| 1 | BLT1 in dendritic cells promotes Th1/Th17 differentiation and its deficiency ameliorates TNBS-induced colitis显示文摘Leukotriene B4(LTB4)synthesis is enhanced in the colonic mucosa in patients with inflammatory bowel disease(IBD).BLT1,a highaffinity receptor for LTB4,exhibits no effect on the progression of dextran sodium sulfate(DSS)-induced colitis,which mostly relies on innate immunity.Here,we reported that BLT1 regulates trinitrobenzene sulfonic acid(TNBS)-induced colitis,which reflects CD4+T-cell-dependent adaptive immune mechanisms of IBD.We found that BLT1 signaling enhanced the progression of colitis through controlling the production of proinflammatory cytokines by dendritic cells(DCs)and modulating the differentiation of Th1 and Th17.BLT1−/−mice displayed an alleviated severity of TNBS-induced colitis with reduced body weight loss and infiltrating cells in the lamina propria.BLT1 deficiency in DCs led to reduced production of proinflammatory cytokines,including IL-6,TNF-α,and IL-12,and these results were further confirmed via treatment with a BLT1 antagonist.The impaired cytokine production by BLT1−/−DCs subsequently led to reduced Th1 and Th17 differentiation both in vitro and in vivo.We further performed a conditional DC reconstitution experiment to assess whether BLT1 in DCs plays a major role in regulating the pathogenesis of TNBS-induced colitis,and the results indicate that BLT1 deficiency in DCs also significantly reduces disease severity.The mechanistic study demonstrated that BLT1-regulated proinflammatory cytokine production through the Gαiβγsubunit-phospholipase Cβ(PLCβ)-PKC pathway.Notably,we found that treatment with the BLT1 antagonist also reduced the production of proinflammatory cytokines by human peripheral blood DCs.Our findings reveal the critical role of BLT1 in regulating adaptive immunity and TNBS-induced colitis,which further supports BLT1 as a potential drug target for adaptive immunity-mediated IBD. | Jinfeng Zhou Weiming Lai Wanjie Yang Juping Pan Hu Shen Yingying Cai Cuixia Yang Ningjia Ma Yue Zhang Ru Zhang Xin Xie Zhongjun Dong Yuan Gao Changsheng Du | 2018 | Cellular & Molecular Immunology2018,15,12: | 9 |
| 2 | Injectable bone cement with magnesium-containing microspheres enhances osteogenesis via anti-inflammatory immunoregulation显示文摘Injectable bone cement is especially useful in minimally invasive surgeries to repair small and irregular bone defects.Amongst different kinds of injectable bone cements,bioactive calcium phosphate bone cement(CPC)has been widely studied due to its biological activity.However,its dense structure and poor biodegradability prevent the ingrowth of living tissue,which leads to undesirable bone regeneration and clinical translation.To address this issue,we prepared bone cement based on Magnesium-containing microspheres(MMSs)that can not only be cured into a 3D porous scaffold but also have controllable biodegradability that continuously provides space for desired tissue ingrowth.Interestingly,magnesium ions released from MMSs cement(MMSC)trigger positive immunomodulation via upregulation of the anti-inflammatory genes IL-10 and M2 macrophage polarization with increased expression of CD206,which is beneficial to osteogenesis.Moreover,the physicochemical properties of MMSC,including heat release,rheology and setting time,can be tuned to meet the requirements of injectable bone cement for clinical application.Using a rat model,we have demonstrated that MMSC promoted osteogenesis via mediation of tissue ingrowth and anti-inflammatory immunomodulation.The study provides a paradigm for the design and preparation of injectable bone cements with 3D porous structures,biodegradability and anti-inflammatory immunoregulation to efficiently promote osteogenesis. | Shenglong Tan Yifan Wang Yingying Du Yin Xiao Shengmin Zhang | 2021 | Bioactive Materials2021,6,10: | 9 |

| 5 | Preparation of silver tin oxide powders by hydrothermal reduction and crystallization显示文摘Silver tin oxide composite powders were synthesized by the hydrothermal method with a silver ammine solution and a Na2SnO3 solution as raw materials. H2C2O4 was used as the co-precipitator of silver ions and tin ions. The co-precipitation conditions were investigated. The results show that the co-precipitate of Ag2C2O4 and Sn(OH)4 is available when the pH value of the solution is 4.27-8.36. Using the obtained precipitate as precursor,the reduction of Ag+ and the crystallization of tin oxide were carried out simultaneously by the hydrothermal method and silver tin oxide composite powders were obtained. The composite powders were characterized by X-ray diffraction (XRD) analysis,scanning electron microscope (SEM),and energy spectrum analysis. | 8 | Targeting slug-mediated non-canonical activation of c-Met to overcome chemo-resistance in metastatic ovarian cancer cells显示文摘Metastasis-associated drug resistance accounts for high mortality in ovarian cancer and remains to be a major barrier for effective treatment. In this study, SKOV3/T4, a metastatic subpopulation of ovarian cancer SKOV3 cells, was enriched to explore potential interventions against metastaticassociated drug resistance. Quantitative genomic and functional analyses were performed and found that slug was significantly increased in the SKOV3/T4 subpopulation and contributed to the high resistance of SKOV3/T4. Further studies showed that slug activated c-Met in a ligand-independent manner due to elevated levels of fibronectin and provoked integrin α V function, which was confirmed by the significant correlation of slug and p-Met levels in 121 ovarian cancer patient samples. Intriguingly,c-Met inhibitor(s) exhibited greatly enhanced anti-cancer effects in slug-positive ovarian cancer models both in vitro and in vivo. Additionally, IHC analyses revealed that slug levels were highly correlated with reduced survival of ovarian cancer patients. | 18 | Tamoxifen attenuates reactive astrocyte-induced brain metastasis and drug resistance through the IL-6/STAT3 signaling pathway显示文摘Brain metastasis affects approxim ately 20%-30%of patients w ith triple-negative breast cancers(TNBCs).Even sm all metastatic lesions in the brain can trigg er severe neurological im pairm ents and result in extrem ely short survival time.Recently,active astrocytes were reported to be associated w ith brain metastases.However,how activated astrocytes regulate the behaviors of disseminated breast cancer cells in the brain remains unknown.In this study,human prim ary astrocytes were stim ulated w ith IL-1β to form active astrocytes to study the cross-talk between strom al cells(astrocytes)and TNBC cells in brain metastases.Our results showed that active astrocytes significantly increase the m alignancy of TNBC cells and prevent them from undergoing apoptosis caused by doxorubicin.We also found that the high level of IL-6 secreted by activated astrocytes was responsible for the drug resistance of breast cancer,which could be abolished by treatm ent of astrocytes with tam oxifen(TAM).The blockage of active astrocyte-derived IL-6 by a neutralizing antibody resulted in the attenuation of drug resistance,consequently enhancing the sensitivity of breast cancer cells to doxorubicin.Furthermore,the possible involved TAM-m odulated drug resistance mechanism may be associated with a decrease in IL-6 expression in astrocytes and the downregulation of MAPK and JAK2/STAT3 signaling in cancer cells.Our data suggested that TAMs might reduce drug resistance through the IL-6/JAK2/STAT3 signaling pathway,providing a possible therapy to treat brain metastasis in TNBCs,as estrogen receptor inhibitors(TAMs,etc.)can cross the blood-brain barrier. | Yongming Xu Yanrong Zhu Yong Yue Shaofeng Pu Junzhen Wu Yingying Lv Dongping Du | 2020 | Acta Biochimica et Biophysica Sinica2020,52,12: | 1 |
