METTL4 is an snRNA m^6Am methyltransferase that regulates RNA splicing显示文摘Dear Editor,Protein coding mRNAs and non-coding RNAs are extensively modifed by chemical modifications,'including methylation of the N^6position of adenosine(m^6A).^2While mRNA internal m^6A methylation is mediated by METTL3 and METTL14,which are members of the MT-A70 family,N^6-methylation of cap-adjacent No,2'-O-dimethyladenosine(m^6Am)is catalyzed by PCIF1.^3-6 Non-coding RNAs are also m^6Am methylated internally,7but the corresponding enzyme remains unknown. Here we provide evidence that METTL4 mediates internal m^6Am methylation of U2 small nuclear RNA(snRNA). | Hao Chen Lei Gu Esteban A.Orellana Yuanyuan Wang Jiaojiao Guo Qi Liu Longfei Wang Zhangfei Shen Hao Wu Richard I.Gregory Yi Xing Yang Shi | 2020 | Cell Research2020,30,6: | 4
LIN28 coordinately promotes nucleolar/ribosomal functions and represses the 2C-like transcriptional program in pluripotent stem cells显示文摘LIN28 is an RNA binding protein with important roles in early embryo development, stem cell differentiation/re-programming,tumorigenesis and metabolism.Previous studies have focused mainly on its role in the cytosol where it interacts with Let-7 microRNA precursors or mRNAs,and few have addressed LIN28's role within the nucleus. Here,we show that LIN28 displays dynamic temporal and spatial expression during murine embryo development.Maternal LIN28 expression drops upon exit from the 2-cell stage, and zygotic LIN28 protein is induced at the forming nucleolus during 4-cell to blas-tocyst stage development,to become dominantly expressed in the cytosol after implantation. In cultured pluripotent stem cells(PSCs),loss of LIN28 led to nucleolar stress and activation of a 2-cell/4-cell-like transcriptional program characterized by the expression of endogenous retrovirus genes.Mechanistically,LIN28 binds to small nucleolar RNAs and rRNA to maintain nucleolar integrity, and its loss leads to nucleolar phase separation defects, ribosomal stress and activation of P53 which in turn binds to and activates 2C transcription factor Dux.LIN28 also resides in a complex containing the nucleolar factor Nucleolin(NCL)and the transcrip-tional repressor TRIM28,and LIN28 loss leads to reduced occupancy of the NCL/TRIM28 complex on the Dux and rDNA loci, and thus de-repressed Dux and reduced rRNA expression.Lin28 knockout cells with nucleolar stress are more likely to assume a slowly cycling, translationally inert and anabolically inactive state, which is a part of previously unappreciated 2C-like transcriptional program. These findings elucidate novel roles for nucleolar LIN28 in PSCs,and a new mechanism linking 2C program and nucleolar functions in PSCs and early embryo development. | Zhen Sun Hua Yu Jing Zhao Tianyu Tan Hongru Pan Yuqing Zhu Lang Chen Cheng Zhang Li Zhang Anhua Lei Yuyan Xu Xianju Bi Xin Huang Bo Gao Longfei Wang Cristina Correia Ming Chen Qiming Sun Yu Feng Li Shen Hao Wu Jianlong Wang Xiaohua Shen George Q.Daley Hu Li Jin Zhang | 2022 | Protein & Cell2022,13,7: | 4
Searching for the Nano-Hertz Stochastic Gravitational Wave Background with the Chinese Pulsar Timing Array Data ReleaseⅠ显示文摘Observing and timing a group of millisecond pulsars with high rotational stability enables the direct detection of gravitational waves(GWs).The GW signals can be identified from the spatial correlations encoded in the times-of-arrival of widely spaced pulsar-pairs.The Chinese Pulsar Timing Array(CPTA)is a collaboration aiming at the direct GW detection with observations carried out using Chinese radio telescopes. This short article serves as a“table of contents”for a forthcoming series of papers related to the CPTA Data Release 1(CPTA DR1)which uses observations from the Five-hundred-meter Aperture Spherical radio Telescope. Here, after summarizing the time span and accuracy of CPTA DR1,we report the key results of our statistical inference finding a correlated signal with amplitude logA_(c)=-14.4_(-2.8)^(+1.0)for spectral index in the range ofα∈[-1.8,1.5]assuming a GW background(GWB)induced quadrupolar correlation. The search for the Hellings–Downs(HD)correlation curve is also presented, where some evidence for the HD correlation has been found that a 4.6σstatistical significance is achieved using the discrete frequency method around the frequency of 14 n Hz.We expect that the future International Pulsar Timing Array data analysis and the next CPTA data release will be more sensitive to the n Hz GWB,which could verify the current results. | Heng Xu Siyuan Chen Yanjun Guo Jinchen Jiang Bojun Wang Jiangwei Xu Zihan Xue RNicolas Caballero Jianping Yuan Yonghua Xu Jingbo Wang Longfei Hao Jingtao Luo Kejia Lee Jinlin Han Peng Jiang Zhiqiang Shen Min Wang Na Wang Renxin Xu Xiangping Wu Richard Manchester Lei Qian Xin Guan Menglin Huang Chun Sun Yan Zhu | 2023 | Research in Astronomy and Astrophysics2023,23,7: | 3
Graph Computing Based Security Constrained Unit Commitment in Hydro-thermal Power Systems Incorporating Pumped Hydro Storage显示文摘This paper proposes a graph computing based mixed integer programming(MIP)framework for solving the security constrained unit commitment(SCUC)problem in hydro-thermal power systems incorporating pumped hydro storage(PHS).The proposed graph computing-based MIP framework considers the economic operations of thermal units, cascade hydropower stations and PHS stations,as well as their technical impacts towards the network security.First, the hydro-thermal power system data and unit information are stored in a graph structure with nodes and edges, which enables nodal and hierarchical parallel computing for the unit commitment(UC)solution calculation and network security analysis.A MIP model is then formulated to solve the SCUC problem with the mathematical models of thermal units, cascade hydropower stations and PHS stations. In addition, two optimization approaches including convex hull reformulation(CHR)and special ordered set(SOS)methods are introduced for speeding up the MIP calculation procedure. To ensure the system stability under the derived UC solution,a parallelized graph power flow(PGPF)algorithm is proposed for the hydro-thermal power system network security analysis. Finally, case studies of the IEEE 118-bus system and a practical 2749-bus hydro-thermal power system are introduced to demonstrate the feasibility and validity of the proposed graph computing-based MIP framework. | Longfei Wei Guangyi Liu Shen Yan Renchang Dai Yachen Tang | 2021 | CSEE Journal of Power and Energy Systems2021,7,3: | 2

Formation, preservation and connectivity control of organic pores in shale显示文摘In view of strong heterogeneity and complex formation and evolution of organic pores, field emission scanning electron microscopy(FESEM),Raman spectrum and fluid injection+CT/SEM imaging technology were used to study the macerals, organic pores and connectivity of organic pores in the lower Paleozoic organic-rich shale samples from Southern China.Combined with the mechanism of hydrocarbon generation and expulsion and pore forming mechanism of organic matter-based activated carbon, the relationships between organic pore development and the organic matter type, hydrocarbon generation process, diagenesis and pore pressure were explored to reveal the controlling factors of the formation, preservation and connectivity of organic pores in shale.(1)The generation of organic pores goes on through the whole hydrocarbon generation process, and is controlled by the type, maturity and decomposition of organic matter;the different hydrocarbon generation components and differential hydrocarbon-generation evolution of kerogen and solid asphalt lead to different pore development characteristics;organic pores mainly develop in solid bitumen and hydrogen-rich kerogen.(2)The preservation of organic pores is controlled by maturity and diagenesis, including the steric hindrance effect of in-situ hydrocarbon retention, rigid mineral framework formed by recrystallization, the coupling mechanism of pore-fluid pressure and shale brittleness-ductility transition.(3)The Ro of 4.0%is the maturity threshold of organic pore extinction, the shale layers with Ro larger than 3.5%have high risk for shale gas exploration, these shale layers have low gas contents,as they were in an open state before uplift, and had high hydrocarbon expulsion efficiency and strong aromatization, thus having the'congenital deficiency'of high maturity and pore densification.(4)The pores in the same organic matter particle have good connectivity;and the effective connectivity between different organic matter pores and inorganic pores and fractures depends on the abundance and distribution of organic matter, and development degree of pores and fractures in the shale;the accumulation, preservation and laminar distribution of different types of organic matter in high abundance is the prerequisite for the development and connection of organic pores, grain margin fractures and bedding fractures in reservoir. | BORJIGIN Tenger LU Longfei YU Lingjie ZHANG Wentao PAN Anyang SHEN Baojian WANG Ye YANG Yunfeng GAO Zhiwei | 2021 | Petroleum Exploration and Development2021,48,4: | 1
Amylin:new insight into pathogenesis, diagnosis, and prognosis of non-insulin-dependent diabetesmellitus-related cardiomyopathy显示文摘Co-secretion with insulin, highly amyloidogenic human amylin is considered to contribute to the initiation and progression of diabetic heart complications, despite other situations such as hypertension and atherosclerosis. In response to insulin resistance, hyperinsulinemia, and consequently hyperamylinemia,is common in prediabetic patients, where highly concentrated amylin is prone to form amylin oligomers, which further assemble into fibrils and amyloids with high b-sheet content. The infusion and deposition of oligomeric amylin in myocytes cause a series of consequences, including cytosolic Ca^(2+)dysregulation,calmodulin activation, myocyte hypertrophy, and ventricular stiffness, eventually leading to heart failure. In this review,we present the latest reports of amylin-related heart complications, provide new insights, and state the underlying pathogenesis, diagnosis, possible treatment, and prevention of diabetic cardiomyopathy. | Jiaying Xie Zhoujie Tong Longfei Shen Yuanyuan Shang Yulin Li Bin Lu Weixuan Ma Wei Zhang Ming Zhong | 2022 | Emergency and Critical Care Medicine2022,2,1: | 0
Cyasterone inhibits IL-1β-mediated apoptosis and inflammation via the NF-κB and MAPK signaling pathways in rat chondrocytes and ameliorates osteoarthritis in vivo显示文摘Osteoarthritis is a prevalent global joint disease, which is characterized by inflammatory reaction and cartilage degradation.Cyasterone,a sterone derived from the roots of Cyathula officinalis Kuan, exerts protective effect against several inflammationrelated diseases. However, its effect on osteoarthritis remains unclear. The current study was designed to investigate the potential antiosteoarthritis activity of cyasterone.Primary chondrocytes isolated from rats induced by interleukin(IL)-1βand a rat model stimulated by monosodium iodoacetate(MIA)were used for in vitro and in vivo experiments, respectively. The results of in vitro experiments showed that cyasterone apparently counteracted chondrocyte apoptosis, increased the expression of collagen II and aggrecan, and restrained the production of the inflammatory factors inducible nitric oxide synthase(iNOS),cyclooxygenase-2(COX-2),a disintegrin and metalloproteinase with thrombospondin motifs-5(ADAMTS-5),metalloproteinase-3(MMP-3),and metalloproteinase-13(MMP13)induced by IL-1βin chondrocytes.Furthermore, cyasterone ameliorated the inflammation and degenerative progression of osteoarthritis potentially by regulating the nuclear factor kappa B(NF-κB)and mitogen-activated protein kinase(MAPK)pathways. For in vivo experiments, cyasterone significantly alleviated the inflammatory response and cartilage destruction of rats induced by monosodium iodoacetate, where dexamethasone was used as the positive control.Overall, this study laid a theoretical foundation for developing cyasterone as an effective agent for the alleviation of osteoarthritis. | TENG Li SHEN Yue QU Yuhan YANG Longfei YANG Yuting JIAN Xi FAN Shengli ZHANG Lele FU Qiang | 2023 | Chinese Journal of Natural Medicines2023,21,2: | 0
Effect of Ga_(2)O_(3)-doping on Properties and Structure of ZBLAN Glass显示文摘It is interesting to explore a novel oxyfluoride glass with good glass stability to be applied in optical communication and optical windows at infrared(IR)wavelength. We demonstrated a new glass of Ga_(2)O_(3)-doped ZrF_(4)-BaF_(2)-LaF_(3)-AlF_(3)-NaF(ZBLAN)glass using a melt-quenched technique. The effect of Ga_(2)O_(3)-doping on glass properties and structure was characterized by differential thermal analysis(DTA),IR spectra,Raman spectra, and X-ray diffraction(XRD).It is found that the glass thermal stability(ΔT)increases by 14% when the addition of Ga_(2)O_(3) reaches 1mol%.With the increase of Ga_(2)O_(3) content, the density and refractive index of the glasses increase.Ga_(2)O_(3)-doping does not affect the IR cut-off edge and maintains the transmittance near 90% in the range of 2.5-5μm,which is almost equal to the undoped sample.Ga_(2)O_(3)-doping hardly changes the initial coordinated structure of Zr^(4+)according to the results of IR spectra and Raman spectra.Ga^(3+)holds in the interstice site of the network coordinated with F^(-)and the part of O_(2)-introduced by Ga_(2)O_(3) is coordinated with Al^(3+)forming Al-O bond. This study offers a new glass composition that may be potentially used in fabricating mid-IR optical fiber and large-size glasses for IR windows. | 崔素杰 李家成 ZHANG Long JIANG Yiguang WANG Zaiyang ZHANG Longfei YUAN Chengfeng SHEN Zongyun 曾惠丹 | 2022 | Journal of Wuhan University of Technology(Materials Science)2022,37,4: | 0
The TMRT K band observations towards 26 infrared dark clouds:NH_3, CCS, and HC_3N显示文摘We present one of the first Shanghai Tian Ma Radio Telescope (TMRT) K Band observations towards a sample of 26 infrared dark clouds (IRDCs).We observed the (1,1),(2,2),(3,3),and (4,4) transitions of NH_(3) together with CCS (2_(1)–>1_(0)) and HC_(3)NJ=2-1,simultaneously.The survey dramatically increases the existing CCS-detected IRDC sample from 8 to 23,enabling a better statistical study of the ratios of carbon-chain molecules (CCM) to N-bearing molecules in IRDCs.With the newly developed hyperfine group ratio (HFGR) method of fitting NH_(3) inversion lines,we found the gas temperature to be between 10 and18 K.The column density ratios of CCS to NH_(3) for most of the IRDCs are less than 10^(-2),distinguishing IRDCs from low-mass star-forming regions. We carried out chemical evolution simulations based on a three-phase chemical model NAUTILUS.Our measurements of the column density ratios between CCM and NH_(3) are consistent with chemical evolutionary ages of 10^(5) yr in the models.Comparisons of the data and chemical models suggest that CCS,HC_(3)N,and NH_(3) are sensitive to the chemical evolutionary stages of the sources. | Jinjin Xie Gary A.Fuller Di Li Longfei Chen Zhiyuan Ren Jingwen Wu Yan Duan Junzhi Wang Juan Li Nicolas Peretto Tie Liu Zhiqiang Shen | 2021 | Science China(Physics,Mechanics & Astronomy)2021,64,7: | 0
Touchable cell biophysics property recognition platforms enable multifunctional blood smart health care显示文摘As a crucial biophysical property, red blood cell(RBC)deformability is pathologically altered in numerous disease states, and biochemical and structural changes occur over time in stored samples of otherwise normal RBCs.However, there is still a gap in applying it further to point-of-care blood devices due to the large external equipment(high-resolution microscope and microfluidic pump),associated operational difficulties, and professional analysis.Herein,we revolutionarily propose a smart optofluidic system to provide a differential diagnosis for blood testing via precise cell biophysics property recognition both mechanically and morphologically.Deformation of the RBC population is caused by pressing the hydrogel via an integrated mechanical transfer device. The biophysical properties of the cell population are obtained by the designed smartphone algorithm.Artificial intelligence-based modeling of cell biophysics properties related to blood diseases and quality was developed for online testing. We currently achieve 100%diagnostic accuracy for five typical clinical blood diseases(90 megaloblastic anemia,78 myelofibrosis,84 iron deficiency anemia,48 thrombotic thrombocytopenic purpura, and 48 thalassemias)via real-world prospective implementation;furthermore, personalized blood quality(for transfusion in cardiac surgery)monitoring is achieved with an accuracy of 96.9%.This work suggests a potential basis for next-generation blood smart health care devices. | Longfei Chen Yantong Liu Hongshan Xu Linlu Ma Yifan Wang Le Yu Fang Wang Jiaomeng Zhu Xuejia Hu Kezhen Yi Yi Yang Hui Shen Fuling Zhou Xiaoqi Gao Yanxiang Cheng Long Bai Yongwei Duan Fubing Wang Yimin Zhu | 2021 | Microsystems & Nanoengineering2021,7,6: | 0
