| 1 | Engineering Broad-Spectrum Bacterial Blight Resistance by Simultaneously Disrupting Variable TALE-Binding Elements of Multiple Susceptibility Genes in Rice显示文摘Xanthomonas oryzae pv.oryzae(Xoo),the causal agent of bacterial blight of rice,employs the transcription activator-like effectors(TALEs)to induce the expression of the OsSWEET family of putative sugar transporter genes,which function in conferring disease susceptibility(S)in rice plants.To engineer broadspectrum bacterial blight resistance,we used CRISPR/Cas9-mediated gene editing to disrupt the TALEbinding elements(EBEs)of two S genes,OsSWEETH and OsSWEET14,in rice cv.Kitaake,which harbors the recessive resistance allele of Xa25/OsSWEET13.The engineered rice line MS14K exhibited broadspectrum resistance to most Xoo strains with a few exceptions,suggesting that the compatible strains may contain new TALEs.We identified two PthXo2-like TALEs,Tal5LN18 and Tal7PX061,as major virulence factors in the compatible Xoo strains LN18 and PX061,respectively,and found that Xoo encodes at least five types of PthXo2-like effectors.Given that PthXo2/PthXo2.1 target OsSlVEETf3 for transcriptional activation,the genomes of 3000 rice varieties were analyzed for EBE variationsin the OsSWEET13 promoter,and 10Xa25-like haplotypes were identified.We found that Tal5LN18 and Tal7PX〇6i bind slightly different EBE sequences in the OsSWEET13 promoter to activate its expression.CRISPR/Cas9 technology was then used to generate InDels in the EBE of the OsSWEET13 promoter in MS14K to creat a new germplasm with three edited OsSWEET EBEs and broad-spectrum resistance against all Xoo strains tested.Collectively,our findings illustrate how to disarm TALE-S co-evolved loci to generate broad-spectrum resistance through the loss of effector-triggered susceptibility in plants. | Zhengyin Xu Xiameng Xu Qiang Gong Ziyang Li Ying Li Sai Wang Yangyang Yang Wenxiu Ma Longyu Liu Bo Zhu Lifang Zou Gongyou Chen | 2019 | Molecular Plant2019,12,11: | 29 |

| 3 | Microstructure and Mechanical Properties of 45 vol.%SiC_p/7075Al Composite显示文摘Microstructure and mechanical behavior of high volume content Si Cp/7xxx Al composites have not been explored yet. Therefore, in the present work, 45 vol.% SiC p/7075 Al composite has been prepared by pressure infiltration method. High density dislocations were found around Si C/Al interface in SiC p/7075 Al composite after water-quenching and aging treatment. Fine dispersed nano-η′ phases were observed after the aging treatment. Adverse to other Si Cp/Al composites prepared by the pressure infiltration method,an interface layer was observed between SiC particles and Al matrix. Furthermore, high-resolution transmission electron microscopy(HRTEM) observation indicated that this interface layer was coherent/semicoherent with that of the SiC particles. 45 vol.% SiC p/7075 Al composite demonstrated high tensile strength(630 MPa) and micro-ductility. | 5 | A genome-wide analysis of SWEET gene family in cotton and their expressions under different stresses显示文摘Background: The SWEET(Sugars will eventually be exported transporters) gene family plays multiple roles in plant physiological activities and development process. It participates in reproductive development and in the process of sugar transport and absorption, plant senescence and stress responses and plant-pathogen interaction. However,thecomprehensive analysis of SWEET genes has not been reported in cotton.Results: In this study, we identified 22, 31, 55 and 60 SWEET genes from the sequenced genomes of Gossypium arboreum, G. raimondii, G. hirsutum and G. barbadense, respectively. Phylogenetic tree analysis showed that the SWEET genes could be divided into four groups, which were further classified into 14 sub-clades. Further analysis of chromosomal location, synteny analysis and gene duplication suggested that the orthologs showed a good collinearity and segmental duplication events played a crucial role in the expansion of the family in cotton. Specific MtN3_slv domains were highly conserved between Arobidopsis and cotton by exon-intron organization and motif analysis. In addition, the expression pattern in different tissues indicated that the duplicated genes in cotton might have acquired new functions as a result of sub-functionalization or neo-funtionalization. The expression pattern of SWEET genes showed that the different genes were induced by diverse stresses. The identification and functional analysis of SWEET genes in cotton may provide more candidate genes for genetic modification.Conclusion: SWEET genes were classified into four clades in cotton. The expression patterns suggested that the duplicated genes might have experienced a functional divergence. | 8 | Tea eggs-inspired high-strength natural polymer hydrogels显示文摘Natural polymer(NP)hydrogels are an irreplaceable class of biomaterials owing to their identified biosafety;however,the intrinsic poor mechanical strengths severely limit their applications as structural tissue engineering scaffolds.Inspired by the stiffening albumen gel of tea eggs,a traditional Chinese snack,high-strength NP hydrogels are constructed by simply soaking in aqueous solution of tea polyphenols(TP),an active ingredient extracted from tea.The TP-treated representative NP hydrogels exhibit considerably enhanced multifaceted mechanical properties with maximum 19-/30-,8.4-,6.1-,72-fold increases in tensile/compressive strengths,Young’s modulus,elongation at break and facture toughness,respectively,compared with pristine hydrogels,primarily due to the hydrogen bonding interactions between TP and NP chains.The TP-treated NP hydrogels can resist different large deformations,which cannot be achieved by their original species at all.In aqueous solution,the TP-treated NP hydrogels can still maintain robust mechanical performances,in spite of somewhat decline in strengths with release of TP,which just favorably affords increased water contents,antibacterial and antioxidant activities.GelMA-TP hydrogel is shown to facilitate wound healing in a full-thickness skin defect model.Importantly,the weak 3D printed GelMA scaffolds are significantly strengthened by TP treatment,broadening the possibility for customizing individualized bioscaffolds. | Tengling Wu Chunyan Cui Chuanchuan Fan Ziyang Xu Yang Liu Wenguang Liu | 2021 | Bioactive Materials2021,6,9: | 3 |

The in situ reaction procedure of the as-cast material was investigated by differential scanning calorimetry (DSC), and phases in the products were detected by X-ray diffraction (XRD). Experimental results showed that TiAl3 was formed first. With an increase in temperature, TiC and Al4C3 were observed, but TiAl3 decreased. In the final product, Al2O3 and TiO2 were observed. | 14 | A robust visual SLAM system in dynamic man-made environments显示文摘This paper presents a robust visual simultaneous localization and mapping(SLAM) system that leverages point and structural line features in dynamic man-made environments. Manhanttan world assumption is considered and the structural line features in such man-made environments provide rich geometric constraint, e.g., parallelism. Such a geometric constraint can be therefore used to rectify 3 D maplines after initialization. To cope with dynamic scenarios, the proposed system are divided into four main threads including 2 D dynamic object tracking, visual odometry, local mapping and loop closing. The 2 D tracker is responsible to track the object and capture the moving object in bounding boxes. In such a case, the dynamic background can be excluded and the outlier point and line features can be effectively removed. To parameterize 3 D lines, we use Pl ¨ucker line coordinates in initialization and projection processes, and utilize the orthonormal representation in unconstrained graph optimization process. | 15 | Insight into the adsorption of tetracycline onto amino and amino–Fe^(3+) gunctionalized mesoporous silica: Effect of functionalized groups显示文摘In order to study the influences of functionalized groups onto the adsorption of tetracycline,we prepared a series of amino and amino–Fe^(3+)complex mesoporous silica adsorbents with diverse content of amino and Fe^(3+)groups(named N,N-SBA15 and Fe-N,N-SBA15).The resulting mesoporous silica adsorbents were fully characterized by X-ray powder diffraction(XRD),Fourier transform infrared spectrometer(FTIR)and N_2adsorption/desorption isotherms.Furthermore,the effects of functionalized groups on the removal of TC were investigated.The results showed that the periodic ordered structure of SBA-15 was maintained after modification of amino/Fe^(3+)groups.The functionalized amino groups decreased the adsorption capacity while the coordinated Fe^(3+)increased the adsorption capacity.The adsorption kinetics of TC fitted pseudo-second-order model well and the equilibrium was achieved quickly.The adsorption isotherms fitted the Langmuir model well and with the Fe^(3+)content increased from 3.93%to 8.26%,the Q_(max)of the adsorbents increased from 102 to 188 mmol/kg.The solution p H affected the adsorption of TC onto amino complex adsorbents slightly while influenced the adsorption onto Fe-amine complex adsorbents greatly.The adsorption of TC on SBA15 and N,N-SBA15 may be related to the formation of outer-sphere surface complexes,while the adsorption of TC onto Fe-N,N-SBA15 was mainly attributed to the inner-sphere surface complexes.This study could offer potential materials that have excellent adsorption behavior for environmental remediation and suggested useful information for the preparing other adsorbents in environmental applications. | Ziyang Zhang Haiyan Li Huijuan Liu | 2018 | Journal of Environmental Sciences2018,30,3: | 2 |

| 19 | Zn/Sr dual ions-collagen co-assembly hydroxyapatite enhances bone regeneration through procedural osteo-immunomodulation and osteogenesis显示文摘The immune microenvironment induced by biomaterials played vital roles in bone regeneration.Hydroxyapatite(HA)and its ion-substituted derivates represent a large class of core inorganic materials for bone tissue engineering.Although ion substitution was proved to be a potent way to grant HA more biological functions,few studies focused on the immunomodulatory properties of ion-doped HA.Herein,to explore the potential osteoimmunomodulatory effects of ion-doped HA,zinc and strontium co-assembled into HA through a collagen template biomimetic way(ZnSr-Col-HA)was successfully achieved.It was found that ZnSr-Col-HA could induce a favorable osteo-immune microenvironment by stimulating macrophages.Furthermore,ZnSr-Col-HA demonstrated a procedural promoting effect on osteogenic differentiation of bone marrow mesenchymal stem cells(BMSCs)in vitro.Specifically,the osteo-immune microenvironment acted as a dominant factor in promoting osteogenic gene expressions at the early stage through OSM signal pathway.Whereas the direct stimulating effects on BMSCs by Zn^(2+)/Sr^(2+) were more effectively at the later stage with Nfatc1/Maf and Wnt signals activated.In vivo study confirmed strong promoting effects of ZnSr-Col-HA on critical-sized cranial defect repair.The current study indicated that such a combined biomaterial design philosophy of dual ion-doping and biomimetic molecular co-assembly to endow HA applicable osteoimmunomodulatory characteristics might bring up a new cutting-edge concept for bone regeneration study. | Zhenyu Zhong Xiaodan Wu Yifan Wang Mengdie Li Yan Li XuLong Liu Xin Zhang Ziyang Lan Jianglin Wang Yingying Du Shengmin Zhang | 2022 | Bioactive Materials2022,7,4: | 2 |
| 20 | Realization of high-performance tri-layer graphene saturable absorber mirror fabricated via a one-step transfer process显示文摘Graphene,as a saturable absorber(SA),has attracted much attention for its application in ultrashort pulse fiber lasers due to its ultrafast interband carrier relaxation and ultra-broadband wavelength operation.Nevertheless,during the stacking process of monolayer graphene layer,the induced nonuniform contact at the interface of graphene layers deteriorate the device performance.Herein,we report the fabrication of graphene saturable absorber mirrors(SAMs)via a one-step transfer process and the realization of the much enlarged modulation depth and the much reduced nonsaturable loss with tri-layer graphene(TLG)than single-layer graphene(SLG)due to the improved uniform contact at the interface.Moreover,the operation of 1550 nm mode-locked Er-doped fiber laser with the TLG SAM exhibits excellent output characteristics of the maximum output power of 9.9 mW,a slope efficiency of 2.4%and a pulse width of 714 fs.Our findings are expected to pave the way toward high-performance ultrashort pulse fiber lasers based on graphene SAs. | Cheng Jiang Xu Wang Jian Liu Jiqiang Ning Changcheng Zheng Xiaohui Li Ziyang Zhang | 2020 | Journal of Semiconductors2020,41,1: | 2 |
