2Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 4A bioactive injectable self-healing anti-inflammatory hydrogel with ultralong extracellular vesicles release synergistically enhances motor functional recovery of spinal cord injury显示文摘The repair and motor functional recovery after spinal cord injury(SCI)remains a worldwide challenge.The inflammatory microenvironment is one of main obstacles on inhibiting the recovery of SCI.Using mesenchymal stem cells(MSCs)derived extracellular vesicles to replace MSCs transplantation and mimic cell paracrine secretions provides a potential strategy for microenvironment regulation.However,the effective preservation and controlled release of extracellular vesicles in the injured spinal cord tissue are still not satisfied.Herein,we fabricated an injectable adhesive anti-inflammatory F127-polycitrate-polyethyleneimine hydrogel(FE)with sustainable and long term extracellular vesicle release(FE@EVs)for improving motor functional recovery after SCI.The orthotopic injection of FE@EVs hydrogel could encapsulate extracellular vesicles on the injured spinal cord,thereby synergistically induce efficient integrated regulation through suppressing fibrotic scar formation,reducing inflammatory reaction,promoting remyelination and axonal regeneration.This study showed that combining extracellular vesicles into bioactive multifunctional hydrogel should have great potential in achieving satisfactory locomotor recovery of central nervous system diseases.Chenggui Wang Min Wang Kaishun Xia Jingkai Wang Feng Cheng Kesi Shi Liwei Ying Chao Yu Haibin Xu Shining Xiao Chengzhen Liang Fangcai Li Bo Lei Qixin Chen 2021Bioactive Materials2021,6,8:9

7Optimization of temporal gate by two-color chirped lasers for the generation of isolated attosecond pulse in soft X rays显示文摘We propose a simple and efficient method to optimize two-color chirped laser pulses by forming a'temporal gate'for the generation of isolated attosecond pulses(IAPs)in soft X rays.We show that the generation process for higher and cutoff harmonics can be effectively limited within the temporal gate,and the harmonic emission interval can be further reduced with the help of phase-matching by only selecting the contribution from shorttrajectory electrons.This two-color gating mechanism is verified by increasing the pulse duration,raising the gas pressure,and extending the target cutoff.Compared to the five-color waveform in Phys.Rev.Lett.102,063003(2009),our waveform can be used to generate the IAP in the long-duration laser pulse while the cutoff energy is higher without the reduction of harmonic yields.Our work provides an alternative temporal gating scheme for the generation of IAPs by simultaneously improving the harmonic conversion efficiency,thus making the attosecond soft X rays an intense and highly time-resolved tabletop light source for future applications.LIWEI HE GUANGLU YUAN KAN WANG WEIJIE HUA CHAO YU CHENG JIN 2019Photonics Research2019,7,12:3
8Release of endogenous hydrogen sulfide in enteric nerve cells suppresses intestinal motility during severe acute pancreatitis显示文摘Previous studies have shown that during severe acute pancreatitis(SAP)attacks,hydrogen sulfide(H2S)is released in the colon.However,the roles played byH2S in regulating enteric nerves remain unclear.In this study,we examined the association between SAP-induced H2S release and loss of intestinal motility,and also explored the relevant mechanism in enteric nerve cells.A rat SAP model was constructed and enteric nerve cells were prepared.Intestinal mobility was evaluated by measuring the number of bowel movements at indicated time points and by performing intestinal propulsion tests.The production of inflammatory cytokines during a SAP attack was quantified by ELISA,and the levels of cystathionine-y-lyase(CSE)and cystathionine-p-synthase(CBS)were examined by immunohistochemistry and western blot analysis.In vivo studies showed that PI3K/Akt/Sp1 signaling in enteric nerve cells was blocked,confirming the mechanism of endogenous H2S formation by western blot analysis and immunofluorescence.Our results also showed that rats with SAP symptoms had reduced intestinal motility.Furthermore,PI3K/Akt/Sp1 signaling was triggered and CSE expression was up-regulated,and these changes were associated with H2S formation in the colon.In addition,propargylglycine reduced the levels of inflammatory cytokines and suppressed the release of H2S.Enteric nerve cells that were incubated with LY294002 and transfected with a Sp7-knockdown vector displayed decreased levels of CSE production,which led to a decrease in H2S production.These results suggest that SAP symptoms suppressed the intestinal motility of rats via the release of H2S in enteric nerve cells,which was dependent on the inflammation-induced PI3K/Akt/Sp1 signaling pathway.Ribin Liao Liwei Xue Zhanrong Qiang Cheng Zhang Ying Liu 2020Acta Biochimica et Biophysica Sinica2020,52,1:2
9Diffuse Intrinsic Pontine Gliomas Exhibit Cell Biological and Molecular Signatures of Fetal Hindbrain-Derived Neural Progenitor Cells显示文摘Diffuse intrinsic pontine glioma(DIPG) is the main cause of brain tumor-related death among children.Until now, there is still a lack of effective therapy with prolonged overall survival for this disease. A typical strategy for preclinical cancer research is to find out the molecular differences between tumor tissue and para-tumor normal tissue, in order to identify potential therapeutic targets. Unfortunately, it is impossible to obtain normal tissue for DIPG because of the vital functions of the pons.Here we report the human fetal hindbrain-derived neural progenitor cells(pontine progenitor cells, PPCs) as normal control cells for DIPG. The PPCs not only harbored similar cell biological and molecular signatures as DIPG glioma stem cells, but also had the potential to be immortalized by the DIPG-specific mutation H3 K27 M in vitro. 10High efficient development of green kinetic hydrate inhibitors via combined molecular dynamic simulation and experimental test approach显示文摘The development of environmental friendly low dose hydrate inhibitors like kinetic hydrate inhibitors(KHIs)is of great significance for the flow assurance in oil&gas production and transportation.In this work,a combined molecular dynamic simulation and experimental verification approach was adopted to increase the efficiency of KHIs development.The inhibition effect of a series of copolymers(N-vinylpyrrolidone and N-acrylate)on hydrate growth was studied by using both molecular dynamics simulation and experimental approaches.The simulation results demonstrated that introduction of hydrophobic ester and butyl group in PVP is beneficial for the inhibition.The length of the alkyl chain of ester group played an important role in improving inhibition performance.PVP-A,the one being introduced butyl ester group into PVP gets the best inhibition effect.In addition,inhibitors can restrict methane bubbles to re-dissolve into the liquid phase,thereby inhibiting the growth of methane hydrate.Increasing the interaction between KHIs and methane can also improve the inhibitory effect of KHIs.The experimental results confirm the reliability of the molecular dynamics simulation.Liwei Cheng Jinlong Cui Jia Li Ran Zhu Bei Liu Shuai Ban Guangjin Chen 2022Green Chemical Engineering2022,3,1:2
11Single-cell transcriptome analysis reveals the regulatory effects of artesunate on splenic immune cells in polymicrobial sepsis显示文摘Sepsis is characterized by a severe and life-threatening host immune response to polymicrobial infection accompanied by organ dysfunction.Studies on the therapeutic effect and mechanism of immunomodulatory drugs on the sepsis-induced hyperinflammatory or immunosuppression states of various immune cells remain limited.This study aimed to investigate the protective effects and underlying mechanism of artesunate(ART)on the splenic microenvironment of cecal ligation and puncture-induced sepsis model mice using single-cell RNA sequencing(scRNA-seq)and experimental validations.The scRNA-seq analysis revealed that ART inhibited the activation of pro-inflammatory macrophages recruited during sepsis.ART could restore neutrophils’chemotaxis and immune function in the septic spleen.It inhibited the activation of T regulatory cells but promoted the cytotoxic function of natural killer cells during sepsis.ART also promoted the differentiation and activity of splenic B cells in mice with sepsis.These results indicated that ART could alleviate the inflammatory and/or immunosuppressive states of various immune cells involved in sepsis to balance the immune homeostasis within the host.Overall,this study provided a comprehensive investigation of the regulatory effect of ART on the splenic microenvironment in sepsis,thus contributing to the application of ART as adjunctive therapy for the clinical treatment of sepsis.Jiayun Chen Xueling He Yunmeng Bai Jing Liu Yin Kwan Wong Lulin Xie Qian Zhang Piao Luo Peng Gao Liwei Gu Qiuyan Guo Guangqing Cheng Chen Wang Jigang Wang 2023Journal of Pharmaceutical Analysis2023,13,7:1
12Development of a bifurcation analysis approach based on gPROMS platform显示文摘A bifurcation analysis approach is developed based on the process simulator gPROMS platform, which can automatically trace a solution path, detect and pass the bifurcation points and check the stability of solutions. The arclength continuation algorithm is incorporated as a process entity in gPROMS to overcome the limit of turning points and get multiple solutions with respect to a user-defined parameter. The bifurcation points are detected through a bifurcation test function τ which is written in C ++ routine as a foreign object connected with gPROMS through Foreign Process Interface. The stability analysis is realized by evaluating eigenvalues of the Jacobian matrix of each steady state solution. Two reference cases of an adiabatic CSTR and a homogenous azeotropic distillation from literature are studied, which successfully validate the reliability of the proposed approach. 19抗瘤中药制剂对结直肠癌T及NK细胞免疫抑制的体外逆转作用(英文)显示文摘Objective: Reversing effects of traditional Chinese medicines on colorectal tumor immunosuppressions of natural killer (NK) cell and T lymphocyte were analyzed to provide evidence on selecting medicines for patients according to the different types of tumor immunosuppression. Methods: Six traditional Chinese medicines, including Arsenious acid (AS), Ligustra-zine hydrochloride (LHC), Astragalus mongholicus bge (AMB), Matrine N-oxide (MOX), Polyporus umbellatus polysaccharide (PUPS) and Artesunate (ART), were enrolled. The reversing effects on suppression of murine splenocyte transformation and NK killing activity were measured by 3-{4,5-dimethyl-2-thiazolyl}-2,5-diphenyl tetrazolium (MTT), and the effects on the suppressed expression of interleukin 2 receptor α (IL-2Rα), CD3ε + ζ + and CD3ε - ζ + were detected by flow cytometry (FCM). The effects on immunosuppressive molecules were measured by enzyme linked immunosorbent assay (ELISA), including transforming growth factor β1 (TGF-β 1), vascular endothelial growth factor (VEGF), interleukin 4 (IL-4), IL-6, IL-10 and pros-taglandin (PG) E 2 . Results: (1) The reversing effects of AMB on the inhibition of NK killing and CD3ε + ζ + expression were the most significant; the effect of LHC on inhibition of CD3ε - ζ + expression was the strongest; the effects of AMB, PUPS and ART on inhibition of transformation were the greatest; and the effect of ART on inhibition of IL-2Rα expression was the strongest. (2) The correlated molecules of these medicines that exerted reversing effects on colorectal tumor immunosuppression were TGF-β 1 and IL-10. AMB had the highest down-regulating effect on the secretion of TGF-β 1 . AS and ART had the highest effects on IL-10. Conclusion: Reversing tumor immunosuppression through the down-regulation of immunosuppressive molecules is one of the novel antitumor mechanisms of traditional Chinese medicines.
