| 1 | IL-10-producing regulatory B cells restrain the T follicular helper cell response in primary Sjögren’s syndrome显示文摘Increased numbers of T follicular helper(Tfh)cells have been implicated in the development of autoimmune diseases including primary Sjögren’s syndrome(pSS),but how the Tfh cell response is regulated during autoimmune pathogenesis remains largely unclear.Here,we first found negative correlations between IL-10^(+)regulatory B(Breg)cell numbers and Tfh cell responses and disease activity in patients with pSS and mice with experimental Sjögren’s syndrome(ESS).Moreover,we detected high expression of IL-10 receptor on Tfh cells and their precursors in both humans and mice.In culture,IL-10 suppressed human and murine Tfh cell differentiation by promoting STAT5 phosphorylation.By using an adoptive transfer approach and two-photon live imaging,we found significantly increased numbers of Tfh cells with enhanced T cell homing into B cell follicles in the draining cervical lymph nodes of RAG-2−/−mice transferred with IL-10-deficient B cells during ESS development compared with those of RAG-2−/−mice transferred with wild-type B cells.In ESS mice,CD19^(+)CD1d^(hi)CD5^(+)Breg cells with decreased IL-10 production exhibited severely impaired suppressive effects on T cell proliferation.Consistently,CD19^(+)CD24^(+)CD38^(hi) Breg cells from pSS patients showed significantly reduced IL-10 production with defective inhibitory function in the suppression of autologous Tfh cell expansion.Furthermore,the adoptive transfer of IL-10-producing Breg cells markedly suppressed the Tfh cell response and ameliorated ESS progression in ESS mice.Together,these findings demonstrate a critical role for IL-10-producing Breg cells in restraining the effector Tfh cell response during pSS development. | Xiang Lin Xiaohui Wang Fan Xiao Kongyang Ma Lixiong Liu Xiaoqi Wang Dong Xu Fei Wang Xiaofei Shi Dongzhou Liu Yan Zhao Liwei Lu | 2019 | Cellular & Molecular Immunology2019,16,12: | 13 |
| 2 | IL-17 sustains the plasma cell response via p38-mediated Bcl-xL RNA stability in lupus pathogenesis显示文摘Recent studies have demonstrated a central role for plasma cells in the development of autoimmune diseases,such as systemic lupus erythematosus(SLE).Currently,both the phenotypic features and functional regulation of autoreactive plasma cells during SLE pathogenesis remain largely unclear.In this study,we first found that a major subset of IL-17 receptor-expressing plasma cells potently produced anti-dsDNA IgG upon IL-17A(IL-17)stimulation in SLE patients and lupus mice.Using a humanized lupus mouse model,we showed that the transfer of Th17 cell-depleted PBMCs from lupus patients resulted in a significantly reduced plasma cell response and attenuated renal damage in recipient mice compared to the transfer of total SLE PBMCs.Moreover,long-term BrdU incorporation in lupus mice detected highly enriched long-lived BrdU+subsets among IL-17 receptor-expressing plasma cells.Lupus mice deficient in IL-17 or IL-17 receptor C(IL-17RC)exhibited a diminished plasma cell response and reduced autoantibody production with attenuated renal damage,while the adoptive transfer of Th17 cells triggered the plasma cell response and renal damage in IL-17-deficient lupus mice.In reconstituted chimeric mice,IL-17RC deficiency resulted in severely impaired plasma cell generation but showed no obvious effect on germinal center B cells.Further mechanistic studies revealed that IL-17 significantly promoted plasma cell survival via p38-mediated Bcl-xL transcript stabilization.Together,our findings identified a novel function of IL-17 in enhancing plasma cell survival for autoantibody production in lupus pathogenesis,which may provide new therapeutic strategies for the treatment of SLE. | Kongyang Ma Wenhan Du Fan Xiao Man Han Enyu Huang Na Peng Yuan Tang Chong Deng Lixiong Liu Yulan Chen Jingjing Li Shiwen Yuan Qin Huang Xiaoping Hong Dajun Hu Xiaoyan Cai Quan Jiang Dongzhou Liu Liwei Lu | 2021 | Cellular & Molecular Immunology2021,18,7: | 6 |
| 3 | Numerical and experimental investigations of liquid mixing in two-stage micro-impinging stream reactors显示文摘A two-stage micro-impinging stream reactor(TS-MISR) that combined a first pre-mixing stage with a second micro-impinging stream reacting stage for continuous multi-component reacting systems has been built from commercial T-junctions and steel micro-capillaries. Both of operating parameters and reactor configurations,such as jet Reynolds number(Rej), volumetric flow ratio(R), the first-stage junction angle(φ), the connecting capillary length(Lc) and connecting capillary diameter(dc), had significant effects on the micromixing efficiency of the reactor. Such effects were investigated for both of the two stage structures, respectively, by experimental and CFD methods and were optimized for the best micromixing performance. | 4 | Integrated Building Envelope Design Process Combining Parametric Modelling and Multi-Objective Optimization显示文摘As an important element in sustainable building design, the building envelope has been witnessing a constant shift in the design approach. Integrating multi-objective optimization(MOO) into the building envelope design process is very promising, but not easy to realize in an actual project due to several factors, including the complexity of optimization model construction, lack of a dynamic-visualization capacity in the simulation tools and consideration of how to match the optimization with the actual design process. To overcome these difficulties, this study constructed an integrated building envelope design process(IBEDP) based on parametric modelling, which was implemented using Grasshopper platform and interfaces to control the simulation software and optimization algorithm. A railway station was selected as a case study for applying the proposed IBEDP, which also utilized a grid-based variable design approach to achieve flexible optimum fenestrations. To facilitate the stepwise design process, a novel strategy was proposed with a two-step optimization, which optimized various categories of variables separately. | 5 | CFD and experimental investigations on the micromixing performance of single countercurrent-flow microchannel reactor显示文摘Microchannel reactors are widely used in different fields due to their intensive micromixing and, thus, high masstransfer efficiency. In this work, a single countercurrent-flow microchannel reactor(S-CFMCR) at the size of ~1 mm was developed by steel micro-capillary and laser drilling technology. Utilizing the Villermaux/Dushman parallel competing reaction, numerical and experimental studies were carried out to investigate the micromixing performance(expressed as the segregation index XS) of liquids inside S-CFMCR at the low flow velocity regime.The effects of various operating conditions and design parameters of S-CFMCR, e.g., inlet Reynolds number(Re),volumetric flow ratio(R), inlet diameter(d) and outlet length(L), on the quality of micromixing were studied qualitatively. It was found that the micromixing efficiency was enhanced with increasing Re, but weakened with the increase of R. Moreover, d and L also have a significant influence on micromixing. CFD results were in good agreement with experimental data. In addition, the visualization of velocity magnitude, turbulent kinetic energy and concentration distributions of various ions inside S-CFMCR was illustrated as well. | 7 | Combined treatment using acupuncture and music therapy on children with cerebral palsy Gross motor function measure comparison in 60 cases显示文摘BACKGROUND: The use of acupuncture has received recognition to effectively treat cerebral palsy. Moreover, music therapy can be used to modify treatment of cerebral palsy. OBJECTIVE: To study the effects of combined treatment using acupuncture and music therapy on gross motor function measure (GMFM) of children with cerebral palsy, compared with acupuncture treatment alone. DESIGN, TIME AND SETTING: Randomized, controlled, clinical study. The experiment was conducted in Shenzhen Hospital of Traditional Chinese Medicine between January 2007 and September 2007. PARTICIPANTS: All children with cerebral palsy in the trial were from the outpatient department of Shenzhen Hospital of Traditional Chinese Medicine. The children were randomly divided into two groups: 30 children in Group B received acupuncture and music therapy, while 30 children in Group A received only acupuncture therapy. METHODS: Subjects in Groups A and B received acupuncture based on syndrome differentiation. The main acupoints were necessary for all participants. At first, flash needling was applied to the acupoints. For the remaining acupoints, the technique of transverse needling was applied to the head acupoints, and perpendicular needling was used for the other points. The inserted needles were twirled and then maintained for 30 minutes. The needle was twirled for one second every other 10 minutes, without reinforcing-reducing techniques. The therapy was performed every other day. The trial consisted of three periods each, and lasted for 84 days. Subjects in Group B received music therapy. They listened to music that they preferred while acupuncture was being performed. Following acupuncture, they were allowed to perform musical activities, such as percussion, singing, and dancing. The music therapy was scheduled for one hour, including listening to music for 30 minutes and music activities for 30 minutes. MAIN OUTCOME MEASURES: The comprehensive functional evaluation scale of cerebral palsy and the GMFM were applied to test symptoms before and after respective treatment. RESULTS: All 60 children were included in final analysis. The general function of all children with cerebral palsy improved, but the general efficiency in Group B was obviously higher than in Group A (93% and 73%, χ2 = 4.32, P < 0.05). GMFM evaluation showed that the function of crawling and kneeling, standing, and walking improved more significantly in Group B than in Group A (P < 0.05), while the function of lying down, rolling, and sitting remained similar between the two groups (P > 0.05). CONCLUSION: The combination of acupuncture and music therapy on the treatment of cerebral palsy is more effective than acupuncture alone. | 15 | Pathogenic analysis of coxsackievirus A10 in rhesus macaques显示文摘Coxsackievirus A10(CV-A10)is one of the etiological agents associated with hand,foot and mouth disease(HFMD)and also causes a variety of illnesses in humans,including pneumonia,and myocarditis.Different people,particularly young children,may have different immunological responses to infection.Current CV-A10 infection animal models provide only a rudimentary understanding of the pathogenesis and effects of this virus.The characteristics of CV-A10 infection,replication,and shedding in humans remain unknown.In this study,rhesus macaques were infected by CV-A10 via respiratory or digestive route to mimic the HFMD in humans.The clinical symptoms,viral shedding,inflammatory response and pathologic changes were investigated in acute infection(1–11 day post infection)and recovery period(12–180 day post infection).All infected rhesus macaques during acute infection showed obvious viremia and clinical symptoms which were comparable to those observed in humans.Substantial inflammatory pathological damages were observed in multi-organs,including the lung,heart,liver,and kidney.During the acute period,all rhesus macaques displayed clinical signs,viral shedding,normalization of serum cytokines,and increased serum neutralizing antibodies,whereas inflammatory factors caused some animals to develop severe hyperglycemia during the recovery period.In addition,there were no significant differences between respiratory and digestive tract infected animals.Overall,all data presented suggest that the rhesus macaques provide the first non-human primate animal model for investigating CV-A10 pathophysiology and assessing the development of potential human therapies. | Suqin Duan Fengmei Yang Yanyan Li Yuan Zhao Li Shi Meng Qin Quan Liu Weihua Jin Junbin Wang Lixiong Chen Wei Zhang Yongjie Li Ying Zhang Jingjing Zhang Shaohui Ma Zhanlong He Qihan Li | 2022 | Virologica Sinica2022,37,4: | 1 |

| 19 | A novel SAXS/XRD/XAFS combined technique for in-situ timeresolved simultaneous measurements显示文摘Synchrotron radiation based combined technique can provide multiple structural information simultaneously,which is an important development direction of structural detection.In this study,a novel small-angle X-ray scattering/X-ray diffraction/X-ray absorption fine structure(SAXS/XRD/XAFS)combined setup was constructed,where an area detector,a curved detector,and a point detector are,respectively,used for the measurements of SAXS,XRD,and XAFS signals.A detailed description about the combined setup was given.A minitype diamond detector coupled to a SAXS beamstop was used to record the transmitted X-ray intensity,making the scattering(SAXS and XRD)signal measurement compatible with the absorption(XAFS)signal measurement,avoiding mechanical switching.The two-way sampling strategy was used to acquire XAFS signals,shortening the non-counting time.The two-way and one-way sampling strategies were discussed.High-frequency sampling scheme was used to collect experimental signals,improving the measurement efficiency and signal-to-noise ratio.A detailed description and discussion about the high-frequency scheme were also given in this paper.Except the rotation of monochromator,there is no mechanical movement in measurements,time resolution may reach the level of seconds.Using this SAXS/XRD/XAFS combined setup,SAXS,XRD,and XAFS signals can be acquired simultaneously.With some in-situ sample environment system,the newlydeveloped combined technique can be used to track the structure evolution in complex fluids.During the formation processes of(BiO)2CO3 and ZnAPO-34 particles,the changes of in-situ experimental data with reaction time demonstrate that SAXS/XRD/XAFS combined technique is feasible to track the dynamic process. | Zhonghua Wu Yunpeng Liu Xueqing Xing Lei Yao Zhongjun Chen Guang Mo Lirong Zheng Quan Cai Hao Wang Jiajun Zhong Yuecheng Lai Lixiong Qian | 2023 | Nano Research2023,16,1: | 0 |
