joint effects of Penta-BDE and heavy metals on Daphnia magna survival,its antioxidant enzyme activities and lipid peroxidation显示文摘The joint toxicity of Penta-BDE(Pe-BDE)and heavy metals including cadmium and copper on Daphnia magna(D.magna)was evaluated on the basis of determining the 48 h survival,antioxidative enzyme responses,and lipid peroxidation.The response was classified as additive,greater than additive,or less than additive by comparing the measured“toxic units,TU”with one.Based on the survival of D.magna,less-than-additive interactions were found in most of mixtures treatments.This may be attributed to the different toxicity mechanism between Pe-BDE and metals.Cu and Cd played a greater role in toxicity than what Pe-BDE did.As for the superoxide dismutase(SOD)and catalase(CAT)activity,most response was less than additive.For the glutathione S-transferases(GST)activity,most of the greater-thanadditive responses were found in the Cu plus Pe-BDE treatments,but the additive responses occurred in Cd plus Pe-BDE treatments and binary metal treatments.For lipid peroxide levels,which were measured as malondialdehyde(MDA)levels,less-than-additive response occurred in the 50%Cd plus 50%Cu and ternary mixture treatments.Results suggested that Pe-BDE,Cd,and Cu could induce different patterns of antioxidant enzyme responses,such as antioxidant/prooxidant responses,depending on their capability to produce reactive oxygen species and antioxidant enzymes to detoxify them. | Baohua TANG Lingyan ZHU Qixing ZHOU | 2011 | Frontiers of Environmental Science & Engineering2011,5,1: | 5 |
| 6 | Different Mortality Effects of Extreme Temperature Stress in Three Large City Clusters of Northern and Southern China显示文摘Extreme temperature events have affected Chinese city residents more frequently and intensively since the early 2000 s, but few studies have identified the impacts of extreme temperature on mortality in different city clusters of China. This study first used a distributed lag,nonlinear model to estimate the county/district-specific effects of extreme temperature on nonaccidental and cardiovascular mortality. The authors then applied a multivariate meta-analysis to pool the estimated effects in order to derive regional temperature–mortality relationship in three large city clusters—the Beijing-Tianjin-Hebei(BTH)region, the Yangtze River Delta(YRD), and the Pearl River Delta(PRD), which represent northern and southern regions. With 0–3 days' lag, the strongest heat-related mortality effect was observed in the BTH region(with relative risk(RR) of 1.29; 95% confidence interval(CI):1.13–1.47), followed by the YRD(RR = 1.25; 95% CI:1.13–1.35) and the PRD(RR = 1.14; 95% CI: 1.01–1.28)areas. With 0–21 days' lag, the cold effect was pronounced in all city clusters, with the highest extreme cold-related mortality risk found in the PRD area(RR = 2.27; 95% CI:1.63–3.16), followed by the YRD area(RR = 1.85; 95%CI: 1.56–2.20) and BTH region(RR = 1.33; 95% CI:0.96–1.83). People in the southern regions tended to be more vulnerable to cold stress, but the northern population was more sensitive to heat stress. | 8 | Prognostic value of ^(18)F-fluorodeoxyglucose positron emission tomography using Deauville criteria in diffuse large B cell lymphoma treated with autologous hematopoietic stem cell transplantation显示文摘Objective: High-dose chemotherapy(HDC) followed by autologous hematopoietic stem cell transplantation(auto-HSCT) plays an important role in improving outcomes of diffuse large B cell lymphoma(DLBCL) patients.18 F-fluorodeoxyglucose(18 F-FDG) positron emission tomography(PET)/computed tomography(CT) has been widely accepted in response assessment and prediction of prognosis in DLBCL. Here, we report the value of 18 FFDG PET/CT pre-and post-HSCT in predicting outcomes of patients with DLBCL.Methods: DLBCL patients who had PET/CT scan before and after HSCT were included. PET results were interpreted based upon Deauville criteria. The prognostic value of 18 F-FDG PET/CT in auto-HSCT was evaluated.Results: Eighty-four patients were enrolled. In univariate analysis, pre-and post-HSCT PET findings were correlated with 3-year progression-free survival(PFS) [hazard ratio(HR)=4.391, P=0.001; HR=7.607, P<0.001] and overall survival(OS)(HR=4.792, P=0.008; HR=26.138, P<0.001). Patients receiving upfront auto-HSCT after firstline treatment had better outcomes than relapsed/refractory DLBCL patients(3-year PFS, P<0.001; 3-year OS,P<0.001). In the relapsed/refractory patients, pre-and post-HSCT PET findings were also associated with 3-year PFS(P=0.003 vs. P<0.001) and OS(P=0.027 vs. P<0.001). A significant correlation was observed between clinical response to chemotherapy before auto-HSCT and outcomes of patients in the entire cohort(3-year PFS, P<0.001;3-year OS, P<0.001) and in the subgroup of 21 patients with positive pre-HSCT PET(3-year PFS, P=0.084; 3-year OS, P=0.240). A significant association between survival and post-HSCT PET findings was observed in multivariate analysis(HR=5.168, P<0.001).Conclusions: PET results before and after HSCT are useful prognostic factors for DLBCL patients receiving HSCT. | 10 | Characterization and Mechanism Elucidation of Dye Adsorption Using Cuprous Selenide Nanoparticles from Aqueous Solutions显示文摘The removal of cationic dyes, methylene blue(MB) and rhodamine B(RB), and anionic dyes, methylorange(MO) and eosin Y(EY), from aqueous solutions by adsorption using Cu2Se nanoparticles(Cu2SeNPs) was stu-died. The effects of the initial pH values, adsorbent doses, contact time, initial dye concentrations, salt concentrations,and operation temperatures on the adsorption capacities were investigated. The adsorption process was better fittedthe Langmuir equation and pseudo-second-order kinetic model, and was spontaneous and endothermic as well. Theadsorption mechanism was probably based on the electrostatic interactions and π-π interactions between Cu2SeNPsand dyes. For an adsorbent of 0.4 g/L of Cu2SeNPs, the adsorption capacities of 23. I(MB), 22.9(RB) and 23.9(EY)mg/g were achieved, respectively, with an initial dye concentration of 10 mg/g(pH=8 for MB and pH=4 for RiB andEY) and a contact time of 120 min. The removal rate of MB was still 70.4% for Cu2SeNPs being reused in the 5thcycle. Furthermore, the recycled Cu2SeNPs produced from selenium nanoparticles adsorbing copper were also an ef-fective adsorbent for the removal of dyes. | 11 | Temperature Effects on Unsaturated Hydraulic Property of Bentonite-Sand Buffer Backfilling Mixtures显示文摘The influence of temperature on the engineered properties of bentonite-sand mixtures (B/S) is of major concern in the design of engineered barriers in underground repositories for high-level radioactive waste disposal. We experimentally studied the influence of temperature on soil unsaturated hydraulic properties related to water holding capacity and permeability of GMZ B/S in China. The vapor equilibrium method and water infiltration apparatus were used to measure the soil water characteristic curve (SWCC) and unsaturated hydraulic conductivity (ku ). The results show that the SWCC under different temperatures from 20 ℃ to 60 ℃ tends to be the same. | 13 | Optimization of Empty Pallets Dispatching Based on Different Transportation Modes显示文摘In the pallet pool system, the differentiation of palletized freight volumes in different regions and seasonal transport of certain goods lead to the imbalance of pallets distribution among regions. It is necessary to improve the utilization of pallets through dispatching. The paper analyzes the factors which affect empty pallets dispatching, it includes carbon emission, transportation time and pallet type based on the pallet pool mode of enterprise alliance. On this basis, the optimization model is established with the goal of minimum total dispatching cost. Then, according to the different influences of railway and highway in transportation cost, the dispatching scheme is analyzed and the transportation mode of empty pallets dispatching is determined. Considering the characteristics of model, Cplex is used to solve it. Finally, a case is used to verify the feasibility and superiority of reasonable empty pallets dispatching in different transportation modes, and the transport characteristics of two transportation modes are analyzed. | 14 | Responses of nutrient biogeochemistry and nitrogen cycle to seasonal upwelling in coastal waters of the eastern Hainan Island显示文摘The coastal upwelling has profound influence on the surrounding ecosystem by supplying the nutrient-replete water to the euphotic zone.Nutrient biogeochemistry was investigated in coastal waters of the eastern Hainan Island in summer 2015 and autumn 2016.From perspectives of nutrient dynamics and physical transport,the nutrient fluxes entered the upper 50 m water depth(between the mixed layer and the euphotic zone)arisen from the upwelling were estimated to be 2.5-5.4 mmol/(m^(2)·d),0.15-0.28 mmol/(m^(2)·d),and 2.2-7.2 mmol/(m^(2)·d)for dissolved inorganic nitrogen(DIN),phosphate(DIP),and dissolved silicate(DSi),respectively,which were around 6-to 12-fold those in the background area.The upwelled nutrients supported an additional plankton growth of(14.70±8.95)mg/m^(2)for chlorophyll a(Chl a).The distributions of nitrateδ^(15)N andδ^(18)O above the 300 m water depth(top of the North Pacific Intermediate Water)were different among the upwelling area,background area in summer,and the stations in autumn,and the difference of environmental and biogeochemical conditions between seasons should be the reason.The higher DIN/DIP concentration ratio,nitrate concentration anomaly,and lower nitrate isotope anomaly(Δ(15,18))in the upper ocean in summer than in autumn indicated the stronger nitrogen fixation and atmospheric deposition,and the following fixed nitrogen regeneration in summer.The higher values of Chl a and nitrateδ^(15)N andδ^(18)O within the euphotic zone in autumn than the background area in summer suggested the stronger nitrate assimilation in autumn.The differences in relatively strength of the assimilation,nitrogen fixation and atmospheric deposition,and the following remineralization and nitrification between the two seasons made the higherδ^(18)O:δ^(15)N and larger difference of enzymatic isotope fractionation factors^(15)εand^(18)εfor nitrate assimilation in summer than in autumn above the North Pacific Tropical Water. | Nan Zhou Sumei Liu Guodong Song Yunyan Zhang Lingyan Wang Xiaoyan Ning | 2022 | Acta Oceanologica Sinica2022,41,6: | 1 |

| 17 | Protective efficacy of Shenge San(参蛤散) on mitochondria in H9c2 cardiomyocytes显示文摘OBJECTIVE: To investigate whether peroxisome proliferator-activated receptor γ-coactivator-1α/nuclear respiratory factor 1(PGC-1α/NRF1) activity can protect mitochondrial function in the setting of cardiac hypertrophy and improve cardiomyocyte energy metabolism. METHODS: Cardiac hypertrophy was modeled in H9c2 cells treated with isoproterenol(ISO) to assess the effects of Shenge San( 参 蛤 散, SGS) on cell viability and mitochondrial membrane potential. We assessed mitochondrial complex m RNA levels and mitochondrial oxidative phosphorylation factor m RNA and protein levels. RESULTS: Compared with the 100 μM ISO group, cell size was significantly decreased in the 0.3 mg/m L SGS and 20 μM ZLN005(PGC-1α activator) groups(P < 0.01). Compared with the SGS(0.3) +ISO group, we observed lower phosphorylated adenosine monophosphateactivated kinase(AMPK) protein levels in the ISO and ZLN005+SGS+ISO groups(P < 0.01). Compared with the compound C group, SGS significantly increased PGC-1α expression in ISO-induced cardiac hypertrophy cells(P < 0.01), and this was inhibited by compound C pretreatment(P < 0.05). Compared with the ISO group, the mitochondrial red-green fluorescence ratio increased in the 0.3 mg/m L SGS group(P < 0.05). m RNA levels of cytochrome c oxidase subunit 1(CO1) in the ISO and compound C groups were lower than those in control group(P < 0.01), and the m RNA levels of CO1 and ATP8 were significantly lower in the ISO and compound C groups versus control(P < 0.01). Compared with the SGS(0.3) +ISO group, ATP synthetase subunit 8(ATP8) m RNA was significantly decreased in the ISO group(P < 0.01) and compound C+SGS+ISO group(P < 0.05). Compared with the SGS(0.3) +ISO group, NRF1 m RNA levels were significantly decreased(P < 0.05) in the ISO and compound C+SGS+ISO groups. CONCLUSIONS: SGS can attenuate ISO-induced cardiomyocyte hypertrophy, restore the decrease in mitochondrial membrane potential, and upregulate PGC-1α/NRF1 levels. | 18 | Preparation and Processing Properties of Nozzle Plate for Aluminum Continuous Strip Casting显示文摘The currently used ceramic nozzles for aluminum deep processing continuous hot rolled strips are featured with good integrity,high strength,good processing property,but non-uniform structure caused by the migration of silica sol. In this work,nozzle materials were prepared with aluminum silicate ceramic fiber,kaolin,and aluminum tripolyphosphate as starting materials,silica sol or silica sol- epoxy resin as the binder. After pulping,casting,curing at different temperatures and hot treatment,the prepared materials were analyzed in terms of migration layer thickness,cold modulus of rupture,and microstructure morphology. | 19 | Complexation with pre-formed“empty”V-type starch for encapsulation of aroma compounds显示文摘Aroma compounds are low-molecular-weight organic volatile molecules and are broadly utilized in the food industry.However,due to their high volatility and evaporative losses during processing and storage,the stabilization of these volatile ingredients using encapsulation is a commonly investigated practice.Complexation of aroma compounds using starch inclusion complex could be a potential approach due to the hydrophobicity of the left-handed single helical structure.In the present study,we used starch of three different V-type structures,namely V,V,and V,to encapsulate six different aroma compounds,including1-decanol(DN),cis-3-hexen-1-ol(HN),4-allylanisole(AN),γ-decalactone(DA),trans-cinnamaldehyde(CA),and citral(CT).The formed inclusion complexes samples were characterized using complementary techniques,including X-ray diffraction(XRD)and differential scanning calorimetry(DSC).The results showed that upon complexation with aroma compounds,all V-subtypes retained their original crystalline structures.However,different trends of crystallinity were observed for each type of the prepared inclusion complexes.Additionally,among three V-type starches,V-type starch formed inclusion complexes with aroma compounds most efficiently and promoted the formation of FormⅡcomplex.This study suggested that the structure of aroma compounds and the type of V starch could both affect the complexation properties. | Jingyi Zhou Lingyan Kong | 2023 | Food Science and Human Wellness2023,12,2: | 0 |
