1AXL is a candidate receptor for SARS-CoV-2 that promotes infection of pulmonary and bronchial epithelial cells显示文摘The current coronavirus disease 2019(COVID-19)pandemic presents a global public health challenge.The viral pathogen responsible,severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),binds to the host receptor ACE2 through its spike(S)glycoprotein,which mediates membrane fusion and viral entry.Although the role of ACE2 as a receptor for SARS-CoV-2 is clear,studies have shown that ACE2 expression is extremely low in various human tissues,especially in the respiratory tract.Thus,other host receptors and/or co-receptors that promote the entry of SARS-CoV-2 into cells of the respiratory system may exist.In this study,we found that the tyrosine-protein kinase receptor UFO(AXL)specifically interacts with the N-terminal domain of SARS-CoV-2 S.Using both a SARS-CoV-2 virus pseudotype and authentic SARS-CoV-2,we found that overexpression of AXL in HEK293T cells promotes SARS-CoV-2 entry as efficiently as overexpression of ACE2,while knocking out AXL significantly reduces SARS-CoV-2 infection in HI 299 pulmonary cells and in human primary lung epithelial cells.Soluble human recombinant AXL blocks SARS-CoV-2 infection in cells expressing high levels of AXL.The AXL expression level is well correlated with SARS-CoV-2 S level in bronchoalveolar lavage fluid cells from COVID-19 patients.Taken together,our findings suggest that AXL is a novel candidate receptor for SARS-CoV-2 which may play an important role in promoting viral infection of the human respiratory system and indicate that it is a potential target for future clinical intervention strategies.Shuai Wang Zongyang Qiu Yingnan Hou Xiya Deng Wei Xu Tingting Zheng Peihan Wu Shaofang Xie Weixiang Bian Chong Zhang Zewei Sun Kunpeng Liu Chao Shan Aifu Lin Shibo Jiang Youhua Xie Qiang Zhou Lu Lu Jing Huang Xu Li 2021Cell Research2021,31,2:39

3Study the ethanol SI/HCCI combustion mode transition by using the fast thermal management system显示文摘In this paper the ethanol homogeneous charge compression ignition (HCCI) is achieved in a modified single cylinder engine by means of a self-developed fast thermal management system (FTMS), and the ethanol SI/HCCI operation regions are defined. It can be concluded that the thermal efficiency is higher and the NOx emission is lower in the HCCI operation region. In addition, the maximum NOx emission drops by 98%. The ethanol SI/HCCI combustion mode transition is conducted in different conditions near the SI/HCCI operation boundaries. It is likely to realize the transition by the utilization of FTMS. However, it is impossible to complete the transition within one operating cycle under current operation conditions. There are fluctuations in engine speed and brake mean effective pressure during the transition process. In order to reduce the fluctuations during the transition, the initial work concerning the effects of the spark ignition on the transition smoothness is carried out and the investigation indicates that the engine speed and brake mean effective pressure fluctuations cannot be eradicated only through spark ignition. 4The Arabidopsis Anaphase-Promoting Complex/Cyclosome Subunit 1 is Critical for Both Female Gametogenesis and Embryogenesis显示文摘Anaphase-promoting complex/cyclosome (APC/C), a multisubunit E3 ligase, plays a critical role in cell cycle control, but the functional characterization of each subunit has not yet been completed. To investigate the function of APC1 in Arabidopsis, we analyzed four mutant alleles of APC1, and found that mutation in APC1 resulted in significantly reduced plant fertility, accumulation of cyclin B, and disrupted auxin distribution in embryos. The three mutant alleles apc1-1, apc1-2 and apc1-3 shared variable defects in female gametogenesis including degradation, abnormal nuclear number, and disrupted polarity of nuclei in the embryo sac as well as in embryogenesis, in which embryos were arrested at multiple stages. All of these defects are similar to those previously identified in apc4. The mutant apc1-4, in which the T-DNA was inserted after the transmembrane domain at the C-terminus, showed much more severe phenotypes; that is, most of the ovules were arrested at the one-nucleate female gametophyte stage (stage FG1). In the apc1 apc4 double mutants, the fertility was further reduced by one-third in apc1-1/+ apc4-1/+, and in some cases no ovules even survived in siliques of apc1-4/+ apc4-1/+. 5Benefit of everolimus as a monotherapy for a refractory breast cancer patient bearing multiple genetic mutations in the PI3K/AKT/mTOR signaling pathway显示文摘A postmenopausal patient with a diagnosis of estrogen receptor(ER)(+), progesterone receptor(PR)(+), and human epidermal growth factor receptor-2(HER2)(-) breast cancer was reported. The patient refused surgery and was resistant to conventional chemotherapy regimens. Computed tomography and the circulating tumor cell test indicated that the patient's tumor burden increased rapidly even after several chemotherapy sessions. Multiple genetic aberrances in the phosphatidylinositol3-kinases(PI3 K) signaling pathway were detected via next-generation sequencing(NGS)-based liquid biopsy, including a p. G1007 R missense mutation in exon 21 of PIK3 CA(33.61%), a p.L70 fs frameshift mutation in exon 3 of phosphatase and tension homolog deleted on chromosome ten(PTEN)(49.14%), and a p. D1542 Y missense mutation in exon 32 of mammalian target of rapamycin(m TOR)(1.66%). Therefore, only the m TOR inhibitor everolimus was administered to the patient. Partial remission(PR) was observed after 2 months, and sustained stable disease(SD) was observed after a year and a half. Subsequent sequencing showed that the mutation ratio of PIK3 CA decreased to 4.17%, and that the PTEN and m TOR mutations disappeared, which revealed the significant curative effect of everolimus. We report the first case of successful monotherapy treatment using everolimus in a patient with advanced breast cancer bearing mutations in genes involved in the PI3 K/ARK/m TOR signaling pathway. 6Development of a large-scale remote sensing ecological index in arid areas and its application in the Aral Sea Basin显示文摘The Aral Sea Basin in Central Asia is an important geographical environment unit in the center of Eurasia.It is of great significance to the ecological protection and sustainable development of Central Asia to carry out dynamic monitoring and effective evaluation of the eco-environmental quality of the Aral Sea Basin.In this study,the arid remote sensing ecological index(ARSEI)for large-scale arid areas was developed,which coupled the information of the greenness index,the salinity index,the humidity index,the heat index,and the land degradation index of arid areas.The ARSEI was used to monitor and evaluate the eco-environmental quality of the Aral Sea Basin from 2000 to 2019.The results show that the greenness index,the humidity index and the land degradation index had a positive impact on the quality of the ecological environment in the Aral Sea Basin,while the salinity index and the heat index exerted a negative impact on the quality of the ecological environment.The eco-environmental quality of the Aral Sea Basin demonstrated a trend of initial improvement,followed by deterioration,and finally further improvement.The spatial variation of these changes was significant.From 2000 to 2019,grassland and wasteland(saline alkali land and sandy land)in the central and western parts of the basin had the worst ecological environment quality.The areas with poor ecological environment quality are mainly distributed in rivers,wetlands,and cultivated land around lakes.During the period from 2000 to 2019,except for the surrounding areas of the Aral Sea,the ecological environment quality in other areas of the Aral Sea Basin has been improved in general.The correlation coefficients between the change in the eco-environmental quality and the heat index and between the change in the eco-environmental quality and the humidity index were–0.593 and 0.524,respectively.Climate conditions and human activities have led to different combinations of heat and humidity changes in the eco-environmental quality of the Aral Sea Basin.However,human activities had a greater impact.The ARSEI can quantitatively and intuitively reflect the scale and causes of large-scale and long-time period changes of the eco-environmental quality in arid areas;it is very suitable for the study of the eco-environmental quality in arid areas.WANG Jie LIU Dongwei MA Jiali CHENG Yingnan WANG Lixin 2021Journal of Arid Land2021,13,1:5
7Novel Video Steganography Algorithm Based on Secret Sharing and Error-Correcting Code for H.264/AVC显示文摘In this paper, we analyze the video steganography technique, which is used to ensure national security and the confidentiality of the information of governmental agencies and enterprises. Videos may be used to transmit secrets and conduct covert communication. As such, we present an algorithm based on a secret sharing scheme and an Error-Correcting Code(ECC), which combines Grey Relational Analysis(GRA) with a partition mode in video compression standard H.264/AVC. First, we process secret information by secret sharing, and then use an ECC to process the obtained information. Moreover, we choose the Discrete Cosine Transform(DCT) blocks using GRA, and then use rules to hide the pretreated information in DCT coefficients of the video frames. Experimental results indicate that our algorithm has good invisibility, better robustness, good anti-steganalysis ability, and little influence on the bit rate of the video carrier. 8Probing the fractional topological charge of a vortex light beam by using dynamic angular double slits显示文摘Vortex beams with fractional topological charge(FTC) have many special characteristics and novel applications.However, one of the obstacles for their application is the difficulty of precisely determining the FTC of fractional vortex beams. We find that when a vortex beam with an FTC illuminates a dynamic angular double slit(ADS), the far-field interference patterns that include the information of the FTC of the beam at the angular bisector direction of the ADS vary periodically. 9OS1 functions in the allocation of nutrients between the endosperm and embryo in maize seeds显示文摘Uncovering the genetic basis of seed develop-ment will provide useful tools for improving both crop yield and nutritional value, However, the genetic regulatory networks of maize (Zea mays) seed development remain largely unknown. The maize opaque endosperm and small germ 1 (osl) mutant has opaque endosperm and a small embryo,Here, we cloned OSi1 and show that it encodes a putative transcription factor containing an RWP-RK domain-Transcriptional analysis indicated that OSi1 expression is elevated in early endosperm development, especially in the basal endosperm transfer layer (BETL), conducting zone (CZ), and central starch endospcrm (CSE) cells. RNA sequencing (RNA-Seq) analysis of the osi mutant revealed sharp downregulation of certain genes in specific cell types, including ZmMRP-1 and Megl in BETL cells and a majority of zein-and starch-related genes in CSE cells. Using a haploid induction system, we show that wild-type endosperm could rescue the smaller size of osi embryo, which suggests that nutrients are allocated by tho wild type endosperm. Therefore, our dsta imply that the network regulated by 051 accomplishes a key step in nutrient allocation between endosperm and embryo within maize seeds. 11Model test and discrete element method simulation of shield tunneling face stability in transparent clay显示文摘The stability of the shield tunneling face is an extremely important factor affecting the safety of tunnel construction.In this study,a transparent clay with properties similar to those of Tianjin clay is prepared and a new transparent clay model test apparatus is developed to overcome the“black box”problem in the traditional model test.The stability of the shield tunneling face(failure mode,influence range,support force,and surface settlement)is investigated in transparent clay under active failure.A series of transparent clay model tests is performed to investigate the active failure mode,influence range,and support force of the shield tunneling face under different burial depth conditions,whereas particle flow code three-dimensional numerical simulations are conducted to verify the failure mode of the shield tunneling face and surface settlement along the transverse section under different burial depth conditions.The results show that the engineering characteristics of transparent clay are similar to those of soft clay in Binhai,Tianjin and satisfy visibility requirements.Two types of failure modes are obtained:the overall failure mode(cover/diameter:C/D£1.0)and local failure mode(C/D≥2.0).The influence range of the transverse section is wider than that of the longitudinal section when C/D≥2.0.Additionally,the normalized thresholds of the relative displacement and support force ratio are 3%-6%and 0.2-0.4,respectively.Owing to the cushioning effect of the clay layer,the surface settlement is significantly reduced as the tunnel burial depth increases.Huayang LEI Yajie ZHANG Yao HU Yingnan LIU 2021Frontiers of Structural and Civil Engineering2021,15,1:2
12Rapid response of brain metastasis to crizotinib in a patient with KLC1-ALK fusion and MET gene amplification positive non-small cell lung cancer:a case report显示文摘Non-small cell lung cancer(NSCLC) ranks as the leading cause of cancer-related death in the world. Brain metastasis(BM) is a common complication of NSCLC, with 25%–40% of patients developing BM during the course of the disease. A significant strategy of local disease control in the central nervous system is radiation therapy. With the development of precision medicine,the concept of treating lung cancer BM has gradually changed. In this case, we performed a surgical procedure to obtain enough tumor tissue for the detection of the target gene and other related experiments after the patient was informed. Finally, we found that the patient had both hepatocyte growth factor receptor(MET) gene amplification and kinesin light chain 1-anaplastic lymphoma kinase fusion(KLC1-ALK) through next-generation sequencing and showed sensitivity to the targeted therapy of crizotinib. The patient exhibited good response. 14Self-cleaning semiconductor heterojunction substrate: ultrasensitive detection and photocatalytic degradation of organic pollutants for environmental remediation显示文摘Emerging technologies in the field of environmental remediation are becoming increasingly significant owing to the increasing demand for eliminating significant amounts of pollution in water,soil,and air.We designed and synthesized MoS 2/Fe 2 O 3 heterojunction nanocomposites(NCs)as multifunctional materials that are easily separated and reused.The trace detection performance of the prepared sample was examined using bisphenol A(BPA)as the probe molecule,with limits of detection as low as 10−9 M;this detection limit is the lowest among all reported semiconductor substrates.BPA was subjected to rapid photocatalytic degradation by MoS_(2)/Fe_(2)O_(3) NCs under ultraviolet irradiation.The highly recyclable MoS_(2)/Fe_(2)O_(3) NCs exhibited photo-Fenton catalytic activity for BPA and good detection ability when reused as a surface-enhanced Raman scattering(SERS)substrate after catalysis.The SERS and photocatalysis mechanisms were proposed while considering the effects of the Z-scheme charge-transfer paths,three-dimensional flower-like structures,and dipole–dipole coupling.Moreover,the prepared MoS_(2)/Fe_(2)O_(3) NCs were successfully applied in the detection of BPA in real lake water and milk samples.Herein,we present insights into the development of MoS_(2)/Fe_(2)O_(3) materials,which can be used as multifunctional materials in chemical sensors and in photocatalytic wastewater treatments for the removal of recalcitrant organic pollutants.Mingyue Hu Yingnan Quan Shuo Yang Rui Su Huilian Liu Ming Gao Lei Chen Jinghai Yang 2020Microsystems & Nanoengineering2020,6,1:1

17Natural infectious behavior of the urediniospores of Melampsora larici-populina on poplar leaves显示文摘The uredinial stage in the life cycle of Melampsora larici-populina on poplar leaves is the most important pathogenic phase. We captured partial phases of uredinial infection in the wild, aiming to reconstruct the process of uredinial ontogeny by using scanning and transmission electron microscope. At the initial infection stage, germ tubes germinated from the echinulate urediniospores. Germ tubes were frequently seen to merge with the leaf surface and cuticle breakage was observed, indicating direct hyphal penetration. Stomata penetration occurred commonly, sometimes with more than one germ tube penetrating the same stoma. Melampsora larici-populina did not form appressoria in the infection process,implying that infectious behavior of this pathogen may differ from the other rust pathogens. In general, germ tubes branched randomly, and no distinct evidence indicated that stoma could induce or orient germ tube branches. However, oriented germ tube growth has been occasionally observed in other studies. The urediniospores collapsed and finally wizened when they became nutrient stressed.
