1Relationship between intestinal microbiota and ulcerative colitis:Mechanisms and clinical application of probiotics and fecal microbiota transplantation显示文摘Ulcerative colitis(UC) is an inflammatory disease that mainly affects the colon and rectum. It is believed that genetic factors, host immune system disorders, intestinal microbiota dysbiosis, and environmental factors contribute to the pathogenesis of UC. however, studies on the role of intestinal microbiota in the pathogenesis of UC have been inconclusive. Studies have shown that probiotics improve intestinal mucosa barrier function and immune system function and promote secretion of anti-inflammatory factors, thereby inhibiting the growth of harmful bacteria in the intestine. Fecal microbiota transplantation(FMT) can reduce bowel permeability and thus the severity of disease by increasing the production of short-chain fatty acids, especially butyrate, which help maintain the integrity of the epithelial barrier. FMT can also restore immune dysbiosis by inhibiting Th1 differentiation, activity of T cells, leukocyte adhesion, and production of inflammatory factors. Probiotics and FMT are being increasingly used to treat UC, but their use is controversial because of uncertain efficacy. 2Anti-inflammatory and anti-thrombogenic effects of atorvastatin in acute ischemic stroke显示文摘Atorvastatin decreases inflammation and thrombogenesis in patients with carotid artery plaque. Atorvastatin is administered to lower lipid levels, but its anti-inflammatory and anti-thrombogenic effects remain unclear. Eighty-nine patients from northeastern China with acute ischemic stroke caused by large-artery atherosclerosis were randomly divided into the study and control groups. All patients received routine treatment, including antiplatelet therapy, circulatory support, and symp-tomatic treatment. The study group (n=43) also received daily atorvastatin 20 mg/d, and the control group (n=46) received daily placebo pills containing glucose. After 4 weeks, the levels of C-reactive protein, fibrinogen, and D-dimer were significantly lower in the study group than in the control group. Decreases in the levels of C-reactive protein, fibrinogen, and D-dimer were not associated with de-creases in the levels of triacylglycerol and low-density lipoprotein cholesterol. 3Discrimination of abiogenic and biogenic alkane gases显示文摘We have combined the analytical data of the carbon isotope distribution pattern, R/Ra and CH4/3He values of abiogenic and biogenic (referring to the thermogenic and bacterial or microbial) alkane gases in China with those of alkane gases from USA, Russia, Germany, Australia and other countries. 4The HuangZaoSi Maize Genome Provides Insights into Genomic Variation and Improvement History of Maize显示文摘Maize is a globally important crop that was a classic model plant for genetic studies. Here, we report a 2.2 Gb draft genome sequence of an elite maize line, HuangZaoSi (HZS). Hybrids bred from HZS-improved lines (HILs) are planted in more than 60% of maize fields in China. Proteome clustering of six completed sequeneed maize genomes show that 638 proteins fall into 264 HZS-specific gene families with the majority of contributions from tandem duplication events. Resequencing and comparative analysis of 40 HZSrelated lines reveals the breeding history of HILs. More than 60% of identified selective sweeps were clustered in identity.by.descent conserved regions, and yield-related genes/QTLs were enriched in HZS characteristic selected regions. Furthermore, we dem on strated that HZS-specific family genes were not uniformly distributed in the genome but enriched in improvement/function.related genomic regions. 5Complement C3 activation regulates the production of tRNA-derived fragments Gly-tRFs and promotes alcohol-induced liver injury and steatosis显示文摘Complement is known to play a role in alcoholic fatty liver disease (AFLD), but the underlying mechanisms are poorly understood, thereby constraining the development of a rational approach for therapeutic intervention in the complement system. C3 deficiency has been shown to impart protective effects against ethanol-induced hepatic steatosis and inflammation. Here we demonstrate a protection effect in wild-type mice by treatment with CR2-Crry, a specific inhibitor of C3 activation. The expression of glycine transfer (t) RNA-derived fragments (Gly-tRFs) is upregulated in ethanol-fed mice and inhibition of Gly-tRFs in vivo decreases chronic ethanol feeding-induced hepatosteatosis without affecting inflammation. The expression of Gly-tRF was downregulated in C3-deficient or CR2-Crry-treated mice, but not in C5-deficient mice;Gly-tRF expression was restored by the C3 activation products C3a or Asp (C3a-des-Arg) via the regulation of CYP2E1. Transcriptome profiling of hepatic tissues showed that Gly-tRF inhibitors upregulate the expression of sirtuin1 (Sirt1) and subsequently affect downstream lipogenesis and β-oxidation pathways. Mechanistically, Gly-tRF interacts with AGO3 to downregulate Sirt1 expression via sequence complementarity in the 3′ UTR. Notably, the expression levels of C3d, CYP2E1 and Gly-tRF are upregulated, whereas Sirt1 is decreased in AFLD patients compared to healthy controls. Collectively, our findings suggest that C3 activation products contribute to hepatosteatosis by regulating the expression of Gly-tRF. 9Deep learning applied to two-dimensional color Doppler flow imaging ultrasound images significantly improves diagnostic performance in the classification of breast masses:a multicenter study显示文摘Background:The current deep learning diagnosis of breast masses is mainly reflected by the diagnosis of benign and malignant lesions.In China,breast masses are divided into four categories according to the treatment method:inflammatory masses,adenosis,benign tumors,and malignant tumors.These categorizations are important for guiding clinical treatment.In this study,we aimed to develop a convolutional neural network(CNN)for classification of these four breast mass types using ultrasound(US)images.Methods:Taking breast biopsy or pathological examinations as the reference standard,CNNs were used to establish models for the four-way classification of 3623 breast cancer patients from 13 centers.The patients were randomly divided into training and test groups(n=1810 vs.n=1813).Separate models were created for two-dimensional(2D)images only,2D and color Doppler flow imaging(2D-CDFI),and 2D-CDFI and pulsed wave Doppler(2D-CDFI-PW)images.The performance of these three models was compared using sensitivity,specificity,area under receiver operating characteristic curve(AUC),positive(PPV)and negative predictive values(NPV),positive(LR+)and negative likelihood ratios(LR-),and the performance of the 2D model was further compared between masses of different sizes with above statistical indicators,between images from different hospitals with AUC,and with the performance of 37 radiologists.Results:The accuracies of the 2D,2D-CDFI,and 2D-CDFI-PW models on the test set were 87.9%,89.2%,and 88.7%,respectively.The AUCs for classification of benign tumors,malignant tumors,inflammatory masses,and adenosis were 0.90,0.91,0.90,and 0.89,respectively(95%confidence intervals[CIs],0.87-0.91,0.89-0.92,0.87-0.91,and 0.86-0.90).The 2D-CDFI model showed better accuracy(89.2%)on the test set than the 2D(87.9%)and 2D-CDFI-PW(88.7%)models.The 2D model showed accuracy of 81.7%on breast masses≤1 cm and 82.3%on breast masses>1 cm;there was a significant difference between the two groups(P<0.001).The accuracy of the CNN classifications for the test set(89.2%)was significantly higher than that of all the radiologists(30%).Conclusions:The CNN may have high accuracy for classification of US images of breast masses and perform significantly better than human radiologists.Trial registration:Chictr.org,ChiCTR1900021375;http://gffzz62a03ee25e6f4a77h99pof9bvcxkq6q95.ffgz.tsg.suse.edu.cn/showproj.aspx?proj=33139.Teng-Fei Yu Wen He Cong-Gui Gan Ming-Chang Zhao Qiang Zhu Wei Zhang Hui Wang Yu-Kun Luo Fang Nie Li-Jun Yuan Yong Wang Yan-Li Guo Jian-Jun Yuan Li-Tao Ruan Yi-Cheng Wang Rui-Fang Zhang Hong-Xia Zhang Bin Ning Hai-Man Song Shuai Zheng Yi Li Yang Guang 2021Chinese Medical Journal2021,,4:11

13Antipyretic Effect of Herba Ephedrae-Ramulus Cinnamomi Herb Pair on Yeast-Induced Pyrexia Rats: A Metabolomics Study显示文摘Objective: To investigate the antipyretic mechanism of Herba Ephedrae(Eph)-Ramulus Cinnamomi(RC) herb pair on yeast-induced pyrexia in rats. Methods: Totally 30 qualified male SD rats were randomly assigned to the normal control(NC) group, the pyrexia model(model) group, the Eph, RC and Eph-RC treatment groups by a random digital table, 6 rats in each group. Each rat received a 20% aqueous suspension of yeast(10 m L/kg) except the NC group. The 3 treatment groups were administered 8.1, 5.4 and 13.5g/kg Eph, RC and Eph-RC respectively at 5 and 12h after yeast injection, the NC group and the model groups were administered equal volume of distilled water. Rectal temperatures were measured at 0,6,8,10,12,15,18,24 and 30h and urine was collected prior to yeast injection and at 6, 10, 18, 24, 30, and 36 h after yeast injection. Then urine metabolomic profiling by gas chromatography tandem mass spectrometry, coupled with multivariate statistical analysis and pattern recognition techniques were used to explore the antipyretic effects of Eph-RC. Partial least squares discriminate analysis was used to analyze the metabolomics dataset including classification and regression in metabolomics plot profiling. Results: Compared with the NC group, rectal temperatures were significantly higher in the model group(P<0.01), while 3 treatment groups decreased significantly compared with the model group(P<0.05 or P<0.01). Rectal temperatures of Eph-RC-treated rats started to go down at 6 h, and markedly decreased at 8, 12, 15, 18 and 24h(P<0.05 or P<0.01), while those of the Eph and RC groups had decreased firstly at 8 h and were markedly lower at 12h(P<0.05 or P<0.01). Seventeen potential biomarkers related to pyrexia were confirmed and identified, including pyruvic acid, L-phenylalanine, L-tyrosine, phenylacetic acid, hippuric acid, succinic acid, citrate and so on. Eight potential alterations of metabolic pathways including phenylalanine metabolism, citrate cycle, tryptophan metabolism, biosynthesis of valine, leucine and isoleucine, were identified in relation to the antipyretic effects of Eph-RC using MetPA software. 14Forest canopy height mapping over China using GLAS and MODIS data显示文摘The Geoscience Laser Altimeter System(GLAS)accurately detects the vertical structural information of a target within its laser spot and is a promising system for the inversion of structural features and other biophysical parameters of forest ecosystems.Since the GLAS footprints are discontinuously distributed with a relativity low density,continuous vegetation height distributions cannot be mapped with a high accuracy using GLAS data alone.The MODIS BRDF product provides more forest structural information than other optical remote sensing data.This study aimed to map forest canopy heights over China from the GLAS and MODIS BRDF data.Firstly,the waveform characteristic parameters were extracted from the GLAS data by the method of wavelet analysis,and the terrain index was calculated using the ASTER GDEM data.Secondly,the model reducing the topographic influence was constructed from the waveform characteristic parameters and terrain index.Thirdly,the final canopy height estimation model was constructed from the neural network combining the canopy height estimated with the GLAS point and the MODIS BRDF data,and applied to get the continuous canopy height map over China.Finally,the map was validated by the measured data and the airborne Li DAR data,and the validation results indicated that forest canopy heights can be estimated with high accuracy from combined GLAS and MODIS data.YANG Ting WANG Cheng LI GuiCai LUO SheZhou XI XiaoHuan GAO Shuai ZENG HongCheng 2015Science China Earth Sciences2015,58,1:8
15Qingkailing Injection(清开灵注射液)for Treatment of Children Pneumonia Induced by Respiratory Syncytial Virus:A Meta-Analysis of Rando mized Controlled Trials显示文摘Objective: To evaluate the efficacy and safety of Qingkailing Injection(清开灵注射液, QKL) for treatment of children pneumonia caused by respiratory syncytial virus(RSV). Methods: Randomized clinical trials(RCTs) comparing QKL with ribavirin injection in the treatment of children pneumonia induced by RSV were searched in Pub Med, Science Direct, Cochrane Library, Chinese VIP database, CNKI and Wanfang databases from their inception to March 2014. Meta-analyses were performed using RevMan 5.2 software. The methodological quality of the selected RCTs was evaluated by the Modified Jadad Score. The primary outcome measures were effective rate and the secondary outcomes were relief time of fever and cough. Results: Seven RCTs with 992 cases published from 2008 to 2013 were identified. The meta-analysis results indicated that QKL was more effective in cure rate [risk ratios(RR)=1.32, 95% CI(1.17, 1.50), P<0.01], total effective rate [RR=1.07, 95% CI(1.02, 1.13), P=0.009] and less fever clearance time [mean difference=–0.73, 95% CI(–1.22, –0.23), P=0.004], compared with ribavirin injection in the treatment of RSV-induced children pneumonia. No dead case was reported in all trials. There were 3 trials mentioned adverse events, 2 reported no obvious adverse event occurred while 1 reported adverse events described as skin hypersensitivity, elevation of ALT, a mild abnormal of hepatic and renal function in both QKL and ribavirin group. Conclusions: QKL was an effective and relatively safe option for the treatment of RSV-induced children pneumonia. 17Direct observation of ultrafast plasmonic hot electron transfer in the strong coupling regime显示文摘Achieving strong coupling between plasmonic oscillators can significantly modulate their intrinsic optical properties.Here,we report the direct observation of ultrafast plasmonic hot electron transfer from an Au grating array to an MoS_(2) monolayer in the strong coupling regime between localized surface plasmons(LSPs)and surface plasmon polaritons(SPPs).By means of femtosecond pump-probe spectroscopy,the measured hot electron transfer time is approximately 40 fs with a maximum external quantum yield of 1.65%.Our results suggest that strong coupling between LSPs and SPPs has synergetic effects on the generation of plasmonic hot carriers,where SPPs with a unique nonradiative feature can act as an‘energy recycle bin’to reuse the radiative energy of LSPs and contribute to hot carrier generation.Coherent energy exchange between plasmonic modes in the strong coupling regime can further enhance the vertical electric field and promote the transfer of hot electrons between the Au grating and the MoS_(2) monolayer.Our proposed plasmonic strong coupling configuration overcomes the challenge associated with utilizing hot carriers and is instructive in terms of improving the performance of plasmonic opto-electronic devices.Hangyong Shan Ying Yu Xingli Wang Yang Luo Shuai Zu Bowen Du Tianyang Han Bowen Li Yu Li Jiarui Wu Feng Lin Kebin Shi Beng Kang Tay Zheng Liu Xing Zhu Zheyu Fang 2019Light(Science & Applications)2019,8,1:7
18Effects of soil salinity on rhizosphere soil microbes in transgenic Bt cotton fields显示文摘With increased cultivation of transgenic Bacillus thuringiensis(Bt) cotton in the saline alkaline soil of China, assessments of transgenic crop biosafety have focused on the effects of soil salinity on rhizosphere microbes and Bt protein residues. In 2013 and 2014, investigations were conducted on the rhizosphere microbial biomass, soil enzyme activities and Bt protein contents of the soil under transgenic Bt cotton(variety GK19) and its parental non-transgenic cotton(Simian 3) cultivated at various salinity levels(1.15, 6.00 and 11.46 dS m^(-1)). Under soil salinity stress, trace amounts of Bt proteins were observed in the Bt cotton GK19 rhizosphere soil, although the protein content increased with cotton growth and increased soil salinity levels. The populations of slight halophilic bacteria, phosphate solubilizing bacteria, ammonifying bacteria, nitrifying bacteria and denitrifying bacteria decreased with increased soil salinity in the Bt and non-Bt cotton rhizosphere soil, and the microbial biomass carbon, microbial respiration and soil catalase, urease and alkaline phosphatase activity also decreased. Correlation analyses showed that the increased Bt protein content in the Bt cotton rhizosphere soil may have been caused by the slower decomposition of soil microorganisms, which suggests that salinity was the main factor influencing the relevant activities of the soil microorganisms and indicates that Bt proteins had no clear adverse effects on the soil microorganisms. 19miR-137 inhibits melanoma cell proliferation through downregulation of GLO1显示文摘Late-stage melanoma is refractory to current therapies. MicroRNAs (miRNAs) can modulate many physiological and pathological processes of melanoma. Studies have demonstrated that miR-137 acts as a tumor suppressor by inhibiting the proliferation of melanoma cells through targeting multiple mRNAs. The glyoxalase system member glyoxalase 1 (GLO1) is the principal scavenging enzyme of methylglyoxal (MG), a toxic byproduct of glycolysis. Using 35S in vivo/vitro labelling analysis for dynamic proteomics (SiLAD), we found that miR-137 downregulated the expression of GLO1 in melanoma cells.Bioinformatics analysis predicted that GLO1 is a direct target of miR-137. This was validated by dual luciferase reporter assay.Quantitative RT-PCR (qRT-PCR) and western blot analysis indicated that miR-137 could decrease endogenous GLO1 expression.Furthermore, siRNA targeting of GLO1 mimicked inhibition of melanoma cell proliferation caused by miR-137 overexpression.Re-expression of GLO1 was able to restore miR-137-mediated suppression of melanoma cell proliferation. 20Degradation and mineralization of aniline by O_3/Fenton process enhanced using high-gravity technology显示文摘The degradation and mineralization of aniline(AN) using ozone combined with Fenton reagent(O_3/Fenton) in a rotating packed bed(RPB) was proposed in this study, and the process(RPB-O_3/Fenton) was compared with conventional O_3/Fenton in a stirred tank reactor(STR-O_3/Fenton) or single ozonation in an RPB(RPB-O_3). Effects of high gravity factor, H_2O_2 dosage, H_2O_2 dosing method and initial p H on the AN mineralization efficiency were investigated in the RPB-O_3/Fenton process. In addition, the behavior of Fe(II) was monitored at different H2 O2 dosing methods and p H values. Finally, the optimal operation conditions were determined with high gravity factor of100, initial pH of 5, Fe(II) concentration of 0.8 mmol·L^(-1) and H_2O_2 dosage of 2.5 ml. Under these conditions, for aniline wastewater at the volume of 1 L and concentration of 200 mg·L^(-1), a fast and thorough decay of AN was conducted in 10 min, and the TOC removal efficiency reached 89% in 60 min. The main intermediates of p-benzoquinone, nitrobenzene, maleic acid and oxalic acid were identified by liquid chromatography/mass spectroscopy(LC/MS), and the degradation pathways of AN in RPB-O_3/Fenton system were proposed based on experimental evidence.
